3Macroscopic superlubricity of potassium hydroxide solution achieved by incorporating in-situ released graphene from friction pairs显示文摘Graphene(G),as a typical two-dimensional material,is often used as an additive for liquid lubricants.However,graphene is mostly added to liquid lubricants in a one-time manner in friction;it mainly exists in the form of multilayer agglomerated structures due to theπ–πstacking between graphene sheets,making it unable to fully exert the synergistic lubrication function.Herein,we propose a new macroscopic superlubric system of graphene/potassium hydroxide(G/KOH)solution;and the graphene additive involved is exfoliated in-situ from graphene/epoxy(G/EP)friction pair by friction,continuously providing freshly-peeled graphene into KOH solution and minimizing the adverse effects of graphene agglomeration.Moreover,the in-situ produced graphene additive has thinner thickness and better anti-aggregation ability,which provide more graphene to accommodate OH−,form more stacked sandwich structures of OH−/graphene/OH−between friction pairs(i.e.,equivalent to a moving pulley block with more wheels),and finally realize superlubricity.This study develops a new liquid superlubric system suitable for alkaline environments,and at the same time proposes a new way to gradually release graphene additives in situ,rather than adding them all at once,deepening the understanding to liquid superlubricity mechanism,and paving the experimental foundation for the practical application of macroscopic superlubricity.Hongyu LIANG Xinjie CHEN Yongfeng BU Meijuan XU Gang ZHENG Kaixiong GAO Xijun HUA Yonghong FU Junyan ZHANG 2023Friction2023,11,4:0
4Synthesis of MgO-doped ordered mesoporous carbons by Mg^(2+)-tannin coordination for efficient isomerization of glucose to fructose显示文摘In-situ MgO-doped ordered mesoporous carbon(OMC@MgO)was fabricated by formaldehyde-free self-assembly method,in which biomass-derived tannin was used as carbon precursor replacing fossil-based phenolics,Mg^(2+)as both cross-linker and precursor of catalytic sites.Up to~20 wt% MgO could be doped in the carbon skeleton with good dispersion retaining well-ordered mesoporous structures,while more MgO content(35 wt%)led to the failing in the formation of ordered mesoporous structure.The OMC@MgO possessed a high specific surface area(298.8 m^(2) g^(-1)),uniform pore size distribution(4.8 nm)and small crystallite size of MgO(1.73 nm)due to the confinement effect of ordered mesoporous structure.Using OMC@MgO as the heterogeneous catalyst,a maximum fructose yield of 32.4% with a selectivity up to 81.1%was achieved from glucose in water(90℃,60 min),which is much higher than that obtained using the MgO doped active carbon via conventional post-impregnation method(26.5%yield with 58.3% selectivity).Higher reaction temperature(>90℃)resulted in decrease of selectivity due to the formation of humins.The designed OMC@MgO displayed tolerant to high initial glucose concentrations(10 wt%)and could remain good recyclability without significant loss of activity for three cycles.Junyan Fu Feng Shen Xiaoning Liu Xinhua Qi 2023Green Energy & Environment2023,8,3:0
5Genomic polymorphisms at the crhr2 locus improve feed conversion efficiency through alleviation of hypothalamus-pituitary-interrenal axis activity in gibel carp(Carassius gibelio)显示文摘Improvement in fish feed conversion efficiency(FCE)is beneficial for sustaining global food fish supplies.Here,we show that a set of polymorphisms at locus of the corticotropin releasing hormone receptor 2(crhr2),which is involved in hypothalamuspituitary-interrenal(HPI)axis signaling,is associated with improved FCE in farmed allogynogenetic gibel carp strain CAS Ⅲ compared with that in the wild gibel carp strain Dongting(DT).This set of polymorphisms downregulates the expression levels of crhr2 mRNA in the brain and pituitary tissues in gibel carp strain CAS Ⅲ compared with those in strain DT.Furthermore,compromised HPI axis signaling is observed in gibel carp strain CAS Ⅲ,such as decreased α-melanocyte stimulating hormone protein levels,plasma cortisol content,and stress responses.Moreover,enhanced activation of protein kinase B/mammalian target of rapamycin complex 1 signaling observed in the muscle tissue of strain CAS Ⅲ in comparison to that in strain DT indicated elevated anabolic metabolism in strain CAS Ⅲ.Thus,these studies demonstrate that the genetic markers associated with compromised HPI axis signaling,such as crhr2,are potentially useful for genetic selection toward improvement in farmed fish growth and FCE,which would reduce fishmeal consumption and thereby indirectly facilitate sustainable fisheries.Chuang Shi Qiyong Lou Beide Fu Junyan Jin Jianfei Huang Yao Lu Xia Jin Jiangyan He Gang Zhai Shouqi Xie Jingou Tong Jianfang Gui Zhan Yin 2022Science China(Life Sciences)2022,65,1:0
