| 1 | Identification of a novel coronavirus causing severe pneumonia in human:a descriptive study显示文摘Background:Human infections with zoonotic coronaviruses(CoVs),including severe acute respiratory syndrome(SARS)-CoV and Middle East respiratory syndrome(MERS)-CoV,have raised great public health concern globally.Here,we report a novel batorigin CoV causing severe and fatal pneumonia in humans.Methods:We collected clinical data and bronchoalveolar lavage(BAL)specimens from five patients with severe pneumonia from Wuhan Jinyintan Hospital,Hubei province,China.Nucleic acids of the BAL were extracted and subjected to next-generation sequencing.Virus isolation was carried out,and maximum-likelihood phylogenetic trees were constructed.Results:Five patients hospitalized from December 18 to December 29,2019 presented with fever,cough,and dyspnea accompanied by complications of acute respiratory distress syndrome.Chest radiography revealed diffuse opacities and consolidation.One of these patients died.Sequence results revealed the presence of a previously unknownβ-CoV strain in all five patients,with 99.8%to 99.9%nucleotide identities among the isolates.These isolates showed 79.0%nucleotide identity with the sequence of SARS-CoV(GenBank NC_004718)and 51.8%identity with the sequence of MERS-CoV(GenBank NC_019843).The virus is phylogenetically closest to a bat SARS-like CoV(SL-ZC45,GenBank MG772933)with 87.6%to 87.7%nucleotide identity,but is in a separate clade.Moreover,these viruses have a single intact open reading frame gene 8,as a further indicator of bat-origin CoVs.However,the amino acid sequence of the tentative receptor-binding domain resembles that of SARS-CoV,indicating that these viruses might use the same receptor.Conclusion:A novel bat-borne CoV was identified that is associated with severe and fatal respiratory disease in humans. | Li-Li Ren Ye-Ming Wang Zhi-Qiang Wu Zi-Chun Xiang Li Guo Teng Xu Yong-Zhong Jiang Yan Xiong Yong-Jun Li Xing-Wang Li Hui Li Guo-Hui Fan Xiao-Ying Gu Yan Xiao Hong Gao Jiu-Yang Xu Fan Yang Xin-Ming Wang Chao Wu Lan Chen Yi-Wei Liu Bo Liu Jian Yang Xiao-Rui Wang Jie Dong Li Li Chao-Lin Huang Jian-Ping Zhao Yi Hu Zhen-Shun Cheng Un-Lin Liu Zhao-Hui Qian Chuan Qin Qi Jin Bin Cao Jian-Wei Wang | 2020 | Chinese Medical Journal2020,,9: | 99 |
| 2 | A rapid advice guideline for the diagnosis and treatment of 2019 novel coronavirus(2019-nCoV) infected pneumonia(standard version)显示文摘In December 2019, a new type viral pneumonia cases occurred in Wuhan, Hubei Province;and then named '2019 novel coronavirus(2019-nCoV)' by the World Health Organization(WHO) on 12 January 2020. For it is a never been experienced respiratory disease before and with infection ability widely and quickly, it attracted the world’s attention but without treatment and control manual. For the request from frontline clinicians and public health professionals of 2019-nCoV infected pneumonia management, an evidence-based guideline urgently needs to be developed. Therefore, we drafted this guideline according to the rapid advice guidelines methodology and general rules of WHO guideline development;we also added the first-hand management data of Zhongnan Hospital of Wuhan University. This guideline includes the guideline methodology, epidemiological characteristics, disease screening and population prevention, diagnosis, treatment and control(including traditional Chinese Medicine), nosocomial infection prevention and control, and disease nursing of the 2019-nCoV. Moreover, we also provide a whole process of a successful treatment case of the severe 2019-nCoV infected pneumonia and experience and lessons of hospital rescue for 2019-nCoV infections. | 6 | Zircon SHRIMP U-Pb geochronology of the Neoproterozoic Chengjiang Formation in central Yunnan Province(SW China) and its geological significance显示文摘The Chengjiang Formation is the earliest continental clastic deposit after the Jinning Orogeny in central Yunnan Province,and therefore its accurate depositional age is significant for understanding the formation and evolution of the Neoproterozoic rift basins in southern China.However,hampered by accuracy of the dating technique,the existing age data for the Chengjiang Formation are not very reliable.A large number of magmatic zircons were obtained from the tuff interbed in the lower part of the Chengjiang Formation in Jinyang area and the bottom part of the Chengjiang Formation in Dongchuan area,central Yunnan Province,and high-precision SHRIMP U-Pb dating was carried out on these zircons.The results show that the weighted mean 206 Pb/238 U ages are 797.8±8.2 and 803.1±8.7 Ma respectively.In combination with related geological evidence,it has been demonstrated that the bottom boundary age of the Chengjiang Formation should be 800±5 Ma,whereas the top boundary age could