| 1 | An in vivo study with an MRI tracer method reveals the biophysical properties of interstitial fluid in the rat brain显示文摘The nature of brain interstitial fluid (ISF) has long been a subject of controversy. Most of the previous studies on brain ISF were carried out in vitro. In the present study, a novel method was developed to characterize ISF in the living rat brain by magnetic resonance (MR) imaging using gadolinium-diethylenetriaminepentaacetic acid (Gd-DTPA) as a tracer. Sprague Dawley rats (n=8) were subjected to MR scanning before and after the introduction of Gd-DTPA into the caudate nucleus. A one-way drainage of brain ISF was demonstrated on the dynamic MR images. According to the traditional diffusion model, the diffusion and clearance rate constants of the tracer within brain extracellular space (ECS) were derived as (3.38±1.07)×10-4 mm 2 s-1 and (7.60±4.18)×10-5 s-1 . Both diffusion and bulk flow contributed to the drainage of ISF from the caudate nucleus, which demonstrated an ISF-cerebrospinal fluid confluence in the subarachnoid space at the lateral and ventral surface of the brain cortex at 3 h after the injection. | 2 | LA-ICP-MS zircon U-Pb dating and phenocryst EPMA of dikes, Guocheng, Jiaodong Peninsula: Implications for North China Craton lithosphere evolution显示文摘Widespread dike swarm, including diorite-, monzonite-porphyry and lamprophyre, intruded in the al- tered breccia gold deposits along basin marginal faults, Guocheng, Jiaodong Peninsula. Petrography exhibits biotite enclaves in amphibole phenocrysts and the presence of acicular apatites in these dikes. Electron probe microanalyses (EPMA) show that the amphibole and clinopyroxene phenocryst’s mantle in diorite porphyry and lamprophyre respectively has sharply higher MgO (Mg#) and Cr2O3 contents in contrast to their cores. The plagioclase phenocryst in monzonite porphyry has reverse zoning. These results indicate that the magma mixing between mantle-derived mafic and crust-derived felsic magmas occurred in the original process of the dikes. Zircon cathodoluminescence (CL) images show well-developed magmatic oscillatory zones and the acquired LA-ICP-MS zircon U-Pb weighted mean 206Pb/238U ages are 114±2 Ma (MSDW=1.5) for monzonite porphyry (GS1) and 116±1 Ma (MSDW=0.8) for diorite porphyry (GS2), respectively. Earlier magmatic events in the northwest Jiaodong Peninsula represented by some inherited or captured zircons also occur in these dikes. Magmatic zircons from GS1 and GS2 display consistent chondrite-normalized REE patterns and Nb/Ta values, implying that they may share a similar or same source. HREE enrichment and obvious negative Eu anomalies of these zircons preclude garnet presented in their source. Our results, combined with preciously pub- lished data, indicate that dike intrusion and gold mineralization among quartz vein, altered tectonite and altered breccia gold deposits are broadly contemporaneous throughout the Jiaodong Peninsula. | 4 | The chromosome-level quality genome provides insights into the evolution of the biosynthesis genes for aroma compounds of Osmanthus fragrans显示文摘Sweet osmanthus(Osmanthus fragrans)is a very popular ornamental tree species throughout Southeast Asia and USA particularly for its extremely fragrant aroma.We constructed a chromosome-level reference genome of O.fragrans to assist in studies of the evolution,genetic diversity,and molecular mechanism of aroma development.A total of over 118 Gb of polished reads was produced from HiSeq(45.1 Gb)and PacBio Sequel(73.35 Gb),giving 100×depth coverage for long reads.The combination of Illumina-short reads,PacBio-long reads,and Hi-C data produced the final chromosome quality genome of O.fragrans with a genome size of 727 Mb and a heterozygosity of 1.45%.The genome was annotated using de novo and homology comparison and further refined with transcriptome data.The genome of O.fragrans was predicted to have 45,542 genes,of which 95.68%were functionally annotated.Genome annotation found 49.35%as the repetitive sequences,with long terminal repeats(LTR)being the richest(28.94%).Genome evolution analysis indicated the evidence of whole-genome duplication 15 million years ago,which contributed to the current content of 45,242 genes.Metabolic analysis revealed that linalool,a monoterpene is the main aroma compound.Based on the genome and transcriptome,we further demonstrated the direct connection between terpene synthases(TPSs)and the rich aromatic molecules in O.fragrans.We identified three new flower-specific TPS genes,of which the expression coincided with the production of linalool.Our results suggest that the high number of TPS genes and the flower tissue-and stage-specific TPS genes expressions might drive the strong unique aroma production of O.fragrans. | Xiulian Yang Yuanzheng Yue Haiyan Li Wenjie Ding Gongwei Chen Tingting Shi Junhao Chen Min S.Park Fei Chen Lianggui Wang | 2018 | Horticulture Research2018,5,1: | 16 |
