| 1 | LncRNAs-directed PTEN enzymatic switch governs epithelial–mesenchymal transition显示文摘Despite the structural conservation of PTEN with dual-specificity phosphatases,there have been no reports regarding the regulatory mechanisms that underlie this potential dual-phosphatase activity.Here,we report that K27-linked polyubiquitination of PTEN at lysines 66 and 80 switches its phosphoinositide/protein tyrosine phosphatase activity to protein serine/threonine phosphatase activity.Mechanistically,high glucose,TGF-β,CTGF,SHH,and IL-6 induce the expression of a long non-coding RNA,GAEA(Glucose Aroused for EMT Activation),which associates with an RNA-binding E3 ligase,MEX3C,and enhances its enzymatic activity,leading to the K27-linked polyubiquitination of PTEN.The MEX3C-catalyzed PTEN^K27-polyUb activates its protein serine/threonine phosphatase activity and inhibits its phosphatidylinositol/protein tyrosine phosphatase activity.With this altered enzymatic activity,PTEN^K27-polyUb dephosphorylates the phosphoserine/threonine residues of TWIST1,SNAI1,and YAP1,leading to accumulation of these master regulators of EMT.Animals with genetic inhibition of PTEN^K27-polyUb,by a single nucleotide mutation generated using CRISPR/Cas9(Pten^K80R/K80R),exhibit inhibition of EMT markers during mammary gland morphogenesis in pregnancy/lactation and during cutaneous wound healing processes.Our findings illustrate an unexpected paradigm in which the lncRNA-dependent switch in PTEN protein serine/threonine phosphatase activity is important for physiological homeostasis and disease development. | Qingsong Hu Chunlai Li Shouyu Wang Yajuan Li Bo Wen Yanyan Zhang Ke Liang Jun Yao Youqiong Ye Heidi Hsiao Tina K.Nguyen Peter K.Park Sergey D.Egranov David H.Hawke Jeffrey R.Marks Leng Han Mien-Chie Hung Bing Zhang Chunru Lin Liuqing Yang | 2019 | Cell Research2019,29,4: | 8 |
| 2 | Robust state-space realizations of digital filters against finite word length errors显示文摘This article deals with two important issues in digital filter implementation:roundoff noise and limit cycles. A novel class of robust state-space realizations, called normal realizations, is derived and characterized. It is seen that these realizations are free of limit cycles. Another interesting property of the normal realizations is that they yield a minimal error propagation gain. The optimal realization problem, defined as to find those normal realizations that minimize roundoff noise gain, is formulated and solved analytically. | 4 | Antidepressant effect of bioactive compounds from Paecilomyces tenuipes in mice and rats显示文摘A bioactive compound from Paecilomyces tenuipes (BCPT) has an inhibitory effect on monoamine oxidase A (MAO-A) in vitro. Researchers have thought that BCPT may be a potential antidepressant. The MAO-A suppressor moclobemide served as a control, and this study investigated the mechanisms of BCPT as an antidepressant. Results demonstrated that BCPT induced significantly increased sucrose intake in chronic unpredictable stressed rats, shortened immobility time in forced swimming mice, improved the scores of blepharoptosis and akinesia in reserpine-treated mice, increased the number of 5-hydroxy tryptophan-induced head-twitches, remarkably enhanced the expression of hippocampus mineralcorticoid receptor and glucocorticoid receptor mRNA, decreased the ratio of mineralcorticoid receptor to glucocorticoid receptor and raised the levels of dopamine, norepinephrine and 5-hydroxytryptamine, while decreasing hydroxyindole acetic acid levels or dihydroxy-phenyl acetic acid in chronic unpredictable stressed rats. Behavioral test results suggested that BCPT potentially had antidepressant-like activity. | 5 | Modulating morphology via side-chain engineering of fused ring electron acceptors for high performance organic solar cells显示文摘In this work, four fused ring electron acceptors(FREAs), 2F-C5, 2F-C6, 2F-C8 and 2F-C10, are developed to investigate the effect of side-chain size on the molecular properties and photovoltaic performance of FREA systematically. The elongation of side-chains in the FREAs not only improves their solubility in the processing solvent, but also enhances their miscibility with the donor PBDB-T. It helps the FREA diffuse into the donor PBDB-T during film-formation, thus