1晚期非小细胞肺癌免疫检查点抑制剂新型生物标志物:血液PD-L1动态检测显示文摘背景免疫检查点抑制剂(immune checkpoint inhibitors,ICIs)具有良好疗效已成为恶性肿瘤治疗领域备受瞩目的翘楚。然而,仅有小部分患者获得长期临床获益。如何选择生物标志物来筛查ICIs治疗优势人群仍是一大热点。方法收集51例晚期恶性肿瘤患者的肿瘤组织和配对血样本,进行PD-L1表达的相关性分析。40例晚期非小细胞肺癌(non-small cell lung cancer,NSCLC)患者接受ICIs治疗,2个月时检测血液PD-L1(bloodPD-L1,bPD-L1)表达的动态变化,包括PD-L1 mRNA、外泌体PD-L1(exosomalPD-L1,exoPD-L1)蛋白和可溶性PD-L1(solublePD-L1,sPD-L1)。应用R软件计算最佳截断值,用于预测最佳客观缓解(best objective response,b OR)、无进展生存期(progression-free survival,PFS)和总生存期(overall survival,OS)。结果51例不同恶性肿瘤患者中,组织PD-L1(tissuePD-L1,tPD-L1)表达阳性患者的PD-L1mRNA表达水平显著升高。40例接受ICIs治疗的晚期NSCLC患者中,bPD-L1 mRNA上调≥2.04的患者b OR、PFS和OS更佳;21例接受ICIs治疗的晚期NSCLC患者中,exoPD-L1上调≥1.86的患者临床获益更大。sPD-L1动态变化与ICIs疗效无显著相关性。PD-L1 mRNA和exoPD-L1联合检测更有利于预测ICIs治疗疗效。结论bPD-L1表达与tPD-L1表达存在正相关;晚期NSCLC患者ICIs治疗初期PD-L1 mRNA和(或)exoPD-L1表达升高,可作为评估临床获益的新型生物标志物。Qiao YANG Mingjing CHEN Jiaoyang GU Kai NIU Xianlan ZHAO Linpeng ZHENG Zihan XU Yongxin YU Feng LI Lingxin MENG Zhengtang CHEN Wenlei ZHUO Luping ZHANG Jianguo SUN 杨峤(译) 孙建国(译) 2021中国肺癌杂志2021,24,12:10
2In Situ Electrochemical Mn(Ⅲ)/Mn(Ⅳ) Generation of Mn(Ⅱ)O Electrocatalysts for High-Performance Oxygen Reduction显示文摘Among various earth-abundant and noble metal-free catalysts for oxygen reduction reaction(ORR),manganese-based oxides are promising candidates owing to the rich variety of manganese valence.Herein,an extremely facile method for the synthesis of cubic and orthorhombic phase coexisting Mn(Ⅱ)O electrocatalyst as an efficient ORR catalyst was explored.The obtained MnO electrocatalyst with oxygen vacancies shows a significantly elevated ORR catalytic activity with a half-wave potential(E1/2) of as high as 0.895 V,in comparison with that of commercial Pt/C(E1/2=0.877 V).More impressively,the MnO electrocatalyst exhibits a marked activity enhancement after test under a constant applied potential for 1000 s thanks to the in situ generation and stable presence of high-valence manganese species(Mn^3+ and Mn^4+) during the electrochemical process,initiating a synergetic catalytic effect with oxygen vacancies,which is proved to largely accelerate the adsorption and reduction of O_2 molecules favoring the ORR activity elevation.Such an excellent ORR catalytic performance of this MnO electrocatalyst is applied in Zn-air battery,which shows an extra-high peak power density of 63.2 mW cm^-2 in comparison with that(47.4 m W cm^-2) of commercial Pt/C under identical test conditions.Han Tian Liming Zeng Yifan Huang Zhonghua Ma Ge Meng Lingxin Peng Chang Chen Xiangzhi Cui Jianlin Shi 2020Nano-Micro Letters2020,12,11:2
3Multimarker Detection of MAGE-1,MAGE-3 and AFP mRNAs by a Real-time Quantitative PCR Assay:a Possible Predictor of Hematogenous Micrometastasis of Hepatocellular Carcinoma显示文摘OBJECTIVE To explore the relationship between multimarker detection of MAGE-1,MAGE-3 and AFP mRNAs in the peripheral blood of patients with hepatocellular carcinoma and micrometastasis using a realtime quantitative-PCR(real-time Q-PCR)assay. METHODS Peripheral blood samples were obtained from control subjects and 86 patients with hepatocellular carcinoma (HCC).Real-time Q-PCR was used to detect MAGE-1,MAGE-3, and AFP mRNAs in the blood cells. RESULTS In 86 tumor specimens,the positivity for MAGE-1, MAGE-3,and AFP genes was respectively 34.9%(30/86),60.5% (52/86)and 69.8%(60/86).All specimens expressed at least one marker.MAGE-1,MAGE-3,and AFP transcripts were detected respectively in 12(14.0%),18(20.1%)and 29(33.7%)of the 86 blood specimens from hepatocellular carcinoma patients,while 45 specimens(52.3%)were positive for at least one marker.In addition,MAGE-1,MAGE-3 and AFP gene transcripts were not detected in any peripheral blood specimens from 25 chronic liver disease patients and 28 normal healthy volunteers.The positive rate correlated with the TNM clinical stages,extrahepatic metastasis and portal vein carcinothrombosis(P<0.05).No correlation was found between tumor size,tumor number, differentiation,serum a-fetoprotein(AFP)and the positive rate. 4Dual synergetic catalytic effects boost hydrogen electric oxidation performance of Pd/W_(18)O_(49)显示文摘Developing anode catalysts of substantially enhanced activity for hydrogen oxidation reaction(HOR)and anti-CO poisoning performance is of great importance for the application of proton exchange membrane fuel cells(PEMFCs).Herein,we report Pd cluster in situ decorated urchin-like W_(18)O_(49)(WO_(2.72))electrocatalysts by a photo-reduction method for high performance HOR.The synthesized Pd-WO_(2.72)-L composite of low loading amount of 0.44 wt.