| 1 | 42,573 cases of hepatectomy in China: a multicenter retrospective investigation显示文摘Hepatectomy is currently routinely performed in most hospitals in China. China owns the largest population of liver diseases and the biggest number of liver resection cases. A nationwide multicenter retrospective investigation involving 112 hospitals was performed, and focused on liver resection for patients with hepatocellular carcinoma(HCC). 42,573 cases of hepatectomy were enrolled, and 18,275 valid cases of liver resection for HCC patients were selected for statistical analysis. The epidemiology of HCC, distribution of hepatectomy, postoperative complications and prognosis were finally analyzed. In the 18,275 HCC patients,81% had hepatitis B virus infection and 10% had hepatitis C virus infection. 38% of the HCC patients had normal Alphafetoprotein(AFP) level, and other 35% had an AFP level lower than 400 ng mL^(-1). In the study period, 97% of the hepatectomy for HCC were treated with open surgery, and 23.81% had vascular exclusion techniques. The operation time was(191.7±105.6) min,the blood loss was(546.0±562.8) m L, and blood transfusion was(543.0±1,035.2) m L. The median survival for HCC patients was 631 days, with 1-, 3-, and 5-year overall survival of 73.2%, 28.8% and 19.6%, respectively. | 2 | Complement C3 activation regulates the production of tRNA-derived fragments Gly-tRFs and promotes alcohol-induced liver injury and steatosis显示文摘Complement is known to play a role in alcoholic fatty liver disease (AFLD), but the underlying mechanisms are poorly understood, thereby constraining the development of a rational approach for therapeutic intervention in the complement system. C3 deficiency has been shown to impart protective effects against ethanol-induced hepatic steatosis and inflammation. Here we demonstrate a protection effect in wild-type mice by treatment with CR2-Crry, a specific inhibitor of C3 activation. The expression of glycine transfer (t) RNA-derived fragments (Gly-tRFs) is upregulated in ethanol-fed mice and inhibition of Gly-tRFs in vivo decreases chronic ethanol feeding-induced hepatosteatosis without affecting inflammation. The expression of Gly-tRF was downregulated in C3-deficient or CR2-Crry-treated mice, but not in C5-deficient mice;Gly-tRF expression was restored by the C3 activation products C3a or Asp (C3a-des-Arg) via the regulation of CYP2E1. Transcriptome profiling of hepatic tissues showed that Gly-tRF inhibitors upregulate the expression of sirtuin1 (Sirt1) and subsequently affect downstream lipogenesis and β-oxidation pathways. Mechanistically, Gly-tRF interacts with AGO3 to downregulate Sirt1 expression via sequence complementarity in the 3′ UTR. Notably, the expression levels of C3d, CYP2E1 and Gly-tRF are upregulated, whereas Sirt1 is decreased in AFLD patients compared to healthy controls. Collectively, our findings suggest that C3 activation products contribute to hepatosteatosis by regulating the expression of Gly-tRF. | 6 | Cloud-based smartphone-assisted chemiluminescent assay for rapid screening of electroactive bacteria显示文摘Electroactive bacteria(EAB)can transfer electrons to exocellular solid acceptors and have widespread applications in the fields of pollutant degradation,biosynthesis,and hydrogen generation.Traditional EAB screening methods can precisely measure the bacterial extracellular electron transfer ability,but they suffer from time-consuming and labor-intensive procedures.The chemiluminescence technique is effective for the rapid detection of hemoglobin but has not been applied for EAB screening.Herein,we utilized a chemiluminescent assay to identify EAB through a multiheme c-type cytochrome(c-Cyt)triggered chemiluminescent reaction.The multiheme c-Cyts-triggered chemiluminescence was conveniently determined by taking photographs with commercial smartphones and cloud-based computing.The acquired image signal shows a significant and linear relationship with the bacterial concentration.Measurement results of the chemiluminescence intensity triggered by different bacterial species with the proposed method are strongly associated with those of the microplate reading method(Pearson’s r of 0.9795;P<0.01)and dissimilatory Fe(Ⅲ)reduction method(Pearson’s r of 0.9628;P<0.01).Furthermore,this cloud-based and smartphone-assisted chemiluminescent assay is instrument-free and easy to operate,and results can be obtained within 2 min.The findings demonstrate that the proposed method can effectively detect EAB.Therefore,it will be an alternative approach for EAB screening and can have promising applications in microbiology,environmental science,and bioenergy. | WEN JunLin HE DaiGui LUO SongQing ZHOU ShunGui YUAN Yong | 2023 | Science China(Technological Sciences)2023,66,3: | 1 |
