| 1 | Comparison of Histological, Microbiological, and Molecular Methods in Diagnosis of Patients with TBLN Having Different Anti-TB Treatment Background显示文摘Objective The influence of anti-tuberculosis(TB) treatment history on tuberculous lymphadenitis(TBLN) diagnosis is unclear. Therefore, this study aims to evaluate the diagnostic methods, including histology, microbiology, and molecular tests, used for TBLN. Methods In this study, suspected patients with TBLN and having different anti-TB treatment background were enrolled. All the samples were tested simultaneously by histology, Ziehl-Neelsen(ZN) staining, mycobacterial culture(culture), Xpert MTB/RIF(xpert), real-time PCR, and high-resolution melting curve PCR(HRM). Thereafter, the performance of these methods on samples with different anti-TB treatment background was assessed. Results In our study, 89 patients were prospectively included 82 patients with TBLN and 7 with other diseases. The overall sensitivities of Xpert, real-time PCR, histology, ZN staining, and culture were 86.6%, 69.5%, 58.5%, 43.9%, and 22.0%, respectively. The anti-TB treatment history revealed dramatic influences on the sensitivity of culture(P < 0.0001). In fact, the treatment that lasted over 3 months also influenced the sensitivity of Xpert(P < 0.05). However, the treatment history did not affect the performance of remaining tests(P > 0.05). For rifampicin drug susceptibility test(DST), the anti-TB treatment showed only significant influence on the success rate of culture DST(P = 0.001), but not on those of Xpert and HRM tests(P > 0.05). | 3 | Cis-2-dodecenoic Acid Mediates Its Synergistic Effect with Triazoles by Interfering with Efflux Pumps in Fluconazole-resistant Candida albicans显示文摘Objective To evaluate the synergy of the Burkholderia signaling molecule cis-2-dodecenoic acid(BDSF) and fluconazole(FLU) or itraconazole(ITRA) against two azole-resistant C. albicans clinical isolates in vitro and in vivo. Methods Minimum inhibitory concentrations(MICs) of antibiotics against two azole-resistant C. albicans were measured by the checkerboard technique, E-test, and time-kill assay. In vivo antifungal synergy testing was performed on mice. Analysis of the relative gene expression levels of the strains was conducted by quantitative reverse-transcription polymerase chain reaction(qR T-PCR). Results BDSF showed highly synergistic effects in combination with FLU or ITRA with a fractional inhibitory concentration index of ≤ 0.08. BDSF was not cytotoxic to normal human foreskin fibroblast cells at concentrations of up to 300 μg/mL. The qR T-PCR results showed that the combination of BDSF and FLU/ITRA significantly inhibits the expression of the efflux pump genes CDR1 and MDR1 via suppression of the transcription factors TAC1 and MRR1, respectively, when compared with FLU or ITRA alone. No dramatic difference in the mR NA expression levels of ERG1, ERG11, and UPC2 was found, which indicates that the drug combinations do not significantly interfere with UPC2-mediated ergosterol levels. In vivo experiments revealed that combination therapy can be an effective therapeutic approach to treat candidiasis. | 4 | Diagnosis,treatment,and prevention of severe acute hepatitis of unknown etiology in children显示文摘Background Severe acute hepatitis of unknown etiology in children has recently exhibited a global trend of concentrated occurrence.This review aimed to summarize the current available information regarding the outbreak of severe acute hepatitis and introduce our hospital’s previous experiences with the diagnosis and treatment of severe acute hepatitis for reference.Data sources Websites including the UK Health Security Agency,European Centre for Disease Prevention and Control,CDC,WHO,and databases including PubMed/Medline,Cochrane Library,Embase and Web of Science were searched for articles on severe acute hepatitis in children.Results As of May 26,2022,a total of 650 cases have been reported in 33 countries;at least 38(6%)children required liver transplantation,and nine(1%)died.Cases are predominantly aged between 3 and 5 years old,and there are no epidemiological links among them.The common manifestations are jaundice,vomiting and pale stools.Adenovirus tested positive in most cases,and SARS-CoV-2 and other viruses were detected in a few cases,but virus particles were not found in liver tissue.Adenovirus immunohistochemistry showed immunoreactivity in the intrasinusoidal lumen from some liver samples.The hierarchical treatment includes symptomatic and supportive therapy,management of coagulation disorders and hepatic encephalopathy,artifcial liver support,and liver transplantation(approximately 6%–10%of cases require liver transplant).Conclusions The etiology of this severe acute hepatitis in children is not clear.The clinical features are severe acute hepatitis with signifcantly elevated liver enzymes.Clinicians need to be alert to children with hepatitis. | Ying‑Hu Chen Jin‑Gan Lou Zi‑Hao Yang Qing‑Jiang Chen Chun‑Zhen Hua Sheng Ye Chen‑Mei Zhang Jie Chen Zong‑Wei Huang Jin‑Dan Yu Zhi‑Gang Gao Qiang Shu | 2022 | World Journal of Pediatrics2022,18,8: | 2 |

