1A LuxR family transcriptional regulator AniF promotes the production of anisomycin and its derivatives in Streptomyces hygrospinosus var.beijingensis显示文摘The protein synthesis inhibitor anisomycin features a unique benzylpyrrolidine system and exhibits potent selective activity against pathogenic protozoa and fungi.It is one of the important effective components in Agricultural Antibiotic120,which has been widely used as naturally-originated agents for treatment of crop decay in China.The chemical synthesis of anisomycin has recently been reported,but the complex process with low productivity made the biosynthesis still to be a vital mainstay in efforts.The biosynthetic gene cluster(BGC)of anisomycin in Streptomyces hygrospinosus var.beijingensis has been identified in our previous work,while poor understanding of the regulatory mechanism limited the yield enhancement via regulation engineering of S.hygrospinosus var.beijingensis.In this study here,we characterized AniF as an indispensable LuxR family transcriptional regulator for the activation of anisomycin biosynthesis.The genetic manipulations of aniF and the real-time quantitative PCR(RT-qPCR)revealed that it positively regulated the transcription of the anisomycin BGC.Moreover,the overexpression of aniF contributed to the improvement of the production of anisomycin and its derivatives.Dissection of the mechanism underlying the function of AniF revealed that it directly activated the transcription of the genes aniR-G involved in anisomycin biosynthesis.Especially,one AniF-binding site in the promoter region of aniR was identified by DNase I footprinting assay and an inverted repeat sequence(5′-GGGC-3′)composed of two 4-nt half sites in the protected region was found.Taken together,our systematic study confirmed the positive regulatory role of AniF and might facilitate the future construction of engineering strains with high productivity of anisomycin and its derivatives.Jufang Shen Lingxin Kong Yan Li Xiaoqing Zheng Qing Wang Weinan Yang Zixin Deng Delin You 2019Synthetic and Systems Biotechnology2019,4,1:10
2晚期非小细胞肺癌免疫检查点抑制剂新型生物标志物:血液PD-L1动态检测显示文摘背景免疫检查点抑制剂(immune checkpoint inhibitors,ICIs)具有良好疗效已成为恶性肿瘤治疗领域备受瞩目的翘楚。然而,仅有小部分患者获得长期临床获益。如何选择生物标志物来筛查ICIs治疗优势人群仍是一大热点。方法收集51例晚期恶性肿瘤患者的肿瘤组织和配对血样本,进行PD-L1表达的相关性分析。40例晚期非小细胞肺癌(non-small cell lung cancer,NSCLC)患者接受ICIs治疗,2个月时检测血液PD-L1(bloodPD-L1,bPD-L1)表达的动态变化,包括PD-L1 mRNA、外泌体PD-L1(exosomalPD-L1,exoPD-L1)蛋白和可溶性PD-L1(solublePD-L1,sPD-L1)。应用R软件计算最佳截断值,用于预测最佳客观缓解(best objective response,b OR)、无进展生存期(progression-free survival,PFS)和总生存期(overall survival,OS)。结果51例不同恶性肿瘤患者中,组织PD-L1(tissuePD-L1,tPD-L1)表达阳性患者的PD-L1mRNA表达水平显著升高。40例接受ICIs治疗的晚期NSCLC患者中,bPD-L1 mRNA上调≥2.04的患者b OR、PFS和OS更佳;21例接受ICIs治疗的晚期NSCLC患者中,exoPD-L1上调≥1.86的患者临床获益更大。sPD-L1动态变化与ICIs疗效无显著相关性。PD-L1 mRNA和exoPD-L1联合检测更有利于预测ICIs治疗疗效。结论bPD-L1表达与tPD-L1表达存在正相关;晚期NSCLC患者ICIs治疗初期PD-L1 mRNA和(或)exoPD-L1表达升高,可作为评估临床获益的新型生物标志物。Qiao YANG Mingjing CHEN Jiaoyang GU Kai NIU Xianlan ZHAO Linpeng ZHENG Zihan XU Yongxin YU Feng LI Lingxin MENG Zhengtang CHEN Wenlei ZHUO Luping ZHANG Jianguo SUN 杨峤(译) 孙建国(译) 2021中国肺癌杂志2021,24,12:10
3Orthogonal design to sift the optimal parameter of Neiguan acupuncture for cerebral infarction显示文摘The individual difference and non-repeatability in acupuncture have not only restricted the development of acupuncture,but have also affected the specificity of acupoints.The present study used instruments to control needle depth,lifting and thrusting frequency,and the duration of acupuncture.Effects of the quantified acupuncture were observed at Neiguan(PC6)with different stimulation parameters.A frequency of 1,2,or 3 Hz and duration of 5,60,or 180 seconds were used to observe cerebral blood flow and ratio of infarct volume recovery.Results showed that stimulation at Neiguan with a frequency of 1 Hz and long duration of 180 seconds or 2/3 Hz and long duration of 5/60seconds significantly increased cerebral blood flow and decreased the ratio of infarct volume.Interactions between frequency and duration play a critical role in quantified acupuncture therapy.Yanan Zhang Sha Yang Xiaonong Fan Shu Wang Nina He Lingxin Li Ding Luo Xuemin Shi 2013Neural Regeneration Research2013,8,28:7
