2The E3 Ubiquitin Ligase gp78 Protects against ER Stress in Zebrafish Liver显示文摘Enhanced endoplasmic reticulum(ER)-associated protein degradation(ERAD) activity by the unfolded protein response(UPR) represents one of the mechanisms for restoring ER homeostasis.In vitro evidence indicates that the mammalian gp78 protein is an E3 ubiquitin ligase that facilitates ERAD by polyubiquitinating and targeting proteins for proteasomal degradation under both physiologic and stress conditions.However,the in vivo function of gp78 in maintaining ER protein homeostasis remains untested.Here we show that like its mammalian counterpart,the zebrafish gp78 is also an E3 ubiquitin ligase as revealed by in vitro ubiquitination assays.Expression analysis uncovered that gp78 is highly expressed in several organs,including liver and brain,of both larval and adult fish.Treatment of larvae or adult fish with tunicamycin induces ER stress and upregulates the expression of several key components of the gp78 ERAD complex in the liver.Moreover,liver-specific overexpression of the dominant-negative form of gp78(gp78-R2M) renders liver more sensitive to tunicamycin-induced ER stress and enhances the expression of sterol response element binding protein(Srebp)-target genes,which was largely suppressed in fish overexpressing wild-type gp78.Together,these data indicate that gp78 plays a critical role in protecting against ER stress in liver.Zhiliang Chen Petek Ballar Yu Fu Jia Luo Shaojun Du Shengyun Fang 2014Journal of Genetics and Genomics2014,41,7:3
3Achieving ultrahigh electrocatalytic NH_(3) yield rate on Fe-doped Bi_(2)WO_(6) electrocatalyst显示文摘NH_(3),derived from electrocatalytic nitrogen reduction reaction(NRR),is promising to satisfy the need of food production and serve as a carbon-free liquid energy carrier in the near future.Yet major challenges remain in enhancing NH_(3) yield rate and conversion efficiency of available electrocatalysts.This work achieved an ultrahigh electrocatalytic NH_(3)yield rate on the_(0.50)Fe-Bi_(2)W0_(6) catalyst by a facile Fe-doped strategy.Up to 289μg·h^(-1)·mg_(cat)^(-1) of NH_(3) formation rate was obtained at-0.75 V vs.RHE,which was reliably quantized by indophenol blue and ^(1)H NMR methods.The impressive result is an order of magnitude higher than that of the reported Fe-and Bi-based catalysts,even more superior than the result of single atom Ru catalyst.The key of the outstanding NRR behaviors on the_(0.50)Fe-Bi_(2)W0_(6) catalyst is the significant hydrogen evolution reaction(HER)suppression and the synergy between Bi and Fe,which can effectively modulate the electron distribution and accelerate the electron transport.This work endows a new insight to further explore the high-performance electrocatalysts toward NRR.Yongqin Liu Liang Huang Youxing Fang Xinyang Zhu Shaojun Dong 2021Nano Research2021,14,8:3

8Regulation of tumor angiogenesis by the microtubule-binding protein CLIP-170显示文摘Angiogenesis,the expansion of preexisting blood vessels,is a complex process required for tumor growth and metastasis.Although current antiangiogenic strategies have shown promising results in several cancer types,identification of additional antiangiogenic targets is required to improve the therapeutic response.Herein,we show that the microtubule-binding protein CLIP-170(cytoplasmic linker protein of 170 kDa)is highly expressed in breast tumor samples and correlates positively with blood vessel density.Depletion of CLIP-170 signifi cantly impaired vascular endothelial tube formation and sprouting in vitro and inhibited breast tumor growth in mice by decreasing tumor vascularization.Our data further show that CLIP-170 is important for the migration but not the proliferation of vascular endothelial cells.In addition,CLIP-170 promotes the polarization of endothelial cells in response to the angiogenic stimulus.These fi ndings thus demonstrate a critical role for CLIP-170 in tumor angiogenesis and suggest its potential as a novel antiangiogenic target.Xiaodong Sun Fang Li Bin Dong Shaojun Suo Min Liu Dengwen Li Jun Zhou 2013Protein & Cell2013,4,4:1

