1Wnt/b-catenin signaling plays an ever-expanding role in stem cell self-renewal,tumorigenesis and cancer chemoresistance显示文摘Wnt signaling transduces evolutionarily conserved pathways which play important roles in initiating and regulating a diverse range of cellular activities,including cell proliferation,calcium homeostasis,and cell polarity.The role of Wnt signaling in controlling cell proliferation and stem cell self-renewal is primarily carried out through the canonical pathway,which is the best-characterized the multiple Wnt signaling branches.The past 10 years has seen a rapid expansion in our understanding of the complexity of this pathway,as many new components of Wnt signaling have been identified and linked to signaling regulation,stem cell functions,and adult tissue homeostasis.Additionally,a substantial body of evidence links Wnt signaling to tumorigenesis of cancer types and implicates it in the development of cancer drug resistance.Thus,a better understanding of the mechanisms by which dysregulation of Wnt signaling precedes the development and progression of human cancer may hasten the development of pathway inhibitors to augment current therapy.This review summarizes and synthesizes our current knowledge of the canonical Wnt pathway in development and disease.We begin with an overview of the components of the canonical Wnt signaling pathway and delve into the role this pathway has been shown to play in stemness,tumorigenesis,and cancer drug resistance.Ultimately,we hope to present an organized collection of evidence implicating Wnt signaling in tumorigenesis and chemoresistance to facilitate the pursuit of Wnt pathway modulators that may improve outcomes of cancers in which Wnt signaling contributes to aggressive disease and/or treatment resistance.Maryam K.Mohammed Connie Shao Jing Wang Qiang Wei Xin Wang Zachary Collier Shengli Tang Hao Liu Fugui Zhang Jiayi Huang Dan Guo Minpeng Lu Feng Liu Jianxiang Liu Chao Ma Lewis L.Shi Aravind Athiviraham Tong-Chuan He Michael J.Lee 2016Genes & Diseases2016,3,1:70

3Pathways of China's PM;air quality 2015–2060 in the context of carbon neutrality显示文摘Clean air policies in China have substantially reduced particulate matter(PM;) air pollution in recent years,primarily by curbing end-of-pipe emissions.However,reaching the level of the World Health Organization(WHO) guidelines may instead depend upon the air quality co-benefits of ambitious climate action.Here,we assess pathways of Chinese PM;air quality from 2015 to 2060 under a combination of scenarios that link global and Chinese climate mitigation pathways(i.e.global 2℃-and 1.5℃-pathways,National Determined Contributions(NDC) pledges and carbon neutrality goals) to local clean air policies.We find that China can achieve both its near-term climate goals(peak emissions) and PM;air quality annual standard(35 μg/m;) by 2030 by fulfilling its NDC pledges and continuing air pollution control policies.However,the benefits of end-of-pipe control reductions are mostly exhausted by 2030,and reducing PM;exposure of the majority of the Chinese population to below 10 μg/m;by 2060 will likely require more ambitious climate mitigation efforts such as China’ s carbon neutrality goals and global1.5℃-pathways.Our results thus highlight that China’s carbon neutrality goals will play a critical role in reducing air pollution exposure to the level of the WHO guidelines and protecting public health.Jing Cheng Dan Tong Qiang Zhang Yang Liu Yu Lei Gang Yan Liu Yan Sha Yu Ryna Yiyun Cui Leon Clarke Guannan Geng Bo Zheng Xiaoye Zhang Steven JDavis Kebin He 2021National Science Review2021,8,12:23

