| 1 | Macrophage-derived small extracellular vesicles promote biomimetic mineralized collagen-mediated endogenous bone regeneration显示文摘Macrophages play an important role in material-related immune responses and bone formation,but the functionality of macrophage-derived extracellular vesicles(EVs)in material-mediated bone regeneration is still unclear.Here,we evaluated intracellular communication through small extracellular vesicles(sEVs)and its effects on endogenous bone regeneration mediated by biomimetic intrafibrillarly mineralized collagen(IMC).After implantation in the bone defect area,IMC generated more neobone and recruited more mesenchymal stem cells(MSCs)than did extrafibrillarly mineralized collagen(EMC).More CD63+CD90+and CD63+CD163+cells were detected in the defect area in the IMC group than in the EMC group.To determine the functional roles of sEVs,extracellular vesicles from macrophages cultured on different mineralized collagen were isolated,and they showed no morphological differences.However,macrophage-derived sEVs in the IMC group showed an enhanced Young’s modulus and exerted beneficial effects on the osteogenic differentiation of bone marrow MSCs by increasing the expression of the osteoblastic differentiation markers BMP2,BGLAP,COL1,and OSX and calcium nodule formation.Mechanistically,sEVs from IMC-treated macrophages facilitated MSC osteogenesis through the BMP2/Smad5 pathway,and blocking sEV secretion with GW4869 significantly impaired MSC proliferative,immunomodulative and osteogenic potential.Taken together,these findings show that macrophage-derived sEVs may serve as an emerging functional tool in biomaterial-mediated endogenous bone regeneration. | Anqi Liu Shanshan Jin Cuicui Fu Shengji Cui Ting Zhang Lisha Zhu Yu Wang Steve G.F.Shen Nan Jiang Yan Liu | 2020 | International Journal of Oral Science2020,12,4: | 5 |
| 2 | Equivalent efficacy study of QL1101 and bevacizumab on untreated advanced non-squamous non-small cell lung cancer patients: a phase 3 randomized, double-blind clinical trial显示文摘Objective:This phase 3 study aimed to test equivalence in efficacy and safety for QL1101,a bevacizumab analogue in Chinese patients with untreated locally advanced non-squamous non-small cell lung cancer(NSCLC).Methods:Eligible patients were randomly assigned 1:1 to receive carboplatin and paclitaxel in combination with either QL1101 or bevacizumab,15 mg/kg every 3-week for 6 cycles.This was followed by maintenance treatment with single agent QL1101 every 3-week.The primary end-point was objective response rate(ORR),with secondary end-points being progression-free survival(PFS),overall survival(OS),disease control rate(DCR),and adverse events(AEs).Results:Of 675 patients,535 eligible patients were randomized to the QL1101 group(n=269)and bevacizumab group(n=266).ORRs were 52.8%and 56.8%,respectively,for the QL1101 and bevacizumab groups,with an ORR hazard ratio 0.93(95%confidence interval:0.8-0131.1).The PFS,OS,DCR,and AEs were comparable between the 2 groups,which remained the same after stratification according to epidermal growth factor receptor mutation or smoking history.Conclusions:QL1101 showed similar efficacy and safety profiles as compared to bevacizumab among Chinese patients with untreated locally advanced non-squamous NSCLC. | Tianqing Chu Jun Lu Minghong Bi Helong Zhang Wu Zhuang Yan Yu Jianhua Shi Zhendong Chen Xiaochun Zhang Qisen Guo Quan Liu Huijuan Wu Jian Fang Yi Hu Xiuwen Wang Cuicui Han Kai Li Baohui Han | 2021 | Cancer Biology & Medicine2021,18,3: | 4 |