4Characterization of the positive SARP family regulator PieR for improving piericidin A1 production in Streptomyces piomogeues var.Hangzhouwanensis显示文摘Piericidin A1,a member ofɑ-pyridone antibiotic,exhibits various biological activities such as antimicrobial,antifungal,and antitumor properties and possesses potent respiration-inhibitory activity against insects due to its competitive binding capacity to mitochondrial complex I.The biosynthetic pathway of piericidin A1 has been reported in Streptomyces piomogeues var.Hangzhouwanensis,while the regulatory mechanism remains poorly understood.In this study,a Streptomyces antibiotic regulatory protein(SARP)family transcriptional regulator PieR was characterized.Genetic disruption and complementation manipulations revealed that PieR positively regulated the production of piericidin A1.Moreover,the overexpression of pieR contributed to the improvement of piericidin A1 productivity.The real-time quantitative PCR(RT-qPCR)was carried out and the data showed that pieR stimulated the transcription of all the biosynthesis-related genes for piericidin A1.In order to explore the regulatory mechanism,electrophoresis mobility shift assays(EMSA)and DNase I footprinting experiments have been conducted.A protected region covering 50 nucleotides within the upstream region of pieR was identified and two 5-nt direct repeat sequences(5′-CCGGA-3′)in the protected region were found.These findings,taken together,set stage for transcriptional control engineering in the view of optimizing piericidin A1 production and thus provide a viable potent route for the construction of strains with high productivity.Yan Li Lingxin Kong Jufang Shen Qing Wang Qian Liu Weinan Yang Zixin Deng Delin You 2019Synthetic and Systems Biotechnology2019,4,1:4
5Multimarker Detection of MAGE-1,MAGE-3 and AFP mRNAs by a Real-time Quantitative PCR Assay:a Possible Predictor of Hematogenous Micrometastasis of Hepatocellular Carcinoma显示文摘OBJECTIVE To explore the relationship between multimarker detection of MAGE-1,MAGE-3 and AFP mRNAs in the peripheral blood of patients with hepatocellular carcinoma and micrometastasis using a realtime quantitative-PCR(real-time Q-PCR)assay. METHODS Peripheral blood samples were obtained from control subjects and 86 patients with hepatocellular carcinoma (HCC).Real-time Q-PCR was used to detect MAGE-1,MAGE-3, and AFP mRNAs in the blood cells. RESULTS In 86 tumor specimens,the positivity for MAGE-1, MAGE-3,and AFP genes was respectively 34.9%(30/86),60.5% (52/86)and 69.8%(60/86).All specimens expressed at least one marker.MAGE-1,MAGE-3,and AFP transcripts were detected respectively in 12(14.0%),18(20.1%)and 29(33.7%)of the 86 blood specimens from hepatocellular carcinoma patients,while 45 specimens(52.3%)were positive for at least one marker.In addition,MAGE-1,MAGE-3 and AFP gene transcripts were not detected in any peripheral blood specimens from 25 chronic liver disease patients and 28 normal healthy volunteers.The positive rate correlated with the TNM clinical stages,extrahepatic metastasis and portal vein carcinothrombosis(P<0.05).No correlation was found between tumor size,tumor number, differentiation,serum a-fetoprotein(AFP)and the positive rate. 16A novel polymer-based nitrocellulose platform for implementing a multiplexed microfluidic paperbased enzyme-linked immunosorbent assay显示文摘Nitrocellose(NC)membranes,as porous paper-like substrates with high protein-binding capabilties,are very popular in the field of point-of-care immunoassays.However,generating robust hydrophobic structures in NC membranes to fabricate microfluidic paper-based analytical devices(μPADs)remains a great challenge.At present,the main method relies on an expensive wax printer In addition,NC membranes very easy to adhere during the printing process due to electrostatic adsorption.Herein,we developed a facile,fast and low-cost strategy to fabricateμPADs in NC membranes by screen-printing polyurethane acrylate(PUA)as a barrier material for defning flow channels and reaction zones.Moreover,hydrophobic barriers based on UV-curable PUA can resist various surfactant solutions and organic