2Correlation between lower gastrointestinal tract symptoms and quality of life in patients with stable chronic obstructive pulmonary disease显示文摘OBJECTIVE:To explore correlations between the symptoms of constipation and abdominal distention and severity of chronic obstructive pulmonary disease(COPD)in patients with stable disease.METHODS:We studied 191 patients with stable COPD(according to defined criteria)in this cross-sectional study from four three-level class A Chinese medicine hospitals in China.We built an Epidata 3.0 database and performed statistical analysis with SPSS,version 17.0.We analyzed correlations between the frequency of lower gastrointestinal tract symptoms(constipation and abdominal distention)and scores for major pulmonary symptoms(cough,sputum and wheezing)based on the St.George's Respiratory Questionnaire(SGRQ),6-minute walking distance(6MWD)and frequency of acute exacerbations of COPD(AECOPD).RESULTS:In addition to their pulmonary symptoms,39.79%and 40.31%of study patients withstable COPD reported constipation and abdominal distention,respectively.Scores for major pulmonary symptoms(cough,sputum and wheezing),AECOPD and SGRQ values in patients with constipation and abdominal distention were significantly greater,and the 6MWD markedly shorter,than in those without them.According to Pearson's correlation analysis,there were strong correlations between these lower gastrointestinal tract symptoms and scores for pulmonary symptoms,SGRQ,6MWD and AECOPD.CONCLUSION:Lower gastrointestinal tract symptoms such as constipation and abdominal distention can adversely affect pulmonary symptoms,frequency of acute exacerbations and quality of life in patients with stable COPD.Yan Sun Fengjie Zheng Yuhang Li Ruohan Wu Yuchao Liu Miao Liu Jinchao Zhang Kuo Gao 2013Journal of Traditional Chinese Medicine2013,33,5:10
3Protective effect of Salvia miltiorrhiza and Carthamus tinctorius extract against lipopolysaccharide-induced liver injury显示文摘AIM:To investigate the hepatoprotective effects and mechanisms of an extract of Salvia miltiorrhiza and Carthamus tinctorius in vivo.METHODS:C57BL/6J mice were randomly assigned to five groups and intraperitoneally administered 0.9% saline,Salvia miltiorrhiza and Carthamus tinctorius extract [Danhong injection(DHI),0.75 and 3 g/kg mixed extract] or reduced glutathione for injection(RGI,300 mg/kg) for 30 min before exposure to lipopolysaccharide(LPS,16 mg/kg). After intraperitoneal LPS stimulation for 90 min or 6 h,the mice were sacrificed by ether anaesthesia,and serum and liver samples were collected. Histological analysis(H&E) and terminal deoxynucleotidyl transferase-mediated d UTP nick end-labelling(TUNEL) staining were performed. Alanine transferase(ALT),aspartate transaminase(AST),total bilirubin(TBil),glutathione-S-transferase(GST),malondialdehyde(MDA),tumour necrosis factor(TNF)-α,interleukin(IL)-6,and caspase-3 levels were measured. Bax,Bcl-2,P-IκBα,IκBα,P-NF-κB p65,and NF-κB p65 protein levels were determined by Western blot. TNF-α,IL-6,caspase-3,Bax and Bcl-2 m RNA expression was measured by real-time reverse transcription-polymerase chain reaction(RT-PCR).RESULTS:Hematoxylin-eosin staining and TUNEL results suggested that DHI(3 g/kg) treatment alleviated inflammatory and apoptotic(P < 0.01) injury in the liver of mice. DHI treatment dose-dependently blunted the abnormal changes in biochemical parameters such as ALT(72.53 ± 2.83 for 3 g/kg,P < 0.01),AST(76.97 ± 5.00 for 3 g/kg,P < 0.01),TBil(1.17 ± 0.10 for 3 g/kg,P < 0.01),MDA(0.81 ± 0.36 for 3 g/kg,P < 0.01),and GST(358.86 ± 12.09 for 3 g/kg,P < 0.01). Moreover,DHI(3 g/kg) remarkably decreased LPS-induced protein expression of TNF-α(340.55 ± 10.18 for 3 g/kg,P < 0.01),IL-6(261.34 ± 10.18 for 3 g/kg,P < 0.01),and enzyme activity of caspase-3(0.93 ± 0.029 for 3 g/kg,P < 0.01). The LPS-induced m RNA expression of TNF-α,IL-6 and caspase-3 was also decreased by DHI. Western blot analysis revealed that DHI antagonised LPS-stimulated decrease of Bcl-2 and increase of Bax protein expression. 