3Combined acoustic radiation force impulse, aminotransferase to platelet ratio index and Forns index assessment for hepatic fibrosis grading in hepatitis B显示文摘AIM: To investigate the combined diagnostic accuracy of acoustic radiation force impulse(ARFI), aspartate aminotransferase to platelet ratio index(APRI) and Forns index for a non-invasive assessment of liver fibrosis in patients with chronic hepatitis B(CHB). METHODS: In this prospective study, 206 patients had CHB with liver fibrosis stages F0-F4 classified by METAVIR and 40 were healthy volunteers were measured by ARFI, APRI and Forns index separately or combined as indicated. RESULTS: ARFI, APRI or Forns index demonstrated a significant correlation with the histological stage(all P < 0.001). According to the AUROC of ARFI and APRI for evaluating fibrotic stages more than F2, ARFI showed an enhanced diagnostic accuracy than APRI(P < 0.05). The combined measurement of ARFI and APRI exhibited better accuracy than ARFI alone when evaluating ≥ F2 fibrotic stage(Z = 2.77, P = 0.006). Combination of ARFI, APRI and Forns index did not obviously improve the diagnostic accuracy compared to the combination of ARFI and APRI(Z = 0.958, P = 0.338). 4A phase I study of different doses and frequencies of pegylated recombinant human granulocyte-colony stimulating factor(PEG rhG-CSF) in patients with standard-dose chemotherapy-induced neutropenia显示文摘Objective: The recommended dose of prophylactic pegylated recombinant human granulocyte-colony stimulating factor(PEG rhG-CSF) is 100 μg/kg once per cycle for patients receiving intense-dose chemotherapy.However, few data are available on the proper dose for patients receiving less-intense chemotherapy. The aim of this phase I study is to explore the proper dose and administration schedule of PEG rhG-CSF for patients receiving standard-dose chemotherapy.Methods:Eligible patients received 3-cycle chemotherapy every 3 weeks.No PEG rhG-CSF was given in the first cycle.Patients experienced grade 3 or 4 neutropenia would then enter the cycle 2 and 3.In cycle 2,patients received a single subcutaneous injection of prophylactic PEG rhG-CSF on d 3,and received half-dose subcutaneous injection in cycle 3 on d 3 and d 5,respectively.Escalating doses(30,60,100 and 200μg/kg)of PEG rhG-CSF were investigated.Results:A total of 26 patients were enrolled and received chemotherapy,in which 24 and 18 patients entered cycle 2 and cycle 3 treatment,respectively.In cycle 2,the incidence of grade 3 or 4 neutropenia for patients receiving single-dose PEG rhG-CSF of 30,60,100 and 200 μg/kg was 66.67%,33.33%,22.22% and 0,respectively,with a median duration less than 1(0–2)d.No grade 3 or higher neutropenia was noted in cycle 3 in all dose cohorts.Conclusions:The pharmacokinetic and pharmacodynamic profiles of PEG rhG-CSF used in cancer patients were similar to those reported,as well as the safety.Double half dose administration model showed better efficacy result than a single dose model in terms of grade 3 neutropenia and above.The single dose of 60 μg/kg,100 μg/kg and double half dose of 30 μg/kg were recommended to the phase Ⅱ study,hoping to find a preferable method for neutropenia treatment.Yan Qin Xiaohong Han Lin Wang Ping Du Jiarui Yao Di Wu Yuanyuan Song Shuxiang Zhang Le Tang Yuankai Shi 2017Chinese Journal of Cancer Research2017,29,5:11

6CiteOpinion: Evidence-based Evaluation Tool for Academic Contributions of Research Papers Based on Citing Sentences显示文摘Purpose:To uncover the evaluation information on the academic contribution of research papers cited by peers based on the content cited by citing papers,and to provide an evidencebased tool for evaluating the academic value of cited papers.Design/methodology/approach:CiteOpinion uses a deep learning model to automatically extract citing sentences from representative citing papers;it starts with an analysis on the citing sentences,then it identifies major academic contribution points of the cited paper,positive/negative evaluations from citing authors and the changes in the subjects of subsequent citing authors by means of Recognizing Categories of Moves(problems,methods,conclusions,etc.),and sentiment analysis and topic clustering.Findings:Citing sentences in a citing paper contain substantial evidences useful for academic evaluation.They can also be used to objectively and authentically reveal the nature and degree of contribution of the cited paper reflected by citation,beyond simple citation statistics.Practical implications:The evidence-based evaluation tool CiteOpinion can provide an objective and in-depth academic value evaluation basis for the representative papers of scientific researchers,research teams,and institutions.Originality/value:No other similar practical tool is found in papers retrieved.Research limitations:There are difficulties in acquiring full text of citing papers.There is a need to refine the calculation based on the sentiment scores of citing sentences.Currently,the tool is only used for academic contribution evaluation,while its value in policy studies,technical application,and promotion of science is not yet tested.Xiaoqiu Le Jingdan Chu Siyi Deng Qihang Jiao Jingjing Pei Liya Zhu Junliang Yao 2019Journal of Data and Information Science2019,4,4:6
