1Clinical and biochemical indexes from 2019-nCoV infected patients linked to viral loads and lung injury显示文摘The outbreak of the 2019-nCoV infection began in December 2019 in Wuhan,Hubei province,and rapidly spread to many provinces in China as well as other countries.Here we report the epidemiological,clinical,laboratory,and radiological characteristics,as well as potential biomarkers for predicting disease severity in 2019-nCoV-infected patients in Shenzhen,China.All 12 cases of the 2019-nCoV-infected patients developed pneumonia and half of them developed acute respiratory distress syndrome (ARDS).The most common laboratory abnormalities were hypoalbuminemia,lymphopenia,decreased percentage of lymphocytes (LYM) and neutrophils (NEU),elevated C-reactive protein (CRP) and lactate dehydrogenase (LDH),and decreased CD8 count.The viral load of 2019-nCoV detected from patient respiratory tracts was positively linked to lung disease severity.ALB,LYM,LYM (%),LDH,NEU (%),and CRP were highly correlated to the acute lung injury.Age,viral load,lung injury score,and blood biochemistry indexes,albumin (ALB),CRP,LDH,LYM (%),LYM,and NEU (%),may be predictors of disease severity.Moreover,the Angiotensin Ⅱlevel in the plasma sample from 2019-nCoV infected patients was markedly elevated and linearly associated to viral load and lung injury.Our results suggest a number of potential diagnosis biomarkers and angiotensin receptor blocker (ARB) drugs for potential repurposing treatment of 2019-nCoV infection.Yingxia Liu Yang Yang Cong Zhang Fengming Huang Fuxiang Wang Jing Yuan Zhaoqin Wang Jinxiu Li Jianming Li Cheng Feng Zheng Zhang Lifei Wang Ling Peng Li Chen Yuhao Qin Dandan Zhao Shuguang Tan Lu Yin Jun Xu Congzhao Zhou Chengyu Jiang Lei Liu 2020Science China(Life Sciences)2020,63,3:157
2Human carboxylesterases:a comprehensive review显示文摘Mammalian carboxylesterases(CEs) are key enzymes from the serine hydrolase superfamily.In the human body, two predominant carboxylesterases(CES1 and CES2) have been identified and extensively studied over the past decade. These two enzymes play crucial roles in the metabolism of a wide variety of endogenous esters, ester-containing drugs and environmental toxicants. The key roles of CES in both human health and xenobiotic metabolism arouse great interest in the discovery of potent CES modulators to regulate endobiotic metabolism or to improve the efficacy of ester drugs. This review covers the structural and catalytic features of CES, tissue distributions, biological functions, genetic polymorphisms, substrate specificities and inhibitor properties of CES1 and CES2, as well as the significance and recent progress on the discovery of CES modulators. The information presented here will help pharmacologists explore the relevance of CES to human diseases or to assign the contribution of certain CES in xenobiotic metabolism. 3Allele-aware chromosome-level genome assembly of Artemisia annua reveals the correlation between ADS expansion and artemisinin yield显示文摘Artemisia annua is the major natural source of artemisinin,an anti-malarial medicine commonly used worldwide.Here,we present chromosome-level haploid maps for two A.annua strains with different artemisinin contents to explore the relationships between genomic organization and artemisinin production.High-fidelity sequencing,optical mapping,and chromatin conformation capture sequencing were used to assemble the heterogeneous and repetitive genome and resolve the haplotypes of A.annua.Approximately 5o,ooo genes were annotated for each haplotype genome,and a triplication event that occurred approximately 58.12million years ago was examined for the first time in this species.A total of 3,903,467-5,193,414 variants(SNPs,indels,and structural variants)were identified in the 1.5-Gb genome during pairwise comparison between haplotypes,consistent with the high heterozygosity of this species.Genomic analyses revealed a correlation between artemisinin concents and the copy number of amorpha-4,11-dienes ynthasegenes.This correlation was further confirmed by resequencing of 36A.annua samples with varied artemisinin contents.Circular consensus sequencing of transcripts facilitated the detection of paralog expression.Collectively,our study provides chromosome-level allele-aware genome assemblies for two A.annua strains and new insights into the biosynthesis of artemisinin and its regulation,which will contribute to conquering malaria worldwide.Baosheng Liao Xiaofeng Shen Li Xiang Shuai Guo Shiyu Chen Ying Meng Yu Liang Dandan Ding Junqi Bai Dong Zhang Tomasz Czechowski Yi Li Hui Yao Tingyu Mai Caroline Howard Chao Sun Haitao Liu Jiushi Liu Jin Pei Jihai Gao Jigang Wang Xiaohui Qiu Zhihai Huang Hongyi Li Ling Yuan Jianhe Wei lan Graham Jiang Xu Boli Zhang Shilin Chen 2022Molecular Plant2022,15,8:9
