2Hydration Mechanism of Sulphoaluminate Cement显示文摘The feasibility of sulphoaluminate cement (SAC) utilization in support mortar was studied. Setting time and strength of as-received sulphoaluminate cement (SAC) paste were examined, hydration kinetics behavior was determined through Isothermal Calorimeter, and hydration mechanism was investigated by X-Ray diffraction analysis (XRD) and field emission scanning electron microscopy analysis(FSEM). Results showed that as-received SAC contained 61% of anhydrous calcium sulfate (3CA·CaSO4) and dicalcium silicate (C2S). The strength after 1 day or 3 days grew to 68.6% or 85.7% of that after 28 days respectively, while most of hydration heat was released within 1 day. The emergency of three exothermic peaks at acceleration stage was found and hydration kinetics model was established choosing the terminal time of the first exothermic peak at accelerating stage as the beginning of accelerating stage. 3The IAA-and ABA-responsive transcription factor CgMYB58 upregulates lignin biosynthesis and triggers juice sac granulation in pummelo显示文摘In citrus,lignin overaccumulation in the juice sac results in granulation and an unpleasant fruit texture and taste.By integrating metabolic phenotyping and transcriptomic analyses,we found 702 differentially expressed genes(DEGs),including 24 transcription factors(TFs),to be significantly correlated with lignin content.CgMYB58 was further identified as a critical R2R3 MYB TF involved in lignin overaccumulation owing to its high transcript levels in Huanong Red-fleshed pummelo(HR,Citrus grandis)fruits.Transient expression of CgMYB58 led to an increase in the lignin content in the pummelo fruit mesocarp,whereas its stable overexpression significantly promoted lignin accumulation and upregulated 19 lignin biosynthetic genes.Among these genes,CgPAL1,CgPAL2,Cg4CL1,and CgC3H were directly modulated by CgMYB58 through interaction with their promoter regions.Moreover,we showed that juice sac granulation in pummelo fruits could be affected by indole-3-acetic acid(IAA)and abscisic acid(ABA)treatments.In HR pummelo,ABA significantly accelerated this granulation,whereas IAA effectively inhibited this process.Taken together,these results provide novel insight into the lignin accumulation mechanism in citrus fruits.We also revealed the theoretical basis via exogenous IAA application,which repressed the expression of CgMYB58 and its target genes,thus alleviating juice sac granulation in orchards.Meiyan Shi Xiao Liu Haipeng Zhang Zhenyu He Hongbin Yang Jiajing Chen Jia Feng Wenhui Yang Youwu Jiang Jia-Long Yao Cecilia Hong Deng Juan Xu 2020Horticulture Research2020,7,1:4
4Orbital control on cyclical organic matter accumulation in Early Silurian Longmaxi Formation shales显示文摘High resolution(939 samples)total organic carbon content(TOC)analyses were conducted on the Shuanghe Section of^152.6 m in the Changning area,Sichuan Basin.The sampling section was divided into two units considering the distinct-different deposit environment and sediments accumulation rate.The lower part(Unit 1)and the peer part(Unit 2)with high resolution sample spacing(0.08–0.4 m)enables the identification of the precession cycle in two sedimentary sequences with distinct different sedimentary accumulation rate.MTM Power spectral analyses on untuned TOC series reveals significant peaks exceeding above the 95%confidence level and shows that both Unit 1 and Unit 2 have recorded Milankovitch cycles of 405 kyr long eccentricity,short eccentricity,obliquity and precession.The floating astronomical time scale(ATS)was constructed on the Shuanghe Section in the Early Silurian(~439.673–444.681 Ma),and which was calibrated by 405 kyr long eccentricity cycles.The total duration of the Wufeng and Longmaxi shales is 5.01 Myr.The floating ATS used for estimating the duration of the graptolite zones and each stage in the study interval.Finally,we postulated two models that could verify the linkage between orbital cycle and organic accumulation.To make sure whether productivity or preservation is the main factor that under long eccentricity control,the phase correlation between the obtained filtered signal and the theoretical orbital solution should be made clear in the further research.Siding Jin Hucheng Deng Xing Zhu Yan Liu Sibing Liu Meiyan Fu 2020Geoscience Frontiers2020,11,2:4
