| 2 | Zanthoxylum-specific whole genome duplication and recent activity of transposable elements in the highly repetitive paleotetraploid Z.bungeanum genome显示文摘Zanthoxylum bungeanum is an important spice and medicinal plant that is unique for its accumulation of abundant secondary metabolites,which create a characteristic aroma and tingling sensation in the mouth.Owing to the high proportion of repetitive sequences,high heterozygosity,and increased chromosome number of Z.bungeanum,the assembly of its chromosomal pseudomolecules is extremely challenging.Here,we present a genome sequence for Z.bungeanum,with a dramatically expanded size of 4.23 Gb,assembled into 68 chromosomes.This genome is approximately tenfold larger than that of its close relative Citrus sinensis.After the divergence of Zanthoxylum and Citrus,the lineage-specific whole-genome duplication event q-WGD approximately 26.8 million years ago(MYA)and the recent transposable element(TE)burst~6.41 MYA account for the substantial genome expansion in Z.bungeanum.The independent Zanthoxylum-specific WGD event was followed by numerous fusion/fission events that shaped the genomic architecture.Integrative genomic and transcriptomic analyses suggested that prominent speciesspecific gene family expansions and changes in gene expression have shaped the biosynthesis of sanshools,terpenoids,and anthocyanins,which contribute to the special flavor and appearance of Z.bungeanum.In summary,the reference genome provides a valuable model for studying the impact of WGDs with recent TE activity on gene gain and loss and genome reconstruction and provides resources to accelerate Zanthoxylum improvement. | Shijing Feng Zhenshan Liu Jian Cheng Zihe Li Lu Tian Min Liu Tuxi Yang Yulin Liu Yonghong Liu He Dai Zujun Yang Qing Zhang Gang Wang Jisen Zhang Huifeng Jiang Anzhi Wei | 2021 | Horticulture Research2021,8,1: | 5 |
| 3 | Depression accelerates gastric cancer invasion and metastasis by inducing a neuroendocrine phenotype via the catecholamine/β_(2)-AR/MACC1 axis显示文摘Background:Depression is a common,easily ignored,accompanied disease of gastric cancer(GC)patients and is often observed with elevated plasma catecholamine levels.Depression frequently promotes GC progression and leads to poor clinical outcomes;however,the molecular mechanisms underlying depression-induced GC progression remain poorly understood.We aimed to study the effects of depression on GC progression and explore possible mechanisms mediating the action of depression-associated catecholamines on GC.Methods:Depression states of GC patients were graded using the Patient Health Questionnaire-9,and plasma catecholamine levels were examined by high performance liquid chromatography coupled with tandem mass spectrometry.Migrative and invasive GC cells were examined using transwell assays,and metastatic GC niches were imaged using bioluminescence technology in a depression mouse model established with chronic unpredictable mild stress.Mouse depression-like behaviors were assessed through sucrose preference,forced swimming,and tail suspension tests.Characteristics of the neuroendocrine phenotype were observed via RT-PCR,Western blotting,flow cytometry,and transmission electron microscopy.Results:Fifty-one GC patients(age:53.61±1.79 years;cancer duration:3.71±0.33 months;depression duration:2.37±0.38 months;male-to-female ratio:1.55:1)were enrolled in the study.Depression grade was significantly higher in GC patients showing higher plasma levels of catecholamines(epinephrine:P=0.018;noradrenaline:P=0.009),higher oncogene metastasis-associated in colon cancer-1(MACC1)level(P=0.018),and metastasis(P<0.001).Further,depression-associated catecholamine specifically bound to the beta-2 adrenergic receptor(β_(2)-AR)and upregulated MACC1 expression,and thus promoting neuroendocrine phenotypic transformation through direct binding betweenMACC1 and synaptophysin.Eventually,the neuroendocrine phenotypic transformation accelerated GC invasion in vitro and metastasis in vivo.However,β_(2)-AR antagonist ICI-118,551 or MACC1 silencing effectively blocked the catecholamineinduced neuroendocrine phenotypic transformation and eliminated depressionenhanced GC migration and invasion.Moreover,β_(2)-AR blocking or MACC1 silencing prevented GC metastasis attributed to a neuroendocrine phenotype in a depression mouse model.Conclusions:Catecholamine-induced neuroendocrine phenotypes of GC cells led to depression-accelerated GC invasion and metastasis via the β_(2)-AR/MACC1 axis,while β_(2)-AR antagonist or MACC1 silencing could reverse it,showing promising potential therapeutic strategies for improving the outcome of GC patients with comorbid depression. | Changqie Pan Jianhua Wu Siting Zheng Huiying Sun Yisheng Fang Zhenhua Huang Min Shi Li Liang Jianping Bin Yulin Liao Jinzhang Chen Wangjun Liao | 2021 | Cancer Communications2021,41,10: | 4 |

Methods:One hundred and seventy-two patients,prospectively,blinded referred for evaluation with four clinical-score systems for suspected deep venous thrombosis,were examined by ultrasonography. Sensitivity,specificity,positive predictive value,nega-tive predictive value and receiver operation curves were calculated for four clinical scores. The difference between areas of the ROC curve for each of the scores was compared with others and reference line. Results:Forty-six of 172 patients had deep venous throm-bosis proven by sonography. The sensitivity,specificity,positive predictive value and negative predictive value for Wells score was 91.3%,57.1%,43.8% and 94.7%,respectively,for Kahn score; 65.2%,71.4%,45.5% and 84.9%,respectively,for St.Andréscore; 63%,38.9%,27.4% and 74.2% respectively,for Constans score; 95.7%,34.9%,34.9% and 95.7% respectively. Area under ROV curve of Constans score was 0.814,which was similar to that of Wells score,then followed by Kahn score and that of St.Andréscore was no difference with the reference line. Conclusion:Based on the results of our study,the sensitivity,negative prediction value and area under ROC curve are larger for Constans score and Wells score in Chinese hospitalized patients than that of Kahn score or St.Andréscore. | 20 | Rational design of a hollow porous structure for enhancing diffusion kinetics of K ions in edge-nitrogen doped carbon nanorods显示文摘The high electrical conductivity makes it possible for one-dimensional(1D)carbon materials to be used as the promising anodes for potassium ion batteries(PIBs),however,the sluggish diffusion kinetics caused by large-sized potassium ions(K^(+))limits their practical applications in energy storage systems.In this work,hollow carbon nanorods were rationally designed as a case to verify the superiority of 1D hollow structure to improve the diffusion kinetics of K^(+).Simultaneously,edge-N(pyridinic-N and pyrrolic-N)atoms were also introduced into 1D hollow carbon structure,which can provide ample active sites and defects in graphitic lattices to adsorb K^(+),providing extra capacitive storage capacity.As expected,the optimized edge-N doped hollow carbon nanorods(ENHCRs)exhibits a high reversible capacity of 544 mAh·g^(−1)at 0.1 A·g^(−1)after 200 cycles.Even at 5 A·g^(−1),it displays a long-term cycling stability with 255 mAh·g^(−1)over 10,000 cycles.The electrochemical measurements confirm that the hollow structure is favorable to improve the transfer kinetics of K^(+)during cycling.And the theoretical calculations demonstrate that edge-N doping can enhance the local electronegativity of graphitic lattices to adsorb much more K^(+),where edge-N doping synergizes with 1D hollow structure to achieve enhanced K^(+)-storage performances. | Ping Niu Yang Yang Zhiqiang Li Gaohui Ding Lingzhi Wei Ge Yao Helin Niu Yulin Min Fangcai Zheng Qianwang Chen | 2022 | Nano Research2022,15,9: | 1 |