4Elevated plasma levels of selective cytokines in COVID-19 patients reflect viral load and lung injury显示文摘A recent outbreak of pneumonia in Wuhan,China was found to be caused by a 2019 novel coronavirus(2019-nCoV or SARS-CoV-2 or HCoV-19).We previously reported the clinical features of 12 patients with2019-n Co V infections in Shenzhen,China.To further understand the pathogenesis of COVID-19 and find better ways to monitor and treat the disease caused by 2019-n Co V,we measured the levels of 48 cytokines in the blood plasma of those 12 COVID-19 patients.Thirty-eight out of the 48 measured cytokines in the plasma of 2019-n Co V-infected patients were significantly elevated compared to healthy individuals.Seventeen cytokines were linked to 2019-nCoV loads.Fifteen cytokines,namely M-CSF,IL-10,IFN-α2,IL-17,IL-4,IP-10,IL-7,IL-1 ra,G-CSF,IL-12,IFN-γ,IL-1α,IL-2,HGF and PDGF-BB,were strongly associated with the lung-injury Murray score and could be used to predict the disease severity of2019-n Co V infections by calculating the area under the curve of the receiver-operating characteristics.Our results suggest that 2019-nCoV infections trigger extensive changes in a wide array of cytokines,some of which could be potential biomarkers of disease severity of 2019-n Co V infections.These findings will likely improve our understanding of the immunopathologic mechanisms of this emerging disease.Our results also suggest that modulators of cytokine responses may play a therapeutic role in combating the disease once the functions of these elevated cytokines have been characterized.Yingxia Liu Cong Zhang Fengming Huang Yang Yang Fuxiang Wang Jing Yuan Zheng Zhang Yuhao Qin Xiaoyun Li Dandan Zhao Shunwang Li Shuguang Tan Zhaoqin Wang Jinxiu Li Chenguang Shen Jianming Li Ling Peng Weibo Wu Mengli Cao Li Xing Zhixiang Xu Li Chen Congzhao Zhou William J Liu Lei Liu Chengyu Jiang 2020National Science Review2020,7,6:6
5High-throughput development and applications of the compositional mechanical property map of the β titanium alloys显示文摘By a combination of the nanoindentation and electron probe microanalysis(EPMA)techniques,the traditional diffusion couple technique is extended to map the mechanical property of β-type Ti alloys over a wide composition range,which can be utilized to develop very versatile novel bio-Ti alloys for hard tissue re placements in arti ficial bones,joints,and dental implants.To create complete single-phase composition ranges of Ti-based bcc solid solution,12 types of bcc Ti-Nb-Zr-Mo/Ti-Nb-Zr-Ta quaternary diffusion couples were fabricated and annealed at 1273 K for 25 h.In this way,the composition-mechanical property relationships in the vast composition space of Ti-based alloys were established using EPMA and nanoindentation probes.Notably,the measured composition-dependent Young’s moduli,hardness,and elastic recovery as well as the derived ratio of hardness to Young’s modulus,and the ratio of the cube of hardness to the square of Young’s modulus,in the developed compositional mechanical property database,were visualized in a five-dimensional scatter plot.This enables an effective tool to screen the Ti-Nb-Zr-based alloys fororthopedic and dental applications according to different clinical requirements,and to rationalize the fundamental mechanical relationships in the rapid development of β-Ti alloys.Jinfeng Ling Dandan Huang Kewu Bai Wei Li Zhentao Yu Weimin Chen 2021Journal of Materials Science & Technology2021,,12:3