9USP19 suppresses inflammation and promotes M2-like macrophage polarization by manipulating NLRP3 function via autophagy显示文摘Macrophage polarization to proinflammatory M1-like or anti-inflammatory M2-like cells is critical to mount a host defense or repair tissue.The exact molecular mechanisms controlling this process are still elusive.Here,we report that ubiquitin-specific protease 19(USP19)acts as an anti-inflammatory switch that inhibits inflammatory responses and promotes M2-like macrophage polarization.USP19 inhibited NLRP3 inflammasome activation by increasing autophagy flux and decreasing the generation of mitochondrial reactive oxygen species.In addition,USP19 inhibited the proteasomal degradation of inflammasome-independent NLRP3 by cleaving its polyubiquitin chains.USP19-stabilized NLRP3 promoted M2-like macrophage polarization by direct association with interferon regulatory factor 4,thereby preventing its p62-mediated selective autophagic degradation.Consistent with these observations,compared to wild-type mice,Usp19−/−mice had decreased M2-like macrophage polarization and increased interleukin-1βsecretion,in response to alum and chitin injections.Thus,we have uncovered an unexpected mechanism by which USP19 switches the proinflammatory function of NLRP3 into an anti-inflammatory function,and suggest that USP19 is a potential therapeutic target for inflammatory interventions.Tao Liu Liqiu Wang Puping Liang Xiaojuan Wang Yukun Liu Jing Cai Yuanchu She Dan Wang Zhi Wang Zhiyong Guo Samuel Bates Xiaojun Xia Junjiu Huang Jun Cui 2021Cellular & Molecular Immunology2021,18,10:15

11Tailoring color emissions from N-doped graphene quantum dots for bioimaging applications显示文摘Unlike inorganic quantum dots,fluorescent graphene quantum dots(GQDs)display excitation-dependent multiple color emission.In this study,we report N-doped GQDs(N-GQDs)with tailored single color emission by tuning p-conjugation degree,which is comparable to the inorganic quantum dot.Starting from citric acid and diethylenetriamine,as prepared N-GQDs display blue,green,and yellow light emission by changing the reaction solvent from water,dimethylformamide(DMF),and solvent free.The X-ray photoelectron spectroscopy,ultraviolet-visible spectra results clearly show the N-GQDs with blue emission(N-GQDs-B)have relatively short effective conjugation length and more carboxyl group because H_(2)O is a polar protic solvent,which tends to donate proton to the reagent to depress the H_(2)O elimination reaction.On the other hand,the polar aprotic solvent(DMF)cannot donate hydrogen,the elimination of H_(2)O is promoted and more nitrogen units enter GQD framework.With the increase of effective p-conjugation length and N content,the emission band of N-GQDS red-shifts to green and yellow.We also demonstrate that N-GQDs could be a potential great biomarker for fluorescent bioimaging.Dan Qu Min Zheng Jing Li Zhigang Xie Zaicheng Sun 2015Light(Science & Applications)2015,4,1:13
12Prognostic evaluation of postoperative adjuvant therapy for operable cervical cancer:10 years'experience of National Cancer Center in China显示文摘Objective: The aim of this study was to investigate the prognostic factors and to evaluate the impact of adjuvant therapy on clinical outcome for early-stage cervical cancer.Methods: The clinical-pathological data of all 1,335 patients with the International Federation of Gynecology and Obstetrics(FIGO) Ib–IIa cervical cancer treated with primary radical surgery at the Chinese National Cancer Center between May 2007 and Dec 2013 were retrospectively reviewed.The median follow-up was 70 months.Results: Of all the patients,61.6% of the cases received adjuvant therapy,with 5-year disease-free survival(DFS)of 92.1% and 5-year overall survival(OS) of 95.0%.In multivariate analysis,differentiation of G3(P<0.05),lymph node metastasis(LNM,P<0.05) and lymphovascular space invasion(LVSI,P<0.05) were independent predictors for OS,while LNM(P<0.05),deep stroma invasion(DSI,P<0.05) and LVSI(P<0.05) were independent factors for DFS.The samples were stratified by histologic type,and cervical squamous cell carcinoma(SCC) was found to share the same independent factors except for differentiation of OS.As to patients with cervical adenocarcinoma/adenosquamous