| 5 | Reproductive management through integration of PGD and MPS-based noninvasive prenatal screening/diagnosis for a family with GJB2-associated hearing impairment显示文摘A couple with a proband child of GJB2(encoding the gap junction protein connexin 26)-associated hearing impairment and a previous pregnancy miscarriage sought for a reproductive solution to bear a healthy child. Our study aimed to develop a customized preconception-to-neonate care trajectory to fulfill this clinical demand by integrating preimplantation genetic diagnosis(PGD), noninvasive prenatal testing(NIPT), and noninvasive prenatal diagnosis(NIPD) into the strategy. Auditory and genetic diagnosis of the proband child was carried out to identify the disease causative mutations. The couple then received in-vitro-fertilization treatment, and eight embryos were obtained for day 5 biopsy. PGD was performed by short-tandem-repeat linkage analysis and Sanger sequencing of GJB2 gene. Transfer of a GJB2 c.235del C heterozygous embryo resulted in a singleton pregnancy. At the 13 th week of gestation, genomic DNA(g DNA) from the trio family and cell-free DNA(cf DNA) from maternal plasma were obtained for assessment of fetal chromosomal aneuploidy and GJB2 mutations. NIPT and NIPD showed the absence of chromosomal aneuploidy and GJB2-associated disease in the fetus, which was later confirmed by invasive procedures and postnatal genetic/auditory diagnosis. | 7 | Numerical simulation on the seismic absorption effect of the cushion in rigid-pile composite foundation显示文摘In order to quantitatively study the seismic absorption effect of the cushion on a superstructure, a numerical simulation and parametric study are carried out on the overall FEA model of a rigid-pile composite foundation in ABAQUS. A simulation of a shaking table test on a rigid mass block is first completed with ABAQUS and EERA, and the effectiveness of the Drucker-Prager constitutive model and the finite-infinite element coupling method is proved. Dynamic time-history analysis of the overall model under frequent and rare earthquakes is carried out using seismic waves from the El Centro, Kobe, and Bonds earthquakes. The different responses of rigid-pile composite foundations and pile-raft foundations are discussed. Furthermore, the influence of thickness and modulus of cushion, and ground acceleration on the seismic absorption effect of the cushion are analyzed. | 8 | Exogenous abscisic acid coordinating leaf senescence and transport of assimilates into wheat grains under drought stress by regulating hormones homeostasis显示文摘Drought at the grain filling stage of wheat will cause premature leaf senescence, thus leading to considerable loss of wheat yield. Therefore, this paper aims to establish a cultivation technology for strong drought resistance, delayed senescence, and yield improvement based on the analysis of hormones homeostasis obtained by applying chemical control substances. Experiments were conducted with two genotypes of wheat. Four water irrigation treatments were applied to impose the water deficit, including well-watered control treatment(WW), mild water deficit(MiWD), moderate water deficit(MoWD), and severe water deficit(SWD). Exogenous abscisic acid(ABA) was sprayed on the plants at the anthesis stage of the wheat. As a result, exogenous ABA reduced initial senescence rate(r0), total duration of chlorophyll(Chltotal), rapid senescence phase(Chlloss), and the accumulated temperature at an inflection point(M) but improved the persistence phase(Chlper) of flag leaves under all of the four treatments. However, exogenous ABA produced inconsistent effects on photoassimilate relocation and grain weight under different treatments. It produced positive regulatory effects on grain weight under WW, MiWD, and MoWD treatments. On the one hand, spraying ABA during the persistence phase of flag leaves reduced the ratios of zeatin to gibberellin(Z/GA3), spermine to spermidine(Spm/Spd), and salicylic acid to ABA(SA/ABA),which prolonged active photosynthesis by stimulating high level of proline(Pro) and increased the activities of antioxidant enzymes, such as superoxide dismutase(SOD), peroxidase(POD), catalase(CAT), and ascorbate peroxidase(APX). Therefore, drought tolerance was enhanced, and more photosynthetic assimilates were accumulated. On the other hand, the rapid senescence phase and the transport rate of assimilates into grains were accelerated, resulting in higher grain weight, yield, and water use efficiency(WUE). However, under SWD treatment, exogenous ABA improved the ratio of SA/ABA, leading to low Pro content and low antioxidant enzyme activity of flag leaves in the rapid loss phase. Meanwhile,drought resistance declined and the transport duration of assimilates into grains was shortened, thus making photosynthetic assimilates redundant. | 11 | Diamond/β-SiC film as adhesion-enhanced interlayer for top diamond coatings on cemented tungsten carbide substrate显示文摘In present work, diamond/β-SiC composite interlayers were deposited on cemented tungsten carbide(WC-6%Co) substrates by microwave plasma enhanced chemical vapor deposition(MPCVD) using H_2,CH_4 and tetramethylsilane(TMS) gas mixtures. The microstructure, chemical bonding, element distribution and crystalline quality of the composite