1Notch1 and Notch4 are markers for poor prognosis of hepatocellular carcinoma显示文摘BACKGROUND: Notch signaling is critical to physiologic angiogenesis and has been implicated in tumor angiogenesis and metastasis. Notch signaling was reported to exert either oncogenic or tumor-suppressive function in hepatocellular carcinoma (HCC) tumorigenesis. However, the prognostic significance of Notch receptors in HCC remains uncertain. In this study, we investigated the roles of Notch receptors in the prognosis of HCC. METHODS: We investigated the expressions of Notch receptors in tumor tissue microarrays of 288 patients with primary HCC who had undergone curative resection using immunohistochemistry. Additionally, prognostic factors of HCC were examined by univariate and multivariate analyses. The median follow-up period was 97.1 months. Tumor recurrence was detected in 189 patients (65.6%), and 99 (34.4%) died of HCC. RESULTS: Cytoplasmic expression of Notch1, cytoplasmic expression of Notch3, coexistent nuclear expression of Notch3, and cytoplasmic Notch4 overexpression were observed in 145 (50.3%), 60 (20.8%), 17 (5.9%), and 172 (59.7%) of the 288 HCCs, respectively. Multivariate analyses revealed that Notch1 expression (P=0.029), Edmondson grade Ⅲ (P=0.038), and higher BCLC stage (P<0.001) were independent predictors of shorter disease-free survival. Cytoplasmic Notch3 expression tended to be an independent predictor of shorter disease-free survival (P=0.055). Notch1 expression (P=0.039), Notch4 overexpression (P=0.012), and higher BCLC stage (P<0.001) were independent predictors of shorter disease-specific survival. On univariate analysis, Notch1 expression tended to show an unfavorable influence on disease-specific survival (P=0.063) and Notch4 overexpression did not show an unfavorable influence on disease-specific survival (P=0.103). CONCLUSIONS: Notch1 expression might be an independent predictor of both shorter disease-free survival and shorter disease-specific survival in HCC patients after curative resection. Notch4 overexpression might be an independent predictor of shorter disease-specific survival. Notch1 could be used as an immunohistochemical biomarker to detect patients with a high-risk of recurrence. 3Scalp electroacupuncture at the Baihui acupoint (DU 20) improves functional recovery in rats with cerebral ischemia Association with increased expression of vascular endothelial growth factors显示文摘In this study, we induced cerebral infarction in rats by occluding the right middle cerebral artery, and tested the effects of electroacupuncture at the Baihui acupoint (DU 20). Motor and sensory function was tested using Garcia’s scale and motor weakness grading, and the expression of vascular endothelial growth factor in the brain was quantified using immunoblotting and immunohistochemistry. We found that scalp electroacupuncture at DU 20 significantly improved motor performance and sensory function in rats with stroke, and this was accompanied by an increased expression of vascular endothelial growth factor in the ischemic brain tissue and peri-ischemic area. 4Prediction of effluent concentration in a wastewater treatment plant using machine learning models显示文摘Of growing amount of food waste, the integrated food waste and waste water treatment was regarded as one of the efficient modeling method. However, the load of food waste to the conventional waste treatment process might lead to the high concentration of total nitrogen(T-N) impact on the effluent water quality. The objective of this study is to establish two machine learning models—artificial neural networks(ANNs) and support vector machines(SVMs), in order to predict 1-day interval T-N concentration of effluent from a wastewater treatment plant in Ulsan, Korea. Daily water quality data and meteorological data were used and the performance of both models was evaluated in terms of the coefficient of determination(R^2), Nash–Sutcliff efficiency(NSE), relative efficiency criteria(d rel). Additionally, Latin-Hypercube one-factor-at-a-time(LH-OAT) and a pattern search algorithm were applied to sensitivity analysis and model parameter optimization, respectively. Results showed that both models could be effectively applied to the 1-day interval prediction of T-N concentration of effluent. SVM model showed a higher prediction accuracy in the training stage and similar result in the validation stage.However, the sensitivity analysis demonstrated that the ANN model was a superior model for 1-day interval T-N concentration prediction in terms of the cause-and-effect relationship between T-N concentration and modeling input values to integrated food waste and waste water treatment. 6Automatic Discontinuity Classification of Wind-turbine Blades Using A-scan-based Convolutional Neural Network显示文摘Recent development trends in wind power generation have increased the importance of the safe operation of wind-turbine blades(WTBs). To realize this objective, it is essential to inspect WTBs for any defects before they are placed into operation. However, conventional methods of fault inspection in WTBs can be rather difficult to implement, since complex curvatures that characterize the WTB structures must ensure accurate and reliable inspection. Moreover, it is considered useful if inspection results can be objectively and consistently classified and analyzed by an automated system and not by the subjective judgment of an inspector. To address this concern,the construction of a pressure-and shape-adaptive phased-array ultrasonic testing platform, which is controlled by a nanoengine operation system to inspect WTBs for internal defects, has been presented in this paper. An automatic classifier has been designed to detect discontinuities in WTBs by using an A-scanimaging-based convolutional neural network(CNN). The proposed CNN classifier design demonstrates a classification accuracy of nearly 99%. 