| 1 | Virus-induced gene silencing(VIGS)in Chinese narcissus and its use in functional analysis of NtMYB3显示文摘Virus-induced gene silencing(VIGS)is a natural defense mechanism of plants,which can cause sequence-specific degradation of viral RNA and is often used for studying the functional genome.At present,the VIGS system mediated by Tobacco rattle virus(TRV)has been successfully established in many plant species.However,the VIGS system in Chinese narcissus had not been reported yet.In this paper,the first use of the TRV-VIGS system in Chinese narcissus is described in detail.By injecting pTRV2-GFP into narcissus leaves,we confirmed that TRV could infect narcissus and move in narcissus plants.After the inoculation of pTRV2-Nt PDS by two methods,phenomena such as photo-bleaching appeared in narcissus leaves and qRT-PCR results proved that PDS gene of narcissus was successfully silenced.Besides,pTRV2-NtMYB3 was inoculated into basal plates of narcissus to study the function of NtMYB3;the results showed that NtMYB3 is an inhibitor of flavonoids biosynthesis,which can increase the content of proanthocyanidins in basal plates of narcissus by repressing the expression level of NtFLS.These results indicate that the TRV-VIGS system has been successfully established in Chinese narcissus and successfully applied to the functional study of NtMYB3. | Ping Zhou Jiayu Peng Minjie Zeng Lixuan Wu Yuxin Fan Lihui Zeng | 2021 | Horticultural Plant Journal2021,7,6: | 4 |
| 2 | Melt-Lithosphere Interaction Controlled Compositional Variations in Mafic Dikes from Fujian Province,Southeastern China显示文摘Late Mesozoic magmatism in southeastern China has been widely considered to be related to the subduction of the Paleo-Pacific Plate.However,it remains controversial whether mafic rocks are derived from the lithosphere or the asthenosphere.Here we present a comprehensive study on mafic dikes from Fujian Province in southeastern China,aiming to understand their source.Two types of mafic rocks have been recognized based on their trace-element features.Type-Ⅰrocks show arc-like trace-elemental characteristics,while type-Ⅱrocks are distinguished by their relatively flat patterns in primitive-mantle-normalized trace-element diagram.Despite such differences between two types of rocks,these mafic dikes show two trends in the plots of^(87)Sr/^(86)Sr(i)versus La/Nb,which can be explained by the influences of crustal contamination and melt-lithospheric mantle interaction,respectively.^(87)Sr/^(86)Sr(i),La/Nb,Sr/Y and Zr/Y ratios of type-I rocks are significantly correlated to the thickness of the underlying lithosphere,and the signals of lithosphere are clearer with increasing lithospheric thickness.This highlights the important influences of melt-lithosphere interaction during their formation.Such observations also indicate that these mafic rocks are more likely to have been originated from the asthenosphere rather than the lithospheric mantle. | Zhuliang Lei Gang Zeng Jianqiang Liu Xiaojun Wang Lihui Chen Xiaoyu Zhang Jinhua Shi | 2021 | Journal of Earth Science2021,32,6: | 1 |
| 3 | The haplotype-resolved autotetraploid genome assembly provides insights into the genomic evolution and fruit divergence in wax apple(Syzygium samarangense(Blume)Merr.and Perry)显示文摘Wax apple(Syzygium samarangense)is an economically important fruit crop with great potential value to human health because of its richness in antioxidant substances.Here,we present a haplotype-resolved autotetraploid genome assembly of the wax apple with a size of 1.59 Gb.Comparative genomic analysis revealed three rounds of whole-genome duplication(WGD)events,including two independent WGDs after WGT-γ.Resequencing analysis of 35 accessions partitioned these individuals into two distinct groups,including 28 landraces and seven cultivated species,and several genes subject to selective sweeps possibly contributed to fruit growth,including the KRP1-like,IAA17-like,GME-like,and FLACCA-like genes.Transcriptome analysis of three different varieties during flower and fruit development identified key genes related to fruit size,sugar content,and male sterility.We found that AP2 also affected fruit size by regulating sepal development in wax apples.The expression of sugar transport-related genes(SWEETs and SUTs)was high in‘ZY’,likely contributing to its high sugar content.Male sterility in‘Tub’was associated with tapetal abnormalities due to the decreased expression of DYT1,TDF1,and AMS,which affected early tapetum development.The chromosome-scale genome and large-scale transcriptome data presented in this study offer new valuable resources for biological research on S.samarangense and shed new light on fruit size control,sugar metabolism,and male sterility regulatory metabolism in wax apple. | Xiuqing Wei Min Chen Xijuan Zhang Yinghao Wang Liang Li Ling Xu Huanhuan Wang Mengwei Jiang Caihui Wang Lihui Zeng Jiahui Xu | 2023 | Horticulture Research2023,10,12: | 0 |
