1Whole-genome sequencing of 508 patients identifies key molecular features associated with poor prognosis in esophageal squamous cell carcinoma显示文摘Esophageal squamous cell carcinoma(ESCC)is a poor-prognosis cancer type with limited understanding of its molecular etiology.Using 508 ESCC genomes,we identified five novel significantly mutated genes and uncovered mutational signature clusters associated with metastasis and patients’outcomes.Several functional assays implicated that NFE2L2 may act as a tumor suppressor in ESCC and that mutations in NFE2L2 probably impaired its tumor-suppressive function,or even conferred oncogenic activities.Additionally,we found that the NFE2L2 mutations were significantly associated with worse prognosis of ESCC.We also identified potential noncoding driver mutations including hotspot mutations in the promoter region of SLC35E2 that were correlated with worse survival.Approximately 5.9%and 15.2%of patients had high tumor mutation burden or actionable mutations,respectively,and may benefit from immunotherapy or targeted therapies.We found clinically relevant coding and noncoding genomic alterations and revealed three major subtypes that robustly predicted patients’outcomes.Collectively,we report the largest dataset of genomic profiling of ESCC useful for developing ESCC-specific biomarkers for diagnosis and treatment.Yongping Cui Hongyan Chen Ruibin Xi Heyang Cui Yahui Zhao Enwei Xu Ting Yan Xiaomei Lu Furong Huang Pengzhou Kong Yang Li Xiaolin Zhu Jiawei Wang Wenjie Zhu Jie Wang Yanchun Ma Yong Zhou Shiping Guo Ling Zhang Yiqian Liu Bin Wang Yanfeng Xi Ruifang Sun Xiao Yu Yuanfang Zhai Fang Wang Jian Yang Bin Yang Caixia Cheng Jing Liu Bin Song Hongyi Li Yi Wang Yingchun Zhang Xiaolong Cheng Qimin Zhan Yanhong Li Zhihua Liu-Show 2020Cell Research2020,30,10:19
2Association of emergency room visits for respiratory diseases with sources of ambient PM2.5显示文摘Previous studies have reported associations of short-term exposure to different sources of ambient fine particulate matter(PM2.5)and increased mortality or hospitalizations for respiratory diseases.Few studies,however,have focused on the short-term effects of source-specific PM2.5 on emergency room visits(ERVs)of respiratory diseases.Source apportionment for PM2.5 was performed with Positive Matrix Factorization(PMF)and generalized additive model was applied to estimate associations between source-specific PM2.5 and respiratory disease ERVs.The association of PM2.5 and total respiratory ERVs was found on lag4(RR=1.011,95%CI:1.002,1.020)per interquartile range(76μg/m3)increase.We found PM2.5 to be significantly associated with asthma,bronchitis and chronic obstructive pulmonary disease(COPD)ERVs,with the strongest effects on lag5(RR=1.072,95%CI:1.024,1.119),lag4(RR=1.104,95%CI:1.032,1.176)and lag3(RR=1.091,95%CI:1.047,1.135),respectively.The estimated effects of PM2.5 changed little after adjusting for different air pollutants.Six primary PM2.5 sources were identified using PMF analysis,including dust/soil(6.7%),industry emission(4.5%),secondary aerosols(30.3%),metal processing(3.2%),coal combustion(37.5%)and traffic-related source(17.8%).Some of the sources were identified to have effects on ERVs of total respiratory diseases(dust/soil,secondary aerosols,metal processing,coal combustion and traffic-related source),bronchitis ERVs(dust/soil)and COPD ERVs(traffic-related source,industry emission and secondary aerosols).Different sources of PM2.5 contribute to increased risk of respiratory ERVs to different extents,which may provide potential implications for the decision making of air quality related policies,rational emission control and public health welfare.Rui Chi Hongyu Li Qian Wang Qiangrong Zhai Daidai Wang Meng Wu Qichen Liu Shaowei Wu Qingbian Ma Furong Deng Xinbiao Guo 2019Journal of Environmental Sciences2019,31,12:8
3In vivo electrochemical recording of continuous change of magnesium in medial vestibular nucleus following vertigo induced by ice water vestibular stimulation显示文摘Vertigo is one of the most common clinical symptoms. However, the chemical processes involved in the pathological mechanism of vertigo remain to be fully understood. In this study, we investigate the dynamic changes in the magnesium (Mg 2+ ) concentration in medial vestibular nucleus (MVN) of guinea pigs following vertigo induced by vestibular ice water stimulation with an electrochemical detection method consisting of in vivo microdialysis and on-line selective electrochemical detection. Electrochemical detection of Mg 2+ was accomplished based on the current enhancement of Mg 2+ towards the electrocatalytic oxidation of NADH at the electrodes modified with the polymerized film of toluidine blue O (TBO). Selectivity for the on-line electrochemical detection