8A simple, flexible and high-throughput cloning system for plant genome editing via CRISPR-Cas system显示文摘CRISPR-Cas9 system is now widely used to edit a target genome in animals and plants. Cas9 protein derived from Streptococcus pyogenes(Sp Cas9) cleaves double-stranded DNA targeted by a chimeric single-guide RNA(sg RNA). For plant genome editing, Agrobacterium-mediated T-DNA transformation has been broadly used to express Cas9 proteins and sg RNAs under the control of Ca MV 35 S and U6/U3 promoter, respectively. We here developed a simple and high-throughput binary vector system to clone a 19 20 bp of sg RNA, which binds to the reverse complement of a target locus, in a large T-DNA binary vector containing an Sp Cas9 expressing cassette. Twostep cloning procedures:(1) annealing two target-specific oligonucleotides with overhangs specific to the Aar I restriction enzyme site of the binary vector; and(2) ligating the annealed oligonucleotides into the two Aar I sites of the vector, facilitate the high-throughput production of the positive clones. In addition, Cas9-coding sequence and U6/U3 promoter can be easily exchanged via the GatewayTMsystem and unique Eco RI/Xho I sites on the vector, respectively. We examined the mutation ratio and patterns when we transformed these constructs into Arabidopsis thaliana and a wild tobacco, Nicotiana attenuata. 9Nitrogen mass balance in a constructed wetland treating piggery wastewater effluent显示文摘The nitrogen changes and the nitrogen mass balance in a free water surface flow constructed wetland(CW) using the four-year monitoring data from 2008 to 2012 were estimated. The CW was composed of six cells in series that include the first settling basin(Cell 1), aeration pond(Cell 2), deep marsh(Cell 3), shallow marsh(Cell 4), deep marsh(Cell 5) and final settling basin(Cell 6). Analysis revealed that the NH+4-N concentration decreased because of ammonification which was then followed by nitrification. The NO-3-N and NO-2-N were also further reduced by means of microbial activities and plant uptake during photosynthesis. The average nitrogen concentration at the influent was 37,819kg/year and approximately 45% of that amount exited the CW in the effluent. The denitrification amounted to 34% of the net nitrogen input, whereas the accretion of sediment was only 7%. The biomass uptake of plants was able to retain only 1% of total nitrogen load. 10Rheological,textural,and functional characteristics of 3D-printed cheesecake containing guava leaf,green tea,and barley sprout powders显示文摘The use of 3D food printing as an alternative to manufacturing customized food is gaining popularity.In this study,functional powders of guava leaf (GL),green tea (GT),and barley sprouts (BS) were tested as edible ink ingredients for printing cheesecake,and their physicochemical and functional properties were evaluated.Particle size and water and oil holding capacities were highest in GL powder,whereas the water-soluble index was highest in BS.In functional powder-added cheesecake,pseudoplastic and shear-thinning behavior was observed which is ideal for 3D printing.The shear modulus (1.87 kPa) of the control cheesecake (without functional powder) was significantly increased to 5 kPa or higher by adding functional powder.The in vitro glycemic index was lowest in GL cheesecake,whereas antioxidant activity and polyphenol content were highest in GT cheesecake.GL and GT powders would be beneficial as an edible ink to improve functional properties,such as antioxidant activity for GT and blood-glucose-lowering effect for BS,with enhanced printability and textural stability of 3D printed cheesecake.Ye-Eun Park Jeong Kim Hyun Woo Kim Jiyeon Chun 2022Food Bioscience2022,47,3:2

16Ig SF11 regulates osteoclast differentiation through association with the scaffold protein PSD-95显示文摘Osteoclasts are multinucleated, giant cells derived from myeloid progenitors. While receptor activator of NF-κB ligand(RANKL)stimulation is the primary driver of osteoclast differentiation, additional signaling further contributes to osteoclast maturation.Here, we demonstrate that immunoglobulin superfamily member 11(Ig SF11), whose expression increases during osteoclast differentiation, regulates osteoclast differentiation through interaction with postsynaptic density protein 95(PSD-95), a scaffold protein with multiple protein interaction domains. Ig SF11 deficiency in vivo results in impaired osteoclast differentiation and bone resorption but no observed defect in bone formation. Consequently, Ig SF11-deficient mice exhibit increased bone mass.Using in vitro osteoclast culture systems, we show that Ig SF11 functions through homophilic interactions. Additionally, we demonstrate that impaired osteoclast differentiation in Ig SF11-deficient cells is rescued by full-length Ig SF11 and that the Ig SF11-PSD-95 interaction requires the 75 C-terminal amino acids of Ig SF11.