6Genetic testing and PGD for unexplained recurrent fetal malformations with MAGEL2 gene mutation显示文摘Birth defects are caused by multiple factors,such as chromosome abnormality,environmental factors,and maternal factors.In this study,we focused on exploring the genetic causes of a non-consanguineous couple who suffered from four times of unsuccessful pregnancy due to unexplained recurrent fetal malformations with similar symptoms and normal chromosome copy number variations.Using trio-whole exome sequencing(trio-WES) for this couple and one of the affected fetuses,we found a mutation,c.1996 delC on the maternal imprinted gene MAGEL2 that was carried by the affected fetus and husband,leading to Schaaf-Yang syndrome.To screen this mutation,we further performed preimplantation genetic diagnosis(PGD) strategy followed by a gene pedigree validation and pathogenicity analysis.After the transfer of a PGD-screened embryo,a normal newborn without previous abnormal symptoms was born(February 15,2019).We present the first data that identified a pathogenic gene(MAGEL2 c.1996 delC) in a fetus with Schaaf-Yang syndrome in the EAS(East Asian) database and overcame this genetic defect by using processed PGD for this couple based on the WES results.Wei Guo Yanli Nie Zhiqiang Yan Xiaohui Zhu Yuqian Wang Shuo Guan Ying Kuo Wenxin Zhang Xu Zhi Yuan Wei Liying Yan Jie Qiao 2019Science China(Life Sciences)2019,62,7:4

8Phase transition and heterogeneous strengthening mechanism in CoCrFeNiMn high-entropy alloy fabricated by laser-engineered net shaping via annealing at intermediate-temperature显示文摘High-entropy alloys(HEAs)have attracted tremendous attention owing to their controllable mechanical properties,whereas additive manufacturing(AM)is an efficient and flexible processing route for novel materials design.However,a profound appraisal of the fundamental material physics behind the strengthening of AM-printed HEAs upon low/intermediate-temperature annealing is essential.In this work,Co CrFe Ni Mn HEAs have been prepared using laser-engineered net shaping(LENS)and subsequently annealed at different temperatures.The Co Cr Fe Ni Mn HEA annealed at intermediate-temperature(873 K)exhibits a strong strain hardening capability,resulting in ultimate strength of 725 MPa and plasticity of 22%.A ternary heterogeneous strengthening mechanism is proposed to explain this phenomenon,in which equiaxed grains,columnar grains,andσprecipitates play different roles during tensile deformation.The resultant excellent strength and ductility can be ascribed to the heterostructure-induced mismatch.The equiaxed grains provide adequate grain boundaries(GBs),which induce dislocation plugging-up and entanglement;the columnar grains induce the onset and arrest of the dislocations for plastic deformation;and theσprecipitates hinder the movement of slip dislocations.The results provide new insights into overcoming the strength-ductility trade-off of LENS-printed HEAs with complex geometries.Yunjian Bai Heng Jiang Kuo Yan Maohui Li Yanpeng Wei Kun Zhang Bingchen Wei 2021Journal of Materials Science & Technology2021,,33:3
9Initiation of Acupoint Molecular Mechanisms for Manual Acupuncture Analgesia—Nuclear FactorκB Signaling Pathway显示文摘Objective To identify the prominent molecular signaling in acupoints and explore their roles in initiating the analgesia effect of manual acupuncture(MA).Method A three-step study was conducted,the experiment 1 was a genome-wide analysis of the tissue at acupoint Zusanli(ST 36),including 12 Wistar rats which were divided into control,control+MA1,and control+MA7 groups.In the experiment 2,the paw withdrawal latency(PWL),immunohistochemistry and Western blot analysis of phospho-nuclear factor kappa B(NFκB)p65(p-p65),phospho-NFκB p50(p-p50)at ST 36 were performed on rats of saline,saline+MA,and complete Freund’s adjuvant(CFA)+MA groups(n=6).In experiment 3,24 rats were divided into saline+DMSO,CFA+DMSO,CFA+DMSO+MA,and CFA+BAY 11–7082+MA groups,the PWL and immunofluorescence assay of NFκB p65 at ST 36 was conducted.Result(1)The gene:inhibitor of NFκB(Nfkbia),interleukin-1β(Il1b),interleukin-6(Il6),chemokine c-x-c motif ligand 1(Cxcl1),monocyte chemoattractant protein-1(MCP-1/Ccl2)expressions in the control+MA7 group were significantly increased(P<0.05 or P<0.01),and the expression of NFκB p65(Rela),NFκB p50(Nfkb1)were increased in the control+MA7 group(P<0.05).(2)CFA+MA groups showed increased PWL from day 1 to 7(P<0.01 vs.CFA),and the Western blot results were consistent with immunohistochemistry,the expression of NFκB p-p65 and NFκB p-p50 were significantly increased in the MA-related groups compared with control and CFA groups(P<0.05).