| 2 | A transgene-free method for rapid and efficient generation of precisely edited pigs without monoclonal selection显示文摘Gene-edited pigs for agricultural and biomedical applications are typically generated using somatic cell nuclear transfer(SCNT).However, SCNT requires the use of monoclonal cells as donors, and the time-consuming and laborious monoclonal selection process limits the production of large populations of gene-edited animals. Here, we developed a rapid and efficient method named RE-DSRNP(reporter RNA enriched dual-sg RNA/CRISPR-Cas9 ribonucleoproteins) for generating gene-edited donor cells. RE-DSRNP takes advantage of the precise and efficient editing features of dual-sg RNA and the high editing efficiency, low off-target effects, transgene-free nature, and low cytotoxic characteristics of reporter RNA enriched RNPs(CRISPR-Cas9ribonucleoproteins), thus eliminating the need for the selection of monoclonal cells and thereby greatly reducing the generation time of donor cells from 3–4 weeks to 1 week, while also reducing the extent of apoptosis and chromosomal aneuploidy of donor cells. We applied RE-DSRNP to produce cloned pigs bearing a deletion edit of the wild-type p53-induced phosphatase 1(WIP1)gene: among 32 weaned cloned pigs, 31(97%) carried WIP1 edits, and 15(47%) were homozygous for the designed fragment deletion, and no off-target event was detected. The WIP1 knockout(KO) pigs exhibited male reproductive disorders, illustrating the utility of RE-DSRNP for rapidly generating precisely edited animals for functional genomics and disease research. | 3 | Water,organic matter,nitrogen and phosphorus contents in sediment of a large-scale mariculture area in the Zhelin Bay of eastern Guangdong Province,China显示文摘The Zhelin Bay is one of the most important bays for large-scale mariculture in Guangdong Province, China. Owing to the increasing human population and the expanding mariculture in the last two decades, the ecological environment has greatly changed with frequent harmful algal blooms. A monthly survey of water content, organic matter (TOM), and various forms of nitrogen and phosphorous in sediment from July 2002 to July 2003 in the bay was conducted. The results showed that the water content was correlated significantly with TOM and various forms of nitrogen and phosphorus and can be used as proxy for quick and rough estimate of these factors in the future surveys. TOM was also correlated significantly with various forms of nitrogen and phosphorus, indicating that it was one of the key factors affecting the concentrations and distributions of nitrogen and phosphorus in the investigated waters. Average total Kjeldhal nitrogen (TkN) content was(1 113.1±382.5)μg/g and average total phosphorus (TP) content was(567.2±223.3)μg/g, and both were much higher than those of similar estuaries in China and elsewhere. Average nitrogen and phosphorus tended to be higher inside than outside the bay, higher at aquaculture than non-aquaculture areas, and higher at fish-cage culture than oyster culture areas, suggesting that large-scale mariculture inside the bay played an important role in the eutrophication of the Zhelin Bay. Various forms of nitrogen and phosphorus concentrations were higher during the warm season (July—September), which was due to the increased decomposition and concentration of organic matter resulted from the fast growth and high mortality of the cultured species. Compared with July 2002, TkN and TP contents were much higher in July 2003, in consonance with the eutrophication of the Zhelin Bay. Because exchangeable phosphorus (Ex-P), iron-bounded phosphorus (Fe-P) and organic phosphorus (OP) combined accounted for 34.3% of the TP and authigenic phosphorus (Au-P) accounted for 49.2% of the TP, biological phosphorus (BP) that includes Ex-P, Fe-P, OP, and a portion of Au-P, thus accounted for 34.3% to 83.5% of the TP in the Zhelin Bay, which was within the percentage range, but with a high absolute value among the estuaries. Au-P was the most important species of phosphorus and accounted for 49.2% of the TP during the investigation. | 6 | Comprehensive evaluation of Actinidia arguta fruit based on the nutrition and taste:67 germplasm native to Northeast China显示文摘Actinidia arguta is a berry fruit with high nutritional values.To evaluate the comprehensive quality,the fruit weight,titratable acid(TAC),soluble solids(SSC),vitamin C,mineral elements,and taste value of67 accessions of A.arguta were measured,and sensory attributes including olfactive intensity,juiciness,sweetness,sourness,and astringency were assessed.Cluster analysis was carried out based on mineral elements,and principal component analysis(PCA)was used for comprehensive score ranking.The result demonstrated that fruit weight ranged from 3.5 g to 24.2 g,SSC ranged from 11.94%to 20.56%,TAC ranged from 0.15%to 1.20%,and vitamin C ranged from 3.51 g/kg to 230.76 g/kg(fresh weight)among the tested samples.There were significant differences in sourness,sweetness,bitterness,and umami among the 67 accessions based on electronic tongue analysis,and this result was rapidly and objectively characterized by the sensory assessment.The dendrogram was obtained and clustered the accessions into two major clusters,and five principal components were extracted by PCA.Overall,the information provided by this study identified the 10 best germplasm resources for fresh-eating and processing fruits.It can provide theoretical reference for nutritional efficacy evaluation and food development of A.arguta. | Mengting Song Heran Xu Guang Xin Changjiang Liu Xiaorong Sun Yinhong Zhi Bin Li Yixiao Shen | 2022 | Food Science and Human Wellness2022,11,2: | 3 |
