| 1 | Type I IFN augments IL-27-dependent TRIM25 expression to inhibit HBV replication显示文摘Hepatitis B virus(HBV)can cause chronic hepatitis B,which may lead to cirrhosis and liver cancer.Type I interferon(IFN)is an approved drug for the treatment of chronic hepatitis B.However,the fundamental mechanisms of antiviral action by type I IFN and the downstream signaling pathway are unclear.TRIM25 is an IFN-stimulated gene(ISG)that has an important role in RIG-I ubiquitination and activation.Whether TRIM25 is induced in liver cells by type I IFN to mediate anti-HBV function remains unclear.Here we report that interleukin-27(IL-27)has a critical role in IFN-induced TRIM25 upregulation.TRIM25 induction requires both STAT1 and STAT3.In TRIM25 knockout HepG2 cells,type I IFN production was consistently attenuated and HBV replication was increased,whereas overexpression of TRIM25 in HepG2 cells resulted in elevated IFN production and reduced HBV replication.More interestingly,we found that TRIM25 expression was downregulated in HBV patients and the addition of serum samples from HBV patients could inhibit TRIM25 expression in HepG2 cells,suggesting that HBV might have involved a mechanism to inhibit antiviral ISG expression and induce IFN resistance.Collectively,our results demonstrate that type I IFN-induced TRIM25 is an important factor in inhibiting HBV replication,and the IFN-IL-27-TRIM25 axis may represent a new target for treating HBV infection. | Guangyun Tan Qingfei Xiao Hongxiao Song Feng Ma Fengchao Xu Di Peng Na Li Xiaosong Wang Junqi Niu Pujun Gao F Xiao-Feng Qin Genhong Cheng | 2018 | Cellular & Molecular Immunology2018,15,3: | 13 |

It was found that with the increase of Ni content, the Charpy impact toughness improved remarkably, and transformed from cleavage brittle fracture to the most ductile fracture. | 4 | Activating Flux Design for Laser Weldingof Ferritic Stainless Steel显示文摘The behaviors of YAG laser welding process of ferritic stainless steel with activating fluxes were investigatedin this study. Some conventional oxides, halides and carbonates were applied in laser welding. The resultsshowed that the effect of oxides on the penetration depth was more remarkable. Most activating fluxes improved thepenetration more effectively at low power than that at high power. The uniform design was adopted to arrange theformula of multicomponent activating fluxes, showing that the optimal formula can make the penetration depth up to2.23 times as large as that without flux, including 50% ZrO2, 12.09% CaCO3, 10.43% CaO and 27.48% MgO. | 5 | Improvement in quality of life in depressed patients following verum acupuncture or electroacupuncture plus paroxetine:A randomized controlled study of 157 cases显示文摘Depressed patients with scores of 17 or more on the 17 items of the Hamilton Depression Rating Scale were treated with the antidepressant drug paroxetine. They also underwent verum acupuncture or electroacupuncture at Baihui (GV20) and Yintang (GV29). The World Health Organization Quality of Life Scale Brief Version showed a significant increase in the total scores of patients who underwent verum acupuncture and electroacupuncture for 6 weeks compared with those who were given paroxetine only; significantly increased physical domain and social relationship scores in verum acupuncture patients compared with paroxetine only; and significantly elevated psychological domain scores with electroacupuncture compared with paroxetine only. | 6 | 青海河湟谷地胃癌病理特征及其变化趋势分析(英文)显示文摘Objective: The aim of our study was to discuss changes in the clinicopathological characteristics of gastric cancer in Hehuang Valley of China in recent 10 years. Methods: A retrospective case-control study was performed on 2379 newly-diagnosed gastric cancer patients. All of them came from Hehuang Valley. The patients were divided into two groups [recent 5 years(R5Y) and late 5 years(L5Y)] from February 2003 to February 2013, and the clinicopathological data were surveyed retrospectively. Results: The constituent ratio of upper 1/3 gastric cancer in R5Y and L5Y was 33.5% and 20.7%, respectively, and it showed a significant difference between the two groups(χ2 = 21.28, P = 0.00). The constituent ratio of squamous carcinoma/adenosquamous carcinoma was 2.7% and 1.6%, respectively, and it also showed a significant difference between two groups(χ2 = 50.91, P = 0.00). The constituent ratio of moderately-poorly differentiated/poorly differentiated gastric carcinoma was 84.2% and 50.2%, respectively, and it showed statistically significant difference(χ2 = 30.28, P = 0.00). The detection rate of early gastric cancer(EGC) was 1.47%(35/2379). The constituent ratio of the types of Borrmann II and Borrmann IV of advanced gastric cancer(AGC) among 2379 cases was 47.6% and 40.8%, respectively, and that still showed statistically significant difference(χ2 = 18.80, P = 0.00). The constituent ratio of diffuse-type of gastric cancer in R5Y and L5Y was 36.2% and 30.8%, respectively, even there was a significant difference(χ2 = 7.49, P = 0.01). Furthermore, there were also significant differences in