7Score model for predicting acute-on-chronic liver failure risk in chronic hepatitis B显示文摘AIM: To establish a clinical scoring model to predict risk of acute-on-chronic liver failure(ACLF) in chronic hepatitis B(CHB) patients.METHODS: This was a retrospective study of 1457 patients hospitalized for CHB between October 2008 and October 2013 at the Beijing Ditan Hospital, Capital Medical University, China. The patients were divided into two groups: severe acute exacerbation(SAE) group(n = 382) and non-SAE group(n = 1075). The SAE group was classified as the high-risk group based on the higher incidence of ACLF in this group than in the non-SAE group(13.6% vs 0.4%). Two-thirds of SAE patients were randomly assigned to risk-model derivation and the other one-third to model validation. Univariate risk factors associated with the outcome were entered into a multivariate logistic regression model for screening independent risk factors. Each variable was assigned an integer value based on the regression coefficients, and the final score was the sum of these values in the derivation set. Model discrimination and calibration were assessed using area under the receiver operating characteristic curve and the Hosmer-Lemeshow test. RESULTS: The risk prediction scoring model includedthe following four factors: age ≥ 40 years, total bilirubin ≥ 171 μmol/L, prothrombin activity 40%-60%, and hepatitis B virus DNA > 107 copies/m L. The sum risk score ranged from 0 to 7; 0-3 identified patients with lower risk of ACLF, whereas 4-7 identified patients with higher risk. The Kaplan-Meier analysis showed the cumulative risk for ACLF and ACLF-related death in the two risk groups(0-3 and 4-7 scores) of the primary cohort over 56 d, and log-rank test revealed a significant difference(2.0% vs 33.8% and 0.8% vs 9.4%, respectively; both P < 0.0001). 10Synergistic effects of magnesium ions and simvastatin on attenuation of high-fat diet-induced bone loss显示文摘Introduction:Magnesium(Mg)has a prophylactic potential against the onset of hyperlipidemia.Similar to statin,Mg is recommended as lipid-lowering medication for hypercholesterolemia and concomitantly exhibits an association with increased bone mass.The combination of statin with Mg ions(Mg^(2+))may be able to alleviate the high-fat diet(HFD)-induced bone loss and reduce the side-effects of statin.This study aimed to explore the feasibility of combined Mg^(2+)with simvastatin(SIM)for treating HFD-induced bone loss in mice and the involving mechanisms.Materials and methods:C57BL/6 male mice were fed with a HFD or a normal-fat diet(NFD).Mice were intraperitoneally injected SIM and/or orally received water with additional Mg^(2+)until sacrificed.Enzyme-linked immunosorbent assay was performed to measure cytokines and cholesterol in serum and liver lysates.Bone mineral density(BMD)and microarchitecture were assessed by micro-computed tomography(μCT)in different groups.The adipogenesis in palmitate pre-treated HepG2 cells was performed under various treatments.Results:μCT analysis showed that the trabecular bone mass was significantly lower in the HFD-fed group than that in NFD-fed group since week 8.The cortical thickness in HFD-fed group had a significant decrease at week 24,as compared with NFD-fed group.The combination of Mg^(2+)and SIM significantly attenuated the trabecular bone loss in HFD-fed mice via arresting the osteoclast formation and bone resorption.Besides,such combination also reduced the hepatocytic synthesis of cholesterol and inhibited matrix metallopeptidase 13(Mmp13)mRNA expression in pre-osteoclasts.Conclusions:The combination of Mg^(2+)and SIM shows a synergistic effect on attenuating the HFD-induced bone loss.Our current formulation may be a cost-effective alternative treatment to be indicated for obesity-related bone loss.Bingyang Dai Xu Li Jiankun Xu Yuwei Zhu Le Huang Wenxue Tong Hao Yao Dick Ho-kiu Chow Ling Qin 2021Bioactive Materials2021,6,8:4

12Clinical Outcome of Autologous Hematopoietic Stem Cell Infusion via Hepatic Artery or Portal Vein in Patients with End-stage Liver Diseases显示文摘Objective To investigate the efficacy of hematopoietic stem cell(HSC) transplantation via the hepatic artery vs.the portal vein for end-stage liver disease(ESLD).Methods Patients with hepatic decompensation were prospectively recruited from September 2010 to September 2012 to receive HSC transplantation via the hepatic artery or the portal vein.Liver function was examined at 3,6,and 12 months after transplantation.Liver biopsy results were analyzed using the Knodell score.Results Eighty patients(58 males and 22 females) were enrolled in the study.The Child-Pugh score was grade B in 69 cases,and grade