5Occurrence,distribution,and potential influencing factors of sewage sludge components derived from nine full-scale wastewater treatment plants of Beijing,China显示文摘Millions of tons of waste activated sludge(WAS) produced from biological wastewater treatment processes cause severe adverse environmental consequences. A better understanding of WAS composition is thus very critical for sustainable sludge management. In this work, the occurrence and distribution of several fundamental sludge constituents were explored in WAS samples from nine full-scale wastewater treatment plants(WWTPs) of Beijing, China. Among all the components investigated, active heterotrophic biomass was dominant in the samples(up to 9478 mg/L), followed by endogenous residues(6736 mg/L),extracellular polymeric substances(2088 mg/L), and intracellular storage products(464 mg/L)among others. Moreover, significant differences(p < 0.05) were observed in composition profiles of sludge samples among the studied WWTPs. To identify the potential parameters affecting the variable fractions of sludge components, wastewater source as well as design and operational parameters of WWTPs were studied using statistical methods. The findings indicated that the component fraction of sewage sludge depends more on wastewater treatment alternatives than on wastewater characteristics among other parameters. A principal component analysis was conducted, which further indicated that there was a greater proportion of residual inert biomass in the sludge produced by the combined system of the conventional anaerobic/anoxic/oxic process and a membrane bioreactor. 6Carrier-free highly drug-loaded biomimetic nanosuspensions encapsulated by cancer cell membrane based on homology and active targeting for the treatment of glioma显示文摘Nanosuspensions,as a new drug delivery system for insoluble drugs,are only composed of a drug and a small amount of stabilizer,which is dispersed in an aqueous solution with high drug-loading,small particle size,high dispersion,and large specific surface area.It can significantly improve the dissolution,bioavailability,and efficacy of insoluble drugs.In this study,paclitaxel nanosuspensions((PTX)NS)were prepared by an ultrasonic precipitation method,with the characteristics of simple preparation and easy repetition.With the help of a homologous targeting mechanism,a kind of glioma C6 cancer cell membrane(CCM)-coated(PTX)NS was developed and modified with DWSW peptide to obtain DWSW-CCM-(PTX)NS with the functions of BBB penetration and tumor targeting.The results showed that the cancer cell membrane could effectively camouflage the nanosuspensions so that it was not cleared by the immune system and could cross the blood-brain-barrier(BBB)and selectively target tumor tissues.Cell uptake experiments and in vivo imaging confirmed that the uptake of DWSW-CCM-(PTX)NS by tumor cells and the distribution in intracranial gliomas increased.Cytotoxicity test and in vivo anti-glioma studies showed that DWSW-CCM-(PTX)NS could significantly inhibit the growth of glioma cells and significantly prolong the survival time of glioma-bearing mice.Finally,the cancer cell membrane coating endowed the nanosuspensions with the biological properties of homologous adhesion and immune escape.This study provides an integrated solution for improving the targeting of nanosuspensions and demonstrates the encouraging potential of biomimetic nanosuspensions applicable to tumor therapy.Yueyue Fan Yuexin Cui Wenyan Hao Mengyu Chen Qianqian Liu Yuli Wang Meiyan Yang Zhiping Li Wei Gong Shiyong Song Yang Yang Chunsheng Gao 2021Bioactive Materials2021,6,12:3

10Advance in germplasm innovation and gene mining of high-oil maize显示文摘As one of the 3 worldwide major grain crops, maize is the main source of food, feed and edible vegetable oil. High-oil maize has been selectively bred for many years due to its high oil content and nutritional quality. However, compared with normal maize, the progress of high-oil maize breeding is hindered nowadays. Main problems are scarce of high-oil maize germplasm, conventional breeding methods, and slow genetic research progress on oil content and fatty acid composition. In this study, germplasm innovation and gene mining of high-oil maize were reviewed to provide theoretical reference for genetic breeding and utilization. Several high-oil basic populations have been developed after more than 100 years of selection, which have served as the unique resources for high-oil maize breeding. They have also been used to dissect genetic architecture of oil biosynthesis in maize kernel. Using linkage and association analyses, many QTLs and loci associated with oil content and fatty acid composition have been identified and mapped. In addition, some candidate genes for QTLs have been validated to regulate maize grain oil content and fatty acid composition. 