leading to the decrease in domain size and domain purity from PBDB-T:2F-C5 to PBDB-T:2F-C10 blend films in sequence. The smaller domain size affords more D/A interfaces to benefit exciton dissociation and inhibit monomolecular recombination. However, severe bimolecular recombination occurs when the domain purity decreases to a critical point. Due to the dual function of the increment of side-chain length, both short-circuit current density(J_(SC)) and fill factor(FF) of devices exhibit an evolution of first increasing then decreasing from 2F-C5, 2F-C6, 2F-C8 to 2F-C10 based OSCs. | 6 | Oral nano-antioxidants improve sleep by restoring intestinal barrier integrity and preventing systemic inflammation显示文摘Sleep deprivation(SD)is a severe public health threat that can cause systemic inflammation and nerve damage.Few effective and side-effect-free drugs are available to address SD.However,the bidirectional communications between the brain and gut provide new strategies for anti-SD therapeutics.Here we explored oral delivery of fullerene nano-antioxidants(FNAO)in the SD model to improve sleep by regulating abnormal intestinal barrier and systemic inflammation via the brain-gut axis.SD caused excessive reactive oxygen species(ROS)production and hyperactive inflammatory responses in the intestines of zebrafish and mouse models,leading to disturbed sleep patterns and reduced brain nerve activity.Of note,based on the property of the conjugatedπbond of the C_(60)structure to absorb unpaired electrons,oral FNAO efficiently reduced the excessive ROS in the intestines,maintained redox homeostasis and intestinal barrier integrity,and ameliorated intestinal and systemic inflammation,resulting in superior sleep improvement.Our findings suggest that maintaining intestinal homeostasis may be a promising avenue for SD-related nerve injury therapy. | Zhanfeng Wu Lei Liu Lei Li Xinran Cao Wang Jia Xiaodan Liao Zhongpu Zhao Hedong Qi Guoqiang Fan Huiqiang Lu Chunying Shu Mingming Zhen Chunru Wang Chunli Bai | 2023 | National Science Review2023,10,12: | 1 |
| 7 | Influencing factors of colostrum exposure to low-level lead and their relationship with early neural development of infants显示文摘BACKGROUND: Exposure to low-level lead has a toxic effect on the development of neonates, which has attracted wide attention. Colostrum lead level can be used as the indication of lead exposure. OBJECTIVE: To observe the relationship of colostrum lead level and the neurobehavioral development of infants. DESIGN: A prospective control observation. SETTING: Center for Maternal and Child Health, Shanxi Provincial Children's Hospital. PARTICIPANTS: Totally 128 neonates of full-term normal delivery, 76 male and 52 female, from Shanxi Provincial Maternal and Child Health Center and Jiexiu Maternal and Child Health Center were involved in this study. All the involved neonates had no peripartal ischemic/hypoxic history or fetus intrauterine developmental lag. Pregnant women had no various acute and chronic diseases in pregnancy, family history of neurological disease or occupational lead exposure. 128 portions of colostrum sample of full-term normal delivery were collected. Informed consents of detected items were obtained from the puerperants and their relatives. METHODS: ① Experimental grouping: Lead level in the colostrum was determined by atomic absorption spectrometry. According to lead level in the colostrum, the neonates were classified into two exposure groups of greater than or equal to 0.24 μmol/L in a high-level lead group and less than 0.24 μ mol/L in a low-level lead group. ② Experimental evaluation: Mental developmental index (MDI) and psychological developmental index (PDI) of 3-month-old infants were evaluated with Bayley Scales of Infant Development (BSID). The relationships of MDI, PDI and colostrum lead level were performed correlation regression analysis; The relationship of colostrum lead level and development was performed multi-factor analysis with family environment and health questionnaires. MAIN OUTCOME MEASURES: ① Evaluation results of MDI and PDI. ② Multi-factor analysis results. RESULTS: Totally 128 neonates were involved in the study. Ten and eleven neonates were lost due to emigration in the high-level lead group and low-level lead group respectively, and