C in the remaining 11 cases.HSC transplantation was performed via the portal vein in 36 patients and via the hepatic artery in 44 patients.ALT levels decreased while serum albumin levels increased significantly in both groups at 6 and 12 months after HSC transplantation(P<0.05 compared with pre-transplantation levels).Total bilirubin levels decreased significantly in both groups at 3,6,and 12 months after HSC transplantation(P<0.05 compared with pre-transplantation levels).Additionally,prothrombin time decreased in both groups at 12 months after HSC transplantation(P<0.05 compared with pre-transplantation level).There were no significant differences in ALT,total bilirubin and prothrombin time between the two groups either before or after transplantation.Moreover,Knodell score decreased significantly at 6 and 12 months.Histological examination showed that liver cell edema,degeneration,necrosis,and inflammation were significantly relieved at 3,6,and 12 months after transplantation.The incidence of portal vein thrombosis,upper gastrointestinal bleeding,and hepatic encephalopathy were 1.25%,3.75%,and 2.5% respectively.The one-year survival rate was 100%.Conclusions Autologous HSC transplantation improves liver function and histology in ESLD patients.The administration route of HSC has no significant impact on the efficacy of transplantation.Xiao-lun Huang Le Luo Lan-yun Luo Hua Xue Ling-ling Wei Yu-tong Yao Hai-bo Zou Xiao-bing Huang Yi-fan Zhu Tian Zhang Ping Xie Mao-zhu Yang Shao-ping Deng 2014Chinese Medical Sciences Journal2014,29,1:4
13A multivariate grey incidence model for different scale data based on spatial pyramid pooling显示文摘In order to solve the problem that existing multivariate grey incidence models cannot be applied to time series on different scales, a new model is proposed based on spatial pyramid pooling.Firstly, local features of multivariate time series on different scales are pooled and aggregated by spatial pyramid pooling to construct n levels feature pooling matrices on the same scale. Secondly,Deng's multivariate grey incidence model is introduced to measure the degree of incidence between feature pooling matrices at each level. Thirdly, grey incidence degrees at each level are integrated into a global incidence degree. Finally, the performance of the proposed model is verified on two data sets compared with a variety of algorithms. 16Received signal strength based indoor positioning algorithm using advanced clustering and kernel ridge regression显示文摘We propose a novel indoor positioning algorithm based on the received signal strength(RSS) fingerprint. The proposed algorithm can be divided into three steps, an offline phase at which an advanced clustering(AC) strategy is used, an online phase of approximate localization at which cluster matching is used, and an online phase of precise localization with kernel ridge regression. Specifically, after offline fingerprint collection and similarity measurement, we employ an AC strategy based on the K-medoids clustering algorithm using additional reference points that are geographically located at the outer cluster boundary to enrich the data of each cluster. During the approximate localization, RSS measurements are compared with the cluster radio maps to determine to which cluster the target most likely belongs. Both the Euclidean distance of the RSSs and the Hamming distance of the coverage vectors between the observations and training records are explored for cluster matching. Then, a kernel-based ridge regression method is used to obtain the ultimate positioning of the target. The performance of the proposed algorithm is evaluated in two typical indoor environments, and compared with those of state-of-the-art algorithms. 17Dihydro-stilbene gigantol relieves CCl4-induced hepatic oxidative stress and inflammation in mice via inhibiting C5b-9 formation in the liver显示文摘In general,anti-inflammatory treatment is considered for multiple liver diseases despite the etiology.But current drugs for alleviating liver inflammation have defects,making it necessary to develop more potent and safer drugs for liver injury.in this study,we screened a series of(dihydro-)stillene or(dihydro-)phenanthrene derivatives extracted from Pholidota chinensis for theirpotential biological activities.Among 31 compounds,the dihydro-stlbene gigantol exerted most potent protective effects onhuman hepatocytes against lithocholic acid toxicity,and exhibited solid antioxidative and anti-inflammatory effect in vitro.In micewith CCl-induced acute liver injury,pre-administration of gigantol(10,20,40 mg kg^-1·d^-1,po,for 7 days)dose-dependentydecreased serum transaminase levels and improved pathological changes in liver tissues.The elevated lipid peroxidation andinflammatory responses in the livers were also signifhcantly alleviated by gigantol.The pharmacokinetic studies showed thatgigantol was highly concentrated in the mouse lvers,which consisted with its efficacy in preventing liver