| 2 | BLT1 in dendritic cells promotes Th1/Th17 differentiation and its deficiency ameliorates TNBS-induced colitis显示文摘Leukotriene B4(LTB4)synthesis is enhanced in the colonic mucosa in patients with inflammatory bowel disease(IBD).BLT1,a highaffinity receptor for LTB4,exhibits no effect on the progression of dextran sodium sulfate(DSS)-induced colitis,which mostly relies on innate immunity.Here,we reported that BLT1 regulates trinitrobenzene sulfonic acid(TNBS)-induced colitis,which reflects CD4+T-cell-dependent adaptive immune mechanisms of IBD.We found that BLT1 signaling enhanced the progression of colitis through controlling the production of proinflammatory cytokines by dendritic cells(DCs)and modulating the differentiation of Th1 and Th17.BLT1−/−mice displayed an alleviated severity of TNBS-induced colitis with reduced body weight loss and infiltrating cells in the lamina propria.BLT1 deficiency in DCs led to reduced production of proinflammatory cytokines,including IL-6,TNF-α,and IL-12,and these results were further confirmed via treatment with a BLT1 antagonist.The impaired cytokine production by BLT1−/−DCs subsequently led to reduced Th1 and Th17 differentiation both in vitro and in vivo.We further performed a conditional DC reconstitution experiment to assess whether BLT1 in DCs plays a major role in regulating the pathogenesis of TNBS-induced colitis,and the results indicate that BLT1 deficiency in DCs also significantly reduces disease severity.The mechanistic study demonstrated that BLT1-regulated proinflammatory cytokine production through the Gαiβγsubunit-phospholipase Cβ(PLCβ)-PKC pathway.Notably,we found that treatment with the BLT1 antagonist also reduced the production of proinflammatory cytokines by human peripheral blood DCs.Our findings reveal the critical role of BLT1 in regulating adaptive immunity and TNBS-induced colitis,which further supports BLT1 as a potential drug target for adaptive immunity-mediated IBD. | Jinfeng Zhou Weiming Lai Wanjie Yang Juping Pan Hu Shen Yingying Cai Cuixia Yang Ningjia Ma Yue Zhang Ru Zhang Xin Xie Zhongjun Dong Yuan Gao Changsheng Du | 2018 | Cellular & Molecular Immunology2018,15,12: | 9 |

| 4 | Identification of the lower limit of high-quality source rocks and its relation to hydrocarbon accumulation——Taking the Beier Sag in the Hailaer Basin as an example显示文摘The theory of 'source rock control' has evolved from source-rock-control hydrocarbon accumulation, to effective source-rock-control hydrocarbon accumulation, and to high-quality source-rock-control hydrocarbon accumulation. However, there are problems, such as whether high-quality source rocks exist or not? What high-quality source rocks are, and how to identify them, are yet to be agreed upon. Aimed at this issue of concern to explorationists, and taking the Beier Sag in the Hailaer Basin as an example, this paper defines the high-quality source rocks and the lower limit for evaluation of high-quality source rocks, by using the inflection point on the relationship curve of hydrocarbon (oil) expulsion, which is calculated by the material balance principle, versus total organic carbon (TOC). The results show that when TOC is low, all source rocks have limited hydrocarbon expulsion and slow growth rate, thus they cannot be high-quality source rocks. However, when TOC rises to some threshold, hydrocarbon expulsion increases significantly with TOC. This inflection point should be the lower limit of high-quality source rocks: those with TOC greater than the inflection-point value are high-quality source rocks. In addition, the lower limit of high-quality source rocks is also related to the type and maturity of organic matters in the source rocks, as well as the mineral components of the source rocks affecting the residual hydrocarbons. Theoretically, the lower limit of high-quality source rocks depends on geological conditions rather than being a constant value. However, for the sake of simplicity and practicability, in this paper TOC=2.0% is regarded as the lower limit of high-quality source rocks. The examination of such standard in the work area indicates that the high-quality source rocks in members K 1 n 2 and K 1 n 1 of the Nantun formation contribute 76% and 82% to oil generation, and 96% and 91% to oil expulsion , respectively. | 5 | Resveratrol induces AMPK and mTOR signaling inhibition-mediated autophagy and apoptosis in multiple myeloma cells显示文摘Resveratrol,a natural compound extracted from the skins of grapes,berries,or other fruits,has been shown to have anti-tumor effects against multiple myeloma(MM)via promoting apoptosis and inhibiting cell viability.In addition to apoptosis,autophagy also plays a significant role in anti-tumor effects.However,whether autophagy is involved in anti-MM activity of resveratrol remains unclear.In this study,human MM cell lines U266,RPMI-8226,and NCI-H929 were treated with resveratrol.Cell Counting Kit-8 assay and colony formation assay were used to measure cell viability.Western blot analysis was used to detect apoptosis-and autophagy-associated proteins.3-Methyladenine(3-MA)was applied to inhibit autophagy.Results showed that resveratrol inhibited cell viability and colony formation via promoting apoptosis and autophagy in MM cell lines U266,RPMI-8226,and NCI-H929.Resveratrol promoted apoptosis-related proteins,Caspase-3 activating poly-ADP-ribose polymerase and Caspase-3 cleavage,and decreased the protein level of Survivin in a dose-dependent manner.Additionally,resveratrol upregulated the levels of LC3 and Beclin1 in a dose-dependent way,indicating that autophagy might be implicated in anti-MM effect of resveratrol.Furthermore,3-MA relieved the cytotoxicity of resveratrol by blocking the autophagic flux.Resveratrol increased the phosphorylation of adenosine monophosphate(AMP)-activated protein kinase and decreased the phosphorylation of mammalian target of rapamycin(mTOR)and its downstream substrates p70S6K and 4EBP1 in a dose-dependent manner,leading to autophagy.Therefore,our results suggest that resveratrol exerts anti-MM effects through apoptosis and autophagy,which can be used as a new therapeutic strategy for MM in clinic. | Ruye Ma Dandan Yu Yu Peng Hongfei Yi Yingcong Wang Taofang Cheng Bingqing Shi Guang Yang Weiming Lai Xiaosong Wu Ye Lu Jumei Shi | 2021 | Acta Biochimica et Biophysica Sinica2021,53,6: | 7 |