11Deep eutectic solvent assisted facile synthesis of low-dimensional hierarchical porous high-entropy oxides显示文摘High-entropy-oxides(HEOs),a new class of solids that contain five or more elemental species,have attracted increasing interests owing to their unique structures and fascinating physicochemical properties.However,it is a huge challenge to construct various nanostructured,especially low-dimensional nanostructured HEOs under the high temperature synthetic conditions.Herein,a facile strategy using glucose-urea deep eutectic solvent(DES)as both a solvent and the carbon source of structure-directed template is proposed for the synthesis of various HEOs with two-dimentional(2D)nanonets and one-dimentional(1D)nanowires,including rock-salt(Co,Cu,Mg,Ni,Zn)O,spinel(Co,Cr,Fe,Mn,Ni)_(3)O_(4),and perovskite La(Co,Cr,Fe,Mn,Ni)O_(3).The as-prepared HEOs possessed five or more uniformly dispersed metal elements,large specific surface areas(more than 25 m^(2)·g^(−1)),and a pure single-phase structure.In addition,high cooling rate(cooling in air or liq-N_(2)-quenching)was indispensable to obtain a single-phase rock-salt(Co,Cu,Mg,Ni,Zn)O because of phase separation caused by copper.By taking advantage of unique features of HEOs,rock-salt(Co,Cu,Mg,Ni,Zn)O can function as a promising candidate for lithium-ion batteries(LIBs)anode material,which achieved excellent cycling stability.This work provides a feasible synthetic strategy for low-dimensional hierarchical HEOs,which creates new opportunities for the stable HEOs being highly active functional materials.Jiale Wei Kai Rong Xiaolong Li Yuchen Wang Zhen-An Qiao Youxing Fang Shaojun Dong 2022Nano Research2022,15,3:1

13Novel dual-band antenna for multi-mode GNSS applications显示文摘A novel dual-band antenna is proposed for mitigating the multi-path interference in the global navigation satellite system(GNSS) applications. The radiation patches consist of a shortedannular-ring reduced-surface-wave(SAR-RSW) element and an inverted-shorted-annular-ring reduced-surface-wave(ISAR-RSW)element. One key feature of the design is the proximity-coupled probe feeds to increase impedance bandwidth. The other is the defected ground structure band rejection filters to suppress the interaction effect between the SAR-RSW and the ISAR-RSW elements. In addition, trans-directional couplers are used to obtain tight coupling. Measurement results indicate that the antenna has a larger than 10 d B return loss bandwidth and a less than 3 d B axial-ratio(AR) bandwidth in the range of(1.164 – 1.255) GHz and(1.552 – 1.610) GHz. 17Hierarchical porous nanosheets of Co-Mn bimetallic oxide from deep eutectic solvent for highly efficient peroxymonosulfate activation显示文摘The development of highly active catalysts is the key to the successful application of sulfate radical(SO_(4)^(·-))-based advanced oxidation processes(AOPs)to wastewater treatment.Herein,bimetallic oxide CoMn_(2)O_(4)hierarchical porous nanosheets(CoMn_(2)O_(4)HPNSs)were constructed using glucose/urea deep eutectic solvent(DES)as sustainable solvent and self-formed sacrificial carbon templates.The prepared CoMn_(2)O_(4)HPNS exhibited outstanding peroxymonosulfate(PMS)activation performance,achieving the rapid degradation of refractory organic compounds in wastewater,including 5-sulfosalicylic acid(100%),acetaminophen(100%),oxtetracycline(100%),and sulfamethoxazole(91%)within 20 min.This excellent performance was attributed not only to the synergistic catalytic effect of Co-Mn bimetals,but also to the hierarchical porous structure which exposes more active sites and accelerates the migration of PMS and organic pollutants.In addition,CoMn_(2)O_(4)HPNS also showed excellent reusability and high stability in multiple cycles of degradation.The active species quenching results and electron paramagnetic resonance measurements revealed that SO_(4)^(·-)greatly contributed to organic degradation,while^(1)O_(2)and·OH also involved.Moreover,a series of other transition metal oxides(Co_(3)O_(4),Fe_(2)O_(3),Mn_(3)O_(4),NiO,and CoFe_(2)O_(4))with hierarchical porous nanosheet structures were successfully fabricated with this method.This study provides a simple,general,and low-cost strategy for fabricating hierarchical porous transition metal oxides,which are promising for the environmental remediation or many other applications in the future.Jiale Wei Kai Rong Yuchen Wang Ling Liu Youxing Fang Shaojun Dong 2023Nano Research2023,16,7:0
