1Small changes in the length of diselenide bond-containing linkages exert great influences on the antitumor activity of docetaxel homodimeric prodrug nanoassemblies显示文摘Homodimeric prodrug-based self-assembled nanoparticles,with carrier-free structure and ultrahigh drug loading,is drawing more and more attentions.Homodimeric prodrugs are composed of two drug molecules and a pivotal linkage.The influence of the linkages on the self-assembly,in vivo fate and antitumor activity of homodimeric prodrugs is the focus of research.Herein,three docetaxel(DTX)homodimeric prodrugs are developed using different lengths of diselenide bond-containing linkages.Interestingly,compared with the other two linkages,the longest diselenide bond-containing linkage could facilitate the self-delivery of DTX prodrugs,thus improving the stability,circulation time and tumor targeting of prodrug nanoassemblies.Besides,the extension of linkages reduces the redox-triggered drug release and cytotoxicity of prodrug nanoassemblies in tumor cells.Although the longest diselenide bond-containing prodrug nanoassemblies possessed the lowest cytotoxicity to 4T1 cells,their stable nanostructure maintained intact during circulation and achieve the maximum accumulation of DTX in tumor cells,which finally“turned the table”.Our study illustrates the crucial role of linkages in homodimeric prodrugs,and gives valuable proposal for the development of advanced nano-DDS for cancer treatment.Lingxiao Li Shiyi Zuo Fudan Dong Tian Liu Yanlin Gao Yinxian Yang Xin Wang Jin Sun Bingjun Sun Zhonggui He 2021Asian Journal of Pharmaceutical Sciences2021,16,3:3
2Newly designed identifying method for ice thickness on highvoltage transmission lines via machine vision显示文摘The icing of transmission lines will bring considerable challenges to the safe operation of the power grid.Therefore,a novel method combines machine vision and machine learning algorithms for identifying the ice thickness on high‐voltage transmission line(HVTL)as proposed herein.First,noise and background interference in the image are filtered,and the grey image is used as input.Then,the algorithms of improved Canny edge detection,Hough transform,improved K‐means clustering,and least‐squares fitting are adopted in turn to locate the edges of conductors.Finally,according to the distance mapping model based on monocular vision,the ice thickness of the conductor is determined by calculating the width difference before and after icing.The experimental results show that the proposed method can accurately locate the edge of the conductor in both field and experimental environments.Moreover,it can ensure ideal effects under different illumination and hardly not be affected by distortion in both horizontal and vertical directions.Besides,the distance mapping model can map the pixel distance to the actual distance with high precision,no matter whether the background is simple or complex,and the calculated ice thickness has only a small deviation compared to the actual value.In addition,the proposed method shows high reliability and effectiveness when various interference such as different backgrounds,uneven icing,height difference changes,conductor movement,contrast changes,and conductor sag occur.Bingjun Weng Wei Gao Weicou Zheng Gengjie Yang 2021High Voltage2021,6,5:3
3Validation and modification of the AJCC 8th TNM staging system for pancreatic ductal adenocarcinoma in a Chinese cohort:A nationwide pancreas data center analysis显示文摘Objective:To validate the 8 th edition of the American Joint Committee on Cancer(AJCC)staging system for pancreatic ductal adenocarcinoma(PDAC)in a Chinese cohort of radically resected patients and to develop a refined staging system for PDAC.Methods:Data were collected from the China Pancreas Data Center(CPDC)for patients with resected PDAC in 2016 and 2017,and cancer-specific survival(CSS)was evaluated using the Kaplan-Meier method and log-rank test.Univariate and multivariate analyses based on Cox regression were performed to identify prognostic factors.The recursive partitioning analysis(RPA),Kaplan-Meier method,and log-rank test were performed on the training dataset to generate a proposed modification for the 8 th TNM staging system utilizing the preoperative carbohydrate antigen(CA)19-9 level.Validation was performed for both staging