| 2 | Urimem,a membrane that can store urinary proteins simply and economically,makes the large-scale storage of clinical samples possible显示文摘By nature,biomarker is the measurable change associated with a physiological or pathophysiological process.Unlike blood which has mechanisms to minimize changes and to keep the internal environment homeostatic,urine is more likely to reflect changes of the body and is a better biomarker source.Because of its potential in biomarker discovery,urinary proteins should be preserved comprehensively as the duration of the patients’corresponding medical records.Here,we propose a method to adsorb urinary proteins onto a membrane we named Urimem.This simple and inexpensive method requires minimal sample handling,uses no organic solvents,and is environmentally friendly.Urine samples were filtered through the membrane,and urinary proteins were adsorbed onto the membrane.The proteins on the membrane were dried and stored in a vacuum bag,which keeps the protein pattern faithfully preserved.The membrane may even permit storage at room temperature for weeks.Using this simple and inexpensive method,it is possible to begin preserving urine samples from all consenting people.Thus,medical research especially biomarker research can be conducted more economically.Even more objective large-scale prospective studies will be possible.This method has the potential to change the landscape of medical research and medical practice. | JIA LuLu LIU XueJiao LIU Liu LI MingXi GAO YouHe | 2014 | Science China(Life Sciences)2014,57,3: | 19 |

| 4 | Specific Recognition of Breast Cancer Cells In Vitro Using Near Infrared-Emitting Long-Persistence Luminescent Zn_3Ga_2Ge_2O_(10):Cr^(3+)Nanoprobes显示文摘In this paper, near-infrared emitting long-persistence luminescent Zn3Ga2Ge2O10:Cr3?(ZGG) nanoparticles with diameters of 30–100 nm and bright luminescence were prepared by a sol–gel synthesis method. After the surface amination, the nanoparticles were further bioconjugated with breast cancer-specific monoclonal antibody(anti-Ep CAM) to form ZGG-Ep CAM nanoprobes which can specifically target breast cancer cell lines(MCF7) in vitro. The results of in vitro images show that the luminescence signals from the cells treated with ZGG-Ep CAM nanoprobes are stronger than those from cells treated with ZGG-unconjugated antibody, indicating that the prepared ZGG-Ep CAM nanoprobes possessed excellent specific recognition capability. Furthermore, due to their long afterglow properties, the imaging could persist more than 1 h. Therefore, these nanoprobes could not only provide a high specificity detection method for cancer cells but also realize the long-time monitoring. | 6 | Securing Multicast Route Discovery for Mobile Ad Hoc Networks显示文摘This paper presents SMRAN, a novel securing multicast route discovery scheme for mobile ad hoc networks. The scheme relies entirely on hash chains based one-time signature mechanism, HORSEI, with very efficient signing and verifying, and we do this by improving the HORSE protocol through the introduction of interme- diate hash joints. The main purpose of SMRAN is to provide source authentication for multicast routing discovery messages in mobile ad hoc networks. SMRAN will construct multicast tree with authentica- tion constrains in ad hoc networks. The performance measure of SMRAN is evaluated using simulator NS2. | 8 | Sol-gel preparation and phosphorescence property of Mn^(2+)-doped zinc boro-silicate glass thin films显示文摘Mn2+-doped zinc borosilicate (ZBSM) glass thin films were first synthesized by sol-gel method. In the experiment, a thin gel film was depos-ited onto quartz glass substrates by dip-coating method and then heat-treated to form a Mn2+-doped zinc borosilicate glass thin film. Long lasting phosphorescence (LLP) and photo-stimulated long lasting phosphorescence (PSLLP) were found in the film sample. According to fluorescence spectra, LLP emission spectra, and PSLLP emission spectra, both LLP and PSLLP emissions are attributed to the energy level transition of 4Eg→4A1g from Mn2+. Both the phosphorescence intensity decay curves contain a fast decay component and another slow decay one. The thermoluminescence (TL) spectra show that the sample has two kinds of traps at least and their energy level values are about 0.8 eV and 1.02 eV, which could be estimated by the Randall and Willcins formula. The infrared absorption spectra (IR) consist of characteristic vi-bration bands