solvents that are generally used in immunoassays and biochemical reactions.To validate the feasibility of this PUA-based NC membrane for immunoassays in point-of-care testing(POCT),we further designed and assembled a rotational paperbased analytical device for implementing a multiplexed enzyme-linked immunosorbent assay(ELISA)in a simple manner.Using the proposed device under the optimal conditions,alpha fetoprotein(AFP)and carcinoembryonic antigen(CEA)could be identified,with limits of detection of 136 pg/mL and 174 pg/mL,respectively,which are below the threshold values of these two cancer biomarkers for clinical diagnosis.We believe that this reliable device provides a promising plaform for the diagnosis of disease based on ELISA or other related bioassays in limited setings or remote regions.Dong Lin Bowei Li Longwen Fu Ji Qi Chunlei Xia Yi Zhang Jiadong Chen Jaebum Choo Lingxin Chen 2022Microsystems & Nanoengineering2022,8,3:0

18Effects of Temperature,Salinity and Stocking Density on Larval Survival and Growth of Reciprocal Crosses Between Two Strains of Pacific Oysters,Crassostrea gigas显示文摘The Pacific oyster Crassostrea gigas is one of the most widely cultivated aquaculture species and contributes signifi-cantly to total seafood production for human beings.However,mass mortality occurred frequently,and in some regions almost all oysters died during seed production and grow-out stage.In order to explore whether hybridization breeding can improve its growth and survival,a complete diallel cross between a selected strain‘Haida No.1’(S)and an orange shell variant(O)of C.gigas was carried out.The larval growth and survival were compared among hybrids and purebred strains at temperatures of 16,20,24,28 and 32℃;salinities of 15,20,25,30 and 35;and stocking densities of 0.5,1,2,4 and 8 larvae mL−1.Under different environments,the hybridization between two strains of C.gigas showed the heterosis of growth and survival.The mean shell height and survival rate of the two reciprocal crosses(OS,SO)were significantly higher than those of the two purebred strains(SS,OO)under all environ-mental conditions.In particular,OS showed greater heterosis than the purebred strains and SO progeny.The results showed that the productive traits of the‘Haida No.1’could be improved by crossing with the orange shell line.Meanwhile,the results from this study also indicated that hybridization between the two strains of C.gigas may be a promising way for breeding new variety with high survival rate.MENG Lingxin XU Chengxun LI Qi 2022Journal of Ocean University of China2022,21,4:0
19Protective effects and microarray-based mechanism of sea cucumber hydrolysates against high-glucose induced nephrotoxicity in mouse glomerulus mesangial cells显示文摘Diabetic nephropathy(DN)is a common type of end-stage renal disease and glomerular mesangial cells(GMCs)are widely used as a cell model for DN.This study firstly investigated the inhibitory effects of the Apostichopus japonicus and Acaudina leucoprocta hydrolysates on cellular growth under high-glucose treatment,better inhibitory effect of A.japonicus hydrolysate was observed compared to that of A.leucoprocta hydrolysate.Subsequently,the global transcription profiles obtained via microarray analysis showed that 6070 and 7015 genes were identified in the A.japonicus and A.leucoprocta groups compared with the model group,respectively.Among them,transcriptions of the slc30a4,slc35dl,tppp3,tp53inpl,bcl-2,apafl,alox12b and adrala genes were restored from the levels of the model group to those of the control group,contributed to cell mitosis and proliferation in both treatment groups.In addition,other apoptosis-related genes,such as bcl-6,clu,foxo3 and akt,showed opposite trends between two groups,which might cause the difference in inhibitory effect.We preliminarily proposed that the regulation effects of A.japonicus and A.leucoprocta on the genes involved in cellular mitosis,proliferation and apoptosis,might contribute to their inhibitory activity on GMCs under high-glucose environment.Lingxin Geng Jiaojiao Han Jun Zhou Ye Li Tinghong Ming Zhen Zhang Chenyang Lu Xiurong Su 2024Journal of Future Foods2024,4,3:0
