| 2 | Bone marrow-derived monocyte infusion improves hepatic fibrosis by decreasing osteopontin,TGF-β1,IL-13 and oxidative stress显示文摘AIM To evaluate the therapeutic effects of bone marrowderived CD11b+CD14+ monocytes in a murine model of chronic liver damage. METHODS Chronic liver damage was induced in C57BL/6 mice by administration of carbon tetrachloride and ethanol for 6 mo. Bone marrow-derived monocytes isolated by immunomagnetic separation were used for therapy. The cell transplantation effects were evaluated by morphometry,biochemical assessment,immunohistochemistry and enzyme-linked immunosorbent assay. RESULTS CD11b+CD14+ monocyte therapy significantly reduced liver fibrosis and increased hepatic glutathione levels. Levels of pro-inflammatory cytokines,including tumor necrosis factor-α,interleukin(IL)-6 and IL-1β,in addition to pro-fibrotic factors,such as IL-13,transforming growth factor-β1 and tissue inhibitor of metalloproteinase-1 also decreased,while IL-10 and matrix metalloproteinase-9 increased in the monocytetreated group. CD11b+CD14+ monocyte transplantation caused significant changes in the hepatic expression of α-smooth muscle actin and osteopontin. | 3 | Role of pentoxifylline in non-alcoholic fatty liver disease in high-fat diet-induced obesity in mice显示文摘AIM:To study pentoxifylline effects in liver and adipose tissue inflammation in obese mice induced by high-fat diet(HFD).METHODS: Male swiss mice(6-wk old) were fed a highfat diet(HFD; 60% kcal from fat) or AIN-93(control diet; 15% kcal from fat) for 12 wk and received pentoxifylline intraperitoneally(100 mg/kg per day) for the last 14 d. Glucose homeostasis was evaluated by measurements of basal glucose blood levels and insulin tolerance test two days before the end of the protocol. Final body weight was assessed. Epididymal adipose tissue was collected and weighted for adiposity evaluation. Liver and adipose tissue biopsies were homogenized in solubilization buffer and cytokines were measured in supernatant by enzyme immunoassay or multiplex kit, respectively. Hepatic histopathologic analyses were performed in sections of paraformaldehyde-fixed, paraffin-embedded liver specimens stained with hematoxylin-eosin by an independent pathologist. Steatosis(macrovesicular and microvesicular), ballooning degeneration and inflammation were histopathologically determined. Triglycerides measurements were performed after lipid extraction in liver tissue. RESULTS: Pentoxifylline treatment reduced microsteatosis and tumor necrosis factor(TNF)-α in liver(156.3 ± 17.2 and 62.6 ± 7.6 pg/mL of TNF-α for non-treated and treated obese mice, respectively; P < 0.05). Serum aspartate aminotransferase levels were also reduced(23.2 ± 6.9 and 12.1 ± 1.6 U/L for nontreated and treated obese mice, respectively; P < 0.05) but had no effect on glucose homeostasis. In obese adipose tissue, pentoxifylline reduced TNF-α(106.1 ± 17.6 and 51.1 ± 9.6 pg/mL for non-treated and treated obese mice, respectively; P < 0.05) and interleukin-6(340.8 ± 51.3 and 166.6 ± 22.5 pg/mL for non-treated and treated obese mice, respectively; P < 0.05) levels; however, leptin(8.1 ± 0.7 and 23.1 ± 2.9 ng/mL for non-treated and treated lean mice, respectively; P < 0.05) and plasminogen activator inhibitor-1(600.2 ± 32.3 and 1508.6 ± 210.4 pg/mL for non-treated and treated lean mice, respectively; P < 0.05) levels increased in lean adipose tissue. TNF-α level in the liver of lean mice also increased(29.6 ± 6.6 and 75.4 ± 12.6 pg/mL for non-treated and treated lean mice, respectively; P < 0.05) while triglycerides presented a tendency to reduction.CONCLUSION: Pentoxifylline was beneficial in obese mice improving liver and adipose tissue inflammation. | 4 | Giant squamous cell carcinoma of the gallbladder: A case report显示文摘BACKGROUND Gallbladder cancer is the most common malignant tumor of the biliary tract. The majority of cases are adenocarcinoma. Squamous cell carcinoma is the histological type present in 12% of all neoplasias accounting for approximately 12% of gallbladder neoplasms. It can occur in its pure form reaching 1%-3% of the tumors. Many patients are at an advanced stage when diagnosed and have bad therapeutic efficacy. CASE SUMMARY A 45-year-old male patient presented with left flank pain for 1 year and irradiated to the mesogastric region. He denied fever, vomiting, and any other intestinal changes. He reported