be ca 725 Ma.By synthetically analyzing the latest age data for the relevant strata,it has been confirmed that the Chengjiang Formation should be correlated with the Suxiong Formation and the Kaijianqiao Formation in western Sichuan Province,the Liantuo Formation in the middle and lower Yangtze,the Hongchicun Formation and the Shangshu Formation in northern Zhejiang Province,the Puling Formation in southern Anhui Province,the Luokedong Formation and the Majianqiao Formation in northwestern Jiangxi Province,the Wuqiangxi Formation in northwestern Hunan Province,the Fanzhao Formation and the Qingshuijiang Formation in southeastern Guizhou Province,and the Sanmenjie Formation and the Gongdong Formation in northern Guangxi Province.Sedimentary cycle analysis shows that the sedimentary filling sequence of the Neoproterozoic rift basins in southern China can be divided into four cycles.Among them,Cycle II began at ca.800 Ma,accompanied by intensive tectonic-thermal events.The zircon U-Pb ages from the bottom of the Chengjiang Formation reported in this paper indicate that the Neoproterozoic Kangdian rift subbasin probably started to develop at ca.800 Ma and therefore missed Cycle I of the Neoproterozoic sedimentary filling sequence in southern China. | JIANG XinSheng WANG Jian CUI XiaoZhuang ZHUO JieWen XIONG GuoQing LU JunZe LIU JianHui | 2012 | Science China Earth Sciences2012,55,11: | 25 |

| 8 | RNA-Seq analysis of yak ovary: improving yak gene structure information and mining reproduction-related genes显示文摘RNA-Seq,a high-throughput(HT)sequencing technique,has been used effectively in large-scale transcriptomic studies,and is particularly useful for improving gene structure information and mining of new genes.In this study,RNA-Seq HT technology was employed to analyze the transcriptome of yak ovary.After Illumina-Solexa deep sequencing,26826516 clean reads with a total of 4828772880 bp were obtained from the ovary library.Alignment analysis showed that 16992 yak genes mapped to the yak genome and 3734 of these genes were involved in alternative splicing.Gene structure refinement analysis showed that7340 genes that were annotated in the yak genome could be extended at the 5′or 3′ends based on the alignments been the transcripts and the genome sequence.Novel transcript prediction analysis identified 6321 new transcripts with lengths ranging from 180 to 14884 bp,and 2267 of them were predicted to code proteins.BLAST analysis of the new transcripts showed that1200?4933 mapped to the non-redundant(nr),nucleotide(nt)and/or SwissProt sequence databases.Comparative statistical analysis of the new mapped transcripts showed that the majority of them were similar to genes in Bos taurus(41.4%),Bos grunniens mutus(33.0%),Ovis aries(6.3%),Homo sapiens(2.8%),Mus musculus(1.6%)and other species.Functional analysis showed that these expressed genes were involved in various Gene Ontology(GO)categories and Kyoto Encyclopedia of Genes and Genomes pathways.GO analysis of the new transcripts found that the largest proportion of them was associated with reproduction.The results of this study will provide a basis for describing the normal transcriptome map of yak ovary and for future studies on yak breeding performance.Moreover,the results confirmed that RNA-Seq HT technology is highly advantageous in improving gene structure information and mining of new genes,as well as in providing valuable data to expand the yak genome information. | LAN DaoLiang XIONG XianRong WEI YanLi XU Tong ZHONG JinCheng ZHI XiangDong WANG Yong LI Jian | 2014 | Science China(Life Sciences)2014,57,9: | 23 |

| 12 | Wnt5a participates in hepatic stellate cell activation observed by gene expression profile and functional assays显示文摘AIM:To identify differentially expressed genes in quiescent and activated hepatic stellate cells(HSCs)and explore their functions.METHODS:HSCs were isolated from the normal Sprague Dawley rats by in suit perfusion of collagenase and pronase and density Nycodenz gradient centrifugation.Total RNA and mRNA of quiescent HSCs,and cultureactivated HSCs were extracted,quantified and reversely transcripted into cDNA.The global gene expression profile was analyzed by microarray with Affymetrix rat genechip.Differentially expressed genes were annotated with Gene Ontology(GO)and analyzed with Kyoto encyclopedia of genes and genomes(KEGG)pathway using the Database for Annotation,Visualization and Integrated Discovery.Microarray data were validated by quantitative real-time polymerase chain reaction(qRTPCR).The function of Wnt5a on human HSCs line LX-2 was assessed with lentivirus-mediated Wnt5a RNAi.The expression of Wnt5a in fibrotic liver of a carbon tetrachloride(CCl4)-induced fibrosis rat model was also analyzed with Western blotting.RESULTS:Of the 28 700 genes represented on this