| 4 | Bevacizumab promotes active biological behaviors of human umbilical vein endothelial cells by activating TG Fpi pathways via off-VEGF signaling显示文摘Objective:Bevacizumab is a recombinant humanized monoclonal antibody that blocks vascular endothelial growth factor(VEGF)with clear clinical benefits.However,overall survival of some cancer types remains low owing to resistance to bevacizumab therapy.While resistance is commonly ascribed to tumor cell invasion induced by hypoxia-inducible factor(HIF),less attention has been paid to the potential involvement of endothelial cells(ECs)in vasculature activated by anti-angiogenic drugs.Methods:Human umbilical vein ECs(HUVECs),bEnd.3 cells,and mouse retinal microvascular ECs(MRMECs)were treated with bevacizumab under conditions of hypoxia and effects on biological behaviors,such as migration and tube formation,examined.Regulatory effects on TGFpi and CD 105(endoglin)were established via determination o f protein and mRNA levels.We further investigated whether the effects of bevacizumab could be reversed using the receptor tyrosine kinase inhibitor anlotinib.Results:Bevacizumab upregulated TGFpi as well as CD 105,a component o f the TGFP receptor complex and an angiogenesis promoter.Elevated CD 105 induced activation of Sm adl/5,the inflammatory pathway and endothelial-mesenchymal transition.The migration ability of HUVECs was enhanced by bevacizumab under hypoxia.Upregulation o f CD 105 was abrogated by anlotinib,which targets multiple receptor tyrosine kinases including VEGFR2/3,FGFR1-4,PD G FRα/β,C-Kit,and RET.Conclusions:Bevacizumab promotes migration and tube formation of HUVECs via activation of the TGFβi pathway and upregulation of CD105 expression.Anlotinib reverses the effects of bevacizumab by inhibiting the above signals. | Xiaoling Zhang Yan Zhang Yanan Jia Tingting Qin Cuicui Zhang Yueya Li Chengmou Huang Zhujun Liu Jing Wang Kai Li | 2020 | Cancer Biology & Medicine2020,17,2: | 3 |
| 5 | Tonoplast-associated calcium signaling regulates manganese homeostasis in Arabidopsis显示文摘Manganese(Mn)is an essential micronutrient in plants.However,excessive Mn absorption in acidic and waterlogged soils can lead to Mn toxicity.Despite their essential roles in Mn homeostasis,transcriptional and post-transcriptional modifications of Mn transporters remain poorly understood.Here,we demonstrated that high-Mn stress induces an obvious Ca^(2+) signature in Arabidopsis.We identified four calcium-dependent protein kinases,CPK4/5/6/11,that interact with the tonoplast-localized Mn and iron(Fe)transporter MTP8 in vitro and in vivo.The cpk4/5/6/11 quadruple mutant displayed a dramatic high-Mn-sensitive phenotype similar to that of the mtp8 mutant.CPKs phosphorylated the N-terminal domain of MTP8 primarily at the Ser31 and Ser32 residues.Transport assays combined with multiple physiological experiments on phospho-dead variant MTP8^(S31/32A)and phospho-mimetic variant MTP8^(S31/32D)plants under different Mn and Fe conditions suggested that Ser31 and Ser32 are crucial for MTP8 function.In addition,genetic analysis showed that CPKs functioned upstream of MTP8.In summary,we identified a tonoplast-associated calcium signaling cascade that orchestrates Mn homeostasis and links Mn toxicity,Ca^(2+) signaling,and Mn transporters.These findings provide new insight into Mn homeostasis mechanisms and Ca^(2+) signaling pathways in plants,providing potential targets for engineering heavy metal toxicity-tolerant plants. | Zhenqian Zhang Dali Fu Zhihui Sun Chuanfeng Ju Cuicui Miao Zhangqing Wang Dixiang Xie Liang Ma Zhizhong Gong Cun Wang | 2021 | Molecular Plant2021,14,5: | 2 |

| 7 | Polyploidy Induction of Pteroceltis tatarinowii Maxim.显示文摘[Objective] This study was conducted to obtain tetraploid Pteroceltis tatarinowii Maxim. with excellent ornamental traits. [Method] The stem apex growing points of Pteroceltis tatarinowii Maxim. were treated with different concentrations of colchicine solution for different hours to figure out a proper method and obtain polyploids. [Result] The most effective induction was obtained by treatment with 0.6%-0.8% colchicine for 72 h with 34.2% mutation rate. | 10 | Peripheral blood indices to predict PFS/OS with anlotinib as a subsequent treatment in advanced small-cell lung cancer显示文摘Objective:In the phase II ALTER-1202(NCT03059797)trial,anlotinib