13A“T.E.S.T.”hydrogel bioadhesive assisted by corneal cross-linking for in situ sutureless corneal repair显示文摘Corneal transplantation is an effective clinical treatment for corneal diseases,which,however,is limited by donor corneas.It is of great clinical value to develop bioadhesive corneal patches with functions of“Transparency”and“Epithelium&Stroma generation”,as well as“Suturelessness”and“Toughness”.To simultaneously meet the“T.E.S.T.”requirements,a light-curable hydrogel is designed based on methacryloylated gelatin(GelMA),Pluronic F127 diacrylate(F127DA)&Aldehyded Pluronic F127(AF127)co-assembled bi-functional micelles and collagen type I(COL I),combined with clinically applied corneal cross-linking(CXL)technology for repairing damaged cornea.The patch formed after 5 min of ultraviolet irradiation possesses transparent,highly tough,and strongly bio-adhesive performance.Multiple cross-linking makes the patch withstand deformation near 600%and exhibit a burst pressure larger than 400 mmHg,significantly higher than normal intraocular pressure(10-21 mmHg).Besides,the slower degradation than GelMA-F127DA&AF127 hydrogel without COL I makes hydrogel patch stable on stromal beds in vivo,supporting the regrowth of corneal epithelium and stroma.The hydrogel patch can replace deep corneal stromal defects and well bio-integrate into the corneal tissue in rabbit models within 4 weeks,showing great potential in surgeries for keratoconus and other corneal diseases by combining with CXL.Meiyan Li Ruoyan Wei Chang Liu Haowei Fang Weiming Yang Yunzhe Wang Yiyong Xian Kunxi Zhang Yong He Xingtao Zhou 2023Bioactive Materials2023,,7:1
14Hydration Mechanism of Silica Fume- sulphoaluminate Cement显示文摘Setting time and strength of sulphoaluminate rapid hardening cement(SAC) incorporated in the presence and absence of silica fume(SF) were determined. Combined with the techniques of isothermal calorimeter, XRD and FSEM, the hydration kinetics of the two systems and the effect mechanism of SF on SAC were investigated. The experimental results showed that SF was proved to be beneficial for SAC system, in terms of setting time and late strength gain. Evidence of accelerator effect of silica fume was found during the first 8 hours of hydration. The formation of AFt was accelerated and the microstructure of the hydration products grew denser with incorporation of SF. SF was proved to play the role of dispersion and setting control at early age and had a greater contribution to later strength due to the increment of crystal nucleation point and the pozzolanic activity. 17Prevalence and risk factors of cardiovascular diseases and psychological distress among female scientists and technicians显示文摘This study aimed to explore the prevalence and risk factors of cardiovascular disease(CVD)and psychological distress among female scientists and technicians in China.Accordingly,we included scientists and technicians from representative research institutions,medical institutions,colleges,universities,and businesses in China,and the data were collected from July 1,2019 to March 31,2021 via online questionnaires.The parameters evaluated in this study included age,sex,marital status,educational background,monthly income,sleep hours,sleep problems,smoking,alcohol consumption,work-related stress,work burnout,cardiovascular symptoms,CVD,family history,and depressive and anxiety symptoms.A total of 14530 scientists and technicians were included,comprising 7144 men and 7386 women.We found 34.9%men and 16.6%women with CVD,35.1%men and 21.4%women with depressive symptoms,28.7%men and 13.8%women with anxiety symptoms,and 22.0%men and9.5%women with CVD combined with depressive or anxiety symptoms.This study focused on the details of women.Younger women(age≤35 years)had the highest prevalence of depressive symptoms(24.9%),anxiety symptoms(16.2%),and comorbidity(11.2%).It was established that,despite traditional risk factors,unmanageable work burnout,depressive symptoms,and anxiety symptoms were associated with a higher risk of CVD in women;insomnia,overwhelming work stress,unmanageable work burnout,and CVD were linked to a higher risk of depressive symptoms and anxiety;insomnia,overwhelming work stress,and unmanageable work burnout were related to CVD combined with depressive or anxiety symptoms.A bidirectional relationship was noted between CVD and depression or anxiety in female scientists and technicians,and insomnia and overwhelming work stress were positively associated with comorbidity.It is suggested that effective measures should be taken to protect female scientists and technicians from CVD and psychological distress.Lijun ZHANG Yanping BAO Guo LI Shuhui TAO Meiyan LIU 2022Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2022,23,12:1
18Cell relay-delivery improves targeting and therapeutic efficacy in tumors显示文摘Cell-mediated drug delivery system(CDDS)has shown great potential for cancer therapy.However,a single cell-mediated drug delivery mechanism has not generally been successful,particularly for systemic administration.To augment the antitumor therapy efficacy,herein,we propose a strategy of cell relay-delivery for the use of artificially damaging/aging erythrocytes to hitchhike on circulating monocytes/macrophages for intratumoral accumulation of anticancer drugs.This biomimetic relay-delivery strategy was derived from the manner in which circulating monocytes/macrophages in body specifically engulf damaged/senescent erythrocytes and actively transmigrate into the tumor bulk.The strategy elegantly combines the natural functions of both cells,which therefore provides a new perspective to challenge current obstacles in drug delivery.According to the strategy,we