the other 107 neonates participated in the final analysis. ① MDI and PDI in the high-level lead group were significantly lower than those in the low-level lead group, respectively (P < 0.01); Regression analysis results showed that two developmental indexes were statistically negatively correlated with colostrum lead level (regression equation y = 1.9+0.01x1,-0.04x2,+0.04x3,+0.03x4). ② Four variables of the factors included by family environment and health questionnaires were taken into equation. Large maternal age, irrational dietary pattern in pregnancy and pollution degree of habitation environment in pregnancy were the risk factors of colostrum lead level (partial regression coefficien t =0.598 4,0.426 8,0.306 7,P < 0.05-0.01), and calcium supplementation in pregnancy was a protective factor (partial regression coefficien t =-0.455 8, P < 0.01). | 11 | Application of mannose-modified fullerene immunomodulator selectively polarizes tumor-associated macrophages potentiating antitumor immunity显示文摘Polarization of tumor associated macrophages(TAMs)has been a promising therapeutic paradigm for tumor.However,how to achieve precise regulation of TAMs and high efficiency of tumor immunotherapy is still a huge challenge.Here,we report dicarboxy fullerene modified with mannose(DCFM)as an immunomodulator to selectively polarize TAMs and prominently boost anti-tumor immunity.The dicarboxy fullerene molecule was synthesized through the Prato reaction and further covalently bonded with mannose,obtaining the DCFM with well-defined structure.Due to the exist of mannose in DCFM,it could accurately recognize mannose receptor in TAMs.Our cellular experiment results showed that mannose modification could notably promote the uptake of DCFM by the immunosuppressive M2-type macrophages that effectively reprogrammed M2-type macrophages into anti-tumor M1-type macrophages,leading to enhance the phagocytosis of tumor cells by macrophages and inhibiting tumor cells migration.Subsequently,we observed that DCFM could significantly distribute into tumor tissues by in vivo fluorescence imaging.Importantly,DCFM exhibited a superior anti-tumor efficiency in the subcutaneous colorectal tumor model.In addition,it showed that DCFM precisely polarized TAMs into M1-type macrophages and actively increased the infiltration of cytotoxic T lymphocytes(CTLs),inducing profound tumor growth inhibition. | Haoyu Wang Lei Li Xinran Cao Mingming Zhen Chunru Wang Chunli Bai | 2023 | Nano Research2023,16,11: | 0 |
| 12 | Correlation of blood lead level with iron, zinc and calcium as well as neonatal neurobehavioral development显示文摘BACKGROUND: Endothelial cell in brain of undeveloped fetus cannot inhibit entrance of lead so as to cause toxic effect. Over-accumulation of lead can cause toxic effect on children’s growth and development, especially on development of nervous system. Lead in fetus body is mainly derived from its mother’s; therefore, blood lead reflects the level of mother’s lead. OBJECTIVE: To analyze the correlations among blood lead level, trace elements, mental development index (MDI) and psychomotor development index (PDI) of 3-month-old children. DESIGN: Contrast observation. SETTING: Maternity and Child Care Hospital, Shanxi Provincial Children Hospital. PARTICIPANTS: A total of 128 neonates with spontaneous delivery were provided by Shanxi Maternity and Child Care Hospital and Jiexiu Municipal Maternity and Child Care Hospital from September 2004 to January 2005. There were 76 boys and 52 girls. All subjects did not have history of ischemia and hypoxia during perinatal period and umbilical blood was accepted to be used in experiments. METHODS: ① Levels of iron, calcium and zinc in umbilical blood: 0.5 mL umbilical blood and 2 mL blood samples were selected from neonates to measure lead content with graphite-oven atom absorption spectrum and contents of iron, calcium and zinc in umbilical blood with atom absorption spectrum. ② Grouping: According to level of blood lead, neonates were divided into high-dosage lead group (blood lead ≥ 0.24 μmol/L) and low-dosage lead group (blood lead < 0.24 μmol/L). ③ Evaluation of neurobehavioral development: Neurobehavioral development of 3-month-old neonates was evaluated with Bayley scale, which was consisted of intelligence