of Si-O-Si, Si-O-Zn, B-O in [BO3], B-O group, and Zn-O in [ZnO4]. Such materials elegantly combine the advantages of chiral surfaces and traditional polymers, and provide a novel solution not only for the investigation of chiral interaction mechanisms but also for the design of biomaterials with diverse applications, such as in tissue engineering and biocompatible materials, bioregulation, chiral separation and chiral sensors. | 10 | Genetically modified CD7-targeting allogeneic CAR-T cell therapy with enhanced efficacy for relapsed/refractory CD7-positive hematological malignancies:a phase I clinical study显示文摘Chimeric antigen receptor(CAR)-T cell therapy against T cell malignancies faces major challenges including fratricide between CAR-T cells and product contamination from the blasts.Allogeneic CAR-T cells,generated from healthy donor T cells,can provide ready-to-use,blast-free therapeutic products,but their application could be complicated by graft-versus-host disease(GvHD)and host rejection.Here we developed healthy donor-derived,CD7-targeting CAR-T cells(RD13-01)with genetic modifications to resist fratricide,GvHD and allogeneic rejection,as well as to potentiate antitumor function.A phase I clinical trial(NCT04538599)was conducted with twelve patients recruited(eleven with T cell leukemia/lymphoma,and one with CD7-expressing acute myeloid leukemia).All patients achieved pre-set end points and eleven proceeded to efficacy evaluation.No dose-limiting toxicity,GvHD,immune effector cell-associated neurotoxicity or severe cytokine release syndrome(grade≥3)were observed.28 days post infusion,81.8%of patients(9/11)showed objective responses and the complete response rate was 63.6%(7/11,including the patient with AML).3 of the responding patients were bridged to allogeneic hematopoietic stem cell transplantation.With a median follow-up of 10.5 months,4 patients remained in complete remission.Cytomegalovirus(CMV)and/or Epstein-Barr virus(EBV)reactivation was observed in several patients,and one died from EBV-associated diffuse large B-cell lymphoma(DLBCL).Expansion of CD7-negative normal T cells was detected post infusion.In summary,we present the first report of a Phase I clinical trial using healthy donor-derived CD7-targeting allogeneic CAR-T cells to treat CD7^(+)hematological malignancies.Our results demonstrated the encouraging safety and efficacy profiles of the RD13-01 allogeneic CAR-T cells for CD7^(+)tumors. | Yongxian Hu Yali Zhou Mingming Zhang Houli Zhao Guoqing Wei Wengang Ge Qu Cui Qitian Mu Gong Chen Lu Han Tingting Guo Jiazhen Cui Xiaoyan Jiang Xiujun Zheng Shuhui Yu Xiaolong Li Xingwang Zhang Mingxi Chen Xiuju Li Ming Gao Kang Wang Cheng Zu Hao Zhang Xiaohong He Yanbin Wang Dongrui Wang Jiangtao Ren He Huang | 2022 | Cell Research2022,32,11: | 2 |

| 13 | Capsid destabilization and epitope alterations of human papillomavirus 18 in the presence of thimerosal显示文摘Thimerosal has been widely used as a preservative in drug and vaccine products for decades.Due to the strong propensity to modify thiols in proteins,conformational changes could occur due to covalent bond formation between ethylmercury(a degradant of thimerosal)and thiols.Such a conformational change could lead to partial or even complete loss of desirable protein function.This study aims to investigate the effects of thimerosal on the capsid stability and antigenicity of recombinant human papillomavirus(HPV)18 virus-like particles(VLPs).Dramatic destabilization of the recombinant viral capsid upon thimerosal treatment was observed.Such a negative effect on the thermal stability of VLPs preserved with thimerosal was shown to be dependent on the thimerosal concentration.Two highly neutralizing antibodies,13H12 and 3C3,were found to be the most sensitive to thimerosal treatment.The kinetics of antigenicity loss,when monitored with 13H12 or 3C3 as probes,yielded two distinctly different sets of kinetic parameters,while the data from both monoclonal antibodies(mAbs)followed a biphasic exponential decay model.The potential effect of thimerosal on protein function,particularly for thiolcontaining proteinaceous active components,needs to be comprehensively characterized during formulation development when a preservative is necessary. | Xiaofen Huang Yike Li Meifeng Nie Mingxi Yue Yufang Li Zhijie Lin Huirong Pan Mujin Fang Ting Wu Shaowei Li Jun Zhang Ningshao Xia Qinjian Zhao | 2021 | Journal of Pharmaceutical Analysis2021,11,5: | 1 |