significantly improved progression-free survival(PFS)and overall survival(OS)in patients with advanced small-cell lung cancer(SCLC)who underwent at least 2 previous chemotherapy cycles,when compared with a placebo group.To identify potential factors for predicting efficacy and prognosis with anlotinib treatment,we analyzed hematological indices at baseline and adverse events(AEs)over the course of anlotinib treatment.Methods:Data were collected from March 2017 to April 2019 from a randomized,double-blind,placebo-controlled,multicenter,phase II trial of anlotinib.Eligible patients were randomly assigned 2:1 to receive anlotinib or placebo until disease progression,intolerable toxicity,or withdrawal of consent.The patients received anlotinib(12 mg)or an analogue capsule(placebo)orally once daily for 14 days every 3 weeks.The hematological indices at baseline and AEs that occurred in the initial 2 treatment cycles were recorded.The Kaplan-Meier test and Cox regression model were used to assess survival differences.Results:A total of 82 patients(81 patients with complete data)were randomly assigned to receive anlotinib,with 38 receiving a placebo as a control.Multivariate analysis indicated that an elevated neutrophil to lymphocyte ratio>7.75 and lactate dehydrogenase>254.65 U/L at baseline were independent risk factors for PFS;basal elevated aspartate aminotransferase>26.75 U/L,neuron specific enolase>18.64 ng/mL,and fibrinogen>4.645 g/L were independent risk factors for OS.During treatment,elevatedγglutamyltransferase and hypophosphatemia were independent predictors for a poor PFS,and elevatedγ-glutamyl transferase and hypercholesterolemia were independent factors for OS.Conclusions:Our study preliminarily defined potential factors that affected the PFS and OS at baseline and during anlotinib treatment in patients with advanced SCLC.Our findings provide a basis for screening the dominant population and for dynamic efficacy monitoring with anlotinib therapy. | Cuicui Zhang Jing Wang Xinyue Wang Zhaoting Meng Ying Cheng Kai Li | 2022 | Cancer Biology & Medicine2022,19,8: | 1 |

| 12 | Spatiotemporal dynamics of cell abundance,colony size and intracellular toxin concentrations of pelagic and benthic Microcystis in Lake Caohai,China显示文摘Lake Caohai has experienced extensive Microcystis blooms in recent years,and to improve its water quality,the local government carried out a series of water control measures.To better understand the dynamics of both pelagic and benthic Microcystis and their characteristics in Lake Caohai,we conducted a 1-year investigation from December 2015to December 2016 to gain a seasonal outlook on the distribution and dynamics of cell abundance,colony size and intracellular microcystins(MCs)of Microcystis.The results indicated that the Microcystis bloom occupied primarily the northeastern region and then moved gradually from lakeshore to lake center.The perennial southwesterly winds and the water inflow from northeast to southwest in Lake Caohai determined the spatiotemporal distribution of pelagic Microcystis.Benthic Microcystis was mainly distributed in the northeastern region in summer,occupied the lake center in autumn and then occupied the southeastern region in winter,determined by the sedimentation of pelagic Microcystis and the death of benthic Microcystis.Small colonies(20-60μm)overwintered more easily in both water column and sediment.The concentrations of intracellular toxin of benthic Microcystis were observed to be significantly higher than those of pelagic Microcystis.This might be because Microcystis synthesized large amount of MCs to acclimate to an unfavorable benthic environment.This knowledge on the dynamics of Microcystis expands our understanding of mechanisms underpinning the formation of Microcystis blooms. | Bing Feng Chunbo Wang Xingqiang Wu Cuicui Tian Meng Zhang Yingying Tian Bangding Xiao | 2019 | Journal of Environmental Sciences2019,31,10: | 1 |

