1Cryospheric Science: research framework and disciplinary system显示文摘Cryospheric changes and their impacts have received increasing concern, extending the research to include the interactions between earth spheres and the impacts of and social adaptation to cryospheric changes. As a result, Cryospheric Science is rapidly developing. However, the research framework of Cryospheric Science has not completely been set up, and the intension and extension of Cryospheric Science are unclear.Following a literature review, the research framework for Cryospheric Science and the discipline components are analysed in this paper. We consider that Cryospheric Science is an inevitable product of international research on the earth and environmental changes as well as on human sustainable development. It will not only strengthen the linkage between the cryosphere and other earth spheres by deepening traditional research areas, but will also bring more focus on the critical roles of the cryosphere in interactions between earth spheres, and will enhance the relationship between cryospheric change and climate, ecology, hydrology, surface environment and sustainable development. The disciplinary tree of Cryospheric Science is developed following the mainstream of change-impacts adaptation. 2The Impacts of Permafrost Change on NPP and Implications:A Case of the Source Regions of Yangtze and Yellow Rivers显示文摘This paper studies the relationship between net primary productivity (NPP) and annual average air temperature (GT) at 0cm above ground in permafrost regions by using revised Chikugo NPP model,cubic spline interpolating functions,and non-linear regression methods.The source regions of the Yangtze and Yellow Rivers were selected as the research areas.Results illustrate that:(1) There is significant non-linear relationship between NPP and GT in various typical years;(2) The maximum value of NPP is 6.17,5.87,7.73,and 5.41 DM·t·hm-2 ·a-1 respectively,and the corresponding GT is 7.1,10.0,21.2,and 8.9 o C respectively in 1980,1990,2000 and 2007;(3) In 1980,the sensitivity of NPP to GT is higher than in 1990,2000 and 2007.This tendency shows that the NPP presents change from fluctuation to an adaptation process over time;(4) During 1980~2007,the accumulated NPP was reduced to 8.05,and the corresponding carrying capacity of theoretical livestock reduced by 11%;(5) The shape of the demonstration region of ecological compensation system,livelihood support system,and science appraisal system in the source regions of Yangtze and Yellow Rivers are an important research for increasing the adaptation capacity and balancing protection and development.FANG Yiping QIN Dahe DING Yongjian YANG Jianping XU Keyan 2011Journal of Mountain Science2011,8,3:9
3Adaptation Management of Mountain Tourism Service: The Case of the Source Regions of the Yangtze and Yellow River显示文摘Mountain areas are often rich in ecological diversity and recreational opportunities. Mountain tourism is thought to be an effective and important means for maintaining and expanding rural economies and, thus, improving the living conditions of rural societies. As mountain tourism service research is a professional field with several disciplines involved, a multi-disciplinary management platform is needed and it facilitates participation in sustainable mountain development by diverse stakeholders. With the source regions of the Yangtze and the Yellow River as a case study, this paper presents a conceptual framework for an adaptation management of mountain tourism services according to technical, policy, social and economic dimensions. 4Harnessing the native type I-B CRISPR-Cas for genome editing in a polyploid archaeon显示文摘Research on CRISPR-Cas(clustered regularly interspaced short palindromic repeats-CRISPR associated protein) systems has led to the revolutionary CRISPR/Cas9 genome editing technique. However, for most archaea and half of bacteria, exploitation of their native CRISPR-Cas machineries may be more straightforward and convenient. In this study, we harnessed the native type I-B CRISPR-Cas system for precise genome editing in the polyploid haloarchaeon Haloarcula hispanica. After testing different designs, the editing tool was optimized to be a single plasmid that carries both the self-targeting miniCRISPR and a 600-800 bp donor. Significantly, chromosomal modifications, such as gene deletion, gene tagging or single nucleotide substitution, were precisely introduced into the vast majority of the transformants. Moreover, we showed that simultaneous editing of two genomic loci could also be readily achieved by one step. 5Thermal behavior simulation of Ni/MH battery显示文摘Thermal behavior of overcharged Ni/MH battery is studied with microcalorimeter. The battery is in-stalled in a special device in a microcalorimeter with a quartz frequency thermometer. Quantity of heat and heat capacity of the battery charged at different state of charge (SOC) at different rates are meas-ured by the microcalorimeter. Based on a series of assumputions, heat transfer equation is set up. Ex-pression of heat generation is attained by curve fitting instead of theoretical calculation. Thermal model is used to simulate thermal behavior of the battery in charging period , results of calculation and ex-periment match very well. The temperature distribution is non-uniform because the poor conductivity limits the heat transfer during charging process. It is difficult to greatly improve the heat conductivity of the battery because it is related to materials inside the battery including electrodes, separators and so on. 14Genomics and proteomics of Apis mellifera filamentous virus isolated from honeybees in China显示文摘Apis mellifera filamentous virus(Am FV)is a large DNA virus that is endemic in honeybee colonies.The genome sequence of the Am FV Swiss isolate(Am FV CH–C05)has been reported,but so far very few molecular studies have been conducted on this virus.In this study,we isolated and purified Am FV(Am FV CN)from Chinese honeybee(Apis mellifera)colonies and elucidated its genomics and proteomics.Electron microscopy showed ovoid purified virions with dimensions of 300–500×210–285 nm,wrapping a 3165×40 nm filamentous nucleocapsid in three figure-eight loops.Unlike Am FV CH–C05,which was reported to have a circular genome,our data suggest that Am FV CN has a linear genome of approximately 493 kb.A total of 197 ORFs were identified,among which36 putative genes including 18 baculoviral homologs were annotated.The overall nucleotide similarity between the CN and CH–C05 isolates was 96.9%.Several ORFs were newly annotated in Am FV CN,including homologs of per os infectivity factor 4(PIF4)and a putative integrase.Phylogenomic analysis placed Am FVs on a separate branch within the newly proposed virus class Naldaviricetes.Proteomic analysis revealed 47 Am FV virionassociated proteins,of which 14 had over 50%sequence coverage,suggesting that they are likely to be main structural proteins.In addition,all six of the annotated PIFs(PIF-0–5)were identified by proteomics,suggesting that they may function as entry factors in Am FV infection.This study provides fundamental information regarding the molecular biology of Am FV.Dahe Yang Jun Wang Xi Wang Fei Deng Qingyun Diao Manli Wang Zhihong Hu Chunsheng Hou 2022Virologica Sinica2022,37,4:1
