1Effect of different seed treatments on dormancy breaking and germination in three species in arid and semi-arid lands显示文摘Seeds of many woody plant species have one of several types of dormancy. They do not germinate unless specific environmental signals are in place or events occur. This study was conducted to evaluate the effects of scarification treatments on seed dormancy and germination of Acacia nilotica (L.) Willd. ex Del., Prosopis juliflora (Sw.) DC. and Dodonaea viscosa (L.) Jacq. The following treatments were applied: T1, untreated seed (control); T2, sulphuric acid (97%) for 45 min; T3, boiling water for 5 min. The seeds were cultured on a Murashige and Skoog (MS) medium after sterilization. The responses of seeds to treatments were compared with each other and with the control treatment. Germination was observed daily for a 30-day period. Results indicated positive responses to treatments, while impermeable seed coats may be responsible for low germination rates in intact seeds as seen experimentally in the untreated control. The highest germination was obtained for P. juliflora and D. viscosa acid-scarified seeds (80.8%-90.8%) and for scarified seeds of A. nilotica (50.2%) boiled in water. 2Immunomodulatory effects of astragalus polysaccharide on Human peripheral blood mononuclear cells co-cultured with cervical cancer cell lineImmunomodulatory effects of astragalus polysaccharide on Human peripheral blood mononuclear cells co-cultured with cervical cancer cell line显示文摘OBJECTIVE:To investigate immunomodulatory effects of Astragalus polysaccharides(APS)on the co-culture of peripheral blood mononuclear cells(PBMCs)with He La cervical cancer cell line.METHODS:To assess the proliferation of PBMCs,carboxyfluorescein succinimidyl ester(CFSE)-labeled PBMCs were co-cultured with He La cells and treated with different concentrations of APS.Supernatants of cell culture were collected for cytokines assay via enzyme-linked immunosorbent assay(ELISA).The impact of APS on the proliferation of PBMCs,induction of regulatory T cells(Tregs),and myeloid-derived suppressor cells(MDSCs)was carried out by flow cytometry.RESULTS:It was observed that APS could increase the proliferation of PBMCs co-cultured with He La cells(P<0.05).However,APS had no significant effects on the induction of Tregs and MDSCs in the co-culture assay(P>0.05).Furthermore,ELISA results demonstrated that APS could decrease IL-10 and TGF-βconcentration(P<0.05).CONCLUSION:The above-mentioned characteristics showed that APS might be able to modulate immune responses and improve anti-tumor effects through increasing the proliferation of PBMCs and decreasing inhibitory cytokines secretion as critical mediators of immune suppression in the tumor microenvironment.Elham Shokati Mohammad-Reza Shokri Kobra Entezami Samaneh Khorrami Mona Amani Morteza Motallebnezhad Elahe Safari 2021Journal of Traditional Chinese Medicine2021,41,5:5
3Cryopreservation for delayed circulating tumor cell isolation is a valid strategy for prognostic association of circulating tumor cells in gastroesophageal cancer显示文摘AIM To demonstrate the feasibility of cryopreservation of peripheral blood mononuclear cells(PBMCs) for prognostic circulating tumor cell(CTC) detection in gastroesophageal cancer.METHODS Using 7.5 m L blood samples collected in EDTA tubes from patients with gastroesopheagal adenocarcinoma, CTCs were isolated by epithelial cell adhesion molecule based immunomagnetic capture using the Iso Flux platform. Paired specimens taken during the same blood draw(n = 15) were used to compare number of CTCs isolated from fresh and cryopreserved PBMCs. Blood samples were processed within 24 h to recover the PBMC fraction, with PBMCs used for fresh analysis immediately processed for CTC isolation. Cryopreservation of PBMCs lasted from 2 wk to 25.2 mo(median 14.6 mo). CTCs isolated from pre-treatment cryopreserved PBMCs(n = 43) were examined for associations with clinicopathological variables and survival outcomes.RESULTS While there was a significant trend to a decrease in CTC numbers associated with cryopreserved specimens(mean number of CTCs 34.4 vs 51.5, P = 0.04), this was predominately in samples with a total CTC count of > 50, with low CTC count samples less affected(P = 0.06). There was no significant association between the duration of cryopreservation and number of CTCs. In cryopreserved PBMCs from patient samples prior to treatment, a high CTC count(> 17) was associated with poorer overall survival(OS)(n = 43, HR = 4.4, 95%CI: 1.7-11.7, P = 0.0013). In multivariate analysis, after controlling for sex, age, stage, ECOG performance status, and primary tumor location, a high CTC count remained significantly associated with a poorer OS(HR = 3.7, 95%CI: 1.2-12.4, P = 0.03). 4Serum immunoglobulin A concentration is a reliable biomarker for liver fibrosis in non-alcoholic fatty liver disease显示文摘AIM:To evaluate the diagnostic accuracy of serum Immunoglobulin A(IgA)for differentiating early stage nonalcoholic fatty liver disease(NAFLD)from nonalcoholic steatohepatitis(NASH).METHODS:All cases had fatty liver change confirmed by ultrasound and aminotransferases of at least twice the normal level.Clinical and biochemical data,including serum IgA,were obtained from 50 histologically proven NAFLD cases and 54 healthy controls.Fasting whole blood samples were obtained from the study population.Immunoturbidimetric methods were used to measure the IgA levels.All NAFLD cases were hospitalized for liver biopsy.Liver specimens were examined for steatosis,steatohepatitis and fibrosis within hepatocytes.Patients were categorized into two groups:NASH and non-NASH.Variables were compared within cases(NASH vs non-NASH)and controls.Cut-off values of serum IgA were evaluated using analysis of receiver operating characteristic(ROC curves).Associations between the variables were tested using calculations of correlation coefficients.Statistical significances were assigned to P values<0.05.RESULTS:The extent of liver fibrosis correlated positively with IgA levels.Subjects with no fibrosis in their liver biopsies had a lower IgA level(301.5±91.2 mg/dL)than subjects with any degree of fibrosis(388.8±140.8 mg/dL),(P=0.01).IgA levels were higher in NASH cases,and its value was significantly higher for higher degrees of fibrosis.Patients with perisinusoidal or pericellular fibrosis had significantly higher levels of IgA(403.5±133.9 mg/dL,418.2±129.5 mg/dL)compared to those without it(301.8±94.9 mg/dL,297.7±91.5 mg/dL),respectively.No significant correlation was found between steatosis grade and serum IgA levels.Based on ROC analysis,the best predictive IgA cutoff value for detecting liver fibrosis was 360mg/dL(61%sensitivity,81%specificity).CONCLUSION:The serum IgA level is useful to evaluate the severity of liver fibrosis and can be used serially for evaluation and follow-up of NAFLD cases.Iradj Maleki Mahmood Reza Aminafshari Tarang Taghvaei Vahid Hosseini Alireza Rafiei Zhila Torabizadeh Maryam Barzin Elahe Orang 2014World Journal of Gastroenterology2014,20,35:4

