| 1 | Effect of herbal medicine Juzentaihoto on hepatic and intestinal heat shock gene expression requires intestinal microflora in mouse显示文摘AIM: To evaluate the role of intestinal microflora in the effects of multi-herbal medicine on gene expression in the gut and liver. METHODS: The multi-herbal medicine Juzentaihoto (JTX) was administered to five germ-free mice and regular mice for 2 wk. Among the results of the comprehensive gene chip analysis of the intestine and liver, we featured heat shock proteins (HSPs) 70 and 105 because their gene expression changed only in the presence of microflora. Real-time RT-PCR was performed to confirm the expression levels of these HSP genes. To determine whether JTX acts directly on the HSP genes, sodium arsenite (SA) was used to induce the heat shock proteins directly. To examine the change of the intestinal microflora with administration of JTX, the terminal restriction fragment polymorphism (T-RFLP) method was used. To identify the changed bacteria, DNA sequencing was performed. RESULTS: Heat shock protein gene expression,documented by gene chip and real-time RT-PCR, changed with the administration of JTX in the regular mice but not in the germ-free mice. JTX did not suppress the direct induction of the HSPs by SA. T-RFLP suggested that JTX decreased unculturable bacteria and increased Lactobacillus johnsoni. These data suggested that JTX changed the intestinal microflora which, in turn, changed HSP gene expression. | 2 | The expression of Spodoptera exigua P450 and UGT genes: tissue specificity and response to insecticides显示文摘Cytochrome P450 and UDP-glucosyltransferase (UGT) as phase I and phase II metabolism enzymes, respectively, play vital roles in the breakdown of endobiotics and xenobiotics. Insects can in crease the expression of detoxificatio n enzymes to cope with the stress from xenobiotics including insecticides. However, the molecular mechanisms for insecticide detoxification in Spodoptera exigua remain elusive, and the genes conferring insecticide metabolisms in this species are less well reported. In this study, 68 P450 and 32 UGT genes were identified. Phylogenetic analysis showed gene expansions in CYP3 and CYP4 clans of P450 genes and UGT33 family of this pest. P450 and UGT genes exhibited specific tissue expression patterns. Insecticide treatments in fat body cells of S. exigua revealed that the expression levels of P450 and UGT genes were significantly influenced by challenges of abamectin, lambda-cyhalothrin, chlorantraniliprole, metaflumizone and indoxacarb. Multiple genes for detoxification were affected in expression levels after insecticide exposures. The results demonstrated that lambda-cyhalothrin, chlorantraniliprole, metaflumizone and indoxacarb induced similar responses in the expression of P450 and UGT genes in fat body cells;eight P450 genes and four UGT genes were co-up-regulated significantly, and no or only a few CYP/UGT genes were down-regulated significantly by these four insecticides. However, abamectin triggered a distinct response for P450 and UGT gene expression;more P450 and UGT genes were down-regulated by abamectin than by the other four compounds. | 3 | 日本工程试验卫星ETS—Ⅶ无人自动交会对接系统的在轨演示显示文摘空间交会对接(RVD)技术是进行航天器在轨修复、空间站补给和设备更换等未来空间活动的必要技术。日本从事空间交会对接技术的研究,尤其受到在21世纪初期通过发射无人有翼往返运输系统(HOPE)实现为α国际空间站(SS)的日本实验舱(JEM)提供后勤支持服务的任务驱动。HOPE-SS交会对接任务的特点是由无人航天器向载人航天器实现RVD。NASDA筑波空间中心从70年代初期就开始从事RVD系统的研究工作,研究重点集中在开发高度自动和安全的RVD系统。目前,由工程试验卫星-Ⅶ(ETS-Ⅶ)在轨演示无人自动RVD系统的工程正在进展之中,ETS-Ⅶ将在1997年由H-Ⅶ运载火箭发射入轨,此次在轨飞行演示试验是NASDA开发RVD技术的第一步,其主要目的在于在不久将来即21世纪初发展自动RVD技术及空间遥操作机器人技术。本文将介绍ETS-ⅦRVD系统的初步研究成果,包括RVD试验计划、试验系统和运行方案。为实现自动和安全RVD系统,需建立星载飞行管理系统(FMS)。本文将描述制导、导航和控制系统(GNC)设计以及FMS策略。工程样机(EM)的设计工作开始于1994年冬天,并拟于1995年初在RVD系统测试中使用。本文也将报告RVD系统的验证确认计划及其地面测试设备。 | Isao Kawano Masaaki Mokuno Hiroshi Horiguchi Koichi Kibe | 1997 | 控制工程(北京)1997,,2: | 2 |