| 7 | Effects of Sini San used alone and in combination with fluoxetine on central and peripheral 5-HT levels in a rat model of depression显示文摘OBJECTIVE: To investigate the effects of Sini San and fluoxetine on the levels of central and peripheral 5-HT in a rat model of depression, and provide new insight into the treatment of depression with integrated Chinese-Western Medicine.METHODS: A rat model of depression was established by chronic mild stress(CMS). Model rats received either Sini San, fluoxetine, a combination of the two drugs, or no drug treatment. Healthy naive rats were used as controls. Open field and sucrose preference tests were used to assess depression-like behavior. ELISA and immunohistochemistry were used to determine central and peripheral levels of 5-HT.RESULTS: In the group with no drug treatment,central 5-HT expression decreased while peripheral5-HT concentrations increased as CMS continued.Four weeks after CMS, Sini San alone was less effective in reducing depression-like behavior than fluoxetine alone or in combination with Sini San,but combined use was more effective than fluoxetine alone. Eight weeks after CMS, Sini San alone or in combination with fluoxetine was more effective in reducing depression-like behavior than fluoxetine alone. Furthermore Sini San and fluoxetine used alone or in combination notably increased central 5-HT expression and decreased peripheral 5-HT levels in the rat model.CONCLUSION:The results of the present study indicate that there is a synergistic action between the two medicines in the treatment of depression. Sini San exhibited a relatively long lag before its effects were observed; however, by eight weeks the Traditional Chinese Medicine appeared at least as effective as fluoxetine. | 8 | White blood cell count and the incidence of hyperuricemia:insights from a community-based study显示文摘Hyperuricemia(HUA)is a risk factor for chronic kidney disease(CKD).The relationship between HUA and white blood cell(WBC)count remains unknown.A sampling survey for CKD was conducted in Sanlin community in 2012 and 2014.CKD was defined as proteinuria in at least the microalbuminuric stage or an estimated GFR of 60 mL/(min·1.73 m2).HUA was defined as serum uric acid>420µmol/L in men and>360µmol/L in women.This study included 1024 participants.The prevalence of HUA was 17.77%.Patients with HUA were more likely to have higher levels of WBC count,which was positively associated with HUA prevalence.This association was also observed in participants without CKD,diabetes mellitus,hyperlipidemia,or obesity.Multivariate logistic regression analysis showed that WBC count was independently associated with the risk for HUA in male and female participants.Compared with participants without HUA,inflammatory factors such as high-sensitivity C-reactive protein,tumor necrosis factor-α,and interleukin 6 increased in participants with HUA.Hence,WBC count is positively associated with HUA,and this association is independent of conventional risk factors for CKD. | Jian Liu Pingyan Shen Xiaobo Ma Xialian Yu Liyan Ni Xu Hao Weiming Wang Nan Chen | 2019 | Frontiers of Medicine2019,13,6: | 3 |
| 9 | Preparation of protein-loaded microspheres with size ≤ 10 μm by electrostatic droplet generation technology显示文摘The development of non-injection route for protein drugs, especially oral administration, has been the main focus of controlled release of drugs. To overcome obstacles unsolved such as enzyme degradation and penetration barrier of intestinal epi- thelium, technologies using microspheres as carrier of protein drugs have been proven potential to realize oral administration. It has been demonstrated that microspheres can not only protect proteins, but also facilitate the penetration and absorption through Peyer’s patches when the size is smaller than 10 μm. Therefore, the objective of this paper is to prepare protein-loaded microspheres with size ≤ 10 μm. Electrostatic droplet generation technology was used with insulin and hemoglobin as drug models and so- dium alginate as microsphere material. By decreas- ing the surface tension of feed solution by adding surfactant, and improving electric field distribution by changing the shape of container and electrode for gelation solution, protein-loaded microspheres with mean size less than 10 μm were successfully pro- duced through needle with diameter of 400 μm. The microspheres showed good sphericity and narrow size distribution. The mean standard variance of size distribution was 1.61. The encapsulation efficiency of proteins was over 70%. Moreover, the significance analysis of factors influencing the size of protein loaded microspheres was carried out through or- thogonal experiments, which showed that output voltage (U), needle diameter (D) and the distance between needle tips to the surface of gelation solu- tion (δ ) influenced significantly the size of micro-spheres. Meanwhile, a new sensitivity analysis method, a general coupling model for steel ground loops, and a partial element equivalent circuit method are proposed to identify and characterize conducted EMI coupling paths. The EMI noise and propagation modeling provide an accurate prediction of conducted EMI in the entire frequency range (0―10 MHz) with good practicability and generality. Finally a new measurement approach is presented to identify the surface current of large dimensional metal shell.
