| 1 | miRNAs and lncRNAs in vascular injury and remodeling显示文摘Vascular injury,remodeling,as well as angiogenesis,are the leading causes of coronary or cerebrovascular disease.The blood vessel functional imbalance trends to induce atherosclerosis,hypertension,and pulmonary arterial hypertension.As several genes have been identified to be dynamically regulated during vascular injury and remodeling,it is becoming widely accepted that several types of non-coding RNA,such as microRNAs(miRNAs)and long non-coding RNAs(lncRNAs),are involved in regulating the endothelial cell and vascular smooth muscle cell(VSMC)behaviors.Here,we review the progress of the extant studies on mechanistic,clinical and diagnostic implications of miRNAs and lncRNAs in vascular injury and remodeling,as well as angiogenesis,emphasizing the important roles of miRNAs and lncRNAs in vascular diseases.Furthermore,we introduce the interaction between miRNAs and lncRNAs,and highlight the mechanism through which lncRNAs are regulating the miRNA function.We envisage that continuous in-depth research of non-coding RNAs in vascular disease will have significant implications for the treatment of coronary or cerebrovascular diseases. | SONG XiaoWei SHAN DongKai CHEN Jian JING Qing | 2014 | Science China(Life Sciences)2014,57,8: | 17 |

| 3 | Exosome-based small RNA delivery:Progress and prospects显示文摘RNA interfering(RNAi), mediated by small interfering RNAs and microRNAs, is currently one of the most promising tools of gene therapy. Small RNAs are capable of inducing specific post-transcriptional gene silencing, providing a potentially effective platform for the treatment of a wide array of diseases. However, similar to other nucleic acid-based drugs,the major hurdle of RNAi therapy is lack of efficient and non-immunogenic delivery vehicles. Currently, viruses, synthetic polymers, and lipid-based carriers are among the most widely studied vehicles for small RNA delivery. However, many drawbacks are reported to be associated with these delivery vehicles. There is a pressing need to replace them with more efficient and better-tolerated approaches. Exosomes secreted from the endocytic compartment of live cells, are a subtype of endogenous extracellular vesicles that transfer genetic and biochemical information among different cells, thus playing an important role in cellcell communication. Recently, accumulating attention has been focused on harnessing exosomes as nanaocarriers for small RNAs delivery. Due to their natural role in shuttling endogenous nucleic acid in our body, exosomes may exhibit higher delivery efficiency, lower immunogenicity, and better compatibility than existing foreign RNA carriers. Importantly,exosomes own intrinsic homing capacity that can guide small RNAs across natural membranous barriers. Moreover, such a capacity can be further improved by adding appropriate targeting moieties. | 4 | The role and clinical implications of microRNAs in hepatocellular carcinoma显示文摘Hepatocellular carcinoma (HCC) is common and one of the most aggressive of all human cancers. Recent studies have indicated that miRNAs, a class of small noncoding RNAs that regulate gene expression post-transcriptionally, directly contribute to HCC by targeting many critical regulatory genes. Several miRNAs are involved in hepatitis B or hepatitis C virus replication and virus-induced changes, whereas others participate in multiple intracellular signaling pathways that modulate apoptosis, cell cycle checkpoints, and growth-factor-stimulated responses. When disturbed, these pathways appear to result in malignant transformation and ultimately HCC development. Recently, miRNAs circulating in the blood have acted as possible early diagnostic markers for HCC. These miRNA also could serve as indicators with respect to drug efficacy and be prognostic in HCC patients. Such biomarkers would assist stratification of HCC patients and help direct personalized therapy. Here, we summarize recent advances regarding the role of miRNAs in HCC development and progression. | 5 | Familial Parkinson's Disease-Associated L166P Mutant DJ-1 is Cleaved by Mitochondrial Serine Protease Omi/HtrA2显示文摘Parkinson's disease(PD) is the most common neurodegenerative movement disorder. Mutations in the DJ-1, including L166 P, are responsible for recessive earlyonset PD. Many lines of evidence have shown that L166 P is not only a loss-of-function mutant, but also a proapoptotic-like protein that results in mitochondrial dysfunction. L166 P has been reported to be unstable and to mislocalize to mitochondria. However, the mechanisms underlying the instability of L166 P compared to wild-type DJ-1 remain largely unknown. Here, we showed that Omi/Htr A2, a mitochondrial serine protease that has also been linked to the