2Identification of the novel recessive gene pi55(t) conferring resistance to Magnaporthe oryzae显示文摘The elite rice cultivar Yuejingsimiao 2 (YJ2) is characterized by a high level of grain quality and yield, and resistance against Magnaporthe oryzae. YJ2 showed 100% resistance to four fungal populations collected from Guangdong, Sichuan, Liaoning, and Heilongjiang Provinces, which is a higher frequency than that shown by the well-known resistance (R) gene donor cultivars such as Sanhuangzhan 2 and 28zhan. Segregation analysis for resistance with F 2 and F 4 populations indicated the resistance of YJ2 was controlled by multiple genes that are dominant or recessive. The putative R genes of YJ2 were roughly tagged by SSR markers, located on chromosomes 2, 6, 8, and 12, in a bulked-segregant analysis using genome-wide selected SSR markers with F 4 lines that segregated into 3 resistant (R):1 susceptible (S) or 1R:3S. The recessive R gene on chromosome 8 was further mapped to an interval 1.9 cM/152 kb in length by linkage analysis with genomic position-ready markers in the mapping population derived from an F 4 line that segregated into 1R:3S. Given that no major R gene was mapped to this interval, the novel R gene was designated as pi55(t). 3miR-124 suppresses multiple steps of breast cancer metastasis by targeting a cohort of pro-metastatic genes in vitro显示文摘Metastasis is a multistep process involving modification of morphology to suit migration, reduction of tumor cell adhesion to the extracellular matrix, increase of cell mobility, tumor cell resistance to anoikis, and other steps. MicroRNAs are well-suited to regulate tumor metastasis due to their capacity to repress numerous target genes in a coordinated manner, thereby enabling their intervention at multiple steps of the invasion-metastasis cascade. In this study, we identified a microRNA exemplifying these attributes, miR-124, whose expression was reduced in aggressive MDA-MB-231 and SK-3rd breast cancer cells. Down-regulation of miR-124 expression in highly aggressive breast cancer cells contributed in part to DNA hypermethylation around the promoters of the three genes encoding miR-124. Ectopic expression of miR-124 in MDA-MB-231 cells suppressed metastasis-related traits including formation of spindle-like morphology, migratory capacity, adhesion to fibronectin, and anoikis. 9Maternal high salt-diet increases offspring's blood pressure with dysfunction of NO/PKGI signaling pathway in heart tissue显示文摘Background:High salt-diets have become significant threats to human health,resulting in hypertension and cardiovascular diseases.Hypertensive disorders during pregnancy are complicated,since the maternal cardiovascular system undergoes extensive physiological changes during pregnancy.High-salt diets during pregnancy can disturb the intrauterine environment and negatively affect fetal development.Therefore,we explored how high-salt diets during pregnancy could affect the offspring.Methods:Rats were divided into three groups and fed with low,normal,and high salt diets.The offspring were separated into three groups after weaning based on dietary salt concentration.The blood pressure and urine protein content of both dams and offspring were measured.To evaluate cardiac function,we used Masson staining and immunodetection to confirm the fibrosis status.Finally,we extracted protein from cardiac tissue to test the expression levels of the Nitric Oxide(NO)/cGMP-dependent protein kinase I(PKGI)pathway and the angiotensin receptor.Results:High-salt diets increased blood pressure,and offspring previously exposed to high-salt environments were predisposed to hypertension.High-salt diets were also found to induce cardiac fibrosis and exacerbate fibrosis in offspring and alter the epithelial-mesenchymal transition(EMT).Under these conditions,the NO/PKGI pathway was activated in cardiac tissue and the type-1angiotensin II receptor(AT1R)was upregulated,though the type-2 angiotensin II receptor(AT2R)had the opposite effect.Conclusion:High-salt diets induce high blood pressure and increase predisposition to hypertension in offspring.They are accompanied by cardiac fibrosis,which could be caused by the activation of NO/PKGI and upregulation of AT1R.Minshan Huang Xiuying Li Luwen Ren Lin Huang Jiahong Pan Jinlin Yao Lili Du Dunjin Chen Jingsi Chen 2022Gynecology and Obstetrics Clinical Medicine2022,2,2:1

20Anti-oxidative effect of zinc in human umbilical cord mesenchymal stem cells显示文摘Human umbilical cord mesenchymal stem cells(HUC-MSCs)are pluripotent and functional in many biological processes,by which releasing secretary factors to promote the self-repairing of damaged tissue or developing into functional cell at local organ.However,there is a high risk that oxidative stress would reduce the pluripotency and factor-secretion during the preparation and transplantation.Therefore,reducing oxidative stress is expected to improve the efficacy of HUC-MSCs therapy.Zinc[Zn] is an essential trace element which involves in the resistance of oxidative stress.To investigate Zn-regulated signaling pathways,we have profiled the gene expression at transcriptome level in primary HUC-MSCs treated with zinc sulfate,followed with GO and KEGG gene enrichment analysis.Zn treatment improved signal pathways for mineral absorption,cell growth,and cell death.Zn deficiency was mimicked by TPEN administration,which suppressed cell proliferation and reduced the expression of HUC-MSCs surface stem cell markers CD73,CD90 and CD105 by flow cytometry.Nuclear factor erythrocyte 2 related factor 2(Nrf2)plays an important role in antioxidant biological processes.In vitro treatment of Zn significantly increased Nrf2 and Sirt3 expression at gene level and protein level respectively.Zn supplementation inhibited TPEN-induced failure of cell survival and reversed the reduction of Nrf2 and Sirt3 expression,which further reduced the production of ROS.Zn successfully presented its antioxidation effect by activating Nrf2/Sirt3 signaling pathway in HUC-MSCs.Zn supplementation may improve the efficacy of HUC-MSCs therapy with reduced oxidative stress.Xiaodan Lu Yifan Lin Xiuying Lin Qiang Zhang Zihang Wang Xuguang Mi Ruobing Wang Xiaofang Zhang Xu Luan Yan Liu Bing Li YanTan Yanqiu Fang 2021Biophysics Reports2021,7,2:0
