| 2 | Analysis of genetic architecture and favorable allele usage of agronomic traits in a large collection of Chinese rice accessions显示文摘Genotyping and phenotyping large natural populations provide opportunities for population genomic analysis and genome-wide association studies(GWAS). Several rice populations have been re-sequenced in the past decade;however, many major Chinese rice cultivars were not included in these studies. Here, we report large-scale genomic and phenotypic datasets for a collection mainly comprised of 1,275 rice accessions of widely planted cultivars and parental hybrid rice lines from China. The population was divided into three indica/Xian and three japonica/Geng phylogenetic subgroups that correlate strongly with their geographic or breeding origins. We acquired a total of 146 phenotypic datasets for 29 agronomic traits under multi-environments for different subpopulations. With GWAS, we identified a total of 143 significant association loci, including three newly identified candidate genes or alleles that control heading date or amylose content. Our genotypic analysis of agronomically important genes in the population revealed that many favorable alleles are underused in elite accessions, suggesting they may be used to provide improvements in future breeding efforts. | 4 | Development of an Inactivated Iridovirus Vaccine Against Turbot Viral Reddish Body Syndrome显示文摘Turbot(Scophthalmus maximus L.) reddish body iridovirus(TRBIV) was propagated in turbot fin cells(TF cells) and inactivated as the TRBIV vaccine with its protection efficiency evaluated in this study.TF cells were cultured in 10% bovine calf serum(BCS)-containing MEM medium(pH7.0) at 22℃,in which TRBIV propagated to a titer as high as 105.6 TCID50 mL-1.The TRBIV was inactivated with 0.1% formalin and formulated with 0.5% aluminum hydroxide.The inactivated vaccine caused neither cytopathogenic effect(CPE) on TF cells nor pathogenic effect on turbots.After being administered with the vaccine twice via muscle injection,the turbot developed high-tittered TRBIV neutralizing antibodies in a dose-dependent manner.The vaccine protected the turbot from dying with an immunoprotection rate of 83.3% as was determined via subcutaneous vaccination in the laboratory and 90.5% via bath vaccination in turbot farms,respectively.The inactivated vaccine was very immunogenic,efficiently preventing tur-bot from death.It holds the potential of being applied in aquaculture. | FAN Tingjun HU Xiuzhong WANG Liyan GENG Xiaofen JIANG Guojian YANG Xiuxia YU Miaomiao | 2012 | Journal of Ocean University of China2012,11,1: | 5 |

The dead time can be reduced by using the ground automatic convert method, hybrid phase-controlled technologies can in theory completely eliminate inrush currents. In this paper, a new system based on hybrid phase-controlled switches is described and termed ground-switching passing neutral section system (GPNSS). The principle for restraining inrush currents is analyzed and strategies are carried out with the dead time limited to 5 ms. The characteristics of the vacuum circuit breaker are illustrated and the closing time window of the transformer is quantified. Through the use of mechanical switches and power electronics, the auxiliary power system may be continuously pow- ered. | 7 | Characterization of a Novel Anti-DR5 Monoclonal Antibody WD1 with the Potential to Induce Tumor Cell Apoptosis显示文摘TNF-related apoptosis-inducing ligand (TRAIL) is a TNF family member capable of inducing apoptosis. Death receptor 5 (DR 5) is a key receptor of TRAIL and plays an important role in TRAIL-induced apoptosis. To prepare monoclonal antibodies (mAbs) against DR5,cDNA encoding soluble DR5 (sDR5) was firstly amplified by reverse transcriptase-polymerase chain reaction (RT-PCR) with specific primers,and then inserted into a prokaryotic expression vector pET-30a. The recombinant plasmid was expressed in Escherichia coli strain BL21 (DE3),and sDR5 was purified by nickel affinity chromatography. As an antigen,sDR5 was used to immunize mice. Hybridomas secreting antibodies against sDR5 were identified. One positive clone was selected to produce antibody,WD1. ELISA and immunofluorescence demonstrated that WD1 could bind recombinant sDR5 and membrane-bound DR5 (mDR5) on Jurkat and Molt-4 cells. ATPLite assays showed that Jurkat and Molt-4 cells were sensitive to the antibody in a dose dependent manner. The Annexin V/PI assays and Giemsa's staining both showed that WD1 could induce Jurkat cell apoptosis efficiently. Transient transfection of 293T cells and indirect immunofluorescence assay demonstrated that mAb (WD1) couldn't cross-react with DR4. Our findings indicated that the novel antibody,WD1 could act as a direct agonist,bind DR5 characteristically,and initiate efficient apoptotic signaling and tumor regression. | 9 | Preparation and luminescence properties of orange-red Ba3Y4O9:Sm3+phosphors显示文摘A novel orange-red emitting Ba_3 Y_4 O_9:Sm^(3+) phosphors were prepared by a high temperature solidstate reaction in air. X-ray diffraction(XRD), photoluminescence spectra, fluorescence decay and temperature-dependent emission spectra were utilized to characterize the structure and luminescence properties. The results show that the excitation spectrum includes a series of linear peaks at350, 367, 382, 410, 424, 445, 470 and 495 nm, respectively. Under 410 nm excitation, the emission peaks were located at 574 nm(~4 G_(5/2)-~6 H_(5/2)), 608 nm(~4 G_(5/2)-~6 H_(7/2)),659 nm(~4 G_(5/2)-~6 H_(9/2)) and722 nm(~4 G_(5/2)-~6 H_(11/2)), respectively. The concentration quenching occurs when x equals 0.08 for Ba_3 Y_(4-x)O_9:xSm^(3+) phosphor and its mechanism