3Role of PRMTs in cancer: Could minor isoforms be leaving a mark?显示文摘Protein arginine methyltransferases(PRMTs) catalyze the methylation of a variety of protein substrates, many of which have been linked to the development, progression and aggressiveness of different types of cancer. Moreover, aberrant expression of PRMTs has been observed in several cancer types. While the link between PRMTs and cancer is a relatively new area of interest, the functional implications documented thus far warrant further investigations into its therapeutic potential. However, the expression of these enzymes and the regulation of their activity in cancer are still significantly understudied. Currently there are nine main members of the PRMT family. Further, the existence of alternatively spliced isoforms for several of these family members provides an additional layer of complexity. Specifically, PRMT1, PRMT2, CARM1 and PRMT7 have been shown to have alternative isoforms and others may be currently unrealized. Our knowledge with respect to the relative expression and the specific functions of these isoforms is largely lacking and needs attention. 4Stem cell-derived exosomes-an emerging tool for myocardial regeneration显示文摘Cardiovascular diseases(CVDs) continue to represent the number one cause of death and disability in industrialized countries. The most severe form of CVD is acute myocardial infarction(AMI), a devastating disease associated with high mortality and disability. In a substantial proportion of patients who survive AMI, loss of functional cardiomyocytes as a result of ischaemic injury leads to ventricular failure, resulting in significant alteration to quality of life and increased mortality. Therefore, many attempts have been made in recent years to identify new tools for the regeneration of functional cardiomyocytes. Regenerative therapy currently represents the ultimate goal for restoring the function of damaged myocardium by stimulating the regeneration of the infarcted tissue or by providing cellsthat can generate new myocardial tissue to replace the damaged tissue. Stem cells(SCs) have been proposed as a viable therapy option in these cases. However, despite the great enthusiasm at the beginning of the SC era, justified by promising initial results, this therapy has failed to demonstrate a significant benefit in large clinical trials. One interesting finding of SC studies is that exosomes released by mesenchymal SCs(MSCs) are able to enhance the viability of cardiomyocytes after ischaemia/reperfusion injury, suggesting that the beneficial effects of MSCs in the recovery of functional myocardium could be related to their capacity to secrete exosomes. Ten years ago, it was discovered that exosomes have the unique property of transferring miRNA between cells, acting as miRNA nanocarriers. Therefore, exosomebased therapy has recently been proposed as an emerging tool for cardiac regeneration as an alternative to SC therapy in the post-infarction period. 13Follow up of infection of chacma baboons with inoculum containing a and non-a genotypes of hepatitis B virus显示文摘AIM: To determine whether one genotype (A or non-A genotypes of HBV) predominated over the other during the course of HBV infection.METHODS: Four baboons were inoculated with HBV. DNA was extracted from serum obtained at monthly intervals postinoculation for 52 weeks and HBV DNA was amplified using primers specific for the core region containing an insert characteristic of genotype A (nt 2 354-2 359, numbering from the EcoRI site). The amplicons were cloned into PCRScriptTM and a minimum of 15 clones per time point were sequenced in both directions.RESULTS: Both genotypes persisted for the entire followup period of 52 weeks. Genotype non-A predominated in two baboons and genotype A in one baboon. 16Preparation of novel methyl methacrylate/fluorinated silsesquioxane copolymer film with low surface energy显示文摘Random copolymers of methyl methacrylate and fluorinated polyhedral oligomeric silsesquioxane(F-POSS) were synthesized and the corresponding thin films were prepared from solvent casting.Their microstructure was confirmed by 1H NMR,elemental analysis and GPC.Separation occurs in the bulk of the film during solvent evaporation which can be evidenced by Transmission Electron Microscopy,TEM,with POSS-rich nanophase sizes from 20 to 50 nm.Nanostructuration is attributed to the self-assembly of F-POSS due to the cluster-cluster interactions resulting from the nature of their ligands,i.e.,cycloaliphatic ligands and perfluorinated chains.Thermogravimetric analysis was used to investigate the thermal degradation temperature.It was shown that when F-POSS content is higher than 2.8 mol%,the incorporation of F-POSS could improve the thermal stability of PMMA significantly.In addition,it was shown that these fluorinated POSS-based copolymer surfaces could reduce the surface energy and could be used to design water-repellant nanocomposite coatings.GALY Jocelyne GéRARD Jean-Franois 2010Science China Chemistry2010,53,9:3
