1Antigenicity of tissues and organs from GGTA1/CMAH/β4GalNT2 triple gene knockout pigs显示文摘Clinical xenotransplantations have been hampered by human preformed antibody-mediated damage of the xenografts.To overcome biological incompatibility between pigs and humans,one strategy is to remove the major antigens[Gal,Neu5 Gc,and Sd(a)]present on pig cells and tissues.Triple gene(GGTAI,CMAH,and β4 GalNT2)knockout(TKO)pigs were produced in our laboratory by CRISPR-Cas9 targeting.To investigate the antigenicity reduction in the TKO pigs,the expression levels of these three xenoantigens in the cornea,heart,liver,spleen,lung,kidney,and pancreas tissues were examined.The level of human IgG/IgM binding to those tissues was also investigated,with wildtype pig tissues as control.The results showed that aGal,Neu5 Gc,and Sd(a)were markedly positive in all the examined tissues in wildtype pigs but barely detected in TKO pigs.Compared to wildtype pigs,the liver,spleen,and pancreas of TKO pigs showed comparable levels of human IgG and IgM binding,whereas corneas,heart,lung,and kidney of TKO pigs exhibited significantly reduced human IgG and IgM binding.These results indicate that the antigenicity of TKO pig is significantly reduced and the remaining xenoantigens on porcine tissues can be eliminated via a gene targeting approach.Ronggen Wang Miaomiao Ruan Runjie Zhang Lei Chen Xiaoxue Li Bin Fang Chu Li Xueyang Ren Jiying Liu Qiang Xiong Lining Zhang Yong Jin Lin Li Rongfeng Li Ying Wang Haiyuan Yang Yifan Dai 2019The Journal of Biomedical Research2019,33,4:3
2Comparison of pH Value, TBA Value, Glycogen and Lactic Acid Content in Muscle of Landrace and Yorkshire under Different Storage Time and Conditions显示文摘In order to assess the meat quality under different storage time and conditions,the pH value,TBA value,glycogen and lactic acid content in muscle of Landrace and Yorkshire were measured. The results showed that within 10 h post slaughtering,post slaughtering time had extremely significant influence on pH value and lactic acid content of Landrace,but had no significant influence on glycogen content and TBA value. On the other hand,post slaughtering time had extremely significant influence on pH value of Yorkshire,but had no significant influence on glycogen,lactic acid content and TBA value. The pH values of Yorkshire at 3,5 and 10 h post slaughtering were 5. 21%( P < 0. 01),5. 64%( P < 0. 05) and 5. 59%( P < 0. 05) higher than those of Landrace,respectively; the lactic acid content in muscle of Landrace at 10 h post slaughtering was 56. 04%( P < 0. 05) higher than that of Yorkshire; the TBA value in muscle of Yorkshire at 10 h post slaughtering was 89. 19%( P < 0. 01) higher than that of Landrace; there was no significant difference in glycogen content between Yorkshire and Landrace. When pork stored at 4℃,storage time had no significant influence on pH value and glycogen content in muscle of Landrace,but had significant influence on drip loss,lactic acid content and TBA value; storage time had no significant influence on pH value,glycogen content and TBA value in muscle of Yorkshire,but extremely significant influence on drip loss and significant influence on lactic acid content. The pH value in muscle of Yorkshire at 24 h post storage at 4℃ was 6. 56%( P <0. 05) higher than that of Landrace; the glycogen contents in muscle of Landrace at 24,48,72,96,120 and 144 h post storage at 4℃ were 84. 90%( P <0. 05),78. 40%( P <0. 01),101. 87%( P <0. 05),83. 80%( P < 0. 05),83. 59%( P < 0. 05) and 67. 25%( P < 0. 01) higher than those of Yorkshire,respectively; the drip loss of Landrace at 72 h post storage and TBA value at 144 h post storage were 55. 70%( P < 0. 05) and 141. 33%( P < 0. 01) higher than those of Yorkshire,respectively; there was no significant difference in glycogen content between Yorkshire and Landrace. When pork stored at -20℃,storage time had significant influence on TBA value in muscle of Landrace,but had no significant influence on pH value,thawing water loss rate,glycogen and lactic acid content; storage time had no significant impact on pH value,thawing water loss rate,glycogen content and TBA value in muscle of Yorkshire,and had significant influence on lactic acid content. 