pathogenesis of PD, contributed to L166 P instability. Omi directly interacted with and cleaved L166 P in mitochondria to decrease the L166 P level. However,Omi did not bind and cleave wild-type DJ-1. Moreover,Omi cleaved L166 P at both serine residues 3 and 121,while L166 P-induced cell death under H_2O_2 treatment was alleviated by over-expression of Omi. | 6 | Cholesterol-tuned liposomal membrane rigidity directs tumor penetration and anti-tumor effect显示文摘Recently, liposomes have been widely used in cancer therapeutics, but their anti-tumor effects are suboptimal due to limited tumor penetration. To solve this problem, researchers have made significant efforts to optimize liposomal diameters and potentials, but little attention has been paid to liposomal membrane rigidity. Herein, we sought to demonstrate the effects of cholesterol-tuned liposomal membrane rigidity on tumor penetration and anti-tumor effects. In this study, liposomes composed of hydrogenated soybean phospholipids(HSPC), 1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[methoxy(polyethylene glycol)-2000](DSPE-PEG2000) and different concentrations of cholesterol were prepared. It was revealed that liposomal membrane rigidity decreased with the addition of cholesterol. Moderate cholesterol content conferred excellent diffusivity to liposomes in simulated diffusion medium, while excessive cholesterol limited the diffusion process. We concluded that the differences of the diffusion rates likely stemmed from the alterations in liposomal membrane rigidity, with moderate rigidity leading to improved diffusion. Next, the in vitro tumor penetration and the in vivo anti-tumor effects were analyzed. The results showed that liposomes with moderate rigidity gained excellent tumor penetration and enhanced anti-tumor effects. | 10 | Simulation of Multi-satellite GNSS Reflected Signals and Design of Simulator显示文摘Using GNSS-R technology for remote sensing of surface parameters has become a new trend in the field of remote sensing.With the rapid development of GNSS-R technology,GNSS-R simulation has become one of the new hot spots.Now the researches of the GNSS-R simulation are all the simulations that consider a single star or a single frequency point,and in actual applications,the signal captured by the receiver is often the reflected signals of multiple stars.In view of this situation,from the perspective of multi-satellite simulation,this paper gives the model of GNSS-R multi-satellite ocean simulation on the basis of analyzing the remote sensing principle,reflection signal model and simulation principle of GNSS-R technology.Based on the GNSS-R multi-satellite ocean simulation model and the fast parallel computing capability of GPU,the GNSS-R multi-satellite ocean simulator was designed.Finally,the direct and reflected signals generated by the GNSS-R multi-satellite simulator were tested and verified.The results show that the positioning result of the direct signal meets the positioning accuracy requirements;The delay-related power results obtained from the simulated two-satellite reflected signals processing are in good agreement with the theoretical model,and the correlation coefficients are all above 0.99;The generated signals are used for GNSS-R height measurement technology,the height measurement error is about 1.4~1.8 m,which is in line with the accuracy of the C/A code ranging receiver;And the parallel operation of the GPU for multi-satellite simulation calculation is 800—900 times higher than the traditional CPU calculation.It proves that the proposed model and the designed simulator are feasible and accurate. | Bowen LI Dongkai YANG Bo ZHANG | 2021 | Journal of Geodesy and Geoinformation Science2021,4,2: | 3 |

| 12 | Molecular characterization and developmental expression patterns of thyroid hormone receptors(TRs) and their responsiveness to TR agonist and antagonist in Rana nigromaculata显示文摘Considering some advantages of Rana nigromaculata as an experimental species, we propose that this species, like Xenopus laevis, could be used to assay thyroid hormone(TH) signaling disrupting actions. To validate the utilizability of R. nigromaculata, we investigated the responsiveness of R. nigromaculata to a TH receptor(TR) agonist(T3) and antagonist(amiodarone) by analyzing expression, based on characterizing TR cDNA and developmental expression patterns. With high levels of identity with the corresponding genes in X. laevis, both TRα and TRβ in R. nigromaculata exhibited roughly similar developmental expression patterns to those of X. laevis, in spite of some species-specific differences. Both TRα and TRβ expression had greater changes in the liver and intestine than in the tail and brain during metamorphosis. T3 exposure for 2 days induced more dramatic