carcinoma(AC/ASC),differentiation was the independent predictor of OS(P<0.05); and LVSI of DFS(P<0.05).Of 236 patients with high-risk factors,there was no significant difference in survival between concurrent chemoradiotherapy(CCRT,n=195),radiotherapy(RT,n=24),and chemotherapy(CT,n=17).Among the 190 patients with LNM who underwent CCRT,124 cases showed improved DFS after sequential CT(P=0.118),with a recurrence rate decrease of 14%,though the difference was not statistically significant.Patients with single intermediate-risk factors like DSI or LVSI were found to partially benefit from adjuvant therapy,but the difference was not statistically significant.Conclusions: LNM,LVSI,DSI and differentiation were found to be independent prognostic factors for operable cervical cancer.Aggressive postoperative adjuvant therapy based on single risk factors in Chinese National Cancer Center could benefit survival.CCRT+CT outperformed CCRT in high-risk patients.For patients with single non-high-risk factor,the role of adjuvant therapy needs to be further discussed.Tong Shu Dan Zhao Bin Li Yating Wang Shuanghuan Liu Pingping Li Jing Zuo Ping Ba i RongZhang Lingying Wu 2017Chinese Journal of Cancer Research2017,29,6:12

14Effect of point application on chronic obstructive pulmonary disease in stationary phase and effects on pulmonary function:A systematic evaluation of randomized controlled trials显示文摘OBJECTIVE:To evaluate clinical efficacy of point application or adjuvant therapy on chronic obstructive pulmonary disease in stationary phase and effects on pulmonary functions.METHODS:Computer retrieved CNKI,VIP,CBM and other databanks and manual operations retrieved correlative literatures to find randomized controlled trials(RCTs) about comparison between point application or adjuvant therapy and no-point-applications for treatment of chronic obstructive pulmonary disease in stationary phase in China.RevMan 5.0 software was used for Meta analysis.RESULTS:Among 3481 cases in the inclusive 32 RCTS,1780 cases were in the test group and 1701 cases in the control group.Meta analysis indicated:1) clinical efficacy:the groups containing point application therapy all were better than the groups of no-point-application;2) force vital capacity(FVC):There was no statistically significant difference between the group of point application plus Western Medicine and the Western Medicine group;3) force expiratory volume 1(FEV1):The groups containing point application therapy were better than the no-point-application;4) FEV1%:the groups of point application plus Western Medicine were better than the Western Medicine groups;5) FEV1/FVC:there was a significant difference between the group of point application plus Chinese drugs and the group of Chinese drug.CONCLUSION:Point application can increase clinical efficacy of chronic obstructive pulmonary disease in stationary phase in varying degrees,and different combinations of point application with Chinese drugs or Western Medicines have incomplete same actions in improvement of pulmonary function and therapeutic effect.Fengsen Li Zhen Gao Jing Jing Dan Xu Halmurat.Upur 2012Journal of Traditional Chinese Medicine2012,32,4:12
15Garcinol inhibits esophageal cancer metastasis by suppressing the p300 and TGF-β1 signaling pathways显示文摘Metastasis causes the main lethality in esophageal cancer patient.Garcinol,a natural compound extracted from Gambogic genera,is a histone acetyltransferase(HAT)inhibitor that has shown anticancer activities such as cell cycle arrest and apoptosis induction.In this study,we investigated the effects of garcinol on the metastasis of esophageal cancer in vitro and in vivo.We found that garcinol(5-15μM)dose-dependently inhibited the migration and invasion of human esophageal cancer cell lines KYSE150 and KYSE450 in wound healing,transwell migration,and Matrigel invasion assays.Furthermore,garcinol treatment dose-dependently decreased the protein