| 7 | Recent advances in chemical durability and mechanical stability of superhydrophobic materials: Multi-strategy design and strengthening显示文摘Inspired by the lotus leaf effect, non-wetting artificial superhydrophobic surfaces demonstrate an enormous potential in numerous fields. However, limited by poor stability and durability, superhydrophobic surfaces are rarely available for practical applications. In this review, based on the wettability mechanisms and failure modes of superhydrophobic surfaces, it is proposed that the construction of highly stable superhydrophobic materials can be approached from four aspects, including structural design, chemical bonding, interfacial-strengthening of hydrophobic materials and substrates, and self-healing. We introduced in detail the design ideas, strengthening approaches, and characterization tools of highly stable superhydrophobic materials from the perspective of multi-strategy design and strengthening, and provided corresponding insights. Eventually, the development, current status, and prospects of highly stable and multifunctional superhydrophobic materials were also presented in detail. | 8 | A Retrospective Study of Treatment Efficacy of 65 Children with Ametroic Amblyopia显示文摘Purpose: To investigate relationship between treatment efficacy and the severity of ametropic amblyopia, the type of anisometropia and patient age. Methods:A total of 65 children with ametropic amblyopia undergoing clinical treatment in the ophthalmology department of Shaoguan Hygienic Hospital of Women and Children between June 2005 and November 2011 were enrolled in this study. The treatment efficacy for those subjects with different severities of ametropic amblyopia, types of anisometropia and ages was recorded. Results: The near-recovery/recovery rate, improvement rate and ineffectiveness rate were 70.8%, 16.9% and 12.3%, respectively. The recovery rates in mild-, moderate- and severe amblyopia groups were 97.1%, 61.1% and 8.3% respectively (P<0.05)..The recovery rate for patients with hyperopic-, astigmatic- and myopic anisometropia were 75.8%, 78.3% and 33.3% respectively. The recovery rates did not differ between hyperopic- and astigmatic- anisometropia children(P>0.05), whereas a statistically significant difference was noted between myopic children compared with their hyperopic and myopic-anisometropia counterparts (all P<0.05). The recovery rate for patients aged 3 to 6 years was 86.8% and 48.1% for those aged between 7 and 10 years (P<0.05). Conclusion: The treatment efficacy in ametropic amblyopia is associated with the severity of amblyopia, type of anisometropia and patient's age. Older patients with more severe amblyopia had poorer treatment efficacy. | 13 | Bioinspired therapeutic platform based on extracellular vesicles for prevention of arterial wall remodeling in hypertension显示文摘Arterial stiffness due to the vessel remodeling is closely linked to raised blood pressure,and its physiopathologic mechanism is still not fully understood.We here aimed to explore whether extracellular vesicle(EV)mediated intercellular communication between endothelium and smooth muscle cell contribute to the blood vessel remodeling under hypertension.We here revealed that the arterial endothelial cells robustly secreted EV,which in turn could be circulated and/or directly taken up by the subendothelial smooth muscle cells(SMC).Under hypertension,the EV secretion increased and the miRNA profile changed significantly mainly due to the raised mechanical force and subsequent enhanced reactive oxygen species generation.Among the miRNA cargos in the EV,miR-320d/423-5p were found increased most significantly.In vivo delivery of miR-320d/423-5p mimics via engineered EV increased their expression in arterial vessels,recapitulating the phenotype in hypertension.In contrast,therapeutic delivery of miR-320d/423-5p inhibitors via engineered EV alleviated the phenotype in spontaneous hypertension rat model.Together,we have found that the injured endothelium due to the raised mechanical force in hypertension contributes to the arterial wall remodeling via the secreted EV.Our study has not only provided novel insights on the mechanism of hypertension associated blood vessel wall remodeling,but also shed light on therapeutic intervention of hypertension associated vascular diseases. | Chen Wang Changyang Xing Zhelong Li Yunnan Liu Qiaoying Li Yixiao Wang Jiao Hu Lijun Yuan Guodong Yang | 2022 | Bioactive Materials2022,7,2: | 1 |
| 14 | Allele-specific genome editing of imprinting genes by preferentially targeting non-methylated loci using Staphylococcus aureus Cas9(SaCas9)显示文摘Allele-specific DNA methylation is the most important imprinting marker localized to differentially methylated regions(DMRs),and aberrant genomic imprinted DNA methylation is associated with some human diseases,including Prader-Willi syndrome and cancer.Thus,the development of an effective strategy for the precise editing of allele-specific methylated genes is essential for the functional clarification of imprinting elements and the correction of imprinting disorders in human diseases.To discover a feasible allele-specific genome editing tool based on the CRISPR/Cas system,which is an efficient genetargeting technique in various organisms,we examined the targeting efficiency of Staphylococcus aureus Cas9(SaCas9)and Streptococcus pyogenes Cas9(SpCas9)in response to DNA methylation interference.We found that the targeting efficiency of SaCas9,but not SpCas9,was enhanced by targeted DNA demethylation using the d Cas9-Tet1 catalytic domain(CD)but suppressed by targeted DNA methylation using Dnmt3l-Dnmt3a-d Cas9.An in vitro cleavage assay further demonstrated that SaCas9 nuclease activity was inhibited by 5-methylcytosine(5mC)in a synthesized Cp G-containing context.Further analysis with Ch IP-Q-PCR demonstrated that the non-methylated sequence targeting of Sa Cas9 depends on the binding preference of SaCas9 to non-methylated sequences.Taking advantage of this feature of SaCas9,we have successfully obtained non-methylated allele-biased targeted embryos/mice for two imprinting genes,H19 and Snrpn,with relatively high efficiencies of 28.6%and 47.4%,respectively.These results indicate that the targeting efficiency of SaCas9 was strongly reduced by DNA methylation.By using SaCas9,we successfully achieved allele-specific genome editing of imprinting genes by preferentially targeting non-methylated loci. | Yajing Liu Jianan Li Changyang Zhou Bin Meng Yu Wei Guang Yang Zongyang Lu Qingmei Shen Yu Zhang Hui Yang Yunbo Qiao | 2019 | Science Bulletin2019,64,21: | 1 |