regional lymph nodes metastasis, perineural infiltration and vascular invasion(P < 0.05). The positive detection rate of HER2, ER and PR was 14.88%, 17.23% and 15.93%, respectively. The constituent ratio of HP in two groups was 43.8% and 36.2% respectively, and it also showed a significant difference(χ2 = 13.51, P = 0.00). Conclusion: The pathogenic sites in gastric cancer change to the upper stomach in Hehuang Valley in recent 10 years, and the detection rate of squamous carcinoma/adenosquamous carcinoma reveals a sharp rise. Borrmann III is still one of the main types of advanced gastric cancer, but the detection rates of Borrmann II and IV are increasing. The main type of gastric cancer is the intestinaltype, but the ratio of diffuse-type is also increasing in recent 10 years. The HP detection rate is 40.65%(967/2379), and it has a slight rise in recent 10 years. The detection rate of poorly differentiated adenocarcinoma is increasing despite the fact that the moderately/moderately-poorly differentiated adenocarcinoma is the main histologic types. High detection rates of lymph nodes metastasis, perineural infiltration and vascular invasion indicate poor prognosis in patients with gastric cancer. | 7 | MULTI-SHOP SCHEDULING PROBLEM显示文摘A new concept of multi-shop (M ) is put forward which contains all basic shops including open shop (O), job shop (J ), flow shop (F ) and hybrid flow shop (H ) so that these basic shop can be scheduled together. Several algorithms including ant colony optimization (ACO), most work remaining (MWR), least work remaining (LWR), longest processing time (LPT) and shortest processing time (SPT) are used for scheduling the M. Numerical experiments of the M adopting data of some car and reC series benchmark instances are tested. The results show that the ACO algorithm has better performance for scheduling the M than the other algorithms, if minimizing the makespan ( C m*ax) is taken as the objective function. As a comparison, the separate shops contained in the M are also scheduled by the ACO algorithm for the same objective function, when the completing time of the jobs in the previous shop is taken as the ready time of these jobs in the following shop. | 11 | Fast-response fluorescent probe with favorable water solubility for highly sensitive imaging of endogenous tyrosinase in living cells and zebrafish model显示文摘Tyrosinase(TYR) is an important polyphenolic oxidase enzyme and usually regards as a biomarker of melano ma cancer.Highly effective tracking TYR activity in vivo will help to study the mechanism of TYR in living organisms and forecasts related diseases.In this study,we present a novel TYR-activatable fluo rescent probe(CHMC-DOPA) for tracking TYR activity in vitro and in vivo.CHMC-DOPA is constructed by incorporating dopamine(DOPA) moiety into a fluorescent chloro-hydroxyl-merocyanine(CHMC)scaffold.Upon exposure to TYR,the dopamine unit in CHMC-DOPA is oxidized to a dopaquinone derivative,and an intramolecular photo-induced electron transfer(PET) process between CHMC fluorophore and o-dopaquinone will take place,the fluorescence of CHMC-DOPA is quenched rapidly.Therefore,the evaluation of TYR activity is established in terms of the relationship between fluorescence quenching efficiency and TYR activity.In our experiments,CHMC-DOPA shows various advantages,such as fast response(8 min),low concentration of TYR activation(0.5 U/mL),good water-solubility,as well as the lowest detection limit(0.003 U/mL) compared with previously reported works.Furthermore,CHMCDO PA also exhibits excellent cell membrane permeability and low cytotoxicity,which is successfully used to monitor endogenous TYR activity in living cancer cells and zebrafish models.CHMC-DOPA performs well,and we anticipate that this newly designed novel platform will provide an alternative for high effective monitoring TYR activity in biosystems. | Zheng Li Xiaofeng Xia Yu You Cuifen Lu Guichun Yang Chao Ma Junqi Nie Qi Sun Shuilin Wu Jun Ren Feiyi Wang | 2021 | Chinese Chemical Letters2021,32,5: | 1 |

| 14 | Junctional and somatic hypermutation-induced CX4C motif is critical for the recognition of a highly conserved epitope on HCV E2 by a human broadly neutralizing antibody显示文摘Induction of broadly neutralizing monoclonal antibodies(bNAbs)that bind to the viral envelope glycoproteins is a major goal of hepatitis C virus(HCV)vaccine research.The study of bNAbs arising in natural infection is essential in this endeavor.We generated a human antibody,8D6,recognizing the E2 protein of HCV isolated from a chronic hepatitis C patient.This antibody shows broadly neutralizing activity,which covers a pan-genotypic panel of cell culture-derived HCV virions(HCVcc).Functional and epitope analyses demonstrated that 8D6 can block the interaction between E2 and CD81 by targeting a highly conserved epitope on E2.We describe how the 8D6 lineage evolved via somatic hypermutation to achieve broad neutralization.We found that the V(D)J recombination-generated junctional and somatic hypermutation-induced disulfide bridge(C-C)motif in the CDRH3 is critical for the broad