injury.Using a label-freequantitative proteomic analysis we revealed that gigantol mainly regulated the immune system process in liver tissues of CCl4-treated mice,and the complement and coagulation cascades was the predominant pathway gigantol markedly inhibited theexpression of complement component C9,which was a key component for the formation of terminal complerment complex(TCC)C5b-9.These results were validated by immunohistochemistry(IHC) or real time-PCR.Confocal microscopy analysis showed thatgigantol significantly inhibited the vascular deposition of TCC in the liver.In conclusion,we demonstrate for the first time that oral administration of gigantol potently relieves liver oxidative stress and inflammation,possibly via a novel mechanism of inhibiting the C5b-9 formation in the liver.Ya-ru Xue Sheng Yao Qian Liu Zhao-liang Peng Qiang-qiang Deng—Bo Liu Zheng-hua Ma Le Wangf Hu Zhou Yang Ye Guo-yu Pan 2020Acta Pharmacologica Sinica2020,41,11:3
18Construction of Rat Model of Hepatic Fibrosis with Blood Stasis Syndrome Integrated with Traditional Chinese Medicine(TCM)Syndrome and Western Medicine Disease显示文摘[Objective]According to the theory of blood stasis in traditional Chinese medicine( TCM),the animal modeling method of hepatic fibrosis integrated with pathology and symptoms was explored and improved,to construct a new type of rat model of hepatic fibrosis with blood stasis syndrome integrated with traditional Chinese and western medicine. [Method]The hepatic fibrosis model of blood stasis with blood stasis syndrome was constructed by jointing multi factors,including intragastric administration of ethanol,high fat and low protein feeding,joint injection of dimethylnitrosamine( DMN),bovine serum albumin( BSA) and norepinephrine( NE). The modeling method was further compared with traditional CC14 single-factor modeling method from the aspects of mortality,blood stasis syndrome of TCM,syndrome grading,general morphology of liver pathology,liver function changes,as well as expression levels of three kinds of collagen( type I and type III collagen,α-SMA) determined by immunohistochemical staining method,and four items of rat hepatic fibrosis( HA,P3 NP,LN,CⅣ content) determined by radio enzyme immunoassay. [Result]In blood stasis group,(1) the mortality of rats was 20%;(2) model rats appeared typical blood stasis syndromes of TCM such as ecchymosis,dark purple tongue,loose stool,and blood stasis syndrome grading was high;(3) fibrosis changes of liver such as dark white surface,dense gray nodules and brittle texture were observed in general morphological examination;(4) serological liver function tests found that ALT and AST of model rats,as well as TBIL,DBIL and IBIL contents increased significantly;(5) immunohistochemical staining demonstrated that the expression levels of three kinds of collagen(type I and type III collagen,α-SMA) increased significantly;(6) four items of rat serum hepatic fibrosis,HA,P3 NP,LN,CⅣ content,increased significantly;(7) the above results in blood stasis syndrome model group(except morality and liver function) were higher than those in CC14 modeling group,and the difference was statistically significant( P < 0. 05). [Conclusion]The new improved modeling method effectively reduces high mortality in traditional CC14 modeling method.In addition to low mortality,the model animal has dual characteristics of disease in western medicine and syndrome in TCM. 20An investigation on application potentiality of microstructure heat sinks with different flow topological morphology显示文摘Microstructure heat sinks have great potential for high heat flux cooling.In this paper,we compared microchannel(MC),micro-pin-fin(MPF),manifold microchannel(MMC),and manifold micro-pin-fin(MMPF)heat sinks to figure out the pros and cons.Fluid flow and manifold unit models are used to study thermal and hydrodynamic performance.The heat sinks with different channel/fin sizes,manifold numbers,and porosities are discussed according to the pressure drop,temperature,thermal resistance,and coefficient of performance.Results show that MMC and MMPF heat sinks are superior to MC and MPF heat sinks,and there are also differences between MMC and MMPF heat sinks.Typically,the MMPF heat sink has lower maximum temperature,temperature non-uniformity,and total thermal resistance R_(tot).In contrast,the MMC heat sink has a lower pressure drop and higher COP.For the MMPF heat sink,at the nozzle width of 6.75μm and the MPF width of 70.71μm(porosity=0.167),it achieves the lowest total thermal resistance of R_(tot)=2.97×10^(-6)K m^(2)/W.Under 10^(3)W/cm^(2)heat flux,the maximum surface temperature rise is 29.74 K,and the maximum temperature difference of the heating surface is 3.15 K.This research initially provides a clear reference on the selection of single-phase cooling microstructures for ultra-high heat flux dissipation.YAO XiaoLe SHI QianLei LIU Qian QIN Le JU Xing XU Chao 2022Science China(Technological Sciences)2022,65,12:2
