1A de novo Genome of a Chinese Radish Cultivar显示文摘Here, we report a high-quality draft genome of a Chinese radish(Raphanus sativus) cultivar. This draft contains 387.73 Mb of assembled scaffolds, 83.93% of the scaffolds were anchored onto nine pseudochromosomes and 95.09% of 43 240 protein-coding genes were functionally annotated. 184.75 Mb(47.65%) of repeat sequences was identified in the assembled genome. By comparative analyses of the radish genome against 10 other plant genomes, 2 275 genes in 780 gene families were found unique to R. sativus. 2FAT1, a direct transcriptional target of E2F1, suppresses cell proliferation, migration and invasion in esophageal squamous cell carcinoma显示文摘Objective: Growing evidence indicates that FAT atypical cadherin 1(FAT1) has aberrant genetic alterations and exhibits potential tumor suppressive function in esophageal squamous cell carcinoma(ESCC). However, the role of FAT1 in ESCC tumorigenesis remains not well elucidated. The aim of this study was to further investigate genetic alterations and biological functions of FAT1, as well as to explore its transcriptional regulation and downstream targets in ESCC.Methods: The mutations of FAT1 in ESCC were achieved by analyzing a combined study from seven published genomic data, while the copy number variants of FAT1 were obtained from an analysis of our previous data as well as of The Cancer Genome Atlas(TCGA) and Cancer Cell Line Encyclopedia(CCLE) databases using the cBioPortal. The transcriptional regulation of FAT1 expression was investigated by chromatin immunoprecipitation(ChIP) and the luciferase reporter assays. In-cell western, Western blot and reverse transcription-quantitative polymerase chain reaction(RT-qPCR) were used to assess the indicated gene expression. In addition, colony formation and Transwell migration/invasion assays were employed to test cell proliferation, migration and invasion.Finally, RNA sequencing was used to study the transcriptomes.Results: FAT1 was frequently mutated in ESCC and was deleted in multiple cancers. Furthermore, the transcription factor E2 F1 occupied the promoter region of FAT1, and depletion of E2 F1 led to a decrease in transcription activity and mRNA levels of FAT1. Moreover, we found that knockdown of FAT1 promoted KYSE30 and KYSE150 cell proliferation, migration and invasion;while overexpression of FAT1 inhibited KYSE30 and KYSE410 cell proliferation, migration and invasion. 30Interacting model of new agegraphic dark energy:observational constraints and age problem显示文摘Many dark energy models fail to pass the cosmic age test because of the old quasar APM 08279+5255 at redshift z=3.91,the ΛCDMmodel and holographic dark energy models being no exception.Inthis paper,we focus onthe topic of the age problem inthe new agegraphic dark energy(NADE)model.We determine the age of the universe in the NADE model by fitting the observational data,including type Ia supernovae(SNIa),baryon acoustic oscillations(BAO)and the cosmic microwave background(CMB).We find that the NADE model also faces the challenge of the age problem caused by the old quasar APM 08279+5255.In order to overcome such a difficulty,we consider the possible interaction between dark energy and dark matter.We show that this quasar can be successfully accommodated in the interacting new agegraphic dark energy(INADE)model at the 2σlevel under the current observational constraints.LI YunHe MA JingZhe CUI JingLei WANG Zhuo ZHANG Xin 2011Science China(Physics,Mechanics & Astronomy)2011,54,8:3
4The selective adsorption of rare earth elements by modified coal fly ash based SBA-15显示文摘Rare earth elements(REE)are strategic resources and the recycling of REE in alternative resources is urgent and gets increasingly attention.However,the separation of REE in these alternative resources is still a challenge due to the low concentration of REE and multi coexisted ions in acidic system.In this study,the species distribution of REE within the pH 0-8.0 was calculated.The SBA-15 originated from coal fly ash was modified by two steps with(3-aminopropyl)triethoxysilane(APTES)and diethylenetriaminepentaacetic dianhydride(DTPADA)to obtain DTPADA-SBA-15 adsorbent,which