levels of p300/CBP(transcriptional cofactors and HATs)and p-Smad2/3 expression in the nucleus,thus impeding tumor cell proliferation and metastasis.Knockdown of p300 could inhibit cell metastasis,but CBP knockdown did not affect the cell mobility.It has been reported that TGF-β1 stimulated the phosphorylation of Smad2/3,which directly interact with p300/CBP in the nucleus,and upregulating HAT activity of p300.We showed that garcinol treatment dose-dependently suppressed TGF-β1-activated Smad and non-Smad pathway,inhibiting esophageal cancer cell metastasis.In a tail vein injection pulmonary metastasis mouse model,intraperitoneal administration of garcinol(20 mg/kg)or 5-FU(20 mg/kg)significantly decreased the number of lung tumor nodules and the expression levels of Ki-67,p300,and p-Smad2/3 in lung tissues.In conclusion,our study demonstrates that garcinol inhibits esophageal cancer metastasis in vitro and in vivo,which might be related to the suppression of p300 and TGF-β1 signaling pathways,suggesting the therapeutic potential of Garcinol for metastatic tumors.Jing Wang Man Wu Dan Zheng Hong Zhang Yue Lv Li Zhang Hong-sheng Tan Hua Zhou Yuan-zhi Lao Hong-xi Xu 2020Acta Pharmacologica Sinica2020,41,1:12
16A preliminary study of the effects of plastic film-mulched raised beds on soil temperature and crop performance of early-sown short-season spring maize(Zea mays L.) in the North China Plain显示文摘To identify a strategy for earlier sowing and harvesting of spring maize(Zea mays L.) in an alternative maize–maize double cropping system, a 2-year field experiment was performed at Quzhou experimental station of China Agricultural University in 2014 and 2015. A short-season cultivar, Demeiya number 1(KX7349), was used in the experiment. Soil temperature to 5 cm depth in the early crop growth stage, crop growth, crop yield, and water use of different treatments(plastic film-mulched raised bed(RF) and flat field without plastic film mulching(CK) in 2014; RF, plastic film-mulched flat field(FF), and CK in 2015)were measured or calculated and compared. Soil temperature in the film-mulched treatments was consistently higher than that in CK(1.6–3.5 °C in average) during the early growth stage. Crops in plastic film-mulched treatments used 214 fewer growing-degree days(GDDs) in 2014 and 262 fewer GDDs in 2015. In 2014, the RF treatment yielded 32.7%higher biomass than CK, although its 9.4% higher grain yield was not statistically significant. Also, RF used 17.9% less water and showed 33.1% higher water use efficiency(WUE) than CK. In 2015, RF and FF showed 56.2% and 49.5% higher yield, 15.0% and 4.5%lower water use(ET), and 63.4% and 75.7% higher WUE, respectively, than CK. RF markedly increased soil temperature in the early crop season, accelerated crop growth, reduced ET,and greatly increased crop yield and WUE. Compared with FF, RF had no obvious effect on crop growth rate, although soil temperature during the period between sowing and stem elongation was slightly increased. However, RF resulted in lower ET and higher WUE than FF. 17中国5地无偿献血者HCV流行特征分析显示文摘目的了解我国献血者丙型肝炎病毒(HCV)的初筛阳性率和确证阳性率。方法采用2种不同ELA试剂对2013年6月-2016年12月我国5家血液中心/中心血站献血者血液标本进行ALT、HBsAg、抗-HCV、抗-HIV、抗-TP进行常规筛查,抗-HCV单试剂或双试剂筛查阳性标本进一步采用MP HCV Blot 3. 0进行确证检测。分别估算首次献血者的HCV流行率及重复献血者的HCV发病率,并采用χ~2检验比较不同人群体、不同献血类型之间HCV确证阳性率。结果在1 276 544人次献血样本中,有3 855 (0. 30%)为抗-HCV ELISA初筛反应性,其中只有30. 18%(904/2 995,)确证为抗-HCV阳性。本次研究中,首次献血者总计648607人次,其中1080份样本确证为抗-HCV阳性,即首次献血者HCV流行率为166. 56 (156. 04—177. 08)/100 000人次,年龄、受教育水平、职业均与HCV确证阳性率相关。重复献血者总计627 937人次,其中126份样本确证为抗-HCV阳性,即重复献血者HCV发病率为13. 04(9. 55—17. 81)/100 000人次,其中重复献血者抗-HCV确证阳性率明显高于单采血小板献血者(0. 02%vs0. 01%)。结论本研究表明我国献血者HCV仍处于较高水平流行状态。尽管现在NAT已成为我国血液检测的常规检测项,但仍需持续有效地对我国输血HCV传播风险进行监测和控制。同时,新的规范中规定可采用1种或2种EIA进行筛查,本意是为了提高检测敏感性,但本研究中EIA筛查阳性样本如此低的确证阳性率提示其特异性差,可能会造成许多合格的献血者丢失。因此,优化献血者HCV筛查策略将有助于防止血液浪费。付萍 Liao Dan 吕运来 张洪斌 何伟兰 黄梅 文秀琼 马红丽 何涛 柯玲 吴秉婷 刘鱼 何苗 Liu Jing Don Brambill 王憬惺 Paul Ness Shan Hua 2018中国输血杂志2018,31,9:10