chip,2566 genes displayed at least a 2-fold increase or decrease in expression at a P<0.01 level with a false discovery rate.Of these,1396 genes were upregulated,while 1170 genes were downregulated in culture-activated HSCs.These differentially expressed transcripts were grouped into 545 GO based on biological process GO terms.The most enriched GO terms included response to wounding,wound healing,regulation of cell growth,vasculature development and actin cytoskeleton organization.KEGG pathway analysis revealed that Wnt5a signaling pathway participated in the activation of HSCs.Wnt5a was significantly increased in cultureactivated HSCs as compared with quiescent HSCs.qRTPCR validated the microarray data.Lentivirus-mediated suppression of Wnt5a expression in activated LX-2 resulted in significantly impaired proliferation,downregulated expressions of typeⅠcollagen and transforming growth factor-β1.Wnt5a was upregulated in the fibrotic liver of a CCl4-induced fibrosis rat model.CONCLUSION:Wnt5a is involved in the activation of HSCs,and it may serve as a novel therapeutic target in the treatment of liver fibrosis. | Wu-Jun Xiong Li-Juan Hu Yi-Cheng Jian Li-Jing Wang Ming Jiang Wei Li Yi He | 2012 | World Journal of Gastroenterology2012,18,15: | 17 |

| 14 | Experimental study on hydration damage mechanism of shale from the Longmaxi Formation in southern Sichuan Basin, China显示文摘As a serious problem in drilling operation,wellbore instability restricts efficient development of shale gas.The interaction between the drilling fluid and shale with hydration swelling property would have impact on the generation and propagation mechanism of cracks in shale formation,leading to wellbore instability.In order to investigate the influence of the hydration swelling on the crack propagation,mineral components and physicochemical properties of shale from the Lower Silurian Longmaxi Formation(LF)were investigated by using the XRD analysis,cation exchange capabilities(CEC)analysis,and SEM observation,and we researched the hydration mechanism of LF shale.Results show that quartz and clay mineral are dominated in mineral composition,and illite content averaged 67%in clay mineral.Meanwhile,CEC of the LF shale are 94.4 mmol/kg.The process of water intruding inside shale along microcracks was able to be observed through high power microscope,meanwhile,the hydration swelling stress would concentrate at the crack tip.The microcracks would propagate,bifurcate and connect with each other,with increase of water immersing time,and it would ultimately develop into macro-fracture.Moreover,the macrocracks extend and coalesce along the bedding,resulting in the rock failure into blocks.Hydration swelling is one of the major causes that lead to wellbore instability of the LF shale,and therefore improving sealing capacity and inhibition of drilling fluid system is an effective measure to stabilize a borehole. | Xiangjun Liu Wei Zeng Lixi Liang Jian Xiong | 2016 | Petroleum2016,2,1: | 14 |

Higher antioxidant enzyme activities and lower malonaldehyde (MDA) content in rice anthers were observed in Jin 23B than in Zhenshan 97B under drought stress. Further, a great increase in the content of soluble sugar in rice anthers of Jin 23B was observed across the whole drought exposure, while Zhenshan 97B showed significant decrease in soluble sugar during 9-12 d after drought stress (DADS). Accordingly, a marked decline of pollen fertility and activity, pollen numbers in an anther and pollen numbers on a stigma was observed in Zhenshan 97B, whereas little difference was found in Jin 23B. | 18 | Opening time and filling pattern of the Neoproterozoic Kangdian Rift Basin,western Yangtze Continent,South China显示文摘The Neoproterozoic Kangdian Rift Basin is a continental rift basin in the western Yangtze Continent.Determining its time of opening and subsequent filling pattern is an important aspect of understanding the formation and evolution of the Yangtze Continent,South China.The Luliang Formation is the early filling in the eastern part of this rift basin,and its deposition age and filling sequence are significant for studies of the regional stratigraphic correlation,opening time,and filling pattern of this basin.Having been correlated to the upper part of the Mesoproterozoic Kunyang Group or to the Neoproterozoic Chengjiang Formation,the age and regional stratigraphic correlation of the Luliang Formation have long been uncertain.This is due to a lack of reliable absolute age constraints.To address this issue,the first zircon SHRIMP U-Pb geochronology has been established for this formation,yielding two high-precision isotopic ages of 818.6±9.2 and 805±14 Ma for the tuff layers at the bottoms of the lower and upper members of the Luliang Formation,respectively.Given the error factor,the bottom age of the lower