1Early Serum HBsAg Kinetics as Predictor of HBsAg Loss in Patients with HBeAg-Negative Chronic Hepatitis B after Treatment with Pegylated Interferonα-2a显示文摘Hepatitis B surface antigen(HBsAg)loss is an ideal treatment endpoint for patients with chronic hepatitis B(CHB).We investigated the predictive value of on-treatment HBsAg levels for HBsAg loss in hepatitis B e antigen(HBe Ag)-negative CHB patients who received 120-week PEG-IFNα-2a treatment.Serum HBV DNA,HBsAg,and anti-HBs levels were assayed at baseline and every 3 months during the treatment.Of 81 patients,12 achieved HBsAg loss,20 achieved HBsAg\100 IU/mL,and 49 maintained HBs Ag C 100 IU/mL.HBsAg loss rate was only 3.7%at 48 weeks,while it reached to 11.1%and 14.8%after treatment of 96 weeks and 120 weeks.The cutoff HBs Ag levels at 12 weeks predicting HBsAg loss at 96 weeks and 120 weeks of treatment were 400 IU/mL and 750 IU/mL,with AUC 0.725 and 0.722,positive predictive value(PPV)29.41%and 30.56%,and negative predictive value(NPV)93.75%and 97.78%,respectively.The cutoff HBsAg levels at 24 weeks predicting HBsAg loss at 96 weeks and 120 weeks of treatment were 174 IU/m L and 236 IU/mL respectively,with AUC 0.925 and 0.922,PPV 40.0%and 46.15%,and both NPV 100%.The predictive ability of the cutoff HBsAg levels at 24 weeks was better than that at 12 weeks for HBs Ag loss at either 96 or 120 weeks(χ~2=3.880,P=0.049 andχ~2=4.412,P=0.036).These results indicate that extended therapy is critical to HBsAg loss in HBe Ag-negative CHB patients during PEG-IFN treatment,and the HBsAg level at 24 weeks can be used to predict HBsAg loss during tailoring PEG-IFN therapy.Minghui Li Lu Zhang Yao Lu Qiqi Chen Huihui Lu Fangfang Sun Zhan Zeng Gang Wan Linqing Zhao Yao Xie 2021Virologica Sinica2021,36,2:15
2Streamflow response to climate variability and human activities in the upper catchment of the Yellow River Basin显示文摘Both sensitivity-based method and simulation method are used to analyze the streamflow response to climate variability and human activities in the upper catchment of the Yellow River Basin (UYRB) in this study. The separation regime of effects from climate variability and human activities is investigated. Results show that the changes of streamflow are more sensitive to precipitation than potential evapotranspiration (PET). Effect of climate variability on streamflow estimated using the sensitivity-based method is weak in the upper catchment of Jimai station, and strong in the upper catchment of Lanzhou station, where the climate effects accounted for about 50% of total streamflow changes. Effects of human activities on streamflow accounted for about 40% in the UYRB, with weaker effects in the upper catchment of Tangnaihai station than those in the upper catchment of Lanzhou station. 3A novel lncRNA, Lnc-OC1, promotes ovarian cancer cell proliferation and migration by sponging miR-34a and miR-34c显示文摘Long non-coding RNAs(lncRNAs) have been reported to be of great importance in tumorigenesis and progression of a variety of cancers. However, the role of lncRNAs in ovarian cancer(OC) remains largely unknown. In the present study, we identified a novel lncRNA, LOC100288181(named as Lnc-OC1), which acted as a key regulator in the development and progression of OC. The combined Gene Expression Omnibus(GEO) database analysis revealed that Lnc-OC1 was significantly upregulated in OC tissues and Kaplan-Meier survival analysis confirmed that high Lnc-OC1 expression was associated with poor prognosis of OC patients. Importantly, we also demonstrated that knockdown of Lnc-OC1 suppressed cell proliferation, colony formation, invasion and migration in vitro and inhibited tumorigenicity in vivo.Mechanistically, Lnc-OC1 repressed the expression of endogenous miR-34 a and miR-34 c as a sponge and vice versa. Moreover, rescue experiments demonstrated