systems in the validation cohort.Results:A total of 1,676 PDAC patients were retrieved,and the median CSS was significantly different between the 8 th TNM groupings,with no significant difference in survival between stage IB and IIA.The analysis of T and N stages demonstrated a better prognostic value in the N category.Multivariate analysis showed that the preoperative serum CA19-9 level was the strongest prognostic indicator among all the independent risk factors.All patients with CA19-9>500 U/mL had similar survival,and we proposed a new staging system by combining IB and IIA and stratifying all patients with high CA19-9 into stage III.The modified staging system had a better performance for predicting CSS than the 8 th AJCC staging scheme.Conclusions:The 8 th AJCC staging system for PDAC is suitable for a Chinese cohort of resected patients,and the N category has a better prognostic value than the T category.Our modified staging system has superior accuracy in predicting survival than the 8 th AJCC TNM staging system.Hao Hu Chang Qu Bingjun Tang Weikang Liu Yongsu Ma Yiran Chen Xuehai Xie Yan Zhuang Hongqiao Gao Xiaodong Tian Yinmo Yang 2021Chinese Journal of Cancer Research2021,33,4:3

8Synergetic lethal energy depletion initiated by cancer cell membrane camouflaged nano-inhibitor for cancer therapy显示文摘Mitochondrial bioenergy plays a vital role in the occurrence and development of cancer.Although strategies to impede mitochondrial energy supply have been rapidly developed,the anticancer efficacy is still far from satisfactory,mainly attributed to the hybrid metabolic pathways of mitochondrial oxidative phosphorylation(OXPHOS)and glycolysis.Herein,we construct a cancer cell membrane camouflaged nano-inhibitor,mTPPa-Sy nanoparticle(NP),which co-encapsulates OXPHOS inhibitor(mitochondrial-targeting photosensitizers:TPPa)and glycolysis inhibitor(syrosingopine(Sy))for synergistically blocking the two different energy pathways.The mTPPa-Sy NPs exhibit precision tumor-targeting due to the high affinity between the biomimic membrane and the homotypic cancer cells.Under laser irradiation,the mitochondrial-targeting TPPa,which is synthesized by conjugating pyropheophorbide a(PPa)with triphenylphosphin,produces excessive reactive oxygen species(ROS)and further disrupts the OXPHOS.Interestingly,OXPHOS inhibition reduces O_(2) consumption and improves ROS production,further constructing a closed-loop OXPHOS inhibition system.Moreover,TPPa-initiated OXPHOS inhibition in combination with the Sytriggered glycolysis inhibition results in lethal energy depletion,significantly suppressing tumor growth even after a single treatment.Our findings highlight the necessity and effectiveness of synergetic lethal energy depletion,providing a prospective strategy for efficient cancer therapy.Fudan Dong Qikun Jiang Lingxiao Li Tian Liu Shiyi Zuo Lin Gao Mengna Fang Yanlin Gao Bingjun Sun Cong Luo Zhonggui He Jin Sun 2022Nano Research2022,15,4:0
9Tumor-suppressive function and mechanism of HOXB13 in right-sided colon cancer显示文摘Right-sided colon cancer(RCC)and left-sided colon cancer(LCC)differ in their clinical and molecular features.An investigation of differentially expressed genes(DEGs)between RCC and LCC could contribute to targeted therapy for colon cancer,especially RCC,which has a poor prognosis.Here,we identified HOXB13,which was significantly less expressed in RCC than in LCC and associated with prognosis in RCC,by using 5 datasets from the Gene Expression Omnibus(GEO).Tissue sample analysis showed that HOXB13 was differentially expressed between normal and only RCC tumor tissues.HOXB13 inhibited colon cancer cell proliferation and induced apoptosis both in vitro and in vivo.Furthermore,we found that HOXB13 might be regulated by DNMT3B and suppress C-myc expression to exert antitumor effects viaβ-catenin/TCF4 signals in RCC.In conclusion,the current study is the first to demonstrate that HOXB13 has a tumor-suppressive effect in RCC.High expression levels of HOXB13 are associated with prolonged overall survival in patients with RCC.The DNMT3B-HOXB13-C-myc signaling axis might be a molecular target for the treatment of RCC.Binbin Xie Bingjun Bai Yuzi Xu Yunlong Liu Yiming Lv Xing Gao Fei Wu Zhipeng Fang Ying Lou Hongming Pan Weidong Han 2019Signal Transduction and Targeted Therapy2019,4,1:0
