| 1 | Immunity-and-matrix-regulatory cells derived from human embryonic stem cells safely and effectively treat mouse lung injury and fibrosis显示文摘Lung injury and fibrosis represent the most significant outcomes of severe and acute lung disorders,including COVID-19.However,there are still no effective drugs to treat lung injury and fibrosis.In this study,we report the generation of clinical-grade human embryonic stem cells(hESCs)-derived immunity-and matrix-regulatory cells(IMRCs)produced under good manufacturing practice requirements,that can treat lung injury and fibrosis in vivo.We generate IMRCs by sequentially differentiating hESCs with serum-free reagents.IMRCs possess a unique gene expression profile distinct from that of umbilical cord mesenchymal stem cells(UCMSCs),such as higher expression levels of proliferative,immunomodulatory and anti-fibrotic genes.Moreover,intravenous delivery of IMRCs inhibits both pulmonary inflammation and fibrosis in mouse models of lung injury,and significantly improves the survival rate of the recipient mice in a dose-dependent manner,likely through paracrine regulatory mechanisms.IMRCs are superior to both primary UCMSCs and the FDA-approved drug pirfenidone,with an excellent efficacy and safety profile in mice and monkeys.In light of public health crises involving pneumonia,acute lung injury and acute respiratory distress syndrome,our findings suggest that IMRCs are ready for clinical trials on lung disorders. | Jun Wu Dingyun Song Zhongwen Li Baojie Guo Yani Xiao Wenjing Liu Lingmin Liang Chunjing Feng Tingting Gao Yanxia Chen Ying Li Zai Wang Jianyan Wen Shengnan Yang Peipei Liu Lei Wang Yukai Wang Liang Peng Glyn Nigel Stacey Zheng Hu Guihai Feng Wei Li Yan Huo Ronghua Jin Ng Shyh-Chang Qi Zhou Liu Wang Baoyang Hu Huaping Dai Jie Hao | 2020 | Cell Research2020,30,9: | 7 |

| 3 | H.sinensis mycelium inhibits epithelial–mesenchymal transition by inactivating the midkine pathway in pulmonary fibrosis显示文摘The medical fungus Hirsutella sinensis has been used as a Chinese folk health supplement because of its immunomodulatory properties.Our previous studies established the antifibrotic action of Hirsutella sinensis mycelium(HSM)in the lung.The epithelial–mesenchymal transition(EMT)is involved in the pathogenesis of idiopathic pulmonary fibrosis.The present study investigates the role of HSM in mediating EMT during the development of pulmonary fibrosis.HSM significantly inhibits bleomycin(BLM)-induced pulmonary fibrosis by blocking the EMT.In addition,the expression levels of midkine are increased in the lungs of the BLM-induced group.Further analysis of the results indicates that the mRNA level of midkine correlated positively with EMT.HSM markedly abrogates the transforming growth factorβ-induced EMT-like phenotype and behavior in vitro.The activation of midkine related signaling pathway is ameliorated following HSM treatment,whereas this extract also caused an effective attenuation of the induction of EMT(caused by midkine overexpression)in vitro.Results further confirm that oral medication of HSM disrupted the midkine pathway in vivo.Overall,findings suggest that the midkine pathway and the regulation of the EMT may be considered novel candidate therapeutic targets for the antifibrotic effects caused by HSM. | Li Lu Haiyan Zhu Hailin Wang Huaping Liang Yayi Hou Huan Dou | 2021 | Frontiers of Medicine2021,15,2: | 3 |