| 10 | Improvement of the SST γ-Re_(θt) transition model for flows along a curved hydrofoil显示文摘The transition in the boundary-layer flow affects the hydrodynamic performances of hydraulic machineries,as the key components in the ship propulsion system.The shear stress transfer(SST)γ-Re_(θt) transition model is an important prediction tool in the boundary layer simulation for hydrofoils.The present paper improves the prediction accuracy of the SST γ-Re_(θt) model for the boundary layers along a curved hydrofoil.The SST γ-Re_(θt) transition model for the flows along a curved hydrofoil is improved by introducing a correction to the transition onset Reynolds number Reθt.First,the transition onset locations for the flows along the hydrofoils of different curvatures are obtained by the large eddy simulation and by using the SST γ-Re_(θt) model.Then,the transition onset Reynolds numbers Reθt in the SST γ-Re_(θt) model is modified to ensure that the predicted boundary layer parameters are consistent with the large eddy simulation(LES)results.The correlation function between the curvature ratio and the modified transition onset Reynolds number is obtained and subsequently used as a correction function in the original SST γ-Re_(θt) model.Three test cases are used to evaluate the performance of the improved SST γ-Re_(θt) model.For the NACA0035 hydrofoil with a large curvature,the predicted results obtained by using the improved SST γ-Re_(θt) model are quite consistent with the experimental data,which indicates the advantages of the improved model in predicting the boundary layer transition along a hydrofoil.In the test cases of the NACA0016 hydrofoil with a mild curvature and the NACA66(mod)-312 hydrofoil,the prediction results of the improved model are in good agreement with the experimental results in terms of the wake region and the boundary layer parameters,which indicates that the improved SST γ-Re_(θt) model can serve as a powerful tool in the design and the optimization of hydraulic machineries such as the waterjet pumps or the naval propellers. | Chang-liang Ye Chao-yue Wang Dan Zi Yuan Tang Bart.P.M.van Esch Fu-jun Wan | 2021 | Journal of Hydrodynamics2021,33,3: | 1 |

| 20 | Leaf Morphology Genes SRL1 and RENL1 Co-Regulate Cellulose Synthesis and Affect Rice Drought Tolerance显示文摘The morphological development of rice(Oryza sativa L.)leaves is closely related to plant architecture,physiological activities,and resistance.However,it is unclear whether there is a co-regulatory relationship between the morphological development of leaves and adaptation to drought environment.In this study,a drought-sensitive,roll-enhanced,and narrow-leaf mutant(renl1)was induced from a semi-rolled leaf mutant(srl1)by ethyl methane sulfonate(EMS),which was obtained from Nipponbare(NPB)through EMS.Map-based cloning and functional validation showed that RENL1 encodes a cellulose synthase,allelic to NRL1/OsCLSD4.The RENL1 mutation resulted in reduced vascular bundles,vesicular cells,cellulose,and hemicellulose contents in cell walls,diminishing the water-holding capacity of leaves.In addition,the root system of the renl1 mutant was poorly developed and its ability to scavenge reactive oxygen species(ROS)was decreased,leading to an increase in ROS after drought stress.Meanwhile,genetic results showed that RENL1 and SRL1 synergistically regulated cell wall components.Our results revealed a theoretical basis for further elucidating the molecular regulation mechanism of cellulose on rice drought tolerance,and provided a new genetic resource for enhancing the synergistic regulation network of plant type and stress resistance,thereby realizing simultaneous improvement of multiple traits in rice. | LIU Dan ZHAO Huibo WANG Zi’an XU Jing LIU Yiting WANG Jiajia CHEN Minmin LIU Xiong ZHANG Zhihai CEN Jiangsu ZHU Li HU Jiang REN Deyong GAO Zhenyu DONG Guojun ZHANG Qiang SHEN Lan LI Qing QIAN Qian HU Songping ZHANG Guangheng | 2024 | Rice science2024,31,1: | 0 |