6Spatial variation assessment of groundwater quality using multivariate statistical analysis(Case Study:Fasa Plain,Iran)显示文摘Groundwater is considered as one of the most important sources for water supply in Iran.The Fasa Plain in Fars Province,Southern Iran is one of the major areas of wheat production using groundwater for irrigation.A large population also uses local groundwater for drinking purposes.Therefore,in this study,this plain was selected to assess the spatial variability of groundwater quality and also to identify main parameters affecting the water quality using multivariate statistical techniques such as Cluster Analysis(CA),Discriminant Analysis(DA),and Principal Component Analysis(PCA).Water quality data was monitored at 22 different wells,for five years(2009-2014)with 10 water quality parameters.By using cluster analysis,the sampling wells were grouped into two clusters with distinct water qualities at different locations.The Lasso Discriminant Analysis(LDA)technique was used to assess the spatial variability of water quality.Based on the results,all of the variables except sodium absorption ratio(SAR)are effective in the LDA model with all variables affording 92.80%correct assignation to discriminate between the clusters from the primary 10 variables.Principal component(PC)analysis and factor analysis reduced the complex data matrix into two main components,accounting for more than 95.93%of the total variance.The first PC contained the parameters of TH,Ca2+,and Mg2+.Therefore,the first dominant factor was hardness.In the second PC,Cl-,SAR,and Na+were the dominant parameters,which may indicate salinity.The originally acquired factors illustrate natural(existence of geological formations)and anthropogenic(improper disposal of domestic and agricultural wastes)factors which affect the groundwater quality.Mehdi Bahrami Elmira Khaksar Elahe Khaksar 2020Journal of Groundwater Science and Engineering2020,8,3:2
7Synthesis of a copolymer carrier for anticancer drug luteolin for targeting human breast cancer cells显示文摘OBJECTIVE: To focus a new chemoprevention approach that uses nanotechnology to deliver luteolin to human breast cancer cells(MCF-7), and its underlying mechanism.METHODS: Water-soluble copolymer-encapsulated nanoparticle-luteolin(CENL) was formulated using the hydrophobic drug luteolin. The ability to load and release he anticancer drug into/from the synthesized hyperbranched polyester was evaluated by high-performance liquid chromatography.The successful synthesis of CENL was supported by analytical techniques including fourier transform infrared spectroscopy, nuclear magnetic resonance spectroscopy, gel permeation chromatography,and dynamic light scattering. Cell viability was measured using the 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide color method. Reactive oxygen species(ROS) were measured using a dichlorofluorescein probe and intracellular calcium(Cai2+) was evaluated with a flu3-AM probe.RESULTS: The results showed that the drug delivery system is stable and that the loading capacity is high. 8Do clinical and paraclinical findings have the power to predict critical conditions of injured patients after traumatic injury resuscitation?Using data mining artificial intelligence显示文摘Purpose The triage and initial care of injured patients and a subsequent right level of care is paramount for an overall outcome after traumatic injury.Early recognition of patients is an important case of such decision-making with risk of worse prognosis.This article is to answer if clinical and paraclinical signs can predict the critical conditions of injured patients after traumatic injury resuscitation.Methods The study included 1107 trauma patients,16 years and older.The patients were trauma victims of Levels I and II triage and admitted to the Rajaee(Emtiaz)Trauma Hospital,Shiraz,in 2014–2015.The cross-industry process for data mining methodology and modeling was used for assessing the best early clinical and paraclinical variables to predict the patients’prognosis.Five modeling methods including the support vector machine,K-nearest neighbor algorithms,Bagging and Adaboost,and the neural network were compared by some evaluation criteria.Results Learning algorithms can predict the deterioration of injured patients by monitoring the Bagging and SVM models with 99%accuracy.The most-fitted variables were Glasgow Coma Scale score,base deficit,and diastolic blood pressure especially after initial resuscitation in the algorithms for overall outcome predictions.Conclusion Data mining could help in triage,initial treatment,and further decision-making for outcome measures in trauma patients.Clinical and paraclinical variables after resuscitation could predict short-term outcomes much better than variables on arrival.With artificial intelligence modeling system,diastolic blood pressure after resuscitation has a greater association with predicting early mortality rather than systolic blood pressure after resuscitation.Artificial intelligence monitoring may have a role in trauma care and should be further investigated.Shahram Paydar Elahe Parva Zahra Ghahramani Saeedeh Pourahmad Leila Shayan Vahid Mohammadkarimi Golnar Sabetian 2021Chinese Journal of Traumatology2021,24,1:2