18Critical evaluation of the glucose oxidase-like activity of gold nanoparticles stabilized by different polymers显示文摘Polymer stabilizers are widely used to synthesize gold nanoparticles(Au NPs)to prevent their aggregation and improve their stability.Although stabilizers are known to greatly influence both the structure and size of Au NPs,limited efforts explore their effects on the activity of Au NPs for biocatalysis.Herein,different polymers are used as stabilizers to synthesize Au NPs.For the glucose oxidase-like activity,we find that without the spatial barrier from stabilizers,naked Au NPs show significantly high catalytic activity as well as the worst stability.Among the polymers,polyacrylic acid-stabilized Au NPs exhibit the highest activity,whose Vmax(0.74μM·s^(−1))is higher than that of the natural glucose oxidase(0.37μM·s^(−1))due to the smallest particle size(<2 nm)and the weak spatial resistance of polyacrylic acid.These variable catalytic results derive from the comprehensive effects including size,steric hindrance,and electronic effect.However,further selectivity and activity tests have exposed shortcomings.They possess universal activities for aldose oxidation,whereas cannot retain activities in typical physiological environments.Our findings highlight the role of polymer stabilizers in imposing effects on the glucose oxidase-like activity of Au NPs and provide a basis for further Au NPs engineering and applications.Lili Xu Jinxing Chen Qian Ma Daiyong Chao Xinyang Zhu Ling Liu Jin Wang Youxing Fang Shaojun Dong 2023Nano Research2023,16,4:0

20Intervention effect of Cigu Xiaozhi prescription(慈菇消脂方)on ceramide lipoapoptosis in non-alcoholic fatty liver disease显示文摘OBJECTIVE:To explore the mechanism of the Chinese medicine Cigu Xiaozhi prescription(慈菇消脂方,CGXZ)in the treatment of the non-alcoholic fatty liver disease(NAFLD)by detoxification and phlegm-reducing,the effect of CGXZ prescription on ceramide-mediated lipid apoptosis in Hep G2 cells with NAFLD.METHODS:The experiment was randomly divided into 6groups:normal control group,model group,CGXZ prescription medicated serum high,medium,and low dose groups,and pioglitazone positive control group.Using 500μmol/L free fatty acid(FFA)mixture to induce Hep G2 cells to establish NAFLD cell model,respectively,with 2%,4%,and 6%concentration of CGXZ prescription medicated serum intervention for 24 h.The changes in organelles and lipid droplet accumulation were observed under the electron microscope.Furthermore,Td T-mediated d UTP Nick-End Labeling method was used to assay hepatocyte apoptosis;Biochemical determination of glutamic-pyruvic transaminase,glutamic oxalacetic transaminase,triglycerides,and FFA levels in Hep G2cells;the content of ceramide was determined by highperformance thin-layer chromatography.Finally,Western Blot and quantitative real-time polymerase chain reaction(q RT-PCR)were used to determine the protein and gene expression levels,such as inducible nitric oxide synthase(i NOS),nuclear factorκB(NF-κB),B cell lymphoma 2(Bcl-2)and Bcl-2-associated X(Bax).RESULTS:Under the electron microscope,the cells in the model group showed moderate-to-severe steatosis,and apoptotic bodies could be seen.The model group had greater improvements in the apoptosis rate(P<0.01),and the levels of ceramide C2 and FFA in the cytoplasm(P<0.01)than the normal control group.The protein expressions of NF-κB,i NOS,and Bax were significantly up-regulated(P<0.05),while the Bcl-2 had no significant change(P>0.05).Compared with the model group,the levels of ceramide C2 and FFA(P<0.01),the protein expressions of NF-κB,i NOS,and Bax(P<0.05)in the CGXZ prescription treatment group and pioglitazone positive control group were significantly decreased;Only the Bcl-2 protein was significantly up-regulated in the high-dose Chinese medicine group(P<0.05).The downregulation of Bax m RNA expression in each Chinese medicine treatment group was significantly better than in the pioglitazone positive control group(P<0.01).CONCLUSIONS:The CGXZ prescription,formulated with the method of detoxification and phlegm,can inhibit lipoapoptosis in the NAFLD cell model by downregulating the levels of ceramide C2 and FFA,which may be achieved by regulating ceramide/i NOS/NF-κB signaling pathway.YANG Shaojun MA Yanhua BAI Zhouxia YU Ye FANG Buwu ZHANG Li WANG Li 2024Journal of Traditional Chinese Medicine2024,44,1:0
