1Hydrophobicity mechanism of non-smooth pattern on surface of butterfly wing显示文摘Twenty-nine species (24 genera, 6 families) of butterflies typical and common in northeast China were selected to make qualitative and quantitative studies on the pattern, hydrophobicity and hydrophobicity mechanism by means of scanning electron microscopy and contact angle measuring system. The scale surface is composed of submicro-class vertical gibbosities and horizontal links. The distance of scale is 48—91 μm, length 65—150 μm, and width 35—70 μm. The distance of submicro-class vertical gib-bosities on scale is 1.06—2.74 μm, height 200—900 nm, and width 200—840 nm. The better hydropho-bicity on the surface of butterfly wing (static contact angle 136.3°—156.6°) is contributed to the co-effects of micro-class scale and submicro-class vertical gibbosities on the wing surface. 2Pilose antler aqueous extract promotes the proliferation and osteogenic differentiation of bone marrow mesenchymal stem cells by stimulating the BMP-2/Smad1, 5/Runx2 signaling pathway显示文摘Peptides from Pilose antler aqueous extract(PAAE) have been shown to stimulate the proliferation and differentiation of bone marrow mesenchymal stem cells(BMSCs). However, the underlying molecular mechanisms are not well understood. Here, PAAE was isolated and purified to explore the molecular mechanisms underlying PAAE’s effects on BMSCs as well as its osteoprotective effects in ovariectomized rats. Our results showed that PAAE promoted proliferation and differentiation of BMSCs to become osteoblasts by enhancing ALP activity and increasing extracellular matrix mineralization. The trabecular microarchitecture of ovariectomized rats was also found to be protected by PAAE. Quantitative reverse transcription-polymerase chain reaction(Quantitative RT-PCR) results suggest that PAAE also increased the expression of osteogenic markers including, alkaline phosphatase(ALP), runt-related transcription factor 2(Runx2), osteocalcin(OCN), bone morphogenetic protein-2(BMP-2), and collagen I(COL-I). Immunoblotting results indicated that PAAE upregulated the levels of BMP-2 and Runx2 and was associated with Smad1/5 phosphorylation. PAAE A at the concentration of 200μg·mL^-1 showed the strongest effect on proliferation and osteogenic differentiation of BMSCs after 48 h. Using matrix-assisted laser desorption/ionization time of flight mass spectrometry(MALDI-TOF MS), we identified the molecular weight of PAAE A and found that it is less than 3000 Da and showed several significant peaks. In conclusion, PAAE activates the BMP-2/Smad1, 5/Runx2 pathway to induce osteoblastic differentiation and mineralization in BMSCs and can inhibit OVX-induced bone loss. 3Super-hydrophobic characteristics of butterfly wing surface显示文摘Many biological surface are hydrophobic because of their complicated composition and surface microstructure. Eleven species (four families) of butterflies were selected to study their micro-, nano-structure and super-hydrophobic characteristic by means of Confocal Light Microscopy, Scanning Electron Microscopy and Contact Angle Measurement. The contact an- gles of water droplets on the butterfly wing surface were consistently measured to be about 150 ? and 100 ? with and without the squamas, respectively. The dust on the surface can be easily cleaned by moving spherical droplets when the inclining angle is larger than 3 ?. It can be concluded that the butterfly wing surface possess a super-hydrophobic, water-repellent, self-cleaning, or “Lotus-effect”characteristic. The contact angle measurement of water droplets on the wing surface with and without the squamas showed that the water-repellent characteristic is a consequence of the microstructure of the squamas. Each water droplet (diameter 2 mm) can cover about 700 squamas with a size of 40 m×80 m of each squama. The regular riblets with a width of 1000 nm to 1500 nm are clearly observed on each single squama. 4Effects of Methanol on Wettability of the Non-Smooth Surface on Butterfly Wing显示文摘The contact angles of distilled water and methanol solution on the wings of butterflies were determined by a visual contact angle measuring system.The scale structures of the wings were observed using scanning electron microscopy,The influence of the scale micro-and ultra-structure on the wettability was investigated.Results show that the contact angle of distilled water on the wing surfaces varies from 134.0° to 159.2°.High hydrophobicity is found in six species with contact angles greater than 150°.The wing surfaces of some species are not only hydrophobic but also resist the wetting by methanol solution with 55% concentration.Only two species in Parnassius can not resist the wetting because the micro-structure(spindle-like shape) and ultra-structure(pinnule-like shape) of the wing scales are remarkably different from that of other species.The concentration of methanol solution for the occurrence of spreading/wetting on the wing surfaces of different species varies from 70% to 95%.After wetting by methanol solution for 10 min,the distilled water contact angle on the wing surface increases by 0.8°-2.1°,showing the promotion of capacity against wetting by distilled water.Yan Fang Gang Sun Qian Cong Guang-hua Chen Lu-quan Ren 2008Journal of Bionic Engineering2008,5,2:19