| 7 | Effects of D2 receptor antagonist haloperidol on hippocampal neuronal apoptosis in a rat model of temporal epilepsy显示文摘BACKGROUND: Dopamine receptors are divided into D1 and D2 subgroups. It has been reported that D2 receptors resist neural toxicity induced by excitatory amino acids and muscarine, and also alleviate epilepsy attacks following pilocarpine treatment. However, it has not yet been established whether D2 receptors regu- late temporal epilepsy. OBJECTIVE: To observe the effects of the D2 antagonist haloperidol on hippocampal neuronal apoptosis and electrical brain activity in a rat model of kainic acid-induced temporal epilepsy. DESIGN, TIME AND SETTING: Randomized grouping and histopathological study were performed at the Neurology Medicine Institute of Zhujiang Hospital, Southern Medical University from August to December 2004. MATERIALS: Twenty-five adult, male, Sprague Dawley rats were selected for the present study. Kainic acid (Sigma, USA) was injected into the right lateral ventricle to establish models of temporal epilepsy. A PowerLab multiplying channel electrophysiolograph was provided by AD Instruments, Australia. METHODS: The rats were randomly divided into 5 groups (n = 5): control, model, haloperidol hippocam-pus, haloperidol striatum, and haloperidol substantia nigra. Temporal epilepsy was established in all rats except the control group. Haloperidol was slowly injected into the hippocampus, striatum and substantia nigra, respectively, in three different injection groups. Normal saline was injected into the right lateral ventricle of the control rats. MAIN OUTCOME MEASURES: Hippocampal apoptosis was observed on the day 3 of treatment using TUNEL staining. Changes in electroencephalogram at 0, 0.5, 2, 6, and 12 hours following treatment onset were observed using a PowerLab multiplying channel electrophysiolograph. Animal behaviors were classified according to the Racine criteria. RESULTS: Twenty-five rats were included in the final analysis. Seizures did not occur in the control group. In the model group, 10 minutes after kainic acid injection to the lateral cerebral ventricle, epilepsy seizures occurred and reached a peak within one hour. Hippocampal neuronal apoptosis occurred following epilepsy, in particular on day 3. Following haloperidol injection, hippocampal neuronal apoptosis increased, in particular in the haloperidol hippocampus group, and was significantly greater than the model group (P < 0.05). CONCLUSION: Results suggested that D2 receptors inhibited temporal epilepsy. | 8 | Surgical treatment of hepatocellular carcinoma with cirrhotic esophageal varices and hypersplenism: a 184 case report显示文摘In treating hepatocellular carcinoma(HCC)patients with advanced cirrhosis,one of the most difficult problems is concomitant esophageal varices and hypersplenism.Whether these conditions should be treated surgically in association with HCC resection is still in debate.To elucidate whether esophageal devascularization or splenectomy is beneficial when simultaneously performed with liver resection in HCC patients with both varices and hypersplenism,HCC patients(n=184)with esophageal varices and hypersplenism received one of the three treatments:simultaneous liver resection and esophageal devascularization(Group I,n=41);simultaneous liver resection and splenectomy(Group II,n=61);liver resection only(Group III,n=82).The incidences of postoperative complications of the three groups were 31.7%,29.5%and 24.4%,respectively,with no significant difference among them.The 5-year tumor-free survival rates for the group I,group II and group III were 34.1%,36.1%and 37.8%,respectively.Variceal bleeding caused death by only 4.2%in group I,but by 14.3%in group II and 23.2%in group III.The survival rates in the group I and the group II were comparable to those in the group III,however,the recurrences of postoperative fatal variceal bleeding in group I and group II were significantly lower than those in group III.The results suggest that HCC patients with esophageal varices and hypersplenism should undergo hepatic resection plus esophageal devascularization or splenectomy if radical resection of HCC can be expected. | JIANG Bin CHEN Xiaoping HUANG Zhiyong ZHANG Zhiwei HE Songqing WANG Shaofa WU Zaide QIU Fazu | 2007 | Frontiers of Medicine2007,1,1: | 0 |