| 4 | Overexpressed NEDD8 as a potential therapeutic target in esophageal squamous cell carcinoma显示文摘Objective:The hyperactivated neddylation pathway plays an important role in tumorigenesis and is emerging as a promising anticancer target.We aimed to study whether NEDD8(neural precursor cell expressed,developmentally down-regulated 8)might serve as a therapeutic target in esophageal squamous cell carcinoma(ESCC).Methods:The clinical relevance of NEDD8 expression was evaluated by using The Cancer Genome Atlas(TCGA)database and tissue arrays.NEDD8-knockdown ESCC cells generated with the CRISPR/Cas9 system were used to explore the anticancer effects and mechanisms.Quantitative proteomic analysis was used to examine the variations in NEDD8 knockdown-induced biological pathways.The cell cycle and apoptosis were assessed with fluorescence activated cell sorting.A subcutaneous-transplantation mouse tumor model was established to investigate the anticancer potential of NEDD8 silencing in vivo.Results:NEDD8 was upregulated at both the mRNA and protein expression levels in ESCC,and NEDD8 overexpression was associated with poorer overall patient survival(mRNA level:P=0.028,protein level:P=0.026,log-rank test).Downregulation of NEDD8 significantly suppressed tumor growth both in vitro and in vivo.Quantitative proteomic analysis revealed that downregulation of NEDD8 induced cell cycle arrest,DNA damage,and apoptosis in ESCC cells.Mechanistic studies demonstrated that NEDD8 knockdown led to the accumulation of cullin-RING E3 ubiquitin ligases(CRLs)substrates through inactivation of CRLs,thus suppressing the malignant phenotype by inducing cell cycle arrest and apoptosis in ESCC.Rescue experiments demonstrated that the induction of apoptosis after NEDD8 silencing was attenuated by DR5 knockdown.Conclusions:Our study elucidated the anti-ESCC effects and underlying mechanisms of NEDD8 knockdown,and validated NEDD8 as a potential target for ESCC therapy. | Jingrong Xian Shiwen Wang Yanyu Jiang Lihui Li Lili Cai Ping Chen Yue Liu Xiaofei Zeng Guoan Chen Chen Ding Robert MHoffman Lijun Jia Hu Zhao Yanmei Zhang | 2022 | Cancer Biology & Medicine2022,19,4: | 0 |

| 7 | Induction of multiple subroutines of regulated necrosis in murine macrophages by natural BH3-mimetic gossypol显示文摘Macrophages are critical sentinel cells armed with multiple regulated necrosis pathways,including pyroptosis,apoptosis followed by secondary necrosis,and necroptosis,and are poised to undergo distinct form(s)of necrosis for tackling dangers of pathogenic infection or toxic exposure.The natural BH3-mimetic gossypol is a toxic phytochemical that can induce apoptosis and/or pyroptotic-like cell death,but what exact forms of regulated necrosis are induced remains largely unknown.Here we demonstrated that gossypol induces pyroptotic-like cell death in both unprimed and lipopolysaccharide-primed mouse bone marrow-derived macrophages(BMDMs),as evidenced by membrane swelling and ballooning accompanied by propidium iodide incorporation and lactic acid dehydrogenase release.Notably,gossypol simultaneously induces the activation of both pyroptotic and apoptotic(followed by secondary necrosis)pathways but only weakly activates the necroptosis pathway.Unexpectedly,gossypol-induced necrosis is independent of nucleotide-binding oligomerization domain-like receptor family pyrin domain containing 3(NLRP3)inflammasome,as neither inhibitor for the NLRP3 pathway nor NLRP3 deficiency protects the macrophages from the necrosis.Furthermore,necrotic inhibitors or even pan-caspase inhibitor alone does not or only partly inhibit such necrosis.Instead,a combination of inhibitors composed of pan-caspase inhibitor IDN-6556,RIPK3 inhibitor GSK′872 and NADPH oxidase inhibitor GKT137831 not only markedly inhibits the necrosis,with all apoptotic and pyroptotic pathways being blocked,but also attenuates gossypol-induced peritonitis in mice.Lastly,the activation of the NLRP3 pathway and apoptotic caspase-3 appears to be independent of each other.Collectively,gossypol simultaneously induces the activation of multiple subroutines of regulated necrosis in macrophages depending on both apoptotic and inflammatory caspases. | Meiyan Zhong Yuanting Huang Bo Zeng Lihui Xu Chunsu Zhong Jiahao Qiu Xunjia Ye Mingye Chen Bo Hu Dongyun Ouyang Xianhui He | 2022 | Acta Biochimica et Biophysica Sinica2022,54,1: | 0 |
