1Interplay between post-translational cyclooxygenase-2 modifications and the metabolic and proteomic profile in a colorectal cancer cohort显示文摘BACKGROUND Colorectal cancer(CRC) is the second most common cause of cancer death worldwide. It is broadly described that cyclooxygenase-2(COX-2) is mainly overexpressed in CRC but less is known regarding post-translational modifications of this enzyme that may regulate its activity, intracellular localization and stability. Since metabolic and proteomic profile analysis is essential for cancer prognosis and diagnosis, our hypothesis is that the analysis of correlations between these specific parameters and COX-2 state in tumors of a high number of CRC patients could be useful for the understanding of the basis of this cancer in humans.AIM To analyze COX-2 regulation in colorectal cancer and to perform a detailed analysis of their metabolic and proteomic profile.METHODS Biopsies from both healthy and pathological colorectal tissues were taken under informed consent from patients during standard colonoscopy procedure in the University Hospital of Bellvitge(Barcelona, Spain) and Germans Trias i Pujol University Hospital(Campus Can Ruti)(Barcelona, Spain). Western blot analysis was used to determine COX-2 levels. Deglycosylation assays were performed in both cells and tumor samples incubating each sample with peptide N-glycosidase F(PNGase F). Prostaglandin E2(PGE2) levels were determined using a specific ELISA. 1 H high resolution magic angle spinning(HRMAS) analysis was performed using a Bruker AVIII 500 MHz spectrometer and proteomic analysis was performed in a nano-liquid chromatography-tandem mass spectrometer(nano LC-MS/MS) using a QExactive HF orbitrap MS.RESULTS Our data show that COX-2 has a differential expression profile in tumor tissue of CRC patients vs the adjacent non-tumor area, which correspond to a glycosylated and less active state of the protein. This fact was associated to a lesser PGE2 production in tumors. These results were corroborated in vitro performing deglycosylation assays in HT29 cell line where COX-2 protein profile was modified after PNGase F incubation, showing higher PGE2 levels. Moreover,HRMAS analysis indicated that tumor tissue has altered metabolic features vs non-tumor counterparts, presenting increased levels of certain metabolites such as taurine and phosphocholine and lower levels of lactate. 12Tracheoesophageal fistulas in coronavirus disease 2019 pandemic:A case report显示文摘BACKGROUND Tracheoesophageal fistulas(TEFs)can be described as a pathological communication between the trachea and the esophagus.According to their origin,they may be classified as benign or malignant.Benign TEFs occur mostly as a consequence of prolonged mechanical ventilation,particularly among patients exposed to endotracheal cuff overinflation.During the severe acute respiratory syndrome coronavirus 2 virus pandemic,the amount of patients requiring prolonged ventilation rose,which in turn increased the incidence of TEFs.CASE SUMMARY We report the cases of 14 patients with different comorbidities such as being overweight,or having been diagnosed with diabetes mellitus or systemic hypertension.The most common symptoms on arrival were dyspnea and cough.In all cases,the diagnosis of TEFs was made through upper endoscopy.Depending on the location and size of each fistula,either endoscopic or surgical treatment was provided.Eight patients were treated endoscopically.Successful closure of the defect was achieved through over the scope clips in two patients,while three of them required endoscopic metal stenting.A hemoclip was used to successfully treat one patient,and it was used temporarily for another patient pended surgery.Surgical treatment was performed in patients with failed endoscopic management,leading to successful defect correction.Two patients died before receiving corrective treatment and four died later on in their clinical course due to infectious complications.CONCLUSION The incidence of TEFs increased during the coronavirus disease 2019 pandemic(from 0.5%to 1.5%).We believe that endoscopic treatment should be considered as an option for this group of patients,since evidence reported in the literature is still a growing area.Therefore,we propose an algorithm to lead intervention in patients presenting with TEFs due to prolonged intubation.Martin Alonso Gomez Zuleta Daniel Mauricio Gallego Ospina Oscar Fernando Ruiz 2022World Journal of Gastrointestinal Endoscopy2022,14,10:0
13Bigger or long-winged male common crossbills exhibit redder carotenoid-based plumage coloration显示文摘Carotenoid-based ornaments are often considered reliable(honest)individual condition signals because their expression implies physiological costs unaffordable for low-quality animals(handicap signals).Recently,it has been suggested that efficient cell respiration is mandatory for producing red ketocarotenoids from dietary yellow carotenoids.This implies that red colorations should be entirely unfalsifiable and independent of expression costs(index signals).In a precedent study,male common crossbills,Loxia curvirostra,showing a red plumage reported higher apparent survival than those showing yellowish-orange colors.The plumage redness in this species is due to ketocarotenoid accumulation in feathers.Here,we correlated the male plumage redness(a 4-level visual score:yellow,patchy,orange,and red)and the body morphology in more than 1,ooo adult crossbills captured in 3 Iberian localities to infer the mechanisms responsible for color evolution.A principal component analysis summarized morphometry of 10 variables(beak,wing,tarsus length,etc.).The overall body size(PC1)and the length of flight feathers regarding body size(Pc3)showed significant positive relationships with plumage redness.Plumage redness was barely correlated with bill shape measures,suggesting no constraint in acquiring carotenoids from pine cones.However,large body sizes or proportionally long flying feathers could help carotenoid acquisition via social competition or increased foraging ranges.Proportionally longer flight feathers might also be associated with a specific cell respiration profile that would simultaneously favor flying capacities and enzymatic transformations needed for ketocarotenoid synthesis.Such a phenotypic profile would agree with the hypothesis of ketocarotenoid-based colors acting as individual quality index signals.Blanca Fernandez-Eslava Daniel Alonso David Galicia Juan Arizaga Carlos Alonso-Alvarez 2023Current Zoology2023,69,2:0
14Yearly variation in the structure and diversity of a non-breeding passerine bird community in a Mediterranean wetland显示文摘Background:The dynamics and structure of many bird communities are changing due to the global warming and changes in the land use and management.The Ebro Valley constitutes one of the chief wintering areas for several passerine species in Iberia,and the reed beds spread across this Valley concentrate huge numbers of birds that use them to roost,while the surrounding agricultural areas serve as main foraging habitats.The aims of this work are(1)to quantify how variable a non-breeding passerine bird community associated to a reed bed area in the Ebro Valley is,and,(2)to test for the effect of weather on possible annual fluctuations in species’proportions at a regional/local scale.Methods:We used for that ringing data collected at Badina de Escudera lagoon(Navarra)over a 12-year period.Results:Though,overall,the assemblage did not vary strongly among years(most similarity values were>0.8),we still found a significant linear effect of year on the proportion of captures of Reed Buntings(Emberiza schoeniclus),i.e.,the dominant species detected in our passerine community.The increasing loss of weight of Reed Buntings in the community was due to a progressive decrease in its abundance.Conclusions:The structure and diversity of passerine community in Badina Escudera was not influenced by meteorological conditions at a local scale,suggesting that local weather would not have an impact on numbers of Reed Buntings in winter.Juan Arizaga Daniel Alonso ArinCrespo Xabier Esparza Efrén Fernández Iógo López David Martín Antonio Vilches 2020Avian Research2020,11,3:0