that the oncogenic function of Lnc-OC1 at least partially depended on suppressing miR-34 a and miR-34 c. 4Salidroside Protects Against 6-Hydroxydopamine-Induced Cytotoxicity by Attenuating ER Stress显示文摘Parkinson's disease(PD) is a neurodegenerative disease characterized by a persistent decline of dopaminergic(DA) neurons in the substantia nigra pars compacta. Despite its frequency, effective therapeutic strategies that halt the neurodegenerative processes are lacking, reinforcing the need to better understand the molecular drivers of this disease. Importantly, increasing evidence suggests that the endoplasmic reticulum(ER)stress-induced unfolded protein response is likely involved in DA neuronal death. Salidroside, a major compound isolated from Rhodiola rosea L., possesses potent antioxidative stress properties and protects against DA neuronal death. However, the underlying mechanisms are not well understood. In the present study, we demonstrate that salidroside prevents 6-hydroxydopamine(6-OHDA)-induced cytotoxicity by attenuating ER stress. Furthermore, treatment of a DA neuronal cell line(SN4741) and primary cortical neurons with salidroside significantly reduced neurotoxin-induced increases in cytoplasmic reactive oxygen species and calcium, both of which cause ER stress, and cleaved caspase-12, which is responsible for ER stress-induced cell death. Together, these results suggest that salidroside protects SN4741 cells and primary cortical neurons from 6-OHDA-induced neurotoxicity by attenuating ER stress. 6Synthesis and optical characterization of Eu^(2+),Tb^(3+)-codoped Sr_3Y(PO_4)_3 green phosphors显示文摘A series of Eu^(2+),Tb^(3+)-codoped Sr_3 Y(PO_4)_3(SYP) green phosphors were synthesized by hightemperature solid-state reaction. Several techniques, such as X-ray diffraction, UV-vis spectrum,and photoluminescence spectrum, were used to investigate the obtained phosphors. The present study investigates in detail photoluminescence excitation and emission properties, energy transfer between the two dopants, and effects of doping ions on optical band gap. SYP:0.05 Eu2+ phosphor shows an intense and broad excitation band ranging from 220 to 400 nm and exhibits a bright green emission band with CIE chromaticity coordinates(0.189, 0.359) under 350 nm excitation. Green emission of SYP:0.03 Tb3+ is intensified by codoping with Eu^(2+), and energy transfer mechanism between them is demonstrated to be a dipole-dipole interaction. Upon 350 nm excitation, SYP:Eu^(2+),Tb^(3+) phosphors exhibits two dominating bands peaking at 466 and 545 nm, which are assigned to 4 f^65 d^1→4 f^7 transition of Eu^(2+) ions and ~5 D_4→~7 F_5 transition of Tb^(3+) ions, respectively. Optimal doping concentrations of Eu^(2+) and Tb^(3+) in the SYP host are 5 mol% and 15 mol%, respectively. 8Genotypic variation in spatiotemporal distribution of canopy light interception in relation to yield formation in cotton显示文摘Background: Within-canopy interception of photosynthetically active radiation(PAR) impacts yield and other agronomic traits in cotton(Gossypium hirsutum L.). Field experiments were conducted to investigate the influence of 6 cotton varieties(they belong to 3 different plant types) on yield, yield distribution, light interception(LI), LI distribution and the relationship between yield formation and LI in Anyang, Henan, in 2014 and 2015.Result: The results showed that cotton cultivars with long branches(loose-type) intercepted more LI than did cultivars with short branches(compact-type), due to increased LI in the middle and upper canopy. Although loose-type varieties had greater LI, they did not yield significantly higher than compact-type varieties, due to decreased harvest index. Therefore, improving the harvest index by adjusting the source-to-sink relationship may further increase cotton yield for loose-type cotton. In addition, there was a positive relationship between reproductive organ biomass accumulation and canopy-accumulated LI, indicating that enhancing LI is important for yield improvement for each cultivar. 