neutralization and binding activity of 8D6.This motif is conserved among a series of broadly neutralizing HCV antibodies,indicating a common binding model.Next,the 8D6 inferred germline(iGL)was reconstructed and tested for its binding affinity and neutralization activity.Interestingly,8D6 iGL-mediated relatively strong inhibition of the 1b genotype PR79L9 strain,suggesting that PR79L9 may serve as a potential natural viral strain that provides E2 sequences that induce bNAbs.Overall,our detailed epitope mapping and genetic studies of the HCV E2-specific mAb 8D6 have allowed for further refinement of antigenic sites on E2 and reveal a new mechanism to generate a functional CDRH3,while its iGL can serve as a probe to identify potential HCV vaccine strains. | Chunyan Yi Jing Xia Lan He Zhiyang Ling Xuesong Wang Yu Yan Jiangjun Wang Xinhao Zhao Weiguo Fan Xiaoyu Sun Ronghua Zhang Sheng Ye Rongguang Zhang Yongfen Xu Liyan Ma Yaguang Zhang Honglin Zhou Zhong Huang Junqi Niu Gang Long Junxia Lu Jin Zhong Bing Sun | 2021 | Cellular & Molecular Immunology2021,18,3: | 0 |
| 15 | Postoperative Radiotherapy and N2 Non-small Cell Lung Cancer Prognosis: A Retrospective Study Based on Surveillance, Epidemiology, and End Results Database显示文摘The purpose of this study is to clarify the significance of postoperative radiotherapy for N2 lung cancer.This study aimed to investigate the effect of postoperative radiotherapy on the survival and prognosis of patients with N2 lung cancer.Data from 12,000 patients with N2 lung cancer were extracted from the Surveillance,Epidemiology,and End Results database(2004-2012).Age at disease onset and 5-year survival rates were calculated.Survival curves were plotted using the Kaplan-Meier method.The univariate log-rank test was performed.Multivariate Cox regression were used to examine factors affecting survival.Patients’median age was 67 years(mean 66.46±10.03).The 5-year survival rate was 12.55%.Univariate analysis revealed age,sex,pathology,and treatment regimen as factors affecting prognosis.In multivariate analysis,when compared to postoperative chemotherapy,postoperative chemoradiotherapy was better associated with survival benefits(hazard ratio[HR]=0.85,95%confidence interval[CI]:0.813-0.898,P<0.001).Propensity score matching revealed that patients who had received postoperative chemoradiotherapy had a better prognosis than did patients who had received postoperative chemotherapy(HR=0.869,95%CI:0.817-0.925,P<0.001).Female patients and patients aged<65 years had a better prognosis than did their counterparts.Patients with adenocarcinoma had a better prognosis than did patients with squamous cell carcinoma.Moreover,prognosis worsened with increasing disease T stage.Patients who had received postoperative chemoradiotherapy had a better prognosis than did patients who had received postoperative chemotherapy.Postoperative radiotherapy was an independent prognostic factor in this patient group. | Yunbo Zhang Liping Zheng Junqi Liu Jinqiu Li Jianguang Zhang Jingjing Ma Yuxiu Song | 2021 | Journal of Oncology Research2021,3,2: | 0 |

| 18 | Deposition of water-soluble inorganic ions in PM_(2.5) in a typical forestry system in Beijing,China显示文摘Background: Rapid economic development in China has resulted in an increase in severe air pollution in city groups such as the Beijing-Tianjin-Hebei Metropolitan Region. PM2.5(fine particles with an aerodynamic equivalent diameter of 2.5 [xm or less) is one of the most important pollutants. The deposition process is an important way of removing particles from the air. To evaluate the effect of an urban forest on atmospheric particle removal, a concentration gradient method was used to measure the deposition velocities of water-soluble inorganics in PM2.5 in two national forest parks in Beijing, China. The following eight water-soluble inorganic ions in PM2.5 were investigated: sodium, ammonium, potassium, magnesium, calcium, chloride, nitrate, and sulfate.Methods: Samples were taken from two sites in Beijing from the 7 th to the 15 th May, 2013. The concentrations of water-soluble inorganic ions were analyzed with ion chromatography. We used the concentration gradient technique to estimate the deposition flux and velocity. To determine the relationships between leaf traits and particle accumulation, typical leaf samples from each selected species were studied using scanning electron microscopy.Results: The total deposition flux and total deposition velocity during the daytime were higher than those at night.Sulfate showed the biggest deposition flux and velocity at both study sites, whereas the other ions showed different trends at each site. Result from higher proportion of coniferous to broadleaved trees, the total deposition flux of the eight ions measured in Jiufeng National Forest Park was greater than that in Olympic Forest Park.Conclusions: The deposition velocity was affected by meteorological conditions such as wind speed, temperature,and humidity. The deposition velocity was also influenced by tree species. The surface of plants is an important factor influencing particle deposition.