scale, motor scale and behavioral scale. Intelligence scale, which was expressed as intelligence development index, contained 163 items, such as adaptability, behavior, language, pilot activities, etc. Motor scale, which was expressed as mental motor development index, contained 81 items, such as vulgar motor, refined motor, etc. The total score was 100 and the standard deviation was 16 points. If MDI and PDI were less than 70 points, neonates were determined as mental retardation. ④ Correlation of blood lead with calcium, iron and zinc in umbilical blood was analyzed with partial correlation coefficient and relation of blood lead with MDI and PDI was detected with linear correlation analysis. MAIN OUTCOME MEASURES: ① Comparisons of iron, calcium and zinc in umbilical blood of neonates in the two groups; ② neurobehavioral scores; ③ correlations among level of blood lead, contents of calcium, iron and zinc in umbilical blood and MDI and PDI. RESULTS: Among all subjects, 28 were lost because of blood coagulation and uncompleted data, and other 100 were involved in the final analysis with 50 in each group. ① Comparisons of iron, calcium and zinc in umbilical blood: Contents of iron and calcium was (2.17±0.51) and (73.6±21.7) mg/L in high-dosage lead group, which was lower than those in low-dosage group [(2.66±0.58), (83.8±24.9) mg/L; t =3.01, 2.72; P < 0.01]. ② Comparisons of MDI and PDI in the two groups: MDI in high-dosage lead group was obviously lower than that in low-dosage group [(103.50±10.36), (109.44±9.71) points; t =2.19, P < 0.01]. ③ Correlation of blood lead with calcium, iron and zinc in umbilical blood: Blood lead was negative correlation with blood iron (partial correlation coefficient = -0.47, P < 0.05). ④ Correlation of blood lead with MDI and PDI: MDI and PDI were negative correlation with blood lead (r = -0.417, -0.327, P < 0.05). | 15 | Synergistic modulation of spin and fluorescence signals in a nano-Saturn assembled by a metallofullerene and cycloparaphenylene nanohoop显示文摘Spin-based interdisciplinary research has attracted considerable attention,and various applications in magnetic memory,quantum science,and precision measuring technology have been reported.In this study,we induced a fluorescence property in a spin-active molecule by supramolecular assembly and realized a synergistic modulation of its spin and fluorescence properties.A Saturn-shaped supramolecular complex was synthesized using a spin-active metallofullerene Sc_(3)C_(2)@C80 with a fluorescent nanohoop of tetra-benzothiadiazole-based[12]cycloparaphenylene(TB[12]CPP),and its spin and fluorescence properties were comprehensively investigated.Temperature-dependent electron paramagnetic resonance(EPR)spectroscopy and fluorescence analyses were conducted.Synchronous changes in the EPR signals and fluorescence peaks were discovered in the temperature range of 170–290 K.Based on nuclear magnetic resonance observations and theoretical calculations,a temperature-dependent host–guest interaction between Sc_(3)C_(2)@C_(80)and a nanohoop was demonstrated,which explained the synchronous changes in the EPR signals and fluorescence peaks for Sc_(3)C_(2)@C_(80)⊂TB[12]CPP.The application of Sc_(3)C_(2)@C_(80)as a molecular spin system to probe the host–guest interaction was also evaluated.These results show that the supramolecular assembly can be used to design advanced spin systems coupled with optical and magnetic behaviors based on paramagnetic and luminescent molecules. | Jie Zhang Zhenlin Qiu Chong Zhao Yuxi Lu Wang Li Linshan Liu Chunru Wang Yuanzhi Tan Taishan Wang | 2023 | Nano Research2023,16,2: | 0 |

| 17 | Lead level in foremilk and neurobehavioral development of neonates显示文摘BACKGROUND: Recently, it has been reported that blood lead level lower than 24 μmol/L can lead to learning and cognitive deficits. OBJECTIVE: To investigate the relationship of lead level in foremilk and early neurobehavioral development of neonates taking lead level in foremilk as lead exposure index. DESIGN: A controlled observation. SETTING: Maternal and Child Health Center, Shanxi Children's Hospital. PARTICIPANTS: Totally 128 neonates of full-term normal delivery, 76 male and 52 female, from Shanxi Provincial Maternal and Child Health Center and Jiexiu Maternal and Child Health Center