against Ca 2+ was achieved by using ethyleneglcol-bis(2-aminoethylether) tetraacetic acid (EGTA) as the selective masking agent for Ca 2+ . The basal level of the extracellular Mg 2+ in the MVN of guinea pigs was determined to be 759.7 ± 176.2 M(n = 16). Upon ice water irrigation of the left external ear canal, the concentration of Mg 2+ in the MVN decreases significantly, reaches 72 ± 6% (n = 8) of the basal level, and maintains for at least 1000 s. Control experiments reveal that neither warm water irrigation of the external ear canal nor ice water irrigation of the auricle induces the decrease in the concentration of Mg 2+ in the MVN. These results demonstrate that the extracellular Mg 2+ in the MVN decreases significantly following vertigo induced by vestibular ice water stimulation. 4Dysregulated ceramides metabolism by fatty acid 2-hydroxylase exposes a metabolic vulnerability to target cancer metastasis显示文摘Whereas it is appreciated that cancer cells rewire lipid metabolism to survive and propagate,the roles of lipid metabolism in metastasis remain largely unknown.In this study,using esophageal squamous cell carcinoma(ESCC)as a pulmonary metastasis model,we find that the enzyme fatty acid 2-hydroxylase(FA2H),which catalyzes the hydroxylation of free fatty acids(FAs),is enriched in a subpopulation of ESCC cells with high metastatic potential,and that FA2H knockdown markedly mitigates metastatic lesions.Moreover,increased FA2H expression is positively associated with poor survival in patients with ESCC.Lipidomics analysis identifies that two dihydroceramides—Cer(d18:0/24:0)and Cer(d18:0/24:1)—are increased in FA2H-depleted metastasizing ESCC cells.Upon administration,Cer(d18:0/24:0)and Cer(d18:0/24:1)impair the formation of overt metastases in a mouse experimental metastasis model.Then,forkhead box protein C2(FOXC2)and FA2H are found to be co-upregulated in metastatic ESCC cell populations and ESCC specimens,and FA2H expression is further experimentally verified to be transcriptionally induced by FOXC2,which is boosted per se by tumour necrosis factorα(TNFα),a critical pro-metastasis cytokine in the tumour microenvironment,in metastasizing cells.Together,these results demonstrate that TNFα-FOXC2-FA2H is a novel signaling axis to promote metastasis,and its downstream dihydroceramide products could be promising drugs to intervene in metastasis.Xuantong Zhou Furong Huang Gang Ma Wenqing Wei Nan Wu Zhihua Liu 2022Signal Transduction and Targeted Therapy2022,7,11:2

6An integrated spectroscopic strategy to trace the geographical origins of emblic medicines: Application for the quality assessment of natural medicines显示文摘Emblic medicine is a popular natural source in the world due to its outstanding healthcare and therapeutic functions.Our preliminary results indicated that the quality of emblic medicines might have an apparent regional variation.A rapid and effective geographical traceability system has not been designed yet.To trace the geographical origins so that their quality can be controlled,an integrated spectroscopic strategy including spectral pretreatment,outlier diagnosis,feature selection,data fusion,and machine learning algorithm was proposed.A featured data matrix(245220)was successfully generated,and a carefully adjusted RF machine learning algorithm was utilized to develop the geographical traceability model.The results demonstrate that the proposed strategy is effective and can be generalized.Sensitivity(SEN),specificity(SPE)and accuracy(ACC)of 97.65%,99.85%and 97.63%for the calibrated set,as well as 100.00%predictive efficiency,were obtained using this spectroscopic analysis strategy.Our study has created an integrated analysis process for multiple spectral data,which can achieve a rapid,nondestructive and green quality detection for emblic medicines originating from seventeen geographical origins.Luming Qi Furong Zhong Yang Chen Shengnan Mao Zhuyun Yan Yuntong Ma 2020Journal of Pharmaceutical Analysis2020,10,4:1

8Mass transfer techniques for large-scaleand high-density microLED arrays显示文摘Inorganic-based micro light-emitting diodes (microLEDs) offer more fascinating properties and unique demands in next-generation displays. However, the small size of the microLED chip (1–100 µm) makes it extremely challenging for high efficiency and low cost to accurately, selectively, integrate millions of microLED chips. Recent impressive technological advances have overcome the drawbacks of traditional pick-and-place techniques when they were utilized in the assembly of microLED display, including the most broadly recognized laser lift-off technique, contact micro-transfer printing (µTP) technique, laser non-contact µTP technique, and self-assembly technique. Herein, we firstly review the key developments in mass transfer technique and highlight their potential value, covering both the state-of-the-art devices and requirements for mass transfer in the assembly of the ultra-large-area display and virtual reality glasses. We begin with the significant challenges and the brief history of mass transfer technique, and expand that mass transfer technique is composed of two major techniques, namely, the epitaxial Lift-off technique and the pick-and-place technique. The basic concept and transfer effects for each representative epitaxial Lift-off and pick-and-place technique in mass transfer are then overviewed separately. 