is ascribed to the dipole-dipole interaction. The chromaticity coordinates of Ba_3 Y_(3.92)O_9:0.08 Sm^(3+) phosphor are in the orange-red region. The temperature-dependent study shows that this phosphor has excellent luminescence thermal-stability.And the luminescence intensity of Ba_3 Y_(3.92)O_9:0.08 Sm^(3+) phosphor at 473 K only declines by about25.75% of its initial intensity. | 10 | Nano-and microphytoplankton community characteristics in brown tide bloom-prone waters of the Qinhuangdao coast,Bohai Sea, China显示文摘Since 2009, the newly recorded species Aureococcus anophagefferens has successively induced brown tides in the Qinhuangdao coastal waters of the Bohai Sea. Here, we focus on the phytoplankton community structure in this region and try to point out the biological background characteristics of the outbreaks of successive brown tides. We analyzed species composition,cell abundance, diversity indices and size-fractionated chlorophyll a(Chl-a) concentration monthly from March 2013 to January2014. The phytoplankton community, as observed by microscopy, underwent succession from nano-celled chained diatoms to dinoflagellates in the spring and summer, and then to micro-celled diatoms and nano-celled chained diatoms as the most prominent groups in the subsequent autumn and winter. Canonical correspondence analysis revealed that changes in the abundance of diatoms were related to nutrient availability, especially of nitrogen, while water temperature was the crucial factor influencing the abundance of dinoflagellates and A. anophagefferens. Considering the contribution of nano-celled chained species to the micro-sized Chl-a,small phytoplankton(<20 μm) composed the majority of total Chl-a and were generally abundant during our study. | 11 | White blood cell count and the incidence of hyperuricemia:insights from a community-based study显示文摘Hyperuricemia(HUA)is a risk factor for chronic kidney disease(CKD).The relationship between HUA and white blood cell(WBC)count remains unknown.A sampling survey for CKD was conducted in Sanlin community in 2012 and 2014.CKD was defined as proteinuria in at least the microalbuminuric stage or an estimated GFR of 60 mL/(min·1.73 m2).HUA was defined as serum uric acid>420µmol/L in men and>360µmol/L in women.This study included 1024 participants.The prevalence of HUA was 17.77%.Patients with HUA were more likely to have higher levels of WBC count,which was positively associated with HUA prevalence.This association was also observed in participants without CKD,diabetes mellitus,hyperlipidemia,or obesity.Multivariate logistic regression analysis showed that WBC count was independently associated with the risk for HUA in male and female participants.Compared with participants without HUA,inflammatory factors such as high-sensitivity C-reactive protein,tumor necrosis factor-α,and interleukin 6 increased in participants with HUA.Hence,WBC count is positively associated with HUA,and this association is independent of conventional risk factors for CKD. | Jian Liu Pingyan Shen Xiaobo Ma Xialian Yu Liyan Ni Xu Hao Weiming Wang Nan Chen | 2019 | Frontiers of Medicine2019,13,6: | 3 |

| 13 | Biosynthesis of antibiotic chuangxinmycin from Actinoplanes tsinanensis显示文摘Chuangxinmycin is an antibiotic isolated from Actinoplanes tsinanensis CPCC 200056 in the1970 s with a novel indole-dihydrothiopyran heterocyclic skeleton. Chuangxinmycin showed in vitro antibacterial activity and in vivo efficacy in mouse infection models as well as preliminary clinical trials.But the biosynthetic pathway of chuangxinmycin has been obscure since its discovery. Herein, we report the identification of a stretch of DNA from the genome of A. tsinanensis CPCC 200056 that encodes genes for biosynthesis of chuangxinmycin by bioinformatics analysis. The designated cxn cluster was then confirmed to be responsible for chuangxinmycin biosynthesis by direct cloning and heterologous expressing in Streptomyces coelicolor M1146. The cytochrome P450 CxnD was verified to be involved in the dihydrothiopyran ring closure reaction by the identification of seco-chuangxinmycin in S. coelicolor M1146 harboring the cxn gene cluster with an inactivated cxn D. Based on these results, a plausible biosynthetic pathway for chuangxinmycin biosynthesis was proposed, by hijacking the primary sulfur transfer system for sulfur incorporation. | 18 | Validated LC–MS/MS method for determination of YH-8, a novel PKnB inhibitor, in rat plasma and its application to pharmacokinetic study显示文摘(E)-Methyl-4-aryl-4-oxabut-2-enoate(YH-8) is a novel PKn B protein kinase inhibitor with good anti-tuberculosis activity. To evaluate its pharmacokinetics in rats, a sensitive and selective high performance liquid chromatography–tandem mass spectrometric(LC–MS/MS) method has been developed and validated for the quantification of YH-8 in rat plasma for the first time. Samples were pre-treated using a liquid–liquid extraction with ethyl acetate and the chromatographic separation was performed on a C18 column by gradient elution with methanol–water as the mobile phase. YH-8 was detected using a tandem mass spectrometer in positive selected reaction monitoring(SRM) mode. Method validation revealed good linearity over the range of1–500 ng/m L for YH-8 with a lower limit of quantification(LLOQ) of 1 ng/m L. Intra- and inter-day precision of YH-8 assay in rat plasma samples were 2.0%–6.8%, with accuracy of the method being 100.69%–106.18%.Stability test showed that when spiked into rat plasma, YH-8 was stable for 12 h at room temperature, for up to15 days at -70℃, and after three freeze-thaw cycles. Extracted samples were found to be stable over 12 h in an auto-sampler.