was applied to the selective adsorption of REE.The results showed that DTPADA-SBA-15 possessed excellent adsorption performance on the selective adsorption of REE,including Eu,Gd,Tb,Nd and Sm,in acidic solution(pH 2)with multi competing ions.The FT-IR and Zeta potential characterization verified that the chemical adsorption through the coordination of O in DTPADA-SBA-15 with REE was dominant at lower pH value.The study of adsorption kinetics indicated that the adsorption of rare earth metal ions followed pseudosecond-order kinetic,of which the adsorption process followed the Langmuir isotherm model.Jinglei Cui Qian Wang Jianming Gao Yanxia Guo Fangqin Cheng 2022Chinese Journal of Chemical Engineering2022,35,7:0
5High-temperature superconductivity up to 223 K in the Al stabilized metastable hexagonal lanthanum superhydride显示文摘As compressed hydrides constantly refresh the records of superconducting critical temperatures(T_(c)) in the vicinity of room temperature, this further reinforces the confidence to find more high-temperature superconducting hydrides. In this process, metastable phases of superhydrides offer enough possibilities to access superior superconducting properties. Here we report a metastable hexagonal lanthanum superhydride(P6_(3)/mmc-LaH_(10)) stabilized at 146 GPa by introducing an appropriate proportion of Al,which exhibits high-temperature superconductivity with T_(c)~178 K, and this value is enhanced to a maximum T_(c)~223 K at 164 GPa. A huge upper critical magnetic field value H_(c2)(0) reaches 223 T at 146 GPa. The small volume expansion of P63/mmc-(La, Al) H_(10) compared with the binary La H_(10) indicates the possible interstitial sites of Al atoms filling into the La–H lattice, instead of forming conventional ternary alloy-based superhydrides. 6Seco-cyclic phorbol derivatives and their anti-HIV-1 activities显示文摘Phorbol esters are recognized for their dual role as anti-HIV-1 agents and as activators of protein kinase C(PKC).The efficacy of phorbol esters in binding with PKC is attributed to the presence of oxygen groups at positions C20,C3/C4,and C9 of phorbol.Concurrently,the lipids located at positions C12/C13 are essential for both the anti-HIV-1 activity and the formation of the PKC-ligand complex.The influence of the cyclopropane ring at positions C13 and C14 in phorbol derivatives on their anti-HIV-1 activity requires further exploration.This research entailed the hydrolysis of phorbol,producing seco-cyclic phorbol derivatives.The anti-HIV-1 efficacy of these derivatives was assessed,and the affinity constant(Kd)for PKC-δprotein of selected seco-cyclic phorbol derivatives was determined through isothermal titration calorimetry.The findings suggest that the chemical modification of cyclopropanols could affect both the anti-HIV-1 activity and the PKC binding affinity.Remarkably,compound S11,with an EC50 of 0.27μmol·L^(−1) and a CC50 of 153.92μmol·L^(−1),demonstrated a potent inhibitory effect on the intermediate products of HIV-1 reverse transcription(ssDNA and 2LTR),likely acting at the viral entry stage,yet showed no affinity for the PKC-δprotein.These results position compound S11 as a potential candidate for further preclinical investigation and for studies aimed at elucidating the pharmacological mechanism underlying its anti-HIV-1 activity.HUANG Xiaolei HUANG Xusheng LI Qirun MA Mengdi CUI Yadong YANG Liumeng WANG Haibo LUO Ronghua CHEN Jinglei YANG Jingxuan LIN Jinrong LI Duxin ZHENG Yongtang ZHANG Jian 2024Chinese Journal of Natural Medicines2024,22,4:0
7Strong gravitational lensing constraints on holographic dark energy显示文摘Strong gravitational lensing(SGL) has provided an important tool for probing galaxies and cosmology. In this paper, we use the SGL data to constrain the holographic dark energy model, as well as models that have the same parameter number, such as the w CDM and Ricci dark energy models. We find that only using SGL is difficult to effectively constrain the model parameters.However, when the SGL data are combined with CBS(CMB+BAO+SN) data, the reasonable estimations can be given and the constraint precision is improved to a certain extent, relative to the case of CBS only.
