| 1 | Determining the critical size of a rabbit rib segmental bone defect model显示文摘In order to establish and standardize the rabbit rib segmental bone defect model,it is of vital importance to determine rabbit rib critical size defect(CSD).According to the general time needed for spontaneous long-bone regeneration,three-month observation period was set to determine the CSD.The rabbit rib segmental bone defects with different sizes from 1 to 5cm with or without periosteum were performed in the eighth rib of 4-month-old male New Zealand rabbits and underwent Xray examinations at the 4th,8th and 12th weeks postoperatively.The gross and histological examinations at postoperative week 12 were evaluated,which showed that the critical sizes in the rabbit rib models with and without periosteum were 5 and 2 cm,respectively.This study provides prerequisite data for establishing rabbit rib CSD model and evaluating bonematerials using this model. | Fengzhen Liu Kun Chen Lei Hou Keyi Li Dawei Wang Bin Zhang Xiumei Wang | 2016 | Regenerative Biomaterials2016,3,5: | 2 |
| 2 | Multiscale ApEn and SampEn in Quantifying Nonlinear Complexity of Depressed MEG显示文摘Depression is a neurophysiological disorder with recurrent dysregulations of self-mental states.Multiscale Approximate entropy(ApEn)and Sample entropy(SampEn)are employed to characterize nonlinear complexity of Magnetoencephalography(MEG)of depressive patients in our contribution.SampEn shares similarities with ApEn while has better distinctions between the MEGs of depression patients and normal people.Test results prove that nonlinear complexity of the depressive MEG is lower than that of the normal subjects,indicating weaker response of depression patients to emotional stimuli,and the optimum discriminations between the depressive and healthy people lie in frontal lobe of brain which is related to emotional regulation.Our findings provide valuable information about depression,highlight the loss of nonlinear complexity in MEG of depressive patient and can be used as clinical diagnostic aids. | YAO Wenpo HU Hui WANG Jun YAN Wei LI Jin HOU Fengzhen | 2019 | Chinese Journal of Electronics2019,28,4: | 2 |
| 3 | Nanobody-based chimeric antigen receptor T cells designed by CRISPR/Cas9 technology for solid tumor immunotherapy显示文摘Chimeric antigen receptor-based T-cell immunotherapy is a promising strategy for treatment of hematological malignant tumors;however,its efficacy towards solid cancer remains challenging.We therefore focused on developing nanobody-based CAR-T cells that treat the solid tumor.CD105 expression is upregulated on neoangiogenic endothelial and cancer cells.CD105 has been developed as a drug target.Here we show the generation of a CD105-specific nanobody,an anti-human CD105 CAR-T cells,by inserting the sequences for anti-CD105 nanobody-linked standard cassette genes into AAVS1 site using CRISPR/Cas9 technology.Co-culture with CD105+target cells led to the activation of anti-CD105 CAR-T cells that displayed the typically activated cytotoxic T-cell characters,ability to proliferate,the production of pro-inflammatory cytokines,and the specific killing efficacy against CD105+target cells in vitro.The in vivo treatment with anti-CD105 CAR-T cells significantly inhibited the growth of implanted CD105+tumors,reduced tumor weight,and prolonged the survival time of tumor-bearing NOD/SCID mice.Nanobody-based CAR-T cells can therefore function as an antitumor agent in human tumor xenograft models.Our findings determined that the strategy of nanobody-based CAR-T cells engineered by CRISPR/Cas9 system has a certain potential to treat solid tumor through targeting CD105 antigen. | Fengzhen Mo Siliang Duan Xiaobing Jiang Xiaomei Yang Xiaoqiong Hou Wei Shi Cueva Jumbo Juan Carlos Aiqun Liu Shihua Yin Wu Wang Hua Yao Zihang Yu Zhuoran Tang Shenxia Xie Ziqiang Ding Xinyue Zhao Bruce D.Hammock Xiaoling Lu | 2021 | Signal Transduction and Targeted Therapy2021,6,3: | 2 |

| 11 | Integrin α_(v)β_(3)-targeted polydopamine-coated gold nanostars for photothermal ablation therapy of hepatocellular carcinoma显示文摘Photothermal therapy(PTT)has emerged as a promising cancer therapeutic method.In this study,Arg-Gly-Asp(RGD)peptide-conjugated polydopamine-coated gold nanostars(Au@PDA-RGD NPs)were prepared for targeting PTT of hepatocellular carcinoma(HCC).A polydopamine(PDA)shell was coated on the surface of gold nanostars by the oxidative self-polymerization of dopamine(termed as Au@PDA NPs).Au@PDA NPs were further functionalized with polyethylene glycol and RGD peptide to improve biocompatibility as well as selectivity toward the HCC cells.Au@PDARGD NPs showed an intense absorption at 822 nm,which makes them suitable for near-infraredexcited PTT.Our results indicated that the Au@PDA-RGD NPs were effective for the PTT therapy of the α_(v)β_(3) integrin receptor-overexpressed HepG2 cells in vitro.Further antitumor mechanism studies showed that the Au@PDA-RGD NPs-based PTT induced human liver cancer cells death via the mitochondrial-lysosomal and autophagy pathways.In vivo experiments showed that Au@PDARGD NPs had excellent tumor treatment efficiency and negligible side effects.Thus,our study showed that Au@PDA-RGD NPs could offer an excellent nanoplatform for PTT of HCC. | Yang Li Ping Hu Xiali Wang Xu Hou Fengzhen Liu Xiaohong Jiang | 2021 | Regenerative Biomaterials2021,8,5: | 0 |
