| 1 | Role of human papillomavirus in oropharyngeal squamous cell carcinoma: A review显示文摘Human papillomavirus(HPV) has been implicated in the pathogenesis of a subset of oropharyngeal squamous cell carcinoma. As a result, traditional paradigms in relation to the management of head and neck squamous cell carcinoma have been changing. Research into HPVrelated oropharyngeal squamous cell carcinoma is rapidly expanding, however many molecular pathological and clinical aspects of the role of HPV remain uncertain and are the subject of ongoing investigation. A detailed search of the literature pertaining to HPV-related oropharyngeal squamous cell carcinoma was performed and information on the topic was gathered. In this article, we present an extensive review of the current literature on the role of HPV in oropharyngeal squamous cell carcinoma, particularly in relation to epidemiology, risk factors, carcinogenesis, biomarkers and clinicalimplications. HPV has been established as a causative agent in oropharyngeal squamous cell carcinoma and biologically active HPV can act as a prognosticator with better overall survival than HPV-negative tumours. A distinct group of younger patients with limited tobacco and alcohol exposure have emerged as characteristic of this HPV-related subset of squamous cell carcinoma of the head and neck. | 10 | A single dose of caffeic acid phenethyl ester prevents initiation in a medium-term rat hepatocarcinogenesis model显示文摘AIM: To study of the effect of caffeic acid phenethyl ester (CAPE) on the initiation period in a medium-term assay of hepatocarcinogenesis. METHODS: Male Wistar rats were subjected to a carcinogenic treatment (CT) and sacrificed at 25th d; altered hepatic foci (AHF) were generated efficiently. To a second group of rats a single 20 mg/kg doses of CAPE was given 12 h before initiation with CT and were sacrificed at 25th d. We evaluated the expression of preneoplastic markers as γ-glutamyltranspeptidase (GGT) and glutathione S-transferase type pi protein (GSTp) by histochemistry, RT-PCR and Western blot analyses, respectively. We measured thiobarbituric acid reactive substances (TBARS) in homogenates of liver and used Unscheduled DNA Synthesis (UDS) assay by incorporation of [3H] thymidine (3HdT) in primary hepatocyte cultures (PHC). RESULTS: At 25th d after CT CAPE reduced the observed increase of GGT+AHF by 84% and liver expression of ggt mRNA by 100%. In case of the GSTp protein, the level was reduced by 90%. As indicative of oxidative stress generated by diethylnitrosamine (DEN) 12 h after its administration, we detected a 68% increase of TBARS. When CAPE was administered before DEN, it completely protected from liver TBARS induction. To have an indication of the sole effect of CAPE on initiation, twocarcinogens were tested in a UDS assay in PHC, we used methyl-n-nitrosoguanidine as a direct carcinogen and DEN, as indirect carcinogen. In this assay, genotoxic damage caused by carcinogens was abolished at 5μM CAPE concentration. | 11 | Epigenetics in the etiology and management of infertility显示文摘BACKGROUND Epigenetic disruptions have been implicated in some cases of infertility and can serve as therapeutic targets.However,the involvement of epigenetics in infertility has not received adequate attention.AIM This study aimed to determine the epigenetic basis of infertility in order to enhance public knowledge.METHODS Relevant articles on the subject were collected from PubMed,RCA,Google Scholar,SpringerLink,and Scopus.The articles were pooled together and duplicates were removed using Endnote software.RESULTS Available information shows that epigenetic mechanisms,mainly DNA methylation,histone modification,and microRNA interference are necessary for normal gametogenesis and embryogenesis.As a result,epigenetic disruptions in genes that control gametogenesis and embryogenesis,such as DDX3X,ADH4,AZF,PLAG1,D1RAS3,CYGB,MEST,JMJD1A,KCNQ1,IGF2,H19,and MTHFR may result in infertility.Aberrant DNA methylation during genomic imprinting and parental epigenetic mark erasures,in particular,may affect the DNA epigenomes of sperm and oocytes,resulting in reproductive abnormalities.in the sperm may also cause reproductive abnormalities.Furthermore,overexpression or repression of certain microRNAs embedded in the ovary,testis,embryo,as well as granulosa cells and oocytes may impair reproduction.Male infertility is characterized by spermatogenesis failure,which includes oligozoospermia,asthenozoospermia,and teratozoospermia,while female infertility is characterized by polycystic ovary syndrome.Some epigenetic modifications can be reversed by deactivating the regulatory enzymes,implying that epigenetic reprogramming could help treat infertility in some cases.For some disorders,epigenetic drugs are available,but none have been formulated for infertility.CONCLUSION Some cases of infertility have an epigenetic etiology and can be treated by reversing the same epigenetic mechanism that caused it.As a result,medical practitioners are urged to come up with epigenetic treatments for infertility that have an epigenetic cause. | Tajudeen Olanrewaju Yahaya Danlami M Bashar Esther O Oladele Ja'afar Umar Daniel Anyebe Abdulrazaq Izuafa | 2022 | World Journal of Medical Genetics2022,10,2: | 0 |