%Pd by Xenon light reduction exhibits markedly high HOR catalytic activity and stability in 0.5 M H_(2)SO_(4),and the specific HOR current density and mass activity of Pd-WO_(2.72)-L are~1.5 and~80 times those of 20 wt.%Pt/C catalyst,respectively.Moreover,excellent anti-CO poisoning ability has also been obtained.The excellent HOR activity and anti-CO poisoning performance of Pd-WO_(2.72)-L have been discussed mainly in terms of the dual synergetic catalytic effects between Pd and WO_(2.72):Pd activation to Pd^(δ+)by the electron transfer from Pd to W promotes the hydrogen adsorption and activation to H*species,which results in largely elevated HOR activity;Pd degradation due to the CO poisoning is effectively prevented by WO_(2.72),which is responsible for the excellent CO-tolerance performance.Lingxin Peng Han Tian Xiangzhi Cui Liang Su Ge Meng Zhonghua Ma Shaowen Cao Jianlin Shi 2021Nano Research2021,14,7:2

7Artemin对胰腺癌细胞系MIA PaCa-2浸润能力的研究(英文)显示文摘Objective:The aim of this study was to analyze the capacity of Artemin promoting the motility and invasiveness of MIA PaCa-2 pancreatic cancer (PAC) cells.Methods:The PAC cell line MIA PaCa-2 was cultured in vitro and studied using Transwell chamber analysis.The motility and invasiveness ability affected by different concentrations of Artemin and its receptor GFRα3 were determined.Expression level of matrix metalloproteinase-2 (MMP-2),epithelial cadherin (E-cadherin) were quantitative analysis using RT-PCR and Western blot in MIA PaCa-2 cells stimulated with Artemin and receptor GFRα3.Results:MIA PaCa-2 PAC cell motility and invasiveness was significantly increased with Artemin and its receptor GFRα3 increasing concentrations than control (P<0.01).150 ng/mL was the best of both the role of concentration.MMP-2 was increased significantly (t=6.35,t=7.32),while E-cadherin was significantly lower (t=4.27,t=5.61),after affected by the 150 ng/mL Artemin and GFRα3,respectively.The difference was statistically significant compared with the control group (P<0.01).Conclusion:Artemin and its receptor GFRα3 can promote PAC cell motility and invasiveness ability and contribute to the aggressive behavior.The mechanism may be related to increased expression of MMP-2 molecule and E-cadherin down-regulation expression.Lingxin Meng Yuhua Chi Xiangxu Wang Zhaojun Ding Ling Mou Wen Cui Yingjie Xue 2012The Chinese-German Journal of Clinical Oncology2012,11,4:0
8Effects of Temperature,Salinity and Stocking Density on Larval Survival and Growth of Reciprocal Crosses Between Two Strains of Pacific Oysters,Crassostrea gigas显示文摘The Pacific oyster Crassostrea gigas is one of the most widely cultivated aquaculture species and contributes signifi-cantly to total seafood production for human beings.However,mass mortality occurred frequently,and in some regions almost all oysters died during seed production and grow-out stage.In order to explore whether hybridization breeding can improve its growth and survival,a complete diallel cross between a selected strain‘Haida No.1’(S)and an orange shell variant(O)of C.gigas was carried out.The larval growth and survival were compared among hybrids and purebred strains at temperatures of 16,20,24,28 and 32℃;salinities of 15,20,25,30 and 35;and stocking densities of 0.5,1,2,4 and 8 larvae mL−1.Under different environments,the hybridization between two strains of C.gigas showed the heterosis of growth and survival.The mean shell height and survival rate of the two reciprocal crosses(OS,SO)were significantly higher than those of the two purebred strains(SS,OO)under all environ-mental conditions.In particular,OS showed greater heterosis than the purebred strains and SO progeny.The results showed that the productive traits of the‘Haida No.1’could be improved by crossing with the orange shell line.Meanwhile,the results from this study also indicated that hybridization between the two strains of C.gigas may be a promising way for breeding new variety with high survival rate.MENG Lingxin XU Chengxun LI Qi 2022Journal of Ocean University of China2022,21,4:0