14Several miRNAs derived from serum extracellular vesicles are potential biomarkers for early diagnosis and progression of Parkinson’s disease显示文摘Background:Blood-based test for predicting disease progression and early diagnosis of Parkinson’s disease(PD)is an unmet need in the clinic.The profiles of microRNAs(miRNAs)are regarded as potential diagnostic biomarkers for human diseases,whereas miRNAs in the periphery are susceptible to the influence of various components.MiRNAs enriched in serum extracellular vesicles(EVs)have demonstrated disease-specific advantages in diagnosis due to their high abundance,stability and resistance to degradation.This study was aimed to screen differentially expressed EV-derived miRNAs between healthy controls and PD patients to aid in diagnosis of PD.Methods:A total of 31 healthy controls and 72 patients with a diagnosis of PD at different Hoehn and Yahr stages in Tangdu Hospital were included.In total,185 differentially expressed miRNAs were obtained through RNA sequencing of serum EVs as well as edgeR and t-test analyses.Subsequently,the weighted gene co-expression network analysis(WGCNA)was utilized to identify the commonly expressed miRNAs in all stages of PD by constructing connections between modules,and specifically expressed miRNAs in each stage of PD by functional enrichment analysis.After aligning these miRNAs with PD-related miRNAs in Human miRNA Disease Database,the screened miRNAs were further validated by receiver operating characteristic(ROC)curves and quantitative real-time polymerase chain reaction(qRT-PCR)using peripheral blood EVs from 40 more participants.Results:WGCNA showed that 4 miRNAs were commonly associated with all stages of PD and 13 miRNAs were specifically associated with different stages of PD.Of the 17 obtained miRNAs,7 were validated by ROC curve analysis and 7 were verified in 40 more participants by qRT-PCR.Six miRNAs were verified by both methods,which included 2 miRNAs that were commonly expressed in all stages of PD and 4 miRNAs that were specifically expressed in different stages of PD.Conclusions:The 6 serum EV-derived miRNAs,hsa-miR-374a-5p,hsa-miR-374b-5p,hsa-miR-199a-3p,hsa-miR-28-5p,hsa-miR-22-5p and hsa-miR-151a-5p,may potentially be used as biomarkers for PD progression and for early diagnosis of PD in populations.Shulei He Lu Huang Ci Shao Tiejian Nie Li Xia Bozhou Cui Fangfang Lu Lin Zhu Bolin Chen Qian Yang 2021Translational Neurodegeneration2021,10,3:1

18Effect of Matrix Modification on Durability of Cementitious Composites in an Acid Rain Environment显示文摘The durability of silane-modifi ed mortar, a cementitious composite, in acid rain environment was investigated given its extensive usage as a structural material. The results indicated that the addition of silane decreased the compressive strength of the cementitious composite. Wetting angle was increased by incorporating silane into the matrix. Decrease in both water absorption ability and coefficient of capillary suction confi rmed hydrophobicity as induced by silane addition. Results of mechanical testing, scanning electron microscopy and X-ray diffraction showed that the sulfuric acid resistance of mortar was enhanced by silane. 