1Identification of genes associated with cotyledon senescence in upland cotton显示文摘In order to unravel the biochemical pathways and understand the molecular mechanisms involved in leaf senescence, suppression subtractive hybridization (SSH) was used to generate a cDNA library enriched for transcripts differentially expressed in developmental senescence cotyledons of upland cotton. After differential screening by membrane- based hybridization and subsequent confirmation by reverse Northern blot analysis, selected 678 clones were sequenced and analyzed. Sequencing of these cDNA fragments reveals that 216 of expressed se- quence tags (ESTs) represented unique genes. Of these 216 cDNAs, 151 clones (69.9%) show signifi- cant homologies to previously known genes, while the remaining 65 do not match any known sequences. 151 unique ESTs are assigned to twelve different categories based on their putative functions gener- ated by BLAST analysis. These SAG-encoded pro- teins are likely to participate in macromolecule deg- radation, nutrient recycling, detoxification of oxidative metabolites, and signaling and regulatory events. The expression pattern of selection of genes was confirmed using northern hybridization. Northern hybridization confirmed several distinct patterns, from expression at a very early stage to the terminal phase of the senescence syndrome. 2Generation of transgenic wheat resistant to wheat yellow mosaic virus and identification of gene silence induced by virus infection显示文摘The plasmid containing the promoter Actl, the coat protein (cp) gene of wheat yellow mosaic virus (WYMV) and the selectable bar gene, was delivered via particle bombardment, directly into immature embryos of a wheat culti-vars. PCR and PCR-RFLP were employed to screen the existence of the cp gene in T0 and T1 generations. Seeds from the positive T1 plants were sowed in fields heavily contaminated with WYMV to detect their resistance. In field trial of virus infection, one of the transgenic wheat lines, P8-T2, exhibited highly disease-resistance. Western blot and RT-PCR analysis showed that the expression level of cp gene in the resistant transgenic line was reduced greatly compared to those susceptible to WYMV infection. 7Karyotype analysis and physical mapping of 45S rDNA in eight species of Sophora, Robinia, and Amorpha显示文摘The karyotype analysis and physical locations of 45S rDNA were carried out by means of fluorescence in situ hybridization in three species,and two forms of Sophora,two species of Robina,and one species of Amorpha.S.japonica L.,S.japonica L.f.oligophylla Franch.,S.japonica L.f.pendula Loud.,and S.xanthantha C.Y.Ma.are all tetraploids with 2n=28.There were four 45S rDNA sites in pericentromeric regions of two pairs of chromosomes in each of them.S.rubriflora Tsoong.is a triploid with 2n=21,and three sites were located in each satellite of group 5 chromosomes.In R.pseudoacacia L.(2n=2x=22),we examined four intensive signals in te-lomeric regions of two pairs of satellite chromosomes.In R.hispida L.(2n=2x=30),there were four other signals in centromeric regions besides those like in R.pseudoacacia.Amorpha fruticosa L.has most chromosomes(2n=40)among the eight materials,however,there were only six 45S rDNA loci and they laid in centromeric regions,and satellites of three pairs of chromosomes.45S rDNA is a valuable chromosomal landmark in karyotype analysis.The distribution and genomic organization of rDNA in the three genera were also discussed.LIU Bo CHEN Chengbin LI Xiulan QI Liwang HAN Suying 2006Frontiers in Biology2006,1,3:0
8Research on the mechanical fault diagnosis method based on sound signal and IEMD-DDCNN显示文摘Purpose–The purpose of this paper is to provide a shorter time cost,high-accuracy fault diagnosis method for water pumps.Water pumps are widely used in industrial equipment and their fault diagnosis is gaining increasing attention.Considering the time-consuming empirical mode decomposition(EMD)method and the more efficient classification provided by the convolutional neural network(CNN)method,a novel classification method based on incomplete empirical mode decomposition(IEMD)and dual-input dual-channel convolutional neural network(DDCNN)composite data is proposed and applied to the fault diagnosis of water pumps.Design/methodology/approach–This paper proposes a data preprocessing method using IEMD combined with mel-frequency cepstrum coefficient(MFCC)and a neural network model of DDCNN.First,the sound signal is decomposed by IEMD to get numerous intrinsic mode functions(IMFs)and a residual(RES).Several IMFs and one RES are then extracted by MFCC features.Ultimately,the obtained features are split into two channels(IMFs one channel;RES one channel)and input into DDCNN.Findings–The Sound Dataset for Malfunctioning Industrial Machine Investigation and Inspection(MIMII dataset)is used to verify the practicability of the method.Experimental results show that decomposition into an IMF is optimal when taking into account the real-time and accuracy of the diagnosis.Compared with EMD,51.52% of data preprocessing time,67.25% of network training time and 63.7%of test time are saved and also improve accuracy.Research limitations/implications–This method can achieve higher accuracy in fault diagnosis with a shorter time cost.Therefore,the fault diagnosis of equipment based on the sound signal in the factory has certain feasibility and research importance.Originality/value–This method provides a feasible method for mechanical fault diagnosis based on sound signals in industrial applications.Haoning Pu Zhan Wen Xiulan Sun Lemei Han Yanhe Na Hantao Liu Wenzao Li 2023International Journal of Intelligent Computing and Cybernetics2023,16,3:0
9High-throughput fluorescent screening of thioredoxin reductase inhibitors to inhibit Mycobacterium tuberculosis显示文摘Tuberculosis(TB)is a chronic infectious disease,which is caused by the pathogen Mycobacterium tuberculosis(Mtb)and reemerged as a global health risk with a significant proportion of multi-drug resistant and extensively drug resistant TB cases.It is very urgent to find some novel high-confidence drug targets in Mtb for discovering the effective anti-TB agents.Thioredoxin reductase(TrxR)has been identified to be a highly viable target for anti-TB drugs for its important role in protecting the pathogen from thiol-specific oxidizing stress,regulating intracellular dithiol/disulfide homeostasis and DNA replication and repair.In the present work,a near-infrared(NIR)fluorescent probe DDAT was developed for the detection of TrxR activity and used to high-throughput screen the TrxR inhibitors from natural products.Two screened TrxR inhibitors from Sappan Lignum and microbial metabolites that were further used to inhibit Mycobacterium tuberculosis.All the results indicate that DDAT is a practical fluorescent molecular tool for the discovery of potential anti-TB drugs.Fei Yan Xin Zhao Ruibo Li Xiuyan Han Qiulong Yan Lei Feng Xiulan Xin Jingnan Cui Xiaochi Ma 2024Chinese Chemical Letters2024,35,3:0