17Prospective evaluation of point-of-care ultrasound for pre-procedure identification of landmarks versus traditional palpation for lumbar puncture显示文摘BACKGROUND: The objective of this study is to determine if point-of-care ultrasound(POCUS) pre-procedure identification of landmarks can decrease failure rate, reduce procedural time, and decrease the number of needle redirections and reinsertions when performing a lumbar puncture(LP).METHODS: This was a prospective, randomized controlled trial comparing POCUS preprocedure identifi cation of landmarks versus traditional palpation for LP in a cohort of patients in the emergency department and intensive care unit.RESULTS: A total of 158 patients were enrolled. 18A new look at auranofin,dextromethorphan and rosiglitazone for reduction of glia-mediated inflammation in neurodegenerative diseases显示文摘Neurodegenerative disorders including Alzheimer's disease are characterized by chronic inflammation in the central nervous system. The two main glial types involved in inflammatory reactions are microglia and astrocytes. While these cells normally protect neurons by providing nutrients and growth factors, disease specific stimuli can induce glial secretion of neurotoxins. It has been hypothesized that reducing glia-mediated inflammation could diminish neuronal loss. This hypothesis is supported by observations that chronic use of non-steroidal anti-inflammatory drugs(NSAIDs) is linked with lower incidences of neurodegenerative disease. It is possible that the NSAIDs are not potent enough to appreciably reduce chronic neuroinflammation after disease processes are fully established. Gold thiol compounds, including auranofin, comprise another class of medications effective at reducing peripheral inflammation. We have demonstrated that auranofin inhibits human microglia- and astrocyte-mediated neurotoxicity. Other drugs which are currently used to treat peripheral inflammatory conditions could be helpful in neurodegenerative disease. 19The tomato HIGH PIGMENT1/DAMAGED DNA BINDING PROTEIN 1 gene contributes to regulation of fruit ripening显示文摘Fleshy fruit ripening is governed by multiple external and internal cues and accompanied by changes in color,texture,volatiles,and nutritional quality traits.While extended shelf-life and increased phytonutrients are desired,delaying ripening via genetic or postharvest means can be accompanied by reduced nutritional value.Here we report that the high pigment 1(hp1)mutation at the UV-DAMAGED DNA BINDING PROTEIN 1(DDB1)locus,previously shown to influence carotenoid and additional phytonutrient accumulation via altered light signal transduction,also results in delayed ripening and firmer texture,resulting at least in part from decreased ethylene evolution.Transcriptome analysis revealed multiple ethylene biosynthesis and signaling-associated genes downregulated in hp1.Furthermore,the hp1 mutation impedes softening of the pericarp,placenta,columella as well as the whole fruit,in addition to reduced expression of the FRUITFUL2(FUL2)MADS-box transcription factor and xyloglucan endotransglucosylase/hydrolase 5(XTH5).These results indicate that DDB1 influences a broader range of fruit development and ripening processes than previously thought and present an additional genetic target for increasing fruit quality and shelf-life.Anquan Wang Danyang Chen Qiyue Ma Jocelyn K.C.Rose Zhangjun Fei Yongsheng Liu James J.Giovannoni 2019Horticulture Research2019,6,1:3
20Trait evolution rates shape continental patterns of species richness in North America's most diverse angiosperm genus (Carex, Cyperaceae)显示文摘Ecological opportunity has been associated with increases in diversification rates across the tree of life.Under an ecological diversification model,the emergence of novel environments is hypothesized to promote morpho-and ecospace evolution.Whether this model holds at the clade level within the most species-rich angiosperm genus found in North America(Carex,Cyperaceae)is yet to be tested.Recent works demonstrate a temporal coupling of climate cooling and widespread colonization of Carex in North America,implicating ecological diversification.In addition,research has consistently found asymmetric patterns of lineage-level diversification in the genus.Why does variation in clade sizes exist in the genus?Is ecological diversification involved?In this study,we tested whether rates of morphological and ecological trait evolution are correlated with clade-level species richness in Carex of North America north of Mexico.We constructed a phylogeny of 477 species-an almost complete regional sample.We estimated rates of evolution of morphological traits,habitat,and climatic niche and assessed whether differences in rates of evolution correlate with species richness differences in replicate non-nested sister clades.Our work demonstrates significant positive correlations between climatic niche rates,habitat and reproductive morphological evolution,and species richness.This coupling of trait and niche evolution and species richness in a diverse,continental clade sample strongly suggests that the ability of clades to explore niche and functional space has shaped disparities in richness and functional diversity across the North American flora region.Our findings highlight the importance of the evolutionary history of trait and niche evolution in shaping continental and regional floras.Jocelyn E.Pender Andrew L.Hipp Marlene Hahn Julian R.Starr 2021Journal of Systematics and Evolution2021,59,4:3
