12,2’,4,4’-四溴二苯醚(BDE-47)对大型溞(Daphnia magna)的毒性效应显示文摘2,2’,4,4’-四溴二苯醚(BDE-47)在水体及各种水生生物(鱼类、海洋哺乳动物以及水生无脊椎动物)中被广泛检出,但BDE-47对水生无脊椎动物毒性效应的研究还处于起步阶段。以大型溞(Daphnia magna)为受试生物,通过急性(48h)和慢性(21d)毒性暴露实验,考察了BDE-47对大型溞活动抑制率、心率、产仔情况和酶活性等指标的影响。结果显示,BDE-47对大型溞活动抑制率的48h-EC50为112.5μg.L-1;高浓度(>100μg.L-1)BDE-47显著诱导提高大型溞的心率。21d慢性暴露实验中,8μg.L-1处理组中大型溞全部死亡;其他各浓度处理组(0.5、1、2、4μg.L-1)中,母溞第1胎产仔时间延后,第1胎子代数量减少,总产仔数量大幅减少,这表明大型溞的繁殖能力受到抑制。BDE-47在一定程度上抑制了母溞胆碱酯酶(ChE)和谷胱甘肽硫转移酶(GST)的活性,大型溞体内的代谢机制没有被诱导,神经活性虽被抑制,但抑制率不高。BDE-47大幅诱导过氧化氢酶(CAT)的活性,并呈现一定的剂量效应关系,相对于ChE和GST,CAT对BDE-47暴露更为敏感,可作为BDE-47对大型溞慢性暴露毒性效应的潜在生物标志物。熊勤犁 史雅娟 吕永龙 李秋爽 Gosens Jorrit 2013生态毒理学报2013,8,1:12
2Factors influencing polychlorinated dibenzo-p-dioxin and polychlorinated dibenzofuran (PCDD/F) emissions and control in major industrial sectors: Case evidence from Shandong Province, China显示文摘Analyzing determinants that influence polychlorinated dibenzo-p-dioxin and polychlorinated dibenzofuran(PCDD/F) emissions is helpful for decision-makers to find effective and efficient ways to mitigate PCDD/F emissions. The PCDD/F emissions and the contributions of the scale effect, structure effect and technology effect to emissions from eight main industrial sectors in2006, 2008 and 2010 in Shandong Province, were calculated in this article. Total PCDD/F emissions in Shandong increased by 52.8% in 2008(614.1 g I-TEQ) and 49.7% in 2010(601.8 g I-TEQ) based on 2006(401.9 g I-TEQ). According to the decomposition method, the largest influencing factor on PCDD/F emission changes was the composition effect(contributed 43.4%in 2008 and 120.6% in 2010 based on 2006), which was also an emission-increasing factor.In this case, the present industrial restructuring policy should be adjusted to control the proportion of production capacities with high emission factors, such as iron ore sintering and steelmaking and the secondary non-ferrous metal sector. The scale effect increased the emissions in 2008(contributed 21.9%) and decreased the emissions in 2010(contributed-28.0%). However, as a source control measure, the excess capacity control policy indeed had a significant role in emission reduction. The main reason for the technology effect(contributed 34.7% in 2008 and 7.4% in 2010 based on 2006) having an emission-increasing role was the weakness in implementing policies for restricting industries with outdated facilities. 6SIZE MATTERS:PITCH DIMENSIONS CONSTRAIN INTERACTIVE TEAM BEHAVIOUR IN SOCCER显示文摘Pitch size varies in official soccer matches and differently sized pitches are adopted for tactical purposes in small-sided training games.Since interactive team behaviour emerges under constraints,the authors evaluate the effect of pitch size(task) manipulations on interactive team behaviour in small-sided soccer games.Pour 4-a-side(plus goalkeepers) small-sided games were played:a reference game(30×20 m),length manipulation(24×20 m),width manipulation(30×16 m),and a combination(24×16 m).Using position data(100Hz),three measures quantifying the teams' interaction were calculated:longitudinal inter-team distance,lateral inter-team distance,and surface area difference.Means and standard deviations,correlations and coupling values were calculated.Running correlations were calculated over a 3-s window to evaluate interaction patterns.As expected,a shorter pitch results in smaller longitudinal inter-team distance,lateral inter-team distance decreased for narrow pitches,and smaller total playing area resulted in decreased surface area.Unanticipated,a crossover effect was present;length and width manipulations also triggered changes in lateral and longitudinal direction respectively.Inter-team distances and surface area difference differed significantly across conditions.Interaction patterns differed across conditions for all measures.So,highly tactically relevant,soccer teams seem to adapt their interactive behaviour according to pitch size in small-sided games.FRENCKEN Wouter VAN DER PLAATS Jorrit VISSCHER Chris LEMMINK Koen 2013Journal of Systems Science & Complexity2013,26,1:1

15Establishment of head and neck squamous cell carcinoma mouse models for cetuximab resistance and sensitivity显示文摘Aim:Acquired resistance to the targeted agent cetuximab poses a significant challenge in finding effective anti-cancer treatments for head and neck squamous cell carcinoma(HNSCC).To accurately study novel combination treatments,suitable preclinical mouse models for cetuximab resistance are key yet currently limited.This study aimed to optimize an acquired cetuximab-resistant mouse model,with preservation of the innate immunity,ensuring intact antibody-dependent cellular cytotoxicity(ADCC)functionality.Methods:Cetuximab-sensitive and acquired-resistant HNSCC cell lines,generated in vitro,were subcutaneously engrafted in Rag2 knock-out(KO),BALB/c Nude and CB17 Scid mice with/without Matrigel or Geltrex.Once tumor growth was established,mice were intraperitoneally injected twice a week with cetuximab for a maximum of 3 weeks.In addition,immunohistochemistry was used to evaluate the tumor and its microenvironment.Results:Despite several adjustments in cell number,cell lines and the addition of Matrigel,Rag2 KO and BALB/C Nude mice proved to be unsuitable for xenografting our HNSCC cell lines.Durable tumor growth of resistant SC263-R cells could be induced in CB17 Scid mice.However,these cells had lost their resistance phenotype in vivo.Immunohistochemistry revealed a high infiltration of macrophages in cetuximab-treated SC263-R tumors.FaDu-S and FaDu-R cells successfully engrafted into CB17 Scid mice and maintained their sensitivity/resistance to cetuximab.Conclusion:We have established in vivo HNSCC mouse models with intact ADCC functionality for cetuximab resistance and sensitivity using the FaDu-R and FaDu-S cell lines,respectively.These models serve as valuable tools for investigating cetuximab resistance mechanisms and exploring novel drug combination strategies.Hannah Zaryouh Ines De Pauw Hasan Baysal Joran Melis Valentin Van den Bossche Christophe Hermans Ho Wa Lau Hide Lambrechts Celine Merlin Cyril Corbet Marc Peeters Jan Baptist Vermorken Jorrit De Waele Filip Lardon An Wouters 2023Cancer Drug Resistance2023,6,4:0
