| 1 | High-intensity focused ultrasound ablation:An effective bridging therapy for hepatocellular carcinoma patients显示文摘AIM:To analyze whether high-intensity focused ultrasound(HIFU) ablation is an effective bridging therapy for patients with hepatocellular carcinoma(HCC).METHODS:From January 2007 to December 2010,49 consecutive HCC patients were listed for liver transplantation(UCSF criteria).The median waiting time for transplantation was 9.5 mo.Twenty-nine patients received transarterial chemoembolization(TACE) as a bringing therapy and 16 patients received no treatment before transplantation.Five patients received HIFU ablation as a bridging therapy.Another five patients with the same tumor staging(within the UCSF criteria) who received HIFU ablation but not on the transplant list were included for comparison.Patients were comparable in terms of Child-Pugh and model for end-stage liver disease scores,tumor size and number,and cause of cirrhosis.RESULTS:The HIFU group and TACE group showed no difference in terms of tumor size and tumor number.One patient in the HIFU group and no patient in the TACE group had gross ascites.The median hospital stay was 1 d(range,1-21 d) in the TACE group and two days(range,1-9 d) in the HIFU group(P < 0.000).No HIFU-related complication occurred.In the HIFU group,nine patients(90%) had complete response and one patient(10%) had partial response to the treatment.In the TACE group,only one patient(3%) had response to the treatment while 14 patients(48%) had stable disease and 14 patients(48%) had progressive disease(P = 0.00).Seven patients in the TACE group and no patient in the HIFU group dropped out from the transplant waiting list(P = 0.559).CONCLUSION:HIFU ablation is safe and effective in the treatment of HCC for patients with advanced cirrhosis.It may reduce the drop-out rate of liver transplant candidate. | Tan To Cheung Sheung Tat Fan See Ching Chan Kenneth SH Chok Ferdinand SK Chu Caroline R Jenkins Regina CL Lo James YY Fung Albert CY Chan William W Sharr Simon HY Tsang Wing Chiu Dai Ronnie TP Poon Chung Mau Lo | 2013 | World Journal of Gastroenterology2013，19，20： | 23 |
| 2 | 小胶质细胞对星形胶质细胞的免疫和神经保护功能的调节作用(英文)显示文摘在中枢神经系统中,小胶质细胞和星形胶质细胞对神经元的发育、功能维持及细胞存活具有重要的作用。但在炎症反应过程中,星形胶质细胞增生的作用及其对神经元的影响目前并不十分清楚。因此,本研究对不同种类的细胞进行培养,给予细菌脂多糖(lipopolysaccharide,LPS)刺激,观察各种细胞的反应。结果显示,星形胶质细胞可以减轻炎症反应的神经元毒性,提示星形胶质细胞增生主要具有神经保护作用。如果去掉上清液中由星形胶质细胞分泌的胶质细胞源性神经营养因子(glial cell line-derived neurotrophic factor,GDNF),这种神经保护作用显著降低。为了研究星形胶质细胞的免疫功能,本研究培养了高浓度的星形胶质细胞,并发现LPS不能诱导肿瘤坏死因子(tumor necrosis factor,TNF)-α及诱导型一氧化氮合酶(inducible nitric oxide synthase,iN OS)/一氧化氮(nitric oxide,NO)(iN OS/NO)释放。但如果在培养的高浓度星形胶质细胞中增加0.5%~1%的小胶质细胞,则可以诱导TNF-α及iN OS/NO的释放。这提示小胶质细胞和星形胶质细胞间的相互作用对于LPS诱导星形胶质细胞释放前炎症因子及GDNF是必须的。我们还发现,小胶质细胞释放的TNF-α可以通过旁分泌作用调节星形胶质细胞的神经保护功能。综上所述,本研究结果提示星形胶质细胞可能不会直接被LPS激活,而是通过与小胶质细胞相互作用被激活,释放神经营养因子,减少因炎症反应激活的小胶质细胞释放的毒性物质对神经元的损害作用。 | Chen SH Oyarzabal EA Sung YF Chu CH Wang Q Chen SL Lu RB Hong JS 唐颖馨 | 2015 | 神经损伤与功能重建2015，10，1： | 9 |
| 3 | Establishment of an orthotopic transplantation tumor model of hepatocellular carcinoma in mice显示文摘AIM:To improve the outcome of orthotopic transplantation in a mouse model,we used an absorbable gelatin sponge(AGS) in nude mice to establish an orthotopic implantation tumor model.METHODS:MHCC-97L hepatocellular carcinoma(HCC)cells stably expressing the luciferase gene were injected into the subcutaneous region of nude mice.One week later,the ectopic tumors were harvested and transplanted into the left liver lobe of nude mice.The AGS was used to establish the nude mouse orthotopic implantation tumor model.The tumor suppressor gene,paired box gene 5(PAX5),which is a tumor suppressor in HCC,was transfected into HCC cells to validate the model.Tumor growth was measured by bioluminescence imaging technology.Semi-quantitative reverse transcription polymerase chain reaction(RT-PCR) and histopathology were used to confirm the tumorigenicity of the implanted tumor from the MHCC-97L cell line.RESULTS:We successfully developed an orthotopic transplantation tumor model in nude mice with the use of an AGS.The success rate of tumor transplantation was improved from 60% in the control group to 100% in the experimental group using AGS.The detection of fluorescent signals showed that tumors grew in all live nude mice.The mice were divided into 3 groups:AGS-,AGS+/PAX5-and AGS+/PAX5 +.Tumor size was significantly smaller in PAX5 transfected nude mice compared to control mice(P < 0.0001).These fluorescent signal results were consistent with observations made during surgery.Pathologic examination further confirmed that the tissues from the ectopic tumor were HCC.Results from RT-PCR proved that the HCC originated from MHCC-97L cells.CONCLUSION:Using an AGS is a convenient and efficient way of establishing an indirect orthotopic liver transplantation tumor model with a high success rate. | Gui-Jun Zhao Li-Xia Xu Eagle SH Chu Ning Zhang Jia-Yun Shen Alatangaole Damirin Xiao-Xing Li | 2012 | World Journal of Gastroenterology2012，18，47： | 4 |