6Hydrogen Sulfide Positively Regulates Abscisic Acid Signaling through Persulf idation of SnRK2.6 in Guard Cells显示文摘The phytohormone abscisic acid(ABA)plays pivotal roles in triggering stomatal closure and facilitating adaptation of plants to drought stress.Hydrogen sulfide(H2S),a small signaling gas molecule,is involved in ABA-dependent stomatal closure.However,how H2S regulates ABA signaling remains largely unclear.Here,we show that ABA induces the production of H2S catalyzed by L-CYSTEINE DESULFHYDRASE1(DES1)in guard cells,and H2S in turn positively regulates ABA signaling through persulfidation of Open Stomata 1(OST1)/SNF1-RELATED PROTEIN KINASE2.6(SnRK2.6).Two cysteine(Cys)sites,Cys131 and Cys137,which are exposed on the surface of SnRK2.6 and close to the activation loop,were identified to be persulfidated,which promotes the activity of SnRK2.6 and its interaction with ABA response element-binding factor 2(ABF2),a transcription factor acting downstream of ABA signaling.When Cys131,Cys137,or both residues in SnRK2.6 were substituted with serine(S),H2S_induced SnRK2_6 activity and SnRK2.6-ABF2 interaction were partially(SnRK2.6c131s and SnRK2.6c137S)or completely(SnRK2.6c131sc137S)compromised.Introduction of SnRK2.6c131s,SnRK2.6c137S,or SnRK2.6c131sc137S into the ost1-3 mutant could not rescue the mutant phenotype:less sensitivity to ABA-and H2S-induced stomatal closure and Ca2+influx as well as increased water loss and decreased drought tolerance.Taken together,our study reveals a novel post-translational regulatory mechanism of ABA signaling whereby H2S persulfidates SnRK2.6 to promote ABA signaling and ABA-induced stomatal closure.Sisi Chen Honglei Jia Xiaofeng Wang Cong Shi Xiao Wang Peiyun Ma Juan Wang Meijuan Ren Jisheng Li 2020Molecular Plant2020,13,5:13

8Nutrient deficiency limits population development,yield formation,and nutrient uptake of direct sown winter oilseed rape显示文摘Direct-sowing establishment method has great significance in improving winter oilseed rape(Brassica napus L.) production and guaranteeing edible oil security in China. However, nutrient responses on direct sown winter oilseed rape(DOR) performance and population development dynamic are still not well understood. Therefore, five on-farm experiments were conducted in the reaches of the Yangtze River(RYR) to determine the effects of nitrogen(N), phosphorus(P), and potassium(K) deficiencies on population density, dry matter production, nutrient uptake, seed yield, and yield components of DOR plants. Four fertilization treatments included the balanced NPK application treatment(NPK, 180 kg N, 39.3 kg P, 100 kg K, and 1.8 kg borax ha–1) and three nutrient deficiency treatments based on the NPK treatment, i.e., –N, –P, and –K. The results indicated that DOR population density declined gradually throughout the growing season, especially at over-wintering and pod-development stages. Nutrient deficiency decreased nutrient concentration in DOR plants, limited dry matter production and nutrient uptake, and thereby exacerbated density reduction during plants growth. The poor individual growth and reduced population density together decreased seed yield in the nutrient deficiency treatment. Averaged across all the experiments, seed yield reduced 61% by N deficiency, 38.3% by P deficiency, and 14.4% by K deficiency. The negative effects of nutrient deficiency on DOR performances followed the order of –N>–P>–K, and the effects were various among different nutrient deficiencies. Although N deficiency improved DOR emergence, but it seriously limited dry matter production and nutrient uptake, which in turn led to substantial plants death and therefore resulted in a very low harvested density. The P deficiency significantly reduced initial density, limited plants growth, and exacerbated density reduction. The K deficiency mainly decreased individual growth and yield, but did not affect density dynamic. 9Chemical characteristics of haze particles in Xi'an during Chinese Spring Festival: Impact of fireworks burning显示文摘Fireworks burning releases massive fine particles and gaseous pollutants, significantly deteriorating air quality during Chinese Lunar New Year(LNY) period. To investigate the impact of the fireworks burning on the atmospheric aerosol chemistry, 1-hr time resolution of PM_(2.5) samples in Xi'an during the winter of 2016 including the LNY were collected and detected for inorganic ions, acidity and liquid water content(LWC) of the fine aerosols. PM_(2.5) during the LNY was 167 ± 87 μg/m^3, two times higher than the China National Ambient Air Quality Standard(75 μg/m^3). K^+(28 wt.% of the total ion mass) was the most abundant ion in the LNY period, followed by SO_4^(2-)(25 wt.%) and Cl-(18 wt.