| 14 | Determination of triglycerides in human serum by near-infrared diffuse reflectance spectroscopy using silver mirror as a substrate显示文摘Near infrared diffuse reflectance spectroscopy(NIRDRS) has gained wide attention due to its convenience for rapid quantitative analysis of complex samples. A method for rapid analysis of triglycerides in human serum using NIRDRS with silver mirror as the substrate is developed. Due to the even and high reflectance of the silver mirror, the spectral response is enhanced and the background interference is reduced.Furthermore, both linear and nonlinear modeling strategies were investigated adopting the partial least squares(PLS) and least squares support vector regression(LS-SVR), continuous wavelet transform(CWT)was used for spectral preprocessing, and variable selection was tried using Monte Carlo uninformative variable elimination(MC-UVE), randomization test(RT) and competitive adaptive reweighted sampling(CARS) for optimization the models. The results show that the determination coefficient(R) between the predicted and reference concentration is 0.9624 and the root mean squared error of prediction(RMSEP) is 0.21. The maximum deviation of the prediction results is as low as 0.473 mmol/L. | 18 | Cryo-EM structure of the human mitochondrial translocase TIM22 complex显示文摘Dear Editor,Mitochondria are essential organelles in cellular metabolism,homeostasis,and apoptosis.1,2 Most mitochondrial proteins are synthesized as precursors in the cytosol and then imported into mitochondria by specific protein translocase complexes,including the translocase of the outer membrane complex(TOM complex),the carrier translocase of the inner membrane complex(TIM22 complex),the presequence translocase of the inner membrane complex(TIM23 complex),the sorting and assembly machinery(SAM complex),and the mitochondrial import complex(MIM complex).3 The TIM22 complex is responsible for the translocation and insertion of hydrophobic membrane proteins,including mitochondrial carrier proteins and translocase subunits(Tim17,Tim22 and Tim23).3 In humans,TIM22 is a 440-kDa complex comprising at least six components:the hypothetical channel-forming protein Tim22,three small Tim proteins(Tim9,Tim10a and Tim10b),Tim29 and acylglycerol kinase(AGK).1 Considering the functional importance of mitochondrial protein import,the TIM22 complex has been linked to many diseases.For example,mutations in the TIM22 gene have been reported to cause early-onset mitochondrial myopathy.4 AGK participates in lipid biosynthesis,and mutations in the AGK gene lead to Sengers syndrome.2 Mutations in the TIMM8A gene(also called DDP1)cause deafness dystonia syndrome. | Liangbo Qi Qiang Wang Zeyuan Guan Yan Wu Cuicui Shen Sixing Hong Jianbo Cao Xing Zhang Chuangye Yan Ping Yin | 2021 | Cell Research2021,31,3: | 1 |

| 20 | Semi-interpenetrating polymer networks toward sulfonated poly(ether ether ketone) membranes for high concentration direct methanol fuel cell显示文摘Low methanol permeability of proton exchange membranes (PEMs) is greatly important for direct methanol fuel cells (DMFCs). Here, sulfonated poly (ether ether ketone) (SPEEK) based semiinterpenetrating polymer networks (semi-IPNs) are successfully prepared by interpenetrating SPEEK into the in-situ synthesized crosslinking networks. The polymeric networks are formed by the covalent bonds between bromobenzyl groups of bro mo methylated poly (phenylene oxide) and amine groups of diamine linkers as well as the ionic bonds between amine species and sulfonated groups. Two linkers without and with sulfonated groups are applied to fabricate the semi-IPNs. The core properties of the membranes, like phase separation, water uptake, proton conductivity and methanol permeability, are systematically studied and compared. The DMFCs assembled by using the semi-IPN membranes display better performance than Nafion 117 in high concentration methanol solutions.