increases of TRβ expression in stage 27 than in stage34 tadpoles but not in stage 42 tadpoles, showing that the responsiveness of R. nigromaculata to TH decreased with development and disappeared at the onset of metamorphic climax.Corresponding to greater changes of TRβ expression in the liver and intestine than in the tail and brain during metamorphosis, the liver and intestine had higher responsiveness to exogenous T3 than the tail and brain. Amiodarone inhibited T3-induced TRβ expression. | 13 | Standard pancreatoduodenectomy versus extended pancreatoduodenectomy with modified retroperitoneal nerve resection in patients with pancreatic head cancer:a multicenter randomized controlled trial显示文摘Background:The extent of pancreatoduodenectomy for pancreatic head cancer remains controversial,and more high-level clinical evidence is needed.This study aimed to evaluate the outcome of extended pancreatoduodenectomy(EPD)with retroperitoneal nerve resection in pancreatic head cancer.Methods:This multicenter randomized trial was performed at 6 Chinese highvolume hospitals that enrolled patients between October 3,2012,and September 21,2017.Four hundred patients with stage I or II pancreatic head cancer and without specific pancreatic cancer treatments(preoperative chemotherapy or chemoradiation)within three months were randomly assigned to undergo standard pancreatoduodenectomy(SPD)or EPD,with the latter followed by dissection of additional lymph nodes(LNs),nerves and soft tissues 270◦on the right side surrounding the superior mesenteric artery and celiac axis.The primary endpoint was overall survival(OS)by intention-to-treat(ITT).The secondary endpoints were disease-free survival(DFS),mortality,morbidity,and postoperative pain intensity.Results:TheR1 ratewas slightly lower with EPD(8.46%)thanwith SPD(12.56%).The morbidity and mortality rates were similar between the two groups.The median OS was similar in the EPD and SPD groups by ITT in the whole study cohort(23.0 vs.20.2 months,P=0.100),while the median DFS was superior in the EPD group(16.1 vs.13.2 months,P=0.031).Patients with preoperative CA19–9<200.0 U/mL had significantly improved OS and DFS with EPD(EPD vs.SPD,30.8 vs.20.9 months,P=0.009;23.4 vs.13.5 months,P<0.001).The EPD group exhibited significantly lower locoregional(16.48%vs.35.20%,P<0.001)andmesenteric LNrecurrence rates(3.98%vs.10.06%,P=0.022).The EPD group exhibited less back pain 6 months postoperation than the SPD group.Conclusions:EPD for pancreatic head cancer did not significantly improve OS,but patients with EPD treatment had significantly improved DFS.In the subgroup analysis,improvements in bothOS and DFS in the EPD armwere observed in patients with preoperative CA19–9<200.0 U/mL.EPD could be used as an effective surgical procedure for patients with pancreatic head cancer,especially those with preoperative CA19–9<200.0 U/mL. | Qing Lin Shangyou Zheng Xianjun Yu Meifu Chen Yu Zhou Quanbo Zhou Chonghui Hu Jing Gu Zhongdong Xu LinWang Yimin Liu Qingyu Liu MinWang Guolin Li He Cheng Dongkai Zhou Guodong Liu Zhiqiang Fu Yu Long Yixiong Li Weilin Wang Renyi Qin Zhihua Li Rufu Chen | 2023 | Cancer Communications2023,43,2: | 3 |
| 14 | Effects of Logging Intensity on Structure and Composition of a Broadleaf-Korean Pine Mixed Forest on Changbai Mountains,Northeast China显示文摘In order to identify a harvesting model which is beneficial for broadleaf-Korean pine mixed forest(BKF) sustainability, we investigated four types of harvested stands which have been logged with intensities of 0(T0, control), 15%(T1, low intensity), 35%(T2, moderate intensity), and 100%(T3, clear-cutting), and examined the impacts of logging intensity on composition and structure of these stands. Results showed that there were no significant differences between T0 and T1 for all structural characteristics, except for density of seeding and large trees. The mean diameter at breast height(DBH, 1.3 m above the ground), stem density and basal area of large trees in T2 were significantly lower than in T0, while the density of seedlings and saplings were significantly higher in T2 than in T0. Structural characteristics in T3 were entirely different from T0. Dominant tree species in primary BKF comprised 93%, 85%, 45% and 10% of the total basal area in T0, T1, T2 and T3, respectively. Three community similarity indices, the Jaccard′s similarity coefficient(CJ); the Morisita-Horn index(CMH); and the Bray-Curtis index(CN), were the highest for T0 and T1, followed by T0 and T2, and T0 and T3, in generally. These results suggest that effects of harvesting on forest composition and structure are related to logging intensities. Low intensity harvesting is conductive to preserving forest structure and composition, allowing it to recover in a short time period.
