| 1 | Evidence of multistage late Quaternary strong earthquakes on typical segments of Longmenshan Active Fault Zone in Sichuan, China显示文摘Investigation of offset landforms and trench excavation are important means to acquire the evidence of multistage activities of active faults. Here we present the result of fault trough investigation in Beichuan County and the Pingtong Town of Pingwu County along the Longmenshan Central Fault Belt, as well as the result from trench excavation at the platform foreslope in Hanwang Town of Mianzhu County on the Longmenshan Front Range Fault Belt. These results show that at least three fault activity events, including the Wenchuan earthquake, occurred in the Beichuan fault trough, at least two, including the Wenchuan earthquake, at Pingtong fault trough, and 2-3 paleoearthquakes in the Hanwang trench. Among these three localities, the times of the last strong earthquake prior to Wenchuan earthquake at Beichuan fault trough and Hanwang trench are close, approximately 6000 years ago, i.e., greater than 5.8 ka and smaller than 6.63 ka ago. | 3 | Holocene Paleoseismic Activity Along the Northern Piedmont Fault of the Hengshan Mountain, South of Datong Basin in Shanxi Province显示文摘Our field investigation indicates that the northern piedmont fault of the Hengshan Mountain has dislocated proluvial fan and terrace I of the gully mouth on the northern border of the Hengshan Mountain. Stratigraphic age data indicates that the surface layer in the middle and rear part of the proluvial fan and on the terrace I of the gully mouth on the mountain border consists of sediments of about 5200~6800a B.P. The Hezhuang and Niucaoyu trenches excavated in the front of Hengshan Mountain revealed that 3 paleoseismic events have occurred along the northern piedmont fault of Hengshan Mountain since the early Holocene. The 3 paleoseismic events occurred in 2260±190~4370±150a BP, nearly 5628±150a BP, and 8083±250~8430±720a BP, respectively, and their recurrence interval is 2313a,2628a and 2471a on average. The coseismic vertical displacement produced by these events is 1 0~3 0m. The elapsed time of the latest movement along the fault has exceeded the recurrence interval of these paleoearthquakes. | 4 | Effects of Different Protein Energy Ratios on Fattening Performance,Slaughter Performance and Meat Quality of Pigs显示文摘[Objectives]Protein energy ratio refers to the proportional relationship between protein and energy levels in animal diets,i.e.,the grams of crude protein corresponding to every megacalorie of energy,which is generally expressed as CP:ME or CP:DE.This study was conducted to investigate the effects of different diets on fattening and slaughter performance and meat quality traits for'L(Large Yorkshire)×L(Landrace)'crossbred pigs.[Methods]Eighteen piglets of L×L crossbred with similar body weights about 51 kg were selected.The piglets were divided into 3 groups randomly and each group was assigned to 3 replicates with 2 piglets in a replicate.Group A was fed diet Ⅰ (control diet),group B was fed the same diet of group A in the first month of the trial but fed diet Ⅱ in second month,and group C consumed diet Ⅲ.All the pigs were fed in the same feeding condition for two months except the different diets during the trial.One pig in similar body weight from each replicate was slaughtered for the determination of slaughter performance and meat quality traits in the end of the trial.[Results]The average daily feed intake(ADFI)of pigs from group B was only(2.32±0.52)kg and significantly lower than those from group A and group C(P<0.05),but no difference was found in average daily gain(ADG) and feed/gain(P>0.05).Also,no differences occurred in the carcass length,back fat thickness,longissimus muscle (LM) area and dressing percentage(P>0.05).In addition,no differences were found in the meat quality traits of shear force,meat color,pH_(45 min),pH_(24 h) and cooking loss(P>0.05).However,the water-holding capacity of meat from group C was(2.58±0.02)ms and significantly lower than that of(2.80±0.20)ms from group A(P<0.05).Although the contents of glutamic acid and cystine in LM from group B was significantly lower than those from group A and group C(P<0.05),no differences occurred in the contents of other amino acids,the total amino acid and total flavor amino acid among the three groups(P>0.05).However,the inosine monophosphate content of LM from group C was only(331.80±11.53)mg/100 g and significantly lower than those of(361.00±6.36)and(366.37±4.80)mg/100 g from group A and B(P<0.05).Even though no differences were found in the contents of DM and CP in LM among the three groups of pigs,the content of intramuscular fat(IMF)in LM from group B and group C was increased by 45.6%and 46.58%respectively from that of group A(P<0.05),but no difference occurred between group B and C(P>0.05).[Conclusions]DietsⅡ and Ⅲ in this study caused no differences in fattening and slaughter performance of L×L crossbred pigs,but the effects on some meat traits were still significant,especially on the improvement of intramuscular fat in experimental pigs.Therefore,they could improve the meat quality of crossbred pigs to a certain extent. | Cui YANG Rongsheng JIANG Yongshao WU Xiying YANG Haihong LIAO Qian QIN Shishan QIN Ying ZHANG Jiahuang YANG Limei QIN Fulin WANG | 2023 | Agricultural Biotechnology2023,12,2: | 0 |