7An Efficient Extraction-Free Method for Oxidative Desulfurization of Model Fuels Employing Cross-Linked Polyionic Liquid Phosphotungstate Catalysts显示文摘An efficient extraction-free oxidative desulfurization(ODS)process using a series of cross-linked polyionic liquid phosphotungstate(CLPIL-PW)catalysts is reported.The cross-linked PILs were prepared with DVB and 1-n-alkyl-3-vinyl imidazole hydrobromide(alkyl=ethyl,butyl,octyl,dodecyl),and were then assembled with phosphotungstic acid(H_(3)PW_(12)O_(40))to form the catalysts.The CLPIL-PWs have been applied to the oxidative removal of dibenzothiophene(DBT)from model oil with H_(2)O_(2) as an oxidant.The effects of ionic liquid(IL)cationic species,varying the DVB/IL molar ratio in the polymerization process,and varying operating conditions were investigated.The CLPIL-PWs were characterized by inductively coupled plasma(ICP)mass spectrometry,elemental analysis,scanning electron microscopy(SEM),Fourier transform infra-red(FTIR)spectroscopy,X-ray diffraction(XRD),^(13)C and^(31)P nuclear magnetic resonance(NMR)spectroscopy.The polydivinylbenzene-co-1-n-octyl-3-vinyl imidazole phosphotungstate(P(DVB-OVIm)PW)exhibited the highest DBT removal efficiency(99.9%)and remarkable recyclability,and could be reused eight times without reducing its activity.Finally,an extraction-free ODS mechanism is proposed.Yuan Dandan Zhang Ling Yin Guoqing Wang Yuanyuan Zhang Jiaojing 2022China Petroleum Processing & Petrochemical Technology2022,24,3:2
8Amphiphilic Dendrimer Vectors for RNA Delivery:State-of-the-Art and Future显示文摘Dendrimers,a special family of polymers,are particularly promising materials for various biomedical applications by virtue of their well-defined dendritic structure and cooperative multivalency.Specifically,in this Account,we present state-of-the-art amphiphilic dendrimers for nucleic acid delivery.Ribonucleic acid(RNA)molecules are fast becoming an important drug modality,particularly since the recent success of mRNA vaccines against COVID-19.Notably,RNA therapeutics offer the unique opportunity to treat diseases at the gene level and address“undruggable”targets.However,RNA therapeutics are not stable and have poor bioavailability,imposing the need for their protection and safe delivery by vectors to the sites-of-action to allow the desired therapeutic effects.Currently,the two most advanced nonviral vectors are based on lipids and polymers,with lipid vectors primarily exploiting the membrane-fusion mechanism and polymer vectors mainly endocytosis-mediated delivery.Notably,only lipid vectors have been advanced through to their clinical use in the delivery of,for example,the first siRNA drug and the first mRNA vaccine.The success of lipid vectors for RNA delivery has motivated research for further innovative materials as delivery vectors.Specifically,we have pioneered lipid/dendrimer conjugates,referred to as amphiphilic dendrimers,for siRNA delivery with the view to harnessing the delivery advantages of both lipid and polymer vectors while enjoying the unique structural features of dendrimers.These amphiphilic dendrimer vectors are lipid/dendrimer hybrids and are thus able to mimic lipid vectors and exploit membranefusion-mediated delivery,while simultaneously retaining the multivalent properties of polymer vectors that allow endocytosis-based delivery.In addition,they have precisely controllable and stable nanosized chemical structures and offer nanotechnology-based delivery.Effective amphiphilic dendrimer vectors share two important elements:chemical hydrophilic entities to bind RNA and RNA complex-stabilizing hydrophobicity.These two combined features allow the encapsulation of RNA within a stable complex before its release into the cytosol following endocytosis.This hydrophilic/hydrophobic balance permitted by the structural features of amphiphilic dendrimers plays a determining role in RNA delivery success.In this Account,we provide a conceptual overview of this exciting field with the latest breakthroughs and key advances in the design of amphiphilic dendrimers for the delivery of siRNA and mRNA.Specifically,we start with a short introduction to siRNA-and mRNA-based therapeutics and their delivery challenges.We then outline the pioneering and representative studies on amphiphilic dendrimer vectors to highlight their historical development and promising features that offer to facilitate the once challenging RNA delivery.We conclude by offering perspectives for the future of amphiphilic dendrimer vectors for nucleic acid delivery in general.Jiaxuan Chen Dandan Zhu Xiaoxuan Liu Ling Peng 2022Accounts of Materials Research2022,3,5:1

