1Programmed death-1 upregulation is correlated with dysfunction of tumor-infiltrating CD8^(+ ) T lymphocytes in human non-small cell lung cancer显示文摘T-cell tolerance is an important mechanism for tumor escape,but the molecular pathways involved in T-cell tolerance remain poorly understood.It remains unknown whether the inhibitory immunoreceptor programmed death-1(PD-1)plays a role in conditions of human non-small cell lung cancer(NSCLC).In this study,we detected PD-1 expression on CD81 T cells from healthy control peripheral blood mononuclear cells(PBMCs)and the PBMCs of NSCLC patients as well as NSCLC tissues.Results showed that tumor-infiltrating CD81 T cells had increased PD-1 expression and impaired immune function,including reducing cytokine production capability and impairing capacity to proliferate.Blockade of the PD-1/PD-L1 pathway by the PD-L1-specific antibody partially restored cytokine production and cell proliferation.These data provide direct evidence that the PD-1/PD-L1 pathway is involved in CD81 T-cell dysfunction in NSCLC patients.Moreover,blocking this pathway provides a potential therapy target in lung cancer.Yan Zhang Shengdong Huang Dejun Gong Yanghua Qin Qian Shen 2010Cellular & Molecular Immunology2010,7,5:48

3SNP-based linkage mapping for validation of adult plant stripe rust resistance QTL in common wheat cultivar Chakwal 86显示文摘Wheat crops in China are constantly challenged by stripe rust. Deployment of cultivars with diverse resistances is the best strategy to control the disease. A recombinant inbred line(RIL) population derived from a cross between the resistant cultivar Chakwal 86 and susceptible landrace Mingxian 169 was studied in multiple environments to examine the underlying genetics and to identify quantitative trait loci(QTL) for stripe rust resistance.One hundred and twenty-eight RILs were genotyped with wheat 35 K SNP array and a genome-wide linkage map with 1480 polymorphic SNP markers, or bins, was constructed.Two major QTL on chromosomes 1BL and 3BS, and one minor QTL on 6BS had significant effects in reducing stripe rust severity. The QTL were validated using composite interval mapping(CIM) and inclusive composite interval mapping(ICIM). These methods explained59.0%–74.1% of the phenotype variation in disease response. The QTL on chromosome 1 BL was confirmed to be Yr29/Lr46 and the one on 3BS was the resistance allele identified in CIMMYT germplasm but was not Yr30/Sr2. The QTL on 6BS probably corresponded to previously known QTL. RILs with combined QTL were more resistant than those with single or no QTL. Kompetitive allele-specific PCR(KASP) assays for the QTL with largest effect QTL on chromosome 3BS were performed on a subset of RILs and 150 unrelated wheat lines. 4Tracking topological entity changes in 3D collaborative modeling systems显示文摘One of the key problems in collaborative geometric modeling systems is topological entity correspondence when topological structure of geometry models on collaborative sites changes.In this article,we propose a solution for tracking topological entity alterations in 3D collaborative modeling environment.We firstly make a thorough analysis and detailed categorization on the alteration properties and causations for each type of topological entity,namely topological face and topological edge.Based on collaborative topological entity naming mechanism,a data structure called TEST (Topological Entity Structure Tree) is introduced to track the changing history and current state of each topological entity,to embody the relationship among topological entities.Rules and algorithms are presented for identification of topological entities referenced by operations for correct execution and model consistency.The algorithm has been verified within the prototype we have implemented with ACIS.Cheng Yuan He Fazhi Huang Zhiyong Cai Xiantao Zhang Dejun 2012Computer Aided Drafting,Design and Manufacturing2012,22,1:2