6Dock4 is required for the maintenance of cochlear hair cells and hearing function显示文摘Auditory hair cells(HCs)are the mechanosensory receptors of the cochlea,and HC loss or malfunction can result from genetic defects.Dock4,a member of the Dock180-related protein superfamily,is a guanine nucleotide exchange factor for Rac1,and previous reports have shown that Dock4 mutations are associated with autism spectrum disorder,myelodysplastic syndromes,and tumorigenesis.Here,we found that Dock4 is highly expressed in the cochlear HCs of mice.However,the role of Dock4 in the inner ear has not yet been investigated.Taking advantage of the piggyBac transposon system,Dock4 knockdown(KD)mice were established to explore the role of Dock4 in the cochlea.Compared to wild-type controls,Dock4 KD mice showed significant hearing impairment from postnatal day 60.Dock4 KD mice showed hair bundle deficits and increased oxidative stress,which eventually led to HC apoptosis,late-onset HC loss,and progressive hearing loss.Furthermore,molecular mechanism studies showed that Rac1/β-catenin signaling was significantly downregulated in Dock4 KD cochleae and that this was the cause for the disorganized stereocilia and increased oxidative stress in HCs.Overall,our work demonstrates that the Dock4/Rac1/β-catenin signaling pathway plays a critical role in the maintenance of auditory HCs and hearing function.Guodong Hong Xiaolong Fu Jieyu Qi Buwei Shao Xuan Han Yuan Fang Shuang Liu Cheng Cheng Chengwen Zhu Junyan Gao Xia Gao Jie Chen Ming Xia Wei Xiong Renjie Chai 2023Fundamental Research2023,3,4:0
7Dietary ribose supplementation improves flesh quality through purine metabolism in gibel carp(Carassius auratus gibelio)显示文摘Since the aquaculture industry is currently observing a deterioration in the flesh quality of farmed fish,the use of nutrients as additives to improve the flesh quality of farmed fish species is a viable strategy.The aim of this study was to investigate the effect of dietary D-ribose(RI)on the nutritional value,texture and flavour of gibel carp(Carassius auratus gibelio).Four diets were formulated containing exogenous RI at 4 gradient levels:0(Control),0.15%(0.15RI),0.30%(0.30RI)and 0.45%(0.45RI).A total of 240 fish(150±0.31 g)were randomly distributed into 12 fibreglass tanks(150 L per tank).Triplicate tanks were randomly assigned to each diet.The feeding trial was carried out in an indoor recirculating aquaculture system for 60 d.After the feeding trial,the muscle and liver of gibel carp were analysed.The results showed that RI supplementation did not result in any negative impact on the growth performance and 0.30RI supplementation significantly increased the whole-body protein content compared to the control group.The contents of collagen and glycogen in muscle were enhanced by RI supplementation.The alterations in the flesh indicated that RI supplementation improved the texture of the flesh in terms of its water-holding capacity and hardness,therefore improving the taste.Dietary RI facilitated the deposition of amino acids and fatty acids in the muscle that contributed to the meaty taste and nutritional value.Furthermore,a combination of metabolomics and expression of key genes in liver and muscle revealed that 0.30RI activated the purine metabolism pathways by supplementing the substrate for nucleotide synthesis and thereby promoting the deposition of flavour substance in flesh.This study offers a new approach for providing healthy,nutritious and flavourful aquatic products.Wanjie Cai Lele Fu Cui Liu Linyue He Haokun Liu Dong Han Xiaoming Zhu Yunxia Yang Junyan Jin Shouqi Xie 2023Animal Nutrition2023,,2:0
8Existing tests vs. novel non-invasive assays for detection of invasive aspergillosis in patients with respiratory diseases显示文摘Background: Although existing mycological tests (bronchoalveolar lavage [BAL] galactomannan [GM], serum GM, serum (1,3)-β-D-glucan [BDG], and fungal culture) are widely used for diagnosing invasive pulmonary aspergillosis (IPA) in non-hematological patients with respiratory diseases, their clinical utility in this large population is actually unclear. We aimed to resolve this clinical uncertainty by evaluating the diagnostic accuracy and utility of existing tests and explore the efficacy of novel sputum-basedAspergillus assays.Methods: Existing tests were assessed in a prospective and consecutive cohort of patients with respiratory diseases in West China Hospital between 2016 and 2019 while novel sputum assays (especially sputum GM andAspergillus-specific lateral-flow device [LFD]) in a case-controlled subcohort. IPA was defined according to the modified European Organization for Research and Treatment of Cancer/Mycoses Study Group criteria. Sensitivity and specificity were computed for each test and receiver operating characteristic (ROC) curve analysis was performed.Results: The entire cohort included 3530 admissions (proven/probable IPA=66, no IPA=3464) and the subcohort included 127 admissions (proven/probable IPA=38, no IPA=89). Sensitivity of BAL GM (≥1.0 optical density index [ODI]: 86% [24/28]) was substantially higher than that of serum GM (≥0.5 ODI: 38% [39/102]) (χ^(2)=19.83,P<0.001), serum BDG (≥70 pg/mL: 33% [31/95]) (χ^(2)=24.65,P<0.001), and fungal culture (33% [84/253]) (χ^(2)=29.38,P<0.001). Specificity varied between BAL GM (≥1.0 ODI: 94% [377/402]), serum GM (≥0.5 ODI: 95% [2130/2248]), BDG (89% [1878/2106]), and culture (98% [4936/5055]). Sputum GM (≥2.0 ODI) had similar sensitivity (84% [32/38]) (Fisher’s exactP=1.000) to and slightly lower specificity (87% [77/89]) (χ^(2)=5.52,P=0.019) than BAL GM (≥1.0 ODI). Area under the ROC curve values were comparable between sputum GM (0.883 [0.812-0.953]) and BAL GM (0.901 [0.824-0.977]) (P=0.734). Sputum LFD had similar specificity (91% [81/89]) (χ^(2)=0.89,P=0.345) to and lower sensitivity (63% [24/38]) (χ^(2)=4.14,P=0.042) than BAL GM (≥1.0 ODI), but significantly higher sensitivity than serum GM (≥0.5 ODI) (χ^(2)=6.95,P=0.008), BDG (χ^(2)=10.43,P=0.001), and fungal culture (χ^(2)=12.70,P<0.001).Conclusions: Serum GM, serum BDG, and fungal culture lack sufficient sensitivity for diagnosing IPA in respiratory patients.