were involved in this study. All the involved neonates had no peripartal ischemic/hypoxic history. The corresponding puerperants were aged (27±5)years. They had no various acute and chronic diseases during pregnancy, and family history of neurological disease as well as occupational lead exposure. Informed consents of detected items were obtained from the puerperants. METHODS: ① Determination of lead level in foremilk: Altogether 128 foremilk samples, 1 mL each, were collected between January and February 2005. The same amount of violet acid was added to each sample. After foremilk was fully dissolved, 0.2 mL solution was taken for determining lead level with atomic absorption spectrometer in graphite stove. The determined process strictly followed the internal quantity control of laboratory and was involved in the blind quality control of Institute of Environmental Health of Chinese Academy. ② Participants grouping: Totally 128 neonates were involved, and the normal reference value of lead level of foremilk was 0.06–0.48 μmol/L. The involved neonates were assigned into high-level lead group (≥ 0.24 μmol/L, n =60) and low-level lead group (< 0.24 μmol/L, n =68). ③ Assessment of neurobehavioral development of neonates: Neurobehavioral development level of neonates who was born 24 to 72 hours was assessed with 20-item neonatal neurobehavioral determination method, which involved behavioral ability (6 items), passive muscular tension (4 items), active muscular tension (4 items), primitive reflection (3 items) and general evaluation (3 items). Each above-mentioned scoring had 3 scales (0,1 and 2 points). The full mark of 20 items was 40 points. Neurological behaviors of neonates might be unabnormal when scoring was < 35 points. MAIN OUTCOME MEASURES: Assessment results of neurobehavioral development of neonates in high- and low-level lead neonates. RESULTS: After lead-level determination, the involved neonates in two groups participated in the final analysis. Neurobehavioral total scores of neonates of high-level lead group were lower than those in the low-level lead group ([35.9±1.3) points vs(.37.7±1.4) points,P < 0.01]. The scores of neonatal erection in high-level lead group were lower than those in low-level lead group [(1.4±0.4) points vs.(1.8±0.5) points,P <0.01], and time for head erection of neonates in the high-level lead group was shortened as compared with that in the low-level lead group [(1.8±1.7) minutes vs.(3.3±2.2) minutes,P < 0.01]. CONCLUSION: 0.24 μmol/L lead level in foremilk has certain relationship with neurobehavioral development. | 18 | A molecular brake hoop for the motion of metal atoms inside fullerene cage显示文摘Molecular machines have attracted extensive attention due to their fancy concept and their potential to influence the science and technology.The dynamic motion of encapsulated metallic clusters is a distinctive character for endohedral metallofullerenes.For the development of molecular rotors based on metallofullerenes,the most challenging issue is how to control the motion of untouchable metallic cluster inside fullerene cage.In this work,we report a molecular brake hoop for the motion of metal atoms inside fullerene cage.A cycloparaphenylene of[12]CPP was employed to hoop the metallofullerene and produce two supramolecular complexes of Sc_(3)N@C_(80)⊂[12]CPP and Sc_(2)C_(2)@C_(82)⊂[12]CPP.Moreover,the temperature-dependent ^(45)Sc nuclear magnetic resonance spectroscopy(NMR)was employed to detect the motion of internal Sc_(3)N and Sc_(2)C_(2) clusters.^(45)Sc NMR results reveal that the[12]CPP can slow down the rotation of internal metallic cluster through host-guest interaction,and thus the[12]CPP can be considered as a molecular brake hoop for the internal metal motion of metallofullerenes.Furthermore,by means of this molecular brake hoop,the motion of metal atoms inside fullerene cage have expanded range of velocity.In addition,theoretical calculations on Sc_(3)N@C_(80)⊂[12]CPP were executed to illustrate the molecular orientation as well as internal Sc_(3)N rotation.This study would promote the research of endohedral metallofullerene as a molecular rotor. | Yuxi Lu Chong Zhao Jie Zhang Wang Li Jiayi Liang Linshan Liu Yongguang Li Chunru Wang Taishan Wang | 2022 | Science China Chemistry2022,65,8: | 0 |
