1Salusins protect myocardium against ischemic injury by alleviating endoplasmic reticulum stress显示文摘Salusins are regulatory peptides that affect cardiovascular function. We previously reported that salusin-α and -β protected cultured cardiomyocytes from serum deprivation-induced cell death through upregulating glucose-regulated protein 78 (GRP78), an endoplasmic reticulum (ER) resident protein whose overexpression acts as a marker and suppressor of ER stress. The present study examined whether salusin-α and -β inhibit ER stress in ischemic myocardium. In a rat model of myocardial infarction created by ligating the left anterior descending coronary artery (LAD), salusin-α or -β was intravenously injected at 5 or 15 nmol kg 1 15 min prior to 2 h of LAD occlusion. The high dose of salusin-α and -β significantly improved heart function and hemodynamics in LAD-occluded rats, but had no effects in sham-operated rats. The arrhythmias caused by LAD occlusion were markedly attenuated by salusin-α and -β. The apoptotic rate in ischemic myocardium was reduced from 31.5%±3.7% to 19.8%±2.2% and 12.3%±2.2%, and the infarct size was reduced from 53.4%±4.0% of the risk area to 26.5%±9.7% and 23.7%±8.9% by 15 nmol kg 1 salusin-α and -β, respectively. Furthermore, salusin-α and -β prevented the activation of GRP78 and ER stress-specific apoptotic effectors caspase-12 and CHOP (C/EBP homologous protein), and attenuated the reduction of an ER stress-associated antiapoptotic protein Bcl-2 in ischemic cardiac tissue. The salusins also inhibited the ER stress induced by tunicamycin in cultured rat H9c2 cardiomyocytes. 2Combined Treatment with Electrocauterization,Carbon Dioxide Laser,and Microneedle Fractional Radiofrequency for Facial Angiofibromas in Tuberous Sclerosis Complex:A Case Report and Literature Review显示文摘Tuberous sclerosis complex is a type of genetic multisystem disease that causes hamartomas in various organs.Facial angiofibromas commonly occur in 80%of patients and are prominently distributed over the cheek,chin,and nasolabial folds with severe disfigurement and emotional distress.Recently,photoelectric devices have been identified for the treatment of angiofibromas with great efficacy and fewer side effects.We report a case of a 42-year-old man with facial angiofibromas,who was treated with a combination of high-frequency electrocauterization,Ultrapulse CO_(2) laser,and microneedle fractional radiofrequency with 7 sessions and a 6-month follow-up.The patient showed great improvement in relation to the elevated lesions and nodules.A low recurrence rate was observed.This is the first study to investigate the efficacy of high-frequency electrocauterization and microneedle fractional radiofrequency in angiofibromas.It may provide an optimal approach for clinicians wherein a combined treatment of various lasers and electric devices is effective for complicated,protuberant,and firm angiofibromas of specific patients.Jiying DONG Shen WANG Min YAN Jing MI Ying ZENG Huyan LIN Yiqiu ZHANG Min YAO 2021Chinese Journal Of Plastic and Reconstructive Surgery2021,3,1:1
3Direct-writing Process and in vivo Evaluation of Prevascularized Composite Constructs for Muscle Tissue Engineering Application显示文摘Tissue engineered skeletal muscle is expected to treat muscle defects caused by trauma and disease.However,designing and manufacturing thick and complex tissue engineered skeletal muscle requires vascularization to ensure its internal cell viability and nutrient supply in vitro.In this article,we developed a set of Direct-Writing(DW)bio-printing procedure to manufacture a prevascularized composite construct with Human Umbilical Vein Endothelial Cell(HUVEC)and C2C12 cells for muscle tissue engineering application.We put the cells into the construct during the DW process to obtain the prevascularization and intend to promote its vascularization in vivo later.The constructs with cells or without cells were implanted respectively into nude mice back for 3 weeks,after which the mice healthily live for all the time and all the implants are tightly bonded to the host.From immunohistochemical analysis,CD31-positive blood vessels existed in the implanted samples with cells are more substantial than those without cells,but the implanted samples with HUVEC and C2C12 cells have much more number of small blood vessels distributing evenly.Moreover,the implants with cells,especially that with HUVEC and C2C12 cells,are able to get better fusion with the host skin and subcutaneous tissues.Histological analysis demonstrates that our DW-based constructs have the potential to be getting to vascularize the tissue engineered muscle.Qin Lian Tingze Zhao Tian Jiao Yige Huyan Heng Gu Lin Gao 2020Journal of Bionic Engineering2020,17,3:1