%). In contrast, NO_3^-(34 wt.%) was the most abundant species in the haze periods(hourly PM32-2.5> 75 μg/m), followed by SO_4(29.2 wt.%) and NH_4^+(16.3 wt.%), while SO_4^(2-)(35 wt.%) was the most abundant species in the clean periods(hourly PM_(2.5)< 75 μg/m^3), followed by NO_3^-(23.1 wt.%) and NH_4^+(11 wt.%). Being different from the acidic nature in the non-LNY periods, aerosol in the LNY period presented an alkaline nature with a pH value of 7.8 ± 1.3. LWC during the LNY period showed a robust linear correlation with K_2SO_4 and KCl, suggesting that aerosol hygroscopicity was dominated by inorganic salts derived from fireworks burning. 12Crop rotation-dependent yield responses to fertilization in winter oilseed rape(Brassica napus L.)显示文摘Differences in soil physical, chemical and biological properties between paddy–upland and continuous upland rotations will influence nutrient relations and crop growth. With the aim of estimating rapeseed yield performance in response to fertilization in rice–rapeseed(RR) and cotton–rapeseed(CR) rotations, on-farm experiments were conducted at 70 sites across Hubei province, central China. The economically optimal fertilizer rates of winter oilseed rape in different rotations were determined. Field experiments showed that previous crops significantly influenced seed yields. Without N fertilization,seed yields were significantly lower for the RR rotation than for the CR rotation. The average yield increase ratio and agronomic efficiency associated with nitrogen(N)fertilization in the RR rotation were 96.6% and 6.56 kg kg- 1, significantly higher than those in the CR rotation. No seed yield differences were detected between the two rotations under phosphorus(P) and potassium(K) fertilization. In contrast to the CR rotation, N fertilizer played a more vital role in maintaining high seed yields in the RR rotation owing to the lower indigenous soil N supply. Compared with local N fertilizer recommendation rates for the RR rotation, on average an additional 18 kg N ha- 1was recommended according to the economically optimal N fertilizer rate(EONFR). In contrast, the EONFR was 14 kg N ha- 1lower than the locally recommended N fertilizer rate for the CR rotation. There were no differences between the two rotations for the average economically optimal P and K fertilization rates. 14High-throughput low-delay MAC protocol for Tera Hertz ultra-high data-rate wireless networks显示文摘To address the problems of the present Tera Hertz medium access control(MAC) protocols such as not updating the time slot requests numbers in time, unreasonable superframe structures and not merging time slot requests from the same pair of nodes, high throughput low delay medium access control(HLMAC), a novel MAC protocol for Tera Hertz ultra-high data-rate wireless networks is proposed. It reduces the data access delay largely with a new superframe structure, from which nodes can get time slot allocation information immediately. The network throughput is also improved with the help of updating time slot requests number and merging time slot requests from the same pair of nodes. 16Mutation analysis of PAX6 gene in a large Chinese family with aniridia显示文摘Background Mutations in PAX6 gene have been shown to be the genetic cause of aniridia, which is a severe panocular eye disease characterised by iris hypoplasia. However, there is no study to do genetic analysis of aniridia, although there are several case reports in China. Here, we describe a mutation analysis of PAX6 in a large Chinese family with aniridia. Methods Genomic DNA from venous blood samples was prepared. Haplotype analysis was performed with two genetic markers (D11S904 and D11S935). Fourteen exons of the PAX6 gene were amplified from genomic DNA. Polymerase chain reaction (PCR) products of each exon were analysed by single strand conformational polymorphism (SSCP). The PCR products having an abnormal pattern were sequenced to confirm the mutation.Results Significant evidence for allele sharing in affected patients was detected suggesting that PAX6 mutation links to aniridia in this family. An extra band corresponding to exon 9 in PAX6 was found by single strand conformational polymorphism analysis in all the aniridia patients in this family, but not detected in the unaffected members. A mutation of C to T was detected by sequencing at the nucleotide 1080 that converts the Arg codon (CGA) to the termination codon (TGA).Conclusions Aniridia is caused by a nonsense mutation of PAX6 gene in the large Chinese kindred. 205-Ethynyl-2′-deoxyuridine as a molecular probe of cell proliferation for high-content siRNA screening assay by “click” chemistry显示文摘Labelling and identification of proliferating cells is important for the study of physiological or pathological processes in high-content screening (HCS) assays. Here we describe ethynyl deoxyuridine (EdU) as a biomarker for the assessment of cell proliferation and clearly demonstrate the feasibility of the EdU-labelling method for use in HCS assays.
