| 1 | The material and biological characteristics of osteoinductive calcium phosphate ceramics显示文摘The discovery of osteoinductivity of calcium phosphate(Ca-P)ceramics has set an enduring paradigm of conferring biological regenerative activity to materials with carefully designed structural characteristics.The unique phase composition and porous structural features of osteoinductive Ca-P ceramics allow it to interact with signaling molecules and extracellular matrices in the host system,creating a local environment conducive to new bone formation.Mounting evidence now indicate that the osteoinductive activity of Ca-P ceramics is linked to their physicochemical and three-dimensional structural properties.Inspired by this conceptual breakthrough,many laboratories have shown that other materials can be also enticed to join the rank of tissue-inducing biomaterials,and besides the bones,other tissues such as cartilage,nerves and blood vessels were also regenerated with the assistance of biomaterials.Here,we give a brief historical recount about the discovery of the osteoinductivity of Ca-P ceramics,summarize the underlying material factors and biological characteristics,and discuss the mechanism of osteoinduction concerning protein adsorption,and the interaction with different types of cells,and the involvement of the vascular and immune systems. | Zhurong Tang Xiangfeng Li Yanfei Tan Hongsong Fan Xingdong Zhang | 2018 | Regenerative Biomaterials2018,5,1: | 20 |
| 2 | A cellular automata evacuation model considering friction and repulsion显示文摘There exist interactions among pedestrians and between pedestrian and environment in evacuation. These interactions include attraction, repulsion and friction that play key roles in human evacuation behaviors, speed and efficiency. Most former evacuation models focus on the attraction force, while repulsion and friction are not well modeled. As a kind of multi-particle self-driven model, the social force model introduced in recent years can represent those three forces but with low simulation efficiency because it is a continuous model with complex rules. Discrete models such as the cellular automata model and the lattice gas model have simple rules and high simulation efficiency, but are not quite suitable for interactions’ simulation. In this paper, a new cellular automata model based on traditional models is introduced in which repulsion and friction are modeled quantitatively. | 3 | Optimizing the CRISPR/Cas9 system for genome editing in grape by using grape promoters显示文摘The efficacy of the CRISPR/Cas9 system in grapevine(Vitis vinifera L.)has been documented,but the optimization of this system,as well as CRISPR/Cas9-mediated multiplex genome editing,has not been explored in this species.Herein,we identified four VvU3 and VvU6 promoters and two ubiquitin(UBQ)promoters in grapevine and demonstrated that the use of the identified VvU3/U6 and UBQ2 promoters could significantly increase the editing efficiency in grape by improving the expression of sgRNA and Cas9,respectively.Furthermore,we conducted multiplex genome editing using the optimized CRISPR/Cas9 vector that contained the conventional multiple sgRNA expression cassettes or the polycistronic tRNA-sgRNA cassette(PTG)by targeting the sugar-related tonoplastic monosaccharide transporter(TMT)family members TMT1 and TMT2,and the overall editing efficiencies were higher than 10%.The simultaneous editing of TMT1 and TMT2 resulted in reduced sugar levels,which indicated the role of these two genes in sugar accumulation in grapes.Moreover,the activities of the VvU3,VvU6,and UBQ2 promoters in tobacco genome editing were demonstrated by editing the phytoene desaturase(PDS)gene in Nicotiana benthamiana leaves.Our study provides materials for the optimization of the CRISPR/Cas9 system.To our knowledge,our simultaneous editing of the grape TMT family genes TMT1 and TMT2 constitutes the first example of multiplex genome editing in grape.The multiplex editing systems described in this manuscript expand the toolbox of grape genome editing,which would facilitate basic research and molecular breeding in grapevine. | Chong Ren Yanfei Liu Yuchen Guo Wei Duan Peige Fan Shaohua Li Zhenchang Liang | 2021 | Horticulture Research2021,8,1: | 6 |