10Effects of iron precursors on the structure and catalytic performance of iron molybdate prepared by mechanochemical route for methanol to formaldehyde显示文摘Mechanochemical synthesis has been applied for many novel material preparations and gained more and more attention due to green and high-efficiency recently. In order to explore the influences of iron precursors on structure and performance of iron molybdate catalyst prepared by mechanochemical route, three typical and cheap iron precursors have been used in preparation of iron molybdate catalyst. Many characterization methods have been employed to obtain the physical and chemical properties of iron molybdate catalyst. Results indicate that iron precursors have the significant impact on the phase composition, crystal morphology and catalytic performance in the conversion of methanol to formaldehyde. It is hard to regulate the phase composition by changing Mo/Fe mole ratios for Fe2(SO4)3 as iron precursor. In addition, as for Fe2(SO4)3, the formaldehyde yield is lower than that from iron molybdate catalyst prepared with Fe(NO3)3·9H2O due to the reduction in Fe2(MoO4)3 phase as active phase. Based on mechanochemical and coprecipitation method, the solvent water could be a key factor for the formation of MoO3 and Fe2(MoO4) for FeCl3·6H2O and Fe2(SO4)3 as precursors. 12Excessive branched-chain amino acid accumulation restricts mesenchymal stem cell-based therapy efficacy in myocardial infarction显示文摘Mesenchymal stem cells(MSCs)delivered into the post-ischemic heart milieu have a low survival and retention rate,thus restricting the cardioreparative efficacy of MSC-based therapy.Chronic ischemia results in metabolic reprogramming in the heart,but little is known about how these metabolic changes influence implanted MSCs.Here,we found that excessive branched-chain amino acid(BCAA)accumulation,a metabolic signature seen in the post-ischemic heart,was disadvantageous to the retention and cardioprotection of intramyocardially injected MSCs.Discovery-driven experiments revealed that BCAA at pathological levels sensitized MSCs to stress-induced cell death and premature senescence via accelerating the loss of histone 3 lysine 9 trimethylation(H3K9me3).A novel mTORC1/DUX4/KDM4E axis was identified as the cause of BCAA-induced H3K9me3 loss and adverse phenotype acquisition.Enhancing BCAA catabolic capability in MSCs via genetic/pharmacological approaches greatly improved their adaptation to the high BCAA milieu and strengthened their cardioprotective efficacy.We conclude that aberrant BCAA accumulation is detrimental to implanted MSCs via a previously unknown metabolite-signaling-epigenetic mechanism,emphasizing that the metabolic changes of the post-ischemic heart crucially influence the fate of implanted MSCs and their therapeutic benefits.Fuyang Zhang Guangyu Hu Xiyao Chen Ling Zhang Lanyan Guo Congye Li Hang Zhao Zhe Cui Xiong Guo Fangfang Sun Dandan Song Wenjun Yan Yunlong Xia Shan Wang Miaomiao Fan Ling Tao 2022Signal Transduction and Targeted Therapy2022,7,7:1
13Molecularly imprinted nanoparticles and their releasing properties, bio-distribution as drug carriers显示文摘Molecular imprinted nanoparticles(MINPs) can memorize the shape and functional group positions complementary to template, which account for the large drug loading capacity and slow drug release behavior as drug carriers. We synthesized MINPs via precipitation polymerization with vinblastine(VBL) as a model drug, and investigated the drug loading,releasing property in vitro and bio-distribution in vivo. The obtained MINPs, from 300 to 450 nm,had smooth surface and favorable dispersibility. The entrapment efficacy and drug loading capacity of VBL loaded MINPs(MINPs-VBL) were 83.25% and 8.72% respectively. In PBS(pH 7.4),MINPs-VBL showed sustained release behavior. The cumulative release percentage reached about 70% during 216 h and no burst release was observed. The releasing behavior of MINPsVBL in vitro conformed to the first-order kinetics model. MINPs-VBL and commercially available vinblastine sulfate injection(VBL injection) were injected via tail vein of SD rats respectively to investigate the bio-distribution. MINPs-VBL group showed higher concentration of VBL in tissues and serum than VBL injection group after 60 min, and the drug level in liver was the highest. 