| 5 | Manufacturing N,O-carboxymethyl chitosan-reduced graphene oxide under freeze-dying for performance improvement of Li-S battery显示文摘Lithium-sulfur(Li-S) batteries can provide far higher energy density than currently commercialized lithium ion batteries, but challenges remain before it they are used in practice.One of the challenges is the shuttle effect that originates from soluble intermediates, like lithium polysulfides. To address this issue, we report a novel laminar composite, N,O-carboxymethyl chitosan-reduced graphene oxide(CC-rGO), which is manufactured via the self-assembly of CC onto GO and subsequent reduction of GO under an extreme condition of 1 Pa and-50°C. The synthesized laminar CC-rGO composite is mixed with acetylene black(AB) and coated on a commercial polypropylene(PP) membrane, resulting in a separator(CC-rGO/AB/PP) that can not only completely suppress the polysulfides penetration, but also can accelerate the lithium ion transportation, providing a Li-S battery with excellent cyclic stability and rate capability. | 7 | microR-1294-5p inhibits glycolytic metabolism of non-small cell lung cancer cells via targeting TMPRSS11B显示文摘Non-smallcell lung cancer(NSCLC)cells intake and consume glucose at high efficiency by aerobic glycolysis to maintain robust cell growth and resist cell death.MicroRNAs(miRNAs)have been known to play pivotal roles in NSCLC development partly through mediating glycolysis.However,only a few miRNAs have been experimentally confirmed as critical regulators of glycolysis in NSCLC.TCGA datasets were analyzed to screen for differentially expressed miRNAs between NSCLC and normal tissues.The function of miR-1294-5p was determined in NSCLC cells by cell proliferation,glucose uptake,lactate release,and Extracellular Acidification Rate(ECAR)assays.The target of miR-1294-5p was predicted by TargetScan and miRDB,which was further validated by flow cytometry analysis,RT-qPCR,western blotting,a dual-luciferase reporter assay,and RNA immunoprecipitation(RIP)assay.In the present study,it was found that miR-1294-5p was a significantly downregulated miRNA in lung adenocarcinoma(LUAD)and lung squamous cell carcinoma(LUSC).The overexpression of miR-1294-5p inhibited glycolysis,lactate export,ECAR,and cell proliferation in NSCLC cells.Analysis with bioinformatic tools,Western Blotting,RT-qPCR,flow cytometry analysis,dual-luciferase reporter assay,and RIP assay showed that miR-1294-5p directly bound to complementary sites in the 3’-Untranslated Region(UTR)of TMPRSS11B resulted in downregulation of TMPRSS11B expression.In addition,transfection of recombinant TMPRSS11B rescued the functions of miR-1294-5p on glycolysis and proliferation of NSCLC cells.The findings provided novel insights for understanding the regulation of glycolytic metabolism in NSCLC. | JI ZHU XIYING BO GENGXI JIANG SHIHUA YAO TIEJUN ZHAO LING CHEN | 2021 | BIOCELL2021,45,3: | 0 |
| 8 | Identification and characterization of stem cells in mammalian esophageal stratified squamous epithelia显示文摘Somatic stem cells are essential for the maintenance of tissue homeostasis. Despite its importance, how the esophageal stratified squamous epithelium executes its self-renewal and maintenance remains elusive. In this study, using 5-bromo-2′-deoxyuridine label-chase in rats in vivo and rat esophageal organoids in vitro together with genome-wide DNA methylation and single-cell RNA sequencing, we identified a slow-cycling/quiescent stem cell population that contained high levels of hemidesmosomes (HDs) and low levels of Wnt signaling localized spatially and randomly at the basal layer of the esophageal epithelium. Pseudotime cell trajectory analysis indicated that tissue cells originated from quiescent basal stem cells in the basal layer. Perturbations of HD component expression and/or Wnt signaling reduced the stem cell population in the basal layer of esophageal keratinocyte organoids, resulting in alterations in the organoid formation rate, size, morphogenesis, and proliferation–differentiation homeostasis. Furthermore, not only high levels of HDs and low levels of Wnt signaling but also an interplay between HD and Wnt signaling defined the stem cells of the basal layer.
