1The adult human testis transcriptional cell atlas显示文摘Human adult spermatogenesis balances spermatogonial stem cell (SSC) self-renewal and differentiation,alongside complex germ cell-niche interactions,to ensure long-term fertility and faithful genome propagation. Here,we performed single-cell RNA sequencing of -6500 testicular cells from young adults. We found five niche/somatic cell types (Leydig,myoid,Sertoli,endothelial,macrophage),and observed germline-niche interactions and key human-mouse differences. Spermatogenesis,including meiosis,was reconstructed computationally,revealing sequential coding,non-coding,and repeat-element transcriptional signatures. Interestingly,we identified five discrete transcriptional/developmental spermatogonial states,including a novel early SSC state,termed State 0. Epigenetic features and nascent transcription analyses suggested developmental plasticity within spermatogonial States. To understand the or丨gin of State 0,we profiled testicular cells from infants,and identified distinct similarities between adult State 0 and infant SSCs. Overall,our datasets describe key transcriptional and epigenetic signatures of the normal adult human testis,and provide new insights into germ cell developmental transitions and plasticity.Jingtao Guo Edward J. 2Photolysis of benzophenone with two-step two-laser excitation显示文摘The first laser excites a molecule to a lower triplet state and another sequential laser excites it resonantly to higher triplet states or makes it ionization. This two-step two-laser method provides a novel way to study the electron transfer or charge transfer of excited molecules. The higher excited benzophenone and its radical cation can be observed under the time-resolved absorption method when it was excited to lower triplet state by one laser and another one exciting it to higher triplet states resonantly. 5Viral RNA-based transfection and expression of enhanced green fluorescent protein in the parasitic protozoan Eimeria stiedae显示文摘Eimeria stiedae(E.stiedae),the most common protozoan pathogen of rabbits,causes coccidiosis.This disease is problematic for rabbit breeders [1].There is little information about the cellular and molecular biology of this pathogen and a lack of genetic tools.Genetic manipulation is necessary to advance our understanding of parasite biology and ultimately develop vaccines and treat diseases.DNA-mediated and viral RNA-mediated transfection systems have successfully been established in many protozoan parasites [2,3].It was discovered that E.stiedae possesses a dsRNA virus(EsRV1),and more recently,the genome of this virus has been sequenced.To date,however,there is no report on the use of viral RNA-mediated transfection for E.stiedae.In this study,we report a transient viral RNA-mediated transfection of E.stiedae using enhanced green fluorescent protein(EGFP)as a reporter gene.Our study provides a valuable genetic tool for studying the biology of E.stiedae.Caiyan Xin Jianhua Li Pengtao Gong Bin Wu Nan Zhang Ju Yang Xuepeng Cai Xichen Zhang 2019Acta Biochimica et Biophysica Sinica2019,51,2:0
6Analysis of products of thymine irradiated by ^(18)O^(8+) ion beam in N_2O saturated aqueous solution显示文摘Some methods of capillary gas chromatography, such as GC, GC-MS,GC-FT-IR, are used to analyze the products of thymine irradiated by 18O8+ ion beam in N2O saturated aqueous solution. From the results of GC-MS the molecular weight of products can be determined, and from the results of GC-FT-IR some molecular structure information of products can be obtained. By this way the products,5,6-Dihydrothyndne, 5-Hydroxyl-5-Methylhydantoin, 5-Hydroxyl-6-Hydrothymine, 5-Hydro-6-Hydroxylthymine, 5-Hydroxymethyluracil, Trans-Thymine glycol, Cis-Thymine glycol and dimers are determined without separation of them from samples. Though these products are as same as those products of thymine irradiated by γ rays in N2O saturated aqueous solution, the mechanism of thymine irradiated by heavy ion beam in aqueous solution is different from that by γ rays.