interlayers were systematically characterized by means of field-emission scanning electron microscopy(FE-SEM), X-ray diffraction(XRD), X-ray photoelectron spectrometer(XPS), electron probe microanalysis(EPMA), Raman spectroscopy and transmission electron microscropy(TEM). The influences of varying TMS flow rates on the diamond/β-SiC composite interlayers were investigated. Through changing the TMS flow rates in the reaction gas, the volume fraction of β-SiC in the composite interlayers were tunable in the range of 12.0%–68.1%. XPS and EPMA analysis reveal that the composite interlayers are composed of C, Si element with little cobalt distribution. The better crystallinity of the diamond in the composite is characterized based on the Raman spectroscopy, which are helpful to deposit top diamond coatings with high quality. Then, the adhesion of top diamond coatings were estimated using Rockwell C indentation analysis, revealing that the adhesion of top diamond coatings on the WC-6%Co substrates can be improved by the interlayers with the diamond/β-SiC composite structures. Comprehensive TEM interfacial analysis exhibits that the cobalt diffusion is weak from WC-6%Co substrate to the composite interlayer. The homogeneous microcrystalline diamond coatings with the most excellent adhesion can be fabricated on the substrates with the composite interlayer with the β-SiC/diamond ratio of about 45%. | 16 | From similarity perspective: a robust collaborative filtering approach for service recommendations显示文摘Collaborative filtering (CF) is a technique commonly used for personalized recommendation and Web service quality-of-service (QoS) prediction. However, CF is vulnerable to shilling attackers who inject fake user profiles into the system. In this paper, we first present the shilling attack problem on CF-based QoS recommender systems for Web services. Then, a robust CF recommendation approach is proposed from a user similarity perspective to enhance the resistance of the recommender systems to the shilling attack. In the approach, the generally used similarity measures are analyzed, and the DegSim (the degree of similarities with top k neighbors) with those measures is selected for grouping and weighting the users. Then, the weights are used to calculate the service similarities/differences and predictions. We analyzed and evaluated our algorithms using WS-DREAM and Movielens datasets. | 17 | The main anammox-based processes, the involved microbes and the novel process concept from the application perspective显示文摘Anammox technology has been widely researched over the past 40-year from the laboratory-scale to full-scale. It is well-known that in actual applications, the solo application of anammox is not feasible. Since both ammonium and nitrite are prerequisites based on the reaction mechanism, the pre-treatment of wastewater is necessary. With the combination of anammox process and other pre-treatment processes to treat the actual wastewater, many types of anammox-based processes have been developed with distinct nitrogen removal performance. Thus, in order to heighten the awareness of researchers to the developments and accelerate the application of these processes to the treatment of actual wastewater, the main anammox-based processes are reviewed in this paper. It includes the partial nitritation/anammox process, the denitratation/anammox (PD/A) process, the denitrifying anaerobic methane oxidation/anammox (DAMO/A) process, and more complex deuterogenic processes. These processes have made the breakthroughs in the application of the anammox technology, such as the combination of nitrification and PD/A process can achieve stability and reliability of nitrogen removal in the treatment of mainstream wastewater, the PD/A process and the DAMO/A have brought about further improvements in the total nitrogen removal efficiency of wastewater. The diversity of functional microbe characteristics under the specific condition indicate the wide application potential of anammox-based processes, and further exploration is necessary. | 19 | Characteristics of Element Migration in the Process of Wall-Rock Alteration in the Shibangou Gold Deposit,Western Henan显示文摘The Shibangou gold deposit in western Henan is associated with irregular quartz veinlets occurring in altered shear zones dissecting a dioritic intrusion. The altered shear zones are characterized by silicification, pyriti-zation, sericitization, chloritization and K-feldspar alteration. Zoning of altered rocks adjacent to the Au-bearing quartz veins is obviously exhibited. Fine-grained sulphides and quartz veinlets of different ages and small-scale fissures are widely distributed in the central part of the altered zones. Major mineralization types in this gold deposit are Au-bearing quartz vehlets and altered rocks in the shear zones. Samples were collected from drilling cores according to the alteration zoning and mineralization type and all samples were analyzed for major and trace elements. Mass balance, volume change (fv=97.3-71.9%) and major element variation sequences are studied in terms of major elements.