| 4 | Zircon SHRIMP U-Pb Dating of the Tourmalinites from Boron-bearing Series of Borate Deposits in Eastern Liaoning and its Geological Implications显示文摘This paper focused on the zircon sensitive high resolution ion micro-probeU-Pb geochronology of the tourmalinites from boron-bearing series of borate deposits in Eastern Liaoning.The zircons commonly have core-rim structures,most cores show oscillatory zoning in cathodoluminescence and plane polarized light images,suggesting a magmatic detrital origin.Ages of the magmatic detrital zircons from the hyalotourmalite samples(N13) and(N14) are 2175±5Ma and 2171±9Ma,respectively.Moreover,metamorphic zircon from the sample(N13) shows an age of 1906±4Ma.Zircon core and rim from the hyalotourmalite sample(N02) record ages of 2171±6Ma and 1889±62Ma,which are explained as indicating the formation and metamorphic ages.Combined with the geological and geochemical studies,it can be concluded that the tourmalinites are formed during sedimentary exhalative mineralizations in the mid-Paleoproterozoic(~2170Ma) and underwent the metamorphism in the late-Paleoproterozoic(~1900Ma).The tourmalinites are the products of submarine acid volcanism in the extension rifting phase of the Liaoji Paleoproterozoic Rift,the rock-forming materials of which are derived from the mantle sources with recycling crustal contamination.The emergence of tourmalinites not only indicates the mid-Paleoproterozoic tectonic-magmatic processes,but also provides impetus,heat and material sources for the mineralization of borate deposits in Eastern Liaoning. | LIU Jingdang XIAO Rongge ZHANG Yanfei FAN Minghui WANG Shengzhi JIA Yuguo WANG Gang LIU Zhixue | 2012 | Acta Geologica Sinica(English Edition)2012,86,1: | 5 |

| 17 | Temporal and spatial stability of the EM/PM molecular subtypes in adult diffuse glioma显示文摘Detailed characterizations of genomic alterations have not identified subtype-specific vulnerabilities in adult gliomas. Mapping gliomas into developmental programs may uncover new vulnerabilities that are not strictly related to genomic alterations. After identifying conserved gene modules co-expressed with EGFR or PDGFRA (EM or PM), we recently proposed an EM/PM classification scheme for adult gliomas in a histological subtype- and grade-independent manner. By using cohorts of bulk samples, paired primary and recurrent samples, multi-region samples from the same glioma, single-cell RNA-seq samples, and clinical samples, we here demonstrate the temporal and spatial stability of the EM and PM subtypes. The EM and PM subtypes, which progress in a subtype-specific mode, are robustly maintained in paired longitudinal samples. Elevated activities of cell proliferation, genomic instability and microenvironment, rather than subtype switching, mark recurrent gliomas. Within individual gliomas, the EM/PM subtype was preserved across regions and single cells. Malignant cells in the EM and PM gliomas were correlated to neural stem cell and oligodendrocyte progenitor cell compartment, respectively. Thus, while genetic makeup may change during progression and/or within different tumor areas, adult gliomas evolve within a neurodevelopmental framework of the EM and PM molecular subtypes. | 20 | Molecular Cloning and Bioinformatics Analysis of kdpD Gene from Vibrio alginolyticus显示文摘In this study,histidine kinase gene kdpD in the two-component regulatory system of Vibrio alginolyticus strain HY9901 was cloned for bioinformatics analysis. Sequence analysis revealed that kdpD gene( GenB ank accession number: KJ544668) was 1 374 bp in length,encoding a putative protein of 457 amino acids. The predicted molecular weight( MW) of KdpE was 51. 60 kD with a theoretical isoelectric point( pI) of 6. 02. Using SignalP 4. 0,TMHMM Server 2. 0 and SoftB erry-Psite software,it was predicted that KdpE protein was equally located in Golgi apparatus,plasma membrane and endoplasmic reticulum( 33. 3%),which did not contain a signal peptide but contained three transmembrane domains. KdpE protein had five casein kinase II phosphorylation sites,five protein kinase C phosphorylation sites and one cA MP- and cG MP-dependent protein kinase phosphorylation site. A phylogenetic tree was constructed by MEGA 5. 0 software,which revealed that KdpD from V. alginolyticus had close genetic relationship with corresponding proteins from V. campbellii and V. parahaemolyticus.