member of the Luliang Formation can be interpreted as ca.820 Ma,corresponding to the bottom age of the lower part of the Banxi Group,which is the early filling of the Neoproterozoic Xianggui Rift Basin,a representative basin of the Neoproterozoic rift basins in the Yangtze Continent,South China.The bottom age of the upper member of the Luliang Formation can be interpreted as ca.800±5 Ma,corresponding to the bottom age of the Chengjiang Formation in the western part of the Neoproterozoic Kangdian Rift Basin and also corresponding to the bottom age of the upper part of the Banxi Group in the Neoproterozoic Xianggui Rift Basin.These ages indicate that the Neoproterozoic Kangdian Rift Basin shares the same opening time and filling sequences as those of the other Neoproterozoic rift basins in South China.Basin analysis shows that the Neoproterozoic Kangdian Rift Basin is a typical half-graben basin,with its main boundary fault in the west and basin center in the east.This basin consisted of mini unidirectional half-graben basins in its juvenile stage and simplified to become a large united half-graben basin in its mature stage,i.e.,a supradetachment basin. | ZHUO JieWen JIANG XinSheng WANG Jian CUI XiaoZhuang XIONG GuoQing LU JunZe LIU JianHui MA MingZhu | 2013 | Science China Earth Sciences2013,56,10: | 12 |
| 19 | Effects of Film-Degraded Rice Controlled Release Fertilizer on the Nitrogen Uptake and Yield of the Hybrid Rice显示文摘Field experiment was carried out to nitrogen release characteristics of rice controlled release fertilizer (RCRF) coated with natural and half natural high molecular materials, and to thereof effects on the nitrogen uptake and yield of early and late hybrid rice from 1999 to 2000, with urea as control.In experimental fields regardless of early and late seasons, the amount of N in the plants increased exponentially at the early growth stage (y=abx) and nonlinearly at the middle and late growth stages (y=a+bx+cx2).Rice controlled release fertilizer lengthened the exponential phase and increased the parameter -b/c during the nonlinear phase. Although the N amount of plants was less in rice controlled release fertilizer plots than in urea plots within 20 days after transplanting, at the heading and maturity stages, the N amount of plants in rice controlled release fertilizer plots was above two times greater than in the urea plots due to the higher N recovery, and the high absorption density of N in the nonlinear phase. Tillers pattern and achievement of the maximum tillering stage varied with fertilizer types. The maximum tillering stage in the urea plots occurred a week earlier than in the rice controlled release fertilizer plots. Yield sink size and potential sink size of the plants reflected the N absorption pattern and the amount of N in the plants. | 20 | DL-3-n-butylphthalide ameliorates diabetes-associated cognitive decline by enhancing PI3K/Akt signaling and suppressing oxidative stress显示文摘DL3-n-Butylphthalide(DLNBP),a small molecular compound extracted from the seeds of Ap/um graveo/ens Linn(Chinese celery),has been shown to exert neuroprotective effects due to its anti-inflammatory,anti-oxidative and anti-apoptotic activities.DL-NBP not only protects against ischemic cerebral injury,but also ameliorates vascular cognitive impairment in dementia patients including AD and PD.In the current study,we investigated whether and how DL-NBP exerted a neuroprotective effect against diabetes-associated cognitive decline(DACD)in db/db mice,a model of type-2 diabetes,db/db mice were orally administered DL-NBP(20,60,120 mg·kg^(-1)·d^(-1))for 8 weeks.Then the mice were subjected to behavioral test,their brain tissue was collected for morphological and biochemical analyses.We showed that oral administration of DL-NBP significantly ameliorated the cognitive decline with improved learning and memory function in Morris water maze testing.Furthermore,DL-NBP administration attenuated diabetes-induced morphological alterations and increased neuronal survival and restored the levels of synaptic protein PSD95,synaptophysin and synapsin-1 as well as dendritic density in the hippocampus,especially at a dose of 60 mg/kg.Moreover,we revealed that DL-NBP administration suppressed oxidative stress by upregulating Nrf2/HO-1 signaling,and increased brain-derived neurotrophic factor(BDNF)expression by activating PI3K/Akt/CREB signaling in the hippocampus.These beneficial effects of DL-NBP were observed in high glucose-treated PC12 cells.Our results suggest that DL-NBP may be a potential pharmacologic agent for the treatment of DACD. | Bei-ni Wang Cheng-biao Wu Zi-miao Chen Pei-pei Zheng Ya-qian Liu Jun Xiong Jing-yu Xu Pei-feng Li Abdullah Al Mamun Li-bing Ye Zhi-long Zheng Yan-qing Wu Jian Xiao Jian Wang | 2021 | Acta Pharmacologica Sinica2021,42,3: | 11 |