19Assessment of respiratory support decision and the outcome of invasive mechanical ventilation in severe COVID-19 with ARDS显示文摘Background:The coronavirus disease 2019(COVID-19)is an ongoing pandemic.Invasive mechanical ventilation(IMV)is essential for the management of COVID-19 with acute respiratory distress syndrome(ARDS).We aimed to assess the impact of compliance with a respiratory decision support system on the outcomes of patients with COVID-19-associated ARDS who required IMV.Methods:In this retrospective,single-center,case series study,patients with COVID-19-associated ARDS who required IMV at Zhongnan Hospital of Wuhan University,China,from January 8th,2020,to March 24th,2020,with the final follow-up date of April 20th,2020,were included.Demographic,clinical,laboratory,imaging,and management information were collected and analyzed.Compliance with the respiratory support decision system was documented,and its relationship with 28-day mortality was evaluated.Results:The study included 46 COVID-19-associated ARDS patients who required IMV.The median age of the 46 patients was 68.5 years,and 31 were men.The partial pressure of arterial oxygen(PaO_(2))/fraction of inspired oxygen(FiO_(2))ratio at intensive care unit(ICU)admission was 104 mmHg.The median total length of IMV was 12.0(interquartile range[IQR]:6.0–27.3)days,and the median respiratory support decision score was 11.0(IQR:7.8–16.0).To 28 days after ICU admission,18(39.1%)patients died.Survivors had a significantly higher respiratory support decision score than non-survivors(15.0[10.3–17.0]vs.8.5(6.0–10.3),P=0.001).Using receiver operating characteristic(ROC)curve to assess the discrimination of respiratory support decision score to 28-day mortality,the area under the curve(AUC)was 0.796(95%confidence interval[CI]:0.657–0.934,P=0.001)and the cut-offwas 11.5(sensitivity=0.679,specificity=0.889).Patients with a higher score(>11.5)were more likely to survive at 28 days after ICU admission(log-rank test,P<0.001).Conclusions:For severe COVID-19-associated ARDS with IMV,following the respiratory support decision and assessing completion would improve the progress of ventilation.With a decision score of>11.5,the mortality at 28 days after ICU admission showed an obvious decrease.Shuhan Cai Fangfang Zhu Hongtao Hu Hui Xiang Dawei Wang Jing Wang Lu Li Xiao Yang Aihua Qin Xin Rao Yun Luo Jianguo Li Kianoush B.Kashani Bo Hu Zhiyong Peng 2022Journal of Intensive Medicine2022,2,2:1
20Seven new 3,4-dihydro-furanocoumarin derivatives from Angelica dahurica显示文摘Objective:To study the chemical constituents of the roots of Angelica dahurica,a well-known Chinese herbal medicine named Baizhi in Chinese.Methods:Compounds were separated by various chromatographies,and the structures of new compounds were elucidated based on the analysis of their spectroscopic and spectrometric data(1D,2D NMR,HRESI MS,IR,and UV).The absolute configurations of new compounds were determined by the calculated electronic circular dichroism and chemical derivatization.The inhibitory activities of all isolates against nitric oxide(NO)production were evaluated using lipopolysaccharide-activated RAW 264.7macrophage cells.Results:Seven new 3,4-dihydro-furanocoumarin derivatives(1a/1b,2a/2b,3a/3b,4)together with a known furanocoumarin(5)were isolated from the roots of A.dahurica.The new compounds included three pairs of enantiomers,(4S,2’’R)-angelicadin A(1a)/(4R,2’’S)-angelicadin A(1b),(4S,2’’S)-angelicadin A(2a)/(4R,2’’R)-angelicadin A(2b),and(4S,2’’S)-secoangelicadin A(3a)/(4R,2’’R)-secoangelicadin A(3b),together with(4R,2’’R)-secoangelicadin A methyl ester(4).The known xanthotoxol(5)inhibited the NO production with the half-maximal inhibitory concentration(IC50)value of(32.8±0.8)μmol/L,but all the new compounds showed no inhibitory activities at the concentration of100μmol/L.Conclusion:This is the first report of the discovery of 3,4-dihydro-furanocoumarins from A.dahurica.The results are not only meaningful for the understanding of the chemical constituents of A.dahurica,but also enrich the reservoir of natural products.Yang Wang Fanyu Shi Zihan Lu Mingliang Zhang Zekun Zhang Fangfang Jia Beibei Zhang Lishan Ouyang Zhixiang Zhu Shepo Shi 2023Chinese Herbal Medicines2023,15,3:1
