| 1 | Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy. | Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen | 2021 | Cancer Communications2021,41,11: | 29 |
| 2 | Genetic and biological characteristics of the globally circulating H5N8 avian influenza viruses and the protective efficacy offered by the poultry vaccine currently used in China显示文摘The H5N8 avian influenza viruses have been widely circulating in wild birds and are responsible for the loss of over 33 million domestic poultry in Europe, Russia, Middle East, and Asia since January 2020. To monitor the invasion and spread of the H5N8 virus in China, we performed active surveillance by analyzing 317 wild bird samples and swab samples collected from 41,172 poultry all over the country. We isolated 22 H5N8 viruses from wild birds and 14 H5N8 viruses from waterfowls. Genetic analysis indicated that the 36 viruses formed two different genotypes: one genotype viruses were widely detected from different wild birds and domestic waterfowls;the other genotype was isolated from a whopper swan. We further revealed the origin and spatiotemporal spread of these two distinct H5N8 virus genotypes in 2020 and 2021. Animal studies indicated that the H5N8 isolates are highly pathogenic to chickens, mildly pathogenic in ducks, but have distinct pathotypes in mice. Moreover, we found that vaccinated poultry in China could be completely protected against H5N8 virus challenge. | 3 | Investigation on Uniaxial Tensile Instability of USIBOR1500 Steel Sheets at Elevated Temperature显示文摘In the sheet forming process with stretch, diffuse instability and localized instability usually occur in the process one after another. The necking rate has a great impact on the instability process and the forming limits of the rate sensitive material, such as USIBOR1500 steel sheets at elevated temperature. The available reports about this steel mainly focus on hot uniaxial tensile, martensitic transformation and forming process, but there are few investigations on instability behavior and forming limits. Based on Inoue Kachiro’s flow stress constitutive model at elevated temperature, combined with Swift’s diffuse instability rule and Hill’s localized instability rule, the relation is theoretically deduced between the diffuse necking rate along with the localized necking rate and the exponent of strain rate sensitivity together with the strain rate in the case of the USIBOR1500 steel sheets at elevated temperature. According to the time–temperature characteristics of the hot stamping process, tensile tests of the steel sheets were carried out on Gleeble3800, and the stress–strain curves were obtained at different temperatures and different strain rates. Then the values of the exponent of strain rate sensitivity and the hardening exponent were obtained through fitting the curves by least squares. The tests also helped to provide the distribution laws of the major strain of the specimens and the uniform strains as well as the necking width. | 5 | Spatial pattern and drivers of urbanization in China's mid-level developing urban agglomeration:A case study of Chang-Zhu-Tan显示文摘Urban agglomeration has become the main form of regional spatial organization in China.While most of the existing studies of urban agglomeration in China have focused on the eastern coastal areas,urban agglomeration with mid-level development in the rest of the country has been overlooke1.To better understand the urbanization process of the mid-level developing urban agglomeration,this study investigated the clustering pattern and the drivers of both urban population and firm dynamics during 2005-2015 in the Changsha-Zhuzhou-Xiangtan(Chang-Zhu-Tan)urban agglomeration of China using the methods ofkernel density estimation and geographic detection.Our results show that centralization was obvious,although decentralization also occurred in Chang-Zhu-Tan,and that the spatial agglomeration was promoted by several factors,such as administrative resources,location advantage,labor cost,and consumption capacity.Some problems hindering the development of this region were also discovered:administrative resources played a critical role in urbanization because small towns and villages did not receive enough attention,and the effect of local policy was not as beneficial as expected.These findings partly explain the relatively slow development of mid-level developing urban agglomerations and have important implications for promoting healthier urbanization. | Yanhua He Yi Lin Guohua Zhou Yixuan Zhu Kai Tang | 2021 | Regional Sustainability2021,2,1: | 6 |