(3)Compared with the CFA+DMSO+MA group,the PWL of the CFA+BAY 11–7082+MA group decreased significantly and continued until day 5 and 7(P<0.05 and P<0.01,respectively),and the NFκB p65 expression of CFA+BAY 11–7082+MA was significantly reduced compared with CFA+DMSO+MA(P<0.01).Conclusion Local NFκB signaling cascade in acupoint caused by MA is an important step in initiating the analgesic effect,which would provide new evidence for the initiation of MA-effect and improve the understanding of the scientific basis of acupuncture analgesia.ZHANG Kuo ZHAO Xue YANG Tao ZHANG Yan-fang YAN Ya-wen XU Zhi-fang GONG Yi-nan LIU Yang-yang GUO Yong-ming GUO Yi 2021Chinese Journal of Integrative Medicine2021,27,12:3

11Overexpression of miR-101 suppresses collagen synthesis by targeting EZH2 in hypertrophic scar fibroblasts显示文摘Background:MicroRNA-101(miR-101)is a tumor suppressor microRNA(miRNA)and its loss is associated with the occurrence and progression of various diseases.However,the biological function and target of miR-101 in the pathogenesis of hypertrophic scars(HS)remains unknown.Methods:We harvested HS and paired normal skin(NS)tissue samples from patients and cultured their fibroblasts(HSF and NSF,respectively).We used quantitative reverse transcriptase polymerase chain reaction(qRT-PCR),fluorescence in situ hybridization(FISH),enzyme-linked immunosorbent assays(ELISA)and Western blot analyses to measure mRNA levels and protein expression of miR-101,enhancer of zeste homolog 2(EZH2),collagen 1 and 3(Col1 and Col3)andα-smooth muscle actin(α-SMA)in different in vitro conditions.We also used RNA sequencing to evaluate the relevant signaling pathways and bioinformatics analysis and dual-luciferase reporter assays to predict miR-101 targets.We utilized a bleomycin-induced fibrosis mouse model in which we injected miR-101 mimics to evaluate collagen deposition in vivo.Results:We found low expression of miR-101 in HS and HSF compared to NS and NSF.Overexpressing miR-101 decreased Col1,Col3 andα-SMA expression in HSF.We detected high expression of EZH2 in HS and HSF.Knockdown of EZH2 decreased Col1,Col3 andα-SMA in HSF.Mechanistically,miR-101 targeted the 3-untranslated region(3UTR)of EZH2,as indicated by the decreased expression of EZH2.Overexpressing EZH2 rescued miR-101-induced collagen repression.MiR-101 mimics effectively suppressed collagen deposition in the bleomycin-induced fibrosis mouse model.Conclusions:Our data reveal that miR-101 targets EZH2 in HS collagen production,providing new insight into the pathological mechanisms underlying HS formation.Jie Li Yan Li Yunchuan Wang Xiang He Jing Wang Weixia Cai Yanhui Jia Dan Xiao Jian Zhang Ming Zhao Kuo Shen Zichao Li Wenbin Jia Kejia Wang Yue Zhang Linlin Su Huayu Zhu Dahai Hu 2021Burns & Trauma2021,9,1:3
12Sparing lung tissue with virtual block method in VMAT planning for locally advanced non-small cell lung cancer显示文摘This study aimed to exploit a new virtual block method to spare normal lung tissue in VMAT planning for patients with locally advanced non-small cell lung cancer(LA-NSCLC).The previous method was used to manually restrict the angle of the beam passing through,which ignored the location and shape of large targets that varied between different slices and did not block the beamlets precisely.Unlike the previous method,this new virtual block method was used to block the beamlets when necessary by closing the multi-leaf collimator(MLC)at prerequisite angles.The algorithm for closing the MLC depended on the thickness of the beamlets passing through the lungs and avoided only the entrance radiation beamlet.Moreover,this block can be automatically contoured.A retrospective study was performed to compare the VMAT plans with and without the virtual block method for 17 LANSCLC patients,named the block plan(B-plan)/non-block plan(N-plan).All cases were selected in this study because of the large tumor size and unmet dose constraints of the lungs.In addition to the maximum dose constraint for the virtual block,B-plans adopted identical optimization parameters to N-plans for each patient.These two types of plans were compared in terms of dosimetric indices and plan scores.The results were statistically analyzed using the Wilcoxon nonparametric signed-rank test.B-plans have advantages in the following dosimetric metrics that