3A Bama miniature pig model of monoallelic TSC1 mutation for human tuberous sclerosis complex显示文摘Tuberous sclerosis complex(TSC)is a dominant genetic neurocutaneous syndrome characterized by multiple organ hamartomas.Although rodent models bearing a germline mutation in either TSC1 or TSC2 gene have been generated,they do not develop pathogenic lesions matching those seen in patients with TSC because of the significant differences between mice and humans,highlighting the need for an improved large animal model of TSC.Here,we successfully generate monoallelic TSC1-modified Bama miniature pigs using the CRISPR/Cas9 system along with somatic cell nuclear transfer(SCNT)technology.The expression of phosphorylated target ribosomal protein S6 is significantly enhanced in the piglets,indicating that disruption of a TSC1 allele activate the mechanistic target of rapamycin(mTOR)signaling pathway.Notably,differing from the mouse TSC models reported previously,the TSC1^(+/−)Bama miniature pig developed cardiac rhabdomyoma and subependymal nodules,resembling the major clinical features that occur in patients with TSC.These TSC1^(+/−)Bama miniature pigs could serve as valuable large animal models for further elucidation of the pathogenesis of TSC and the development of therapeutic strategies for TSC disease.Xiaoxue Li Tingdong Hu Jiying Liu Bin Fang Xue Geng Qiang Xiong Lining Zhang Yong Jin Xiaorui Liu Lin Li Ying Wang Rongfeng Li Xiaochun Bai Haiyuan Yang Yifan Dai 2020Journal of Genetics and Genomics2020,47,12:2

6Combined Treatment with Electrocauterization,Carbon Dioxide Laser,and Microneedle Fractional Radiofrequency for Facial Angiofibromas in Tuberous Sclerosis Complex:A Case Report and Literature Review显示文摘Tuberous sclerosis complex is a type of genetic multisystem disease that causes hamartomas in various organs.Facial angiofibromas commonly occur in 80%of patients and are prominently distributed over the cheek,chin,and nasolabial folds with severe disfigurement and emotional distress.Recently,photoelectric devices have been identified for the treatment of angiofibromas with great efficacy and fewer side effects.We report a case of a 42-year-old man with facial angiofibromas,who was treated with a combination of high-frequency electrocauterization,Ultrapulse CO_(2) laser,and microneedle fractional radiofrequency with 7 sessions and a 6-month follow-up.The patient showed great improvement in relation to the elevated lesions and nodules.A low recurrence rate was observed.This is the first study to investigate the efficacy of high-frequency electrocauterization and microneedle fractional radiofrequency in angiofibromas.It may provide an optimal approach for clinicians wherein a combined treatment of various lasers and electric devices is effective for complicated,protuberant,and firm angiofibromas of specific patients.Jiying DONG Shen WANG Min YAN Jing MI Ying ZENG Huyan LIN Yiqiu ZHANG Min YAO 2021Chinese Journal Of Plastic and Reconstructive Surgery2021,3,1:1

10Improving maize growth and development simulation by integrating temperature compensatory effect under plastic film mulching into the AquaCrop model显示文摘Temperature compensatory effect, which quantifies the increase in cumulative air temperature from soil temperature increase caused by mulching, provides an effective method for incorporating soil temperature into crop models. In this study, compensated temperature was integrated into the AquaCrop model to investigate the capability of the compensatory effect to improve assessment of the promotion of maize growth and development by plastic film mulching(PM). A three-year experiment was conducted from2014 to 2016 with two maize varieties(spring and summer) and two mulching conditions(PM and non-mulching(NM)), and the AquaCrop model was employed to reproduce crop growth and yield responses to changes in NM, PM, and compensated PM. A marked difference in soil temperature between NM and PM was observed before 50 days after sowing(DAS) during three growing seasons. During sowing–emergence and emergence–tasseling, the increase in air temperature was proportional to the compensatory coefficient, with spring maize showing a higher compensatory temperature than summer maize. Simulation results for canopy cover(CC) were generally in good agreement with the measurements, whereas predictions of aboveground biomass and grain yield under PM indicated large underestimates from 60 DAS to the end of maturity. Simulations of spring maize biomass and yield showed general increase based on temperature compensation, accompanied by improvement in modeling accuracy, with RMSEs decreasing from 2.5 to 1.6 t ha^(-1)and from 4.1 t to 3.4 t ha^(-1). Improvement in biomass and yield simulation was less pronounced for summer than for spring maize, implying that crops grown during low-temperature periods would benefit more from the compensatory effect.