| 9 | Toxic Effects of Three Heavy Metallic Ions on Rana zhenhaiensis Tadpoles显示文摘Heavy metal pollution is widespread in some areas of China and results in contamination of land, water, and air with which all living organisms interact. In this study, we used three heavy metallic ions(Cu2+, Pb2+ and Zn2+) to assess their toxicity effects on mortality, blood biomarker and growth traits(body length and body mass) of Rana zhenhaiensis tadpoles. The results showed that the toxicity levels of the three metallic ions were different when conducted with different experiment designs. For acute toxicity tests, Cu2+ was the most toxic with the highest tadpole mortality. The mortalities of tadpoles showed significant differences among the treatments at the same exposure time endpoints(24, 48, 72 and 96h). Results from repeated measures ANOVA indicated that metallic ion concentration, exposure time and their interactions significantly affected the mortalities of R. zhenhaiensis tadpoles. Also, the toxicity effects of all binary combinations of the three metallic ion treatments showed synergism. The half lethal concentrations(LC50) decreased with increasing exposure time during the experimental period, and the safe concentration(SC) values of Cu2+, Pb2+ and Zn2+ were different from each other. Combined and compared LC50 values with previous data reported, it is suggestes that the toxicity levels of metal pollution to anuran tadpoles should be species-and age-related. For blood biomarker tests, Zn2+ was the most toxic with the highest total frequencies of abnormal erythrocytic nucleus. All three metallic ions caused higher abnormal erythrocytic nucleus compared with control groups. In a chronic toxicity test, Pb2+ was the most toxic with lowest growth traits. Survival rate(except for 18 days), total body length and body mass showed significant differences among the treatments. | 10 | Effects of Predation by Invasive Western Mosquitofish(Gambusia affinis) on Survival of Eggs,Embryos and Tadpoles of Pelophylax nigromaculatus and Duttaphrynus melanostictus in South China显示文摘Alien species are one of the most serious threats to the decline and extinction of native amphibian populations. In this study, we examined the predation of invasive Western Mosquitofish Gambusia affinis on the eggs, embryos, and tadpoles of Duttaphrynus melanostictus and Pelophylax nigromaculatus in south China. Our results suggested that the survival of eggs and embryos remaining in the egg capsules of P. nigromaculatus and D. melanostictus was significantly higher than those removed from the egg capsule at 12-h intervals within 72 h in the presence of G. affinis. The survival of P. nigromaculatus eggs and embryos without egg capsules was significantly lower than those of D. melanostictus without egg capsules. The survival of P. nigromaculatus eggs and embryos with egg capsules was significantly higher than those of D. melanostictus with egg capsules from 24 h to 72 h except for 12 h. The survival of D. melanostictus tadpoles was significantly higher than that of P. nigromaculatus tadpoles in the presence of G. affinis. The survival of Gosner stage 26 tadpoles of P. nigromaculatus was significantly higher than that of Gosner stage 30 tadpoles from 12 h to 60 h, but there were no significant differences at 72 h. In contrast, the survival of Gosner stage 26 tadpoles of D. melanostictus was significantly lower than that of Gosner stage 30 tadpoles within 72 h, recording every 12 h. The increasing temperature caused a significant increase in predation by G. affinis on P. nigromaculatus eggs and embryos. The outer jelly capsule surrounding anurans eggs might serve as a mechanical defense against predation by G. affinis due to its large diameter, relatively stationary state and unpalatability. The differences in the vulnerability of P. nigromaculatus and D. melanostictus embryos and tadpoles to G. affinis probably due to differences in the unpalatability, black skin and activity. | 16 | Injectable and thermosensitive hydrogels mediating a universal macromolecular contrast agent with radiopacity for noninvasive imaging of deep tissues显示文摘It is very challenging to visualize implantable medical devices made of biodegradable polymers in deep tissues.Herein,we designed a novel macromolecular contrast agent with ultrahigh radiopacity(iodinate content>50%)via polymerizing an iodinated trimethylene carbonate monomer into the two ends of poly(ethylene glycol)(PEG).A set of thermosensitive and biodegradable polyester-PEG-polyester triblock copolymers with varied polyester compositions synthesized by us,which were soluble in water at room temperature and could spontaneously form hydrogels at body temperature,were selected as the demonstration materials.The addition of macromolecular contrast agent did not obviously compromise the injectability and thermogelation properties of polymeric hydrogels,but conferred them with excellent X-ray opacity,enabling visualization of the hydrogels at clinically relevant depths through X-ray fluoroscopy or Micro-CT.In a mouse model,the 3D morphology of the radiopaque hydrogels after injection into different target sites was visible using Micro-CT imaging,and their injection volume could be accurately obtained.Furthermore,the subcutaneous degradation process of a radiopaque hydrogel could be non-invasively monitored in a real-time and quantitative manner.In particular,the corrected degradation curve based on Micro-CT imaging well matched with the degradation profile of virgin polymer hydrogel determined by the gravimetric method.These findings indicate that the macromolecular contrast agent has good universality for the construction of various radiopaque polymer hydrogels,and can nondestructively trace and quantify their degradation in vivo.Meanwhile,the present methodology developed by us affords a platform technology for deep tissue imaging of polymeric materials. | Xiaohui Wu Xin Wang Xiaobin Chen Xiaowei Yang Qian Ma Guohua Xu Lin Yu Jiandong Ding | 2021 | Bioactive Materials2021,6,12: | 1 |