13Novel Incentive Scheme to Motivate Flexible Customers for Phase Balancing显示文摘Phase imbalance is a significant and widespread problem in low voltage(LV,415V)networks.This paper develops a novel incentive scheme that encourages flexible customers to engage in phase balancing.On the technical side,this incentive scheme is based on a centralized control algorithm utilizing the inherent fexibility of customers'AC/DC converters to achieve phase balancing at the substation side of LV networks.On the incentive side,first,this paper calculates the benefits from phase balancing.It then defines a rebalancing contribution index that quantifies the flexible customer's contribution to addressing the predominant imbalance-induced consequence,e.g,capacity wastes,and energy losses.According to the contribution indices and the benefits,the incentive scheme rewards flexible customers correspondingly and guides them to prioritize the predominant consequence of phase imbalance.Case studies demonstrate that:1)the remunerations paid to the flexible customers primarily depends on their contributions to addressing the predominant imbalance-induced consequences for the network in question;2)the incentive scheme does not discriminate against small/medium-sized flexible customers who are dedicated to phase balancing,thus promoting inclusiveness.Lurui Fang Kang Ma Furong Li Fei Xue 2022CSEE Journal of Power and Energy Systems2022,8,4:0

15Multi-Attribute Couplings-Based Euclidean and Nominal Distances for Unlabeled Nominal Data显示文摘Learning unlabeled data is a significant challenge that needs to han-dle complicated relationships between nominal values and attributes.Increas-ingly,recent research on learning value relations within and between attributes has shown significant improvement in clustering and outlier detection,etc.However,typical existing work relies on learning pairwise value relations but weakens or overlooks the direct couplings between multiple attributes.This paper thus proposes two novel and flexible multi-attribute couplings-based distance(MCD)metrics,which learn the multi-attribute couplings and their strengths in nominal data based on information theories:self-information,entropy,and mutual information,for measuring both numerical and nominal distances.MCD enables the application of numerical and nominal clustering methods on nominal data and quantifies the influence of involving and filtering multi-attribute couplings on distance learning and clustering perfor-mance.Substantial experiments evidence the above conclusions on 15 data sets against seven state-of-the-art distance measures with various feature selection methods for both numerical and nominal clustering.Lei Gu Furong Zhang Li Ma 2023Computers, Materials & Continua2023,,6:0
16Altered Retinal Dopamine Levels in a Melatonin-proficient Mouse Model of Form-deprivation Myopia显示文摘Reduced levels of retinal dopamine,a key regulator of eye development,are associated with experimental myopia in various species,but are not seen in the myopic eyes of C57BL/6 mice,which are deficient in melatonin,a neurohormone having extensive interactions with dopamine.Here,we examined the relationship between form-deprivation myopia(FDM)and retinal dopamine levels in melatonin-proficient CBA/CaJ mice.We found that these mice exhibited a myopic refractive shift in form-deprived eyes,which was accompanied by altered retinal dopamine levels.When melatonin receptors were pharmacologically blocked,FDM could still be induced,but its magnitude was reduced,and retinal dopamine levels were no longer altered in FDM animals,indicating that melatonin-related changes in retinal dopamine levels contribute to FDM.Thus,FDM is mediated by both dopamine level-independent and melatonin-related dopamine level-dependent mechanisms in CBA/CaJ mice.The previously reported unaltered retinal dopamine levels in myopic C57BL/6 mice may be attributed to melatonin deficiency.Kang-Wei Qian Yun-Yun Li Xiao-Hua Wu Xue Gong Ai-Lin Liu Wen-Hao Chen Zhe Yang Ling-Jie Cui Yun-Feng Liu Yuan-Yuan Ma Chen-Xi Yu Furong Huang Qiongsi Wang Xiangtian Zhou Jia Qu Yong-Mei Zhong Xiong-Li Yang Shi-Jun Weng 2022Neuroscience Bulletin2022,38,9:0