14Rapid bioluminescence assay for monitoring rat CES1 activity and its alteration by traditional Chinese medicines显示文摘In traditional Chinese medicine herbs(TCM),including Radix Salviae Miltiorrhizae(Danshen),Radix Puerariae Lobatae(Gegen),Radix Angelicae Sinensis(Danggui),and Rhizoma Chuanxiong(Chuanxiong)are widely used for the prevention and treatment of cardiovascular diseases and also often co-administered with Western drugs as a part of integrative medicine practice.Carboxylesterase 1(CES1)plays a pivotal role in the metabolisms of pro-drugs,Since(S)-2-(2-(6-dimethylamino)-benzothiazole)-4,5-dihydrothiazole-4-carboxylate(NLMe)has recently been identified by us as a selective CES1 bioluminescent sensor,we developed a rapid method using this substrate for the direct measurement of CES1 activity in rats.This bioluminescence assay was applied to determine CES1 activity in rat tissues after a two-week oral administration of each of the four herbs noted above.The results demonstrated the presence of CES1 enzyme in rat blood and all tested tissues with much higher enzyme activity in the blood,liver,kidney and heart than that in the small intestine,spleen,lung,pancreas,brain and stomach.In addition,the four herbs showed tissue-specific effects on rat CES1 expression.Based on the CES1 biodistribution and its changes after treatment in rats,the possibility that Danshen,Gegen and Danggui might alter CES1 activities in human blood and kidney should be considered.In summary,a selective and sensitive bioluminescence assay was developed to rapidly evaluate CES1 activity and the effects of orally administered TCMs in rats.Jun Zhang Dandan Wang Liwei Zou Min Xiao Yufeng Zhang Ziwei Li Ling Yang Guangbo Ge Zhong Zuo 2020Journal of Pharmaceutical Analysis2020,10,3:1

17A Study on the Ultrasonic Extraction and Antioxidant Activity of Lychee Pericarp Polysaccharides显示文摘The aim was to optimize ultrasonic extraction conditions and study antioxidant activities of lychee pericarp polysaccharides. The effects of extraction temperature,time,ultrasonic power,solid-liquid ratio on polysaccharides yield were investigated by single-factor experiment and orthogonal experiment to obtain optimum extraction conditions; the antioxidant activities of lychee pericarp polysaccharides were evaluated by the total antioxidant activity. Optimum extraction process was under extraction temperature of 80 ℃,extraction time of 2. 5 h,ultrasonic power of 180 W,solid-liquid ratio of 1∶ 25 g / m L; the total antioxidant activity was increased with the increasing concentration of polysaccharides. The extraction process is simple and efficient. 20Cancer cell membrane-coated bacterial ghosts for highly efficient paclitaxel delivery against metastatic lung cancer显示文摘Chemotherapy is one of the major approaches for the treatment of metastatic lung cancer,although it is limited by the low tumor delivery efficacy of anticancer drugs.Bacterial therapy is emerging for cancer treatment due to its high immune stimulation effect;however,excessively generated immunogenicity will cause serious inflammatory response syndrome.Here,we prepared cancer cell membrane-coated liposomal paclitaxel-loaded bacterial ghosts(LP@BG@CCM)by layer-by-layer encapsulation for the treatment of metastatic lung cancer.The preparation processes were simple,only involving film formation,electroporation,and pore extrusion.LP@BG@CCM owned much higher 4T1 cancer cell toxicity than LP@BG due to its faster fusion with cancer cells.In the 4T1 breast cancer metastatic lung cancer mouse models,the remarkably higher lung targeting of intravenously injected LP@BG@CCM was observed with the almost normalized lung appearance,the reduced lung weight,the clear lung tissue structure,and the enhanced cancer cell apoptosis compared to its precursors.Moreover,several major immune factors were improved after administration of LP@BG@CCM,including the CD4^(+)/CD8a^(+)T cells in the spleen and the TNF-α,IFN-γ,and IL-4 in the lung.LP@BG@CCM exhibits the optimal synergistic chemo-immunotherapy,which is a promising medication for the treatment of metastatic lung cancer.Dandan Ling Xueli Jia Ke Wang Qiucheng Yan Bochuan Yuan Lina Du Miao Li Yiguang Jin 2024Acta Pharmaceutica Sinica B2024,14,1:0