a weight loss of 10 kg in a period of 7 mo. He denied alcoholism, smoking, drug use, or prior illness. Computed tomography of the abdomen showed in the gallbladder fossa a voluminous mesogastric heterogeneous collection that had a thick and irregular capsule with liquid and gaseous contents. A predominantly hypoattenuating rounded material with partially calcified margins measuring about 2.0 cm related to gallstone was also emphasized. No lymphadenomegalies or free fluid was observed in the abdominal cavity. Patient underwent laparotomy where a huge tumor was observed affecting the transverse colon and gallbladder. This mass was resected en bloc removing gallbladder and transverse colon together with corresponding mesocolon and regional lymphadenectomy. There were no complications in the postoperative period. Although oncological treatment was performed, the patient died 6 mo after surgery. CONCLUSION Squamous cell carcinoma represents a rare disease. Patients often present with large, bulky tumors with involvement of adjacent organs. | 7 | Influence of donor age on the differentiation and division capacity of human adipose-derived stem cells显示文摘BACKGROUND Human adipose-derived stromal/stem cells(hASCs)are one of the most useful types of mesenchymal stromal/stem cells,which are adult multipotent cells with great therapeutic potential for the treatment of several diseases.However,for successful clinical application,it is critical that high-quality cells can be obtained.Diverse factors seem to be able to influence cell quality and performance,especially factors related to donors’intrinsic characteristics,such as age.Nevertheless,there is no consensus regarding this characteristic,and there is conflicting information in the literature.AIM To investigate the growth kinetics and differentiation potential of adipose-derived stem cells isolated from the lipoaspirates of elderly and young donors.METHODS hASCs were harvested from liposuctioned adipose tissue obtained from female donors(aged 20-70 years).Cells were distributed into two groups according to age range:old hASCs(oASCs,≥55 years,n=9)and young hASCs(yASCs,≤35 years,n=9).For each group,immunophenotypic characterization was performed by flow cytometry.Population doubling time was assessed over seven days.For adipogenic potential evaluation,lipid deposits were assessed after 7 d,14 d and 21 d of adipogenic induction.Osteogenic potential was verified by analyzing cell mineralization after 14 d,21 d and 28 d of osteogenic induction.mRNA expression of PPARγ2,CEBPA and Runx2 were detected by quantitative reverse transcription polymerase chain reaction.RESULTS hASCs were successfully obtained,cultured,and grouped according to their age:yASCs(26.33±4.66 years old)and oASCs(64.78±4.58 years old).After maintenance of the cells in culture,there were no differences in morphology between cells from the young and old donors.Additionally,both groups showed classical immunophenotypic characteristics of mesenchymal stem/stromal cells.The average doubling time indicated that yASCs(4.09±0.94 d)did not significantly differ from oASCs(4.19±1.29 d).Concerning differentiation potential,after adipogenic and osteogenic induction,yASCs and oASCs were able to differentiate to greater levels than the noninduced control cells.However,no differences were found in the differentiation efficiency of yASCs and oASCs in adipogenesis or osteogenesis.Additionally,the mRNA expression of PPARγ2,CEBPA and Runx2 were similar in yASCs and oASCs.CONCLUSION Our findings suggest that age does not seem to significantly affect the cell division or adipogenic or osteogenic differentiation ability of adipose-derived stem cells isolated from lipoaspirates. | Cintia DS Horinouchi María Julia Barisón Anny W Robert Crisciele Kuligovski Alessandra M Aguiar BrunoDallagiovanna | 2020 | World Journal of Stem Cells2020,12,12: | 2 |

| 12 | Microstructural evaluation of initial dissolution of aluminosilicate particles and formation of geopolymer material显示文摘The rheological behavior of microparticulate structures of geopolymers was studied.The materials were developed from alternative sources of aluminosilicate and were activated in different concentrations using 5,10,and 15M sodium hydroxide,and sodium silicate,which were added in a 2:1ratio (by volume).The solid constituents used were bottom ash/metakaolin and neat metakaolin in a 2:1blend.