have statistical significance(p<0.05):(1)lower V_(5)/V_(10)/D_(mean)/normal tissue complication probability(NTCP)of total lungs;(2)reductions in V_(5)/V_(10)for the contralateral lung;(3)decrease in Dmean/V_(40)of the heart;(4)decrease in esophagus V_(40);(5)reductions in Dmean,V_(5)/V_(10) of normal tissue.B-plans(82.51±7.07)achieved higher-quality scores than N-plans(80.74±7.22).The new virtual block spared the lungs as well as other normal structures in VMAT planning for LA-NSCLC.Thus,the block method may decrease the risk of radiation-related toxicity in patients.Jia-Yun Chen Da-Quan Wang Xiao-Dong Zhang Qi Fu Xue-Na Yan Kuo Men Jian-Rong Dai Nan Bi 2022Nuclear Science and Techniques2022,33,4:3
13Methylation of IRAK3 is a novel prognostic marker in hepatocellular carcinoma显示文摘AIM: To examine the methylation levels of interleukin-1 receptor-associated kinase 3(IRAK3) and GLOXD1 and their potential clinical applications in hepatocellular carcinoma(HCC).METHODS: m RNA expression and promoter methylation of IRAK3 and GLOXD1 in HCC cells were analyzed by reverse transcription-polymerase chain reaction(RTPCR) and methylation-specific PCR(MSP), respectively. Using pyrosequencing results, we further established a quantitative MSP(Q-MSP) system for the evaluation of IRAK3 and GLOXD1 methylation in 29 normal controls and 160 paired HCC tissues and their adjacent nontumor tissues. We also calculated Kaplan-Meier survival curves to determine the applications of gene methylation in the prognosis of HCC.RESULTS: IRAK3 and GLOXD1 expression was partially restored in several HCC cell lines after treatment with 5-aza-2′-deoxycytidine(DNA methyltransferase inhibitor; 5DAC). A partial decrease in the methylated band was also observed in the HCC cell lines after 5DAC treatment. Using GLOXD1 as an example, we found a significant correlation between the data obtained from the methylation array and from pyrosequencing. The methylation frequency of IRAK3 and GLOXD1 in HCC tissues was 46.9% and 63.8%, respectively. Methylation of IRAK3 was statistically associated with tumor stage. Moreover, HCC patients with IRAK3 methylation had a trend toward poor 3-year disease-free survival(P < 0.05). CONCLUSION: IRAK3 and GLOXD1 were frequently methylated in HCC tissues compared to normal controls and nontumor tissues. IRAK3 methylation was associated with tumor stage and poor prognosis of patients. 17Primary retroperitoneal mucinous cystadenoma:Report of a case and review of the literature显示文摘Primary retroperitoneal mucinous cystadenomas (RMCs) are very rare and their biological behavior and histogenesis remain speculative. An accurate preoperative diagnosis of these tumors is difficult because no effective diagnostic measures have been established. We describe a 29-year-old woman with abdominal pain and a palpable mass. Computed tomography (CT) of the abdomen revealed a retroperi-toneal cystic mass which was resected successfully at laparotomy. Histopathological examination of the resected mass confirmed the diagnosis of RMC. 18A Novel Design of Thermoelectric Generator for Automotive Waste Heat Recovery显示文摘With progressively stringent fuel consumption regulations,many researchers and engineers are focusing on the employment of waste heat recovery technologies for automotive applications.Regarded as a promising method of waste heat recovery,the thermoelectric generator(TEG)has been given increasing attention over the whole automotive industry for the last decade.In this study,we first give a brief review of improvements in thermoelectric materials and heat exchangers for TEG systems.We then present a novel design for a concentric cylindrical TEG system that addresses the existing weaknesses of the heat exchanger.In place of the typical square-shaped thermoelectric module,our proposed concentric cylindrical TEG system uses an annular thermoelectric module and employs the advantages of the heat pipe to enhance the heat transfer in the radial direction.The simulations we carried out to verify the performance of the proposed system showed better power output among the existing TEG system,and a comparison of water-inside and gas-inside arrangements showed that the water-inside concentric cylindrical TEG system produced a higher power output.Kuo Huang Yuying Yan Bo Li Yong Li Kai Li Jun Li 2018Automotive Innovation2018,1,1:2