developed biotinylated erythrocyte-poly(lactic-co-glycolic acid)(PLGA)nanoparticle hybrid DDSs(bE-NPs)using avidin-biotin coupling.In such a system,biotinylated erythrocytes can mimic the natural property of damaged/senescent erythrocytes,while PLGA NPs are capable of encapsulating anticancer drugs and promoting sustained drug release.Anticancer drugs can effectively target tumor sites by two steps.First,by using biotinylated erythrocytes as the carrier,the drug-loaded PLGA NPs could be specifically phagocytized by monocytes/macrophages.Second,by taking advantage of the tumor-tropic property of monocytes/macrophages,the drug-loaded PLGA NPs could be efficiently transported into the tumor bulk.After encapsulating vincristine(VIN)as the model drug,bE-NPs exhibited the most favorable antitumor effects in vitro and in vivo by the cell relay-delivery effect.These results demonstrate that the cell relay-delivery provides a potential method for improving tumor treatment efficacy.Ye Feng Qianqian Liu Yi Li Yang Han Meng Liang Hao Wang Qing Yao Yuli Wang Meiyan Yang Zhiping Li Wei Gong Yang Yang Chunsheng Gao 2021Bioactive Materials2021,6,6:0
19Peptide nanotube loaded with a STING agonist,c-di-GMP,enhance cancer immunotherapy against melanoma显示文摘The activation of the stimulating factor of the interferon gene(STING)pathway can enhance the immune response within the tumor.Cyclic diguanylate monophosphate(c-di-GMP)is a negatively charged,hydrophilic STING agonist,however,its effectiveness is limited due to the poor membrane permeability and low bioavailability.Herein,we introduced KL-7 peptide derived from Aβamyloid fibrils that can self-assemble to form nanotubes to load and deliver c-di-GMP,which significantly enhanced c-di-GMP’s effectiveness and then exhibited a robust“in situ immunity”to kill melanoma cells.KL-7 peptide nanotube,also called PNT,was loaded with negatively charged c-di-GMP via electrostatic interaction,which prepared a nanocomposite named c-di-GMP-PNT.Treatment of RAW 264.7 cells(leukemia cells in mouse macrophage)with c-di-GMP-PNT markedly stimulated the secretion of IL-6 and INF-βalong with phospho-STING(Ser365)protein expression,indicating the activation of the STING pathway.In the unilateral flank B16-F10(murine melanoma cells)tumor-bearing mouse model,compared to PNT and cdi-GMP,c-di-GMP-PNT can promote the expression of INF-β,TNF-α,IL-6,and IL-1β.At the same time,up-regulated CD4 and CD8 active T cells kill tumors and enhance the immune response in tumor tissues,resulting in significant inhibition of tumor growth in tumor-bearing mice.More importantly,in a bilateral flank B16-F10 tumor model,both primary and distant tumor growth can also be significantly inhibited by c-di-GMP-PNT.Moreover,c-di-GMP-PNT demonstrated no obvious biological toxicity on the main organs(heart,liver,spleen,lung,and kidney)and biochemical indexes of mice.In summary,our study provides a strategy to overcome the barriers of free c-di-GMP in the tumor microenvironment and c-di-GMP-PNT may be an attractive nanomaterial for anti-tumor immunity.Ziyuan Zhang Juan Liu Min Xiao Quanfeng Zhang Zhonghua Liu Meiyan Liu Peng Zhang Youlin Zeng 2023Nano Research2023,16,4:0
20Psp2,a novel regulator of autophagy that promotes autophagy-related protein translation显示文摘Macroautophagy/autophagy defines an evolutionarily conserved catabolic process that targets cytoplasmic components for lysosomal degradation.The process of autophagy from initiation to closure is tightly executed and controlled by the concerted action of autophagy-related(Atg)proteins.Although substantial progress has been made in characterizing transcriptional and post-translational regulation of ATG/Atg genes/proteins,little is known about the translational control of autophagy.Here we report that Psp2,an RGG motif protein,positively regulates autophagy through promoting the translation of Atg1 and Atg13,two proteins that are crucial in the initiation of autophagy.During nitrogen starvation conditions,Psp2 interacts with the 5′UTR of ATG1 and ATG13 transcripts in an RGG motif-dependent manner and with eIF4E and eIF4G2,components of the translation initiation machinery,to regulate the translation of these transcripts.Deletion of the PSP2 gene leads to a decrease in the synthesis of Atg1 and Atg13,which correlates with reduced autophagy activity and cell survival.Furthermore,deactivation of the methyltransferase Hmt1 constitutes a molecular switch that regulates Psp2 arginine methylation status as well as its mRNA binding activity in response to starvation.These results reveal a novel mechanism by which Atg proteins become upregulated to fulfill the increased demands of autophagy activity as part of translational reprogramming during stress conditions,and help explain how ATG genes bypass the general block in protein translation that occurs during starvation.Zhangyuan Yin Xu Liu Aileen Ariosa Haina Huang Meiyan Jin Katrin Karbstein Daniel J